Quality Characteristics of Miljeonbyeong Added with Carrot and Spinach Juice (채소 즙을 첨가한 밀전병의 품질 특성)

  • Quality characteristics of griddled miljeonbyeong containing vegetable juice were assessed by conducting sensory tests and measuring texture and color. When varying amounts of carrot/or spinach juice was added to miljeonbyeong, the sensory preferences of appearance and texture were higher in miljeonbyeong containing a small amount of carrot and spinach vegetable juice, and the preferences of smell and taste were higher in miljeonbyeong containing a large amount of carrot and spinach juice. Overall, preference was highest in miljeonbyeong supplemented with 30 g carrot juice and 10 g spinach juice. Overall preference correlated significantly with appearance (r=0.44) and texture (r=0.47). In miljeonbyeong containing vegetable juice, tensile strength decreased significantly with increasing amounts of carrot juice (34.98~30.82) or spinach juice (40.84~37.07). In all miljeonbyeong samples, tensile strength increased slightly in 1 hour but rapidly increased after 2 hours. Lightness progressively decreased. A-value increased (3.46~9.63) and b-value increased with increasing amounts of vegetable juice added. No difference in the b-value increase was evident between carrot juice (15.19~26.43) and spinach juice(18.19~24.40). Tensile strength was markedly lower in miljeonbyeong amended with carrot juice (34.98~30.82g) or spinach juice (39.05~32.67 g), which contained fat-soluble pigment, as compared with vegetable-free miljeonbyeong (44.81 g), but the addition of beet juice (45.30~44.78 g) containing water-soluble pigment did not lower the tensile strength of miljeonbyeong compared to vegetable juice. In order to improve the texture of miljeonbyeong containing vegetable juice, tensile strength was measured in miljeonbyeong prepared with 30 g carrot juice or 10 g of spinach juice, which produced the highest sensory test scores, and additionally amended with potato starch, corn starch, glutinous rice flour, strong flour or yam powder with a consequent reduction in the content of wheat flour. Improvement was noted in carrot juice-containing samples prepared with 6 g corn starch, 15 g potato starch and 15 g hard wheat flour, and in spinach juice-containing samples prepared with 6 g corn starch, 6 g potato starch, 6 g hard wheat flour and 15 g dried yam powder.

  • The use of plasma has increased in bio-application field in recent years. Particularly, water treated by arc discharge or atmospheric pressure plasma has been actively utilized in bio-industry. In this study, we have developed a plasma activated water generating system. For this system, two kinds of plasma sources; dielectric barrier discharge (DBD) plasma and arc discharge plasma have been used. The discharge energy was calculated using the breakdown voltage and current, and the emission spectrum was measured to investigate the generated reactive species. We also analyzed the amount of reactive oxygen and nitrogen species in water using the chemical methods and nitric oxide sensor. Finally, the influence of plasma generated reactive species on the germination and growth of spinach (Spinacia oleracea) was investigated. Spinach is a green leafy vegetable that contains a large amount of various physiologically active organic compounds. However, it is characterized with a low seed germination rate.

Effects of Potassium-Cyanoethylstarch (K-CES) and Polyacrylamide (PAM) on Growth of Spinach (Spinacia oleracea L.) (토양내 K-CES와 PAM처리가 시금치의 생육에 미치는 영향)

  • The objective of this study was to observe the effects of potassium-cyanoethylstarch (K-CES), which is a natural polymer derivate,. and polyacrylamide (PAM), which is a synthetic polymer, on the growth of spinach under environment-controlled biotron. K-CES was made from tapioca starch, and introductions of acrylonitrile was confirmed by FT-IR. Water absorbing capacities (WAC) of K-CES and PAM were 37.6 and 47.8 times, respectively. WAC of PAM was slightly higher than K-CES, WAC of PAM m saline solutions were more reduced than those of K-CES. It was introduced that K-CES treatment was more effective than PAM in higher salinity of water and soil. Dry super absorptive polymers were put into solution, and the solution was applied to soil as an soil conditioner. Both amendments, K-CES and PAM, increased the vegetative growth of the spinach when applied at 0.1% of soil weight. In general, the most favorable results for early vegetative growth of the spinach came from the PAM treatment, followed by the K-CES treatment.

Effects of Cooking Method on the Vitamin and Mineral Contents in Frequently Used Vegetables (조리 방법이 상용채소의 비타민 및 무기질 함량에 미치는 영향 -시금치, 양배추, 콩나물, 당근을 중심으로 -)

  • Purpose: We have investigated for the purpose of studying change of vitamins and minerals in frequently used vegetables by low moisture cooking method. Methods: Vitamin B complex, vitamin C and mineral (Mg, Ca, K, Zn, Fe) contents are analyzed in vegetables such as spinach, cabbage, bean sprouts and carrot which are heat-treated with degrees of water contents. Low moisture cooking method represents heat-treated vegetable (LM experiment group) with 25 wt.% water content for the vegetable weight while general blanching method includes heat-treated vegetable (GB experiment group) with 500-1,000 wt.% water content for the vegetable weight. Results: Retention rate of vitamin B1 (thiamin) in the LM experiment group is relatively high (87.50-95.68%) and dosen't show considerable differences from raw vegetables. On the other hand, the retention rate of vitamin $B_1$ in GB group's cabbage and spinach dramatically decreased to 19.46-25.00%. Retention rate of vitamin B2 (riboflavin) is sustained stably in LM experiment group (75.00-87.50% in bean sprouts, cabbage and spinach), represents that low moisture cooking method has relatively higher contents in vitamin $B_2$. The LM experiment group has 71.43-85.71% on retention rate of niacin while the GB group shows relatively lower niacin (57.14-64.58%), represents conflicting results from the general idea that vitamin $B_3$ is relatively stable in heat treatment and blanching. Retention rate of pantothenic acid is considerably higher in both LM and GB experiment group, indicates vitamin pantothenic acid is relatively stable in the various cooking condition such as amount of water and heat treatment. In the experiment under various water contents, vitamin C is not detected in both bean sprouts and carrots, while the contents in cabbage and spinach are 19.87 mg/100 g and 26.65 mg/100 g respectively. In the same experiment, the retention rate of Vitamin C in LM experiment group (91.65%, 92.23%) is considerably higher than GB group (58.08%, 61.61%). Retention rate of Mg, K, Fe in the LM experiment group is relatively higher than GB group. Conclusion: Resultingly, the observations suggests that minimum water quantity and minimum heat treatment processes should be established in cooking vegetables which have soluble vitamins and minerals.

Preliminary Results of Extraction, Separation and Quantitation of Arsenic Species in Food and Dietary Supplements by HPLC-ICP-MS

  • Various extraction procedures were investigated using reference materials and samples to evaluate extraction efficiency and effectiveness. Inductively coupled plasma mass spectrometry (ICP-MS) was used to measure total arsenic and to quantitate arsenic species when coupled to an HPLC (high pressure liquid chromatography). Arsenic species were extracted from rice flour (NIST SRM 1568a) with water/methanol mixtures using accelerated solvent extraction (ASE). Total arsenic extraction efficiency ranged from 42 to 64%, for water and various methanol concentrations. From spinach (NIST SRM 1570), freeze-dried apple, and rice flour (NIST SRM 1568a), arsenic species were extracted with trifluoroacetic acid (TFA) at 100 ${^{\circ}C}$. Total arsenic extraction efficiency was 90% for spinach, 75% for freeze-dried apple, and 83% for rice flour. Enzymatic extraction with alpha-amylase and sonication resulted in extraction efficiency of 104% for rice flour, 98% for freeze-dried apple, and 7% for spinach. Chromatograms of arsenic species extracted by the optimum extraction methods were obtained, and the species were quantified. Arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA), and monomethylarsonic acid (MMA) were found in the apple sample, and DMA and As(V) in the rice flour sample. As(V) and MMA were found in three herbal dietary supplement samples.

Changes in Color of Spinach Leaves by Blanching (데치기에 따른 시금치 잎의 변색)

  • The changes in color of spinach leaves upon blanching in boiling water and by microwave oven were investigated. The noticible changes in color of leaves were observed above $70^{\circ}C$. The values of L (lightness), a (redness) and b (yellowness) were decreased upon blanching. The value of the under surface of leaves was decreased by 2~3 times compared to that of the upper surface. The noticible color changes occured from 15 sec and the absorbance at 432 nm of water increased rapidly from 2 min of blanching in boiling water. Salt (0.5 and 0.9%) decreased the absorbance at 432 nm of water. The changes of L and b of the under surface of leaves upon heating with a microwave oven were less compared with those in boiling water. Based on the color differences of leaves upon blanching in boiling water, the optimum ratio of spinach to water was 1:4 or 1:5.

The Degradation Patterns of Two Pesticides in Spinach by Cultivation, Storage and Washing (시금치의 생육, 저장기간 및 세척에 따른 잔류농약 분해특성)

  • Two pesticides commonly used in spinach were subjected to a field residue trial to ensure safety of terminal residues in the harvest. The residual patterns of two pesticides, which were Azoxystrobin and chlorpyrifos were examined after applying with the recommended dose and their $DT_{50}$ were calculated. Also degradation patterns of pesticides at storage $4^{\circ}C$ were compared to those at $20^{\circ}C$, and removal rates of pesticides by washing spinach with water were measured. Biological half-lives of azoxystrobin and chlorpyrifos were 3.2~3.8 and 3.8~4.7 days, respectively. During the storage period, the degradation patterns were appeared more obviously at $20^{\circ}C$ than $4^{\circ}C$. Removal rates of azoxystrobin and chlorpyrifos were 9.6~90.0% and 17.7~85.8% by various washing methods.

Comparison of the Chemotaxis Potential of Bacteria Isolated from Spinach Roots and Nonrhizosphere Soil

  • In order to investigate the role of bacterial chemotaxis in root colonization, the chemotaxis potential of bacteria isolated from spinach roots was compared with that of bacteria from nonhizosphere soil, with reference to the plant age (1,000 isolates), soil moisture conditons (1,400 isolates), and part of the root (200 isolates). The % CT (% occurrence of chemotaxis (+) isolates among total bacterial isoltes) of the root isolates significantlyfluctuated during the plant growth period, reaching a maximum after 10-15 days of growth. At this time period, the maximum % CT for the root isolates was around 70-80% CT under a soil moisture 50% WFP (% volume of water-filled pores in total soil pores), and then gradually reduced with an increasing % WFP. The results of the chemotaxis potential of each of the 100 islates from the spinach roots and nonrhizosphere soil under various % WFP demonstrated that the % CT of the root isolates were significantly higher than those of solates from the nonrhizosphere soil under a wide range of soil moisture content (35-80% WFP). Furthermore, the % CT value (80%) from the upper root was significantly higher than tht (55%) from the lower root. Compared with the % CT values of the roots, the values from the nonrhizosphere soil did not significantly vary relative to the plant age of % WFP. These results indicate that chemotaxis would appear to be a major factor in bacterial root colonization.

Antimutagenic Effect of Green-Yellow Vegetables toward Aflatoxin $B_1$, and 4-Nitroquinoline-1-oxide (아플라톡신 $B_1$과 4-NQO에 대한 녹황색 채소류의 항돌연변이 효과)

  • The antimutagenic effects of green-yellow vegetables toward aflatoxin B$_1$(AFB$_1$) and 4-nitroquinoline-1-ox-ide (4-NQO) using the Ames assay system with Salmonella typhimurium TA98 and TA100 were studied. Forty six to fifty percent of the methanol extracts of the vegetable samples inhibited the mutagenicity induced by AFB$_1$in TA98 and TA100. Perilla leaf, lettuce, broccoli, crown daisy, water dropwort, small water dropwort, red pepper, red pepper leaves, amaranth, spinach and radish root were significantly reduced the mutagenicity of AFB$_1$(p< 0.01). Whereas 25 out of 27 samples (93%) exhibited antimutagenicity toward a direct mutagen of 4-NQO (p< 0.01. 0.05). The samples which showed the strong antimutagenicity (>60%) were cabbage, kale, lettuce, broccoli, mustard leaf, green red pepper, green sweet pepper, spinach, amaranth, soybean sprout and immature pumpkin. The juices from the several samples also showed antimu-tagenic activity toward AFB$_1$. Cabbage, perilla leaf, small water dropwort and spinach reduced TAT100 revertants dose dependently in the range of 50-500$m\ell$/plate, however, cucumber and carrot showed little effect.

Effect of EFD-1 and PC-10 deicers on Growth of Wheat, Barley and Spinach (대체제설제인 EFD-1과 PC-10의 밀, 보리, 시금치 생육에 미치는 영향)

  • BACKGROUND: During the winter, the use of deicers was rapidly increased for reduction of traffic accidents as well as injuries in Korea, whose components are largely comprised of calcium chloride and sodium chloride. Recently, to reduce the adverse effects of chloride-deciers such as pollution of water and soil, and decrease of agricultural productivity, the attention of eco-friendly deciers have been increased. This study aimed to investigate biological effects of magnesium chloride deicer (PC-10) and organic acid deicer (EFD-1) against wheat, barley, and spinach. METHODS AND RESULTS: We examined the effect of two deciers, PC-10 and EFD-1 on the seed germination and growth of wheat, barley and spinach. EFD-1 showed higher suppression of the germination than PC-10 among tested crop seeds. In demage index of the seedlings of the crops, there was no symptoms in spinach such as spotting and color change of leaves. EFD-1 showed much stronger inhibitory effect on the germination of tested crop seeds than PC-10 when crops were exposed continuously to tested deciers in soils. The growth and shoot and root in examined crops was relatively higher in PC-10 treatment than in EFD-1 treatment when compared to the control. The biomass decrease was found in all examined conditions of deciers. PC-10 showed 23-35% reduction of biomass whereas EFD-1 exhibited 39-84% loss in all examined crops at over 2% concentration. CONCLUSION: These results suggest that the effects of deicers used in this study by inputting into soil against growing tested crops cause the reduction of germination of seeds, growth, and biomass compared to the control.
