Intramolecular Esterification by Lipase Powder in Microaqueous Cycohexane (미소 수용 Cyclohexange 중에서 분말 Lipase에 의한 분자내 에스테르화반응)

  • The effects of substrate concentration, enzyme concentration, reaction temperature, and water content were investigated in intramolecular esterification. This study used cyclohexane as organic solvent, power lipase as enzyme, and benzyl alcohol and octanoic acid as substrate. The initial reaction rate was found to be proportional to enzyme concentration; followed Michaelis-Menten equation for octanoic acid; and was inhibited by benzyl alcohol . The observed initial reaction rate first increased, then decreased with increasing reaction temperature, giving rise to the maximum rate at 20$\circ$. The drop in the reaction rate at higher temperature was to partition equilibrium change of substrate between organic solvent and hydration layer of enzyme molecule in addition to the deactivation by enzyme denaturation. Water layer surrounding enzyme molecule seemed to activate in organic solvent and the realistic reaction was done in the water layer. In the enzymatic reaction in organic solvent, the initial reaction rate was influenced by partition quilibrium of substrate, so the optimum condition of substrate concentration, enzyme concentration, reaction temperature, and water content would give a good design tool.

Enzyme-Catalyzed Henry Reaction in Choline Chloride-Based Deep Eutectic Solvents

  • The enzyme-catalyzed Henry reaction was realized using deep eutectic solvents (DESs) as a reaction medium. The lipase from Aspergillus niger (lipase AS) showed excellent catalytic activity toward the substrates aromatic aldehydes and nitromethane in choline chloride:glycerol at a molar ratio of 1:2. Addition of 30 vol% water to DES further improved the lipase activity and inhibited DES-catalyzed transformation. A final yield of 92.2% for the lipase AS-catalyzed Henry reaction was achieved under optimized reaction conditions in only 4 h. In addition, the lipase AS activity was improved by approximately 3-fold in a DES-water mixture compared with that in pure water, which produced a final yield of only 33.4%. Structural studies with fluorescence spectroscopy showed that the established strong hydrogen bonds between DES and water may be the main driving force that affects the spatial conformation of the enzyme, leading to a change in lipase activity. The methodology was also extended to the aza-Henry reaction, which easily occurred in contrast to that in pure water. The enantioselectivity of both Henry and aza-Henry reactions was not found. However, the results are still remarkable, as we report the first use of DES as a reaction medium in a lipase-catalyzed Henry reaction.

  • This study was to experiment, through sewage sludge treatment by Vacuum Drying Method, variation of water content with reaction pressure, reaction time, reaction temperature. The result are as follows; The water content decreased with the same reaction temperature and reaction time at lower pressure and 360~40 mmHg (a close vacuum) showed lower water content at low reaction temperature and short reaction time. The water content rapidly decreased with the same reaction pressure and time at low reaction temperature (above $120^{\circ}C$).

  • ThThe purpose of this study is to investigate the compressive strength properties of fly ash using water eluted from recycled coarse aggregate. When fly ash come into contact with water, they have not a autonomously chemical reaction. But fly ash is a pozzolan reaction when fly ash come into contact with water and calcium hydroxide(Ca(OH)2) in alkaline environment. For that reason, if water eluted from recycled coarse aggregate use mixture water, fly ash is expected to reaction of pozzolan reaction property in early stage. According to the experimentation result, ICP-MS analysis showed water eluted from recycled coarse aggregate has a high alkali-ash value of pH of 12 and over. And mixing ratio 30% fly ash mortar using water eluted from recycled coarse aggregate showed a similar strength of plain mortar due to the pozzolan reaction. Also, poor strength in initial age, disadvantage of mortar using fly ash, can be improved as hydration in early age is expedited due to calcium hydroxide(Ca(OH)2) and unhydrated cement component eluted from recycled aggregate mortar.

  • A facile and precise batch oxidation reaction system allows continuous monitoring of the oxidation rate and cumulated oxygen conversion of xylenes, and the side reactions to carbon monoxide and carbon dioxide may also be studied. The oxidation reaction can be analyzed precisely with the rate and amount of oxygen consumed. The reaction reveals that 4-carboxybenzaldehyde is an unstable intermediate of p-xylene oxidation as the reaction proceeds instantaneously from p-toluic acid to TPA (terephthalic acid). The alkali metals accelerate oxidation, even though they retard the reaction initially. The oxidation rate increases with decreasing water concentration. However, in the later part of reaction, the reactivity decreases a bit if the water concentration is very low. This retarding effect of water can be overcome partly by the addition of potassium. The oxidation of o-xylene, compared with the oxidation of p-xylene and m-xylene, proceeds quite fast initially, however, the oxidation rate of xylene isomers in the later stage of reaction is in the order of p-xylene > mxylene > o-xylene.

  • The industrial techniques of ultrasound have been used in the various fields, such as cleaning, medical surgery, emulsification, cell disruption etc. Especially the application of cell disruption was interested in the field of disinfection process in water by ultrasonic irradiation. It has been recognized that the ultrasounds are irradiated in aqueous solution, cavitation bubbles are generated and shock waves of high temperature and pressure are emitted as the bubbles are developed and finally broken, which function as a energy source to promote reaction efficiencies of various kinds of chemical reactions such as disinfection reaction in water. Therefore, this study was performed to apply the ultrasound for the disinfection method of infected drinking raw water and to discuss the limiting factors such as pH, sample volume and reaction temperature influenced on the removal efficiency of E. coli from experimental analysis of the results obtained in bench-scale plant. For the experiments to measure the influence of reaction parameters in the ultrasonic disinfection process, escalated reactivity of aqueous solutions was excellent when pH in aqueous solution was low, and sample volume was small. On the contrary, the reactivity of disinfection became elevated when reaction temperature was high. It was found that the rate constant of disinfection reaction was applied by Chick's law, reaction kinetics of Chick's law was irreversible and pseudo-first order at all the tested conditions.As a conclusion it appeared that the technology using ultrasonic irradiation can be applied to the treatment of disinfection in infected water which are difficult to be treated by conventional methods.

An Influence of Water Ingestion into Engine Cylinder on the Joint Reaction Force of the Connecting Rod (엔진 실린더 내 물 유입이 커넥팅로드 조인트반력에 미치는 영향)

  • This paper focus on investigating the influence of the amount of water ingestion and the engine speed on the joint reaction force of the connecting rod in engine. The connecting rod was modelled by MSC/PATRAN, the modal informations of it were obtained by the DMAP module in the MSC/NASTRAN, and the dynamic force history was computed through the flexible multibody dynamic simulation in DADS. To analyze the joint reaction force acting on the connecting rod, the 48 cases were investigated. The engine speed varies with 200, 700, 1600, 2400rpm and the volumetric ratio of water to the combustion chamber varies with 0, 10, 20, ..., 90, 95 and 97.5% . As the engine speed decreases and the amount of water ingestion increases, the joint reaction force increase. Especially when the amount of water ingestion exceeds the 70% of the volume of the combution chamber, the joint reaction force acting on the connecting rod is over the design strength.

Reaction Path Modelling on Geochemical Evolution of Groundwater and Formation of Secondary Minerals in Water-Gneiss Reaction System (편마암-물 반응계에서 지하수의 지화학적 진화 및 이차광물 생성에 관한 반응경로 모델링)

  • The reaction path of water-gneiss in 200m borehole at the Soorichi site of Yugu Myeon, Chungnam was simulated by the EQ3NR/EQ6 program. Mineral composition of borehole core and fracture-filling minerals, and chemical composition of groundwater was published by authors. In this study, chemical evolution of groundwater and formation of secondary minerals in water-gneiss system was modelled on the basis of published results. The surface water was used as a starting solution for reaction. Input parameters for modelling such as mineral assemblage and their volume percent, chemical composition of mineral phases, water/rock ratio reactive surface area, dissolution rates of mineral phases were determined by experimental measurement and model fit. EQ6 modelling of the reaction path in water-gneiss system has been carried out by a flow-centered flow through open system which can be considered as a suitable option for fracture flow of groundwater. The modelling results show that reaction time of 133 years is required to reach equilibrium state in water-gneiss system, and evolution of present groundwater will continue to pH 9.45 and higher na ion concentration. The secondary minerals formed from equeous phase are kaolinite, smectite, saponite, muscovite, mesolite, celadonite, microcline and calcite with uincreasing time. Modeling results are comparatively well fitted to pH and chemical composition of borehole groudwater, secondary minerals identified and tritium age of groundwater. The EQ6 modelling results are dependent on reliability of input parameters: water-rock ratio, effective reaction surface area and dissolution rates of mineral phases, which are difficult parameters to be measured.

  • The reaction between cyanide ion and triphenyl methane dyes is subject to marked catalysis by cationic micelles of cetyltrimethyl ammonium bromide(CTABr) and retarded by anionic micelles of sodium lauryl sulfate(NaLS). Added salts, anions inhibit the catalysis by CTABr, and cations, especially $Zn^{++},\;Cd^{++}$ decrease the retardation of the reaction rates in the presence of NaLS. The kinetic effects of the ionic micelles are much larger in water than in ethanol-water, methanol-water, propanol-water and acetone-water, but strange solvent effects, acceleration the reaction rates, was found in the reaction with malachite green in water-methanol system.

On-line Conversion Estimation for Solvent-free Enzymatic Esterification System with Water Activity Control

  • On-line conversion estimation of enzymatic esterification reactions in solvent-free media was investigated. In principle, conversion to ester can be determined from the amount of water produced by the reaction, because water is formed as a by-product in a stoichiometric manner. In this study, we estimated the water production rate only from some measurements of relative humidity and water balances without using any analytical methods. In order to test the performance of the on-line conversion estimation, the lipase-catalyzed esterification of n-capric acid and n-decal alcohol in solvent-free media was performed whilst controlling water activity at various values. The reaction conversions estimated on-line were similar to those determined by offline gas chromatographic analysis. However, when the water activity was controlled at higher values, discrepancies between the estimated conversion values and the measured values became significant. The deviation was found to be due to the inaccurate measurement of the water content in the reaction medium during the initial stages of the reaction. Using a digital filter, we were able to improve the accuracy of the on-line conversion estimation method considerably. Despite the simplicity of this method, the on-line estimated conversions were in good agreement with the off-line measured values.
