한국 울릉도 근해의 비브리오속의 분리연구 (Studies on the Isolation of Vibrio Species from Sea Water and Marine Products at Ulleung Island of Korea in 1985)

  • Authors studies on the isolation of non-sucrose fermentation Vibrio species such as V. parahaemolyticus, V. vulnificus and V. damsela from sea water, shellfishes and various algae at Ulleung island in the east of Korea on summer in 1985. Authors carried out test for isolated strains to biochemical characteristics, halophilism, hemolysis and serological reaction. The results obtained were as follows: 1. Strains isolated from total specimens were 25 strains of V. parahaemolyticus, 2 strains of V. vulnificus and 1 strain of V. damsela, respectively. 2. Isolated frequency of Vibrio species from sea water and marine products was lower than on specimens in the southern sea of Korea from 1980 to 1985. 3. On hemolysis reaction on blood agar media using human and rabbit erythrocytes, 14 strains among 25 strains of V. parahaemolyticus isolated were positive, and V. vulnificus and V. damsela were positive, respectively. 4. The distributions of serotypes of V. parahaemolyticus isolated were from O1 group to O10 group, and from K 10 to K 69. The results of K serotypes were as follows ; serotype K 10 and K 33 were 2 strains, serotype K 59 and K 69 were 1 strain, respectively. And 2 strains of V. vulnificus isolated were not agglutinated by antiserum of V. vulnificus. Therefore, the probability is that 2 strains of V. vulnificus isolated were other serotypes distributed on sea weater and marine products of Ulleung island of Korea.

Osmotic Shock에 의한 Vibrio vulnificus 사멸에 관한 연구 (Bactericidal Effect of Osmotic Shock Against Vibrio vulnificus)

  • As a process to establish an effective preventive measure of V. vulnificus septicemia, bactericidal effect of distilled water against V. vulnificus was studied. When about $2.0{\times}10^7\;CFU/ml$ of V. vulnificus was inoculated in distilled water, a dramatic decrease in the number of viable bacteria by 5 to $6LOG_{10}$ was observed in 5 minutes. Bactericidal kinetic curves could be divided into the first rapid killing phase until 1 minute and the later slow killing phase after then, showing the heterogeneity of the bacterial population inoculated. When V. vulnificus was inoculated in saline solutions having various salinities, significant decrease in the number of viable bacteria was noted only at salinities under 0.2%. The higher was the concentration of NaCl, the greater was the degree of protection against osmotic shock. When glucose, NaCl, $MgCl_2$, and $CaCl_2$ were diluted with deionized water to give same osmolarities and V. vulnificus was inoculated in each of them to compare the bactericidal curves plotted during the first 5 minutes after inoculation, the protection efficiencies were in the order of $MgCl_2>CaCl_2{\gg}NaCl{\gg}glucose$. Above results indicate that treatment(or thorough washing) of contaminated sea animals or other products with distilled water can be used as a preventive measure of V. vulnificus septicemia, and divalent cations can protect V. vulnifcus to osmotic shock with high efficiency.

Vibrio damsela의 분리연구 (Studies on the Isolation of Vibrio damsela)

  • Authors studied on the isolation of V. damsela from sea water, fish and shellfish at the Keoje Hae keumkang on the southern sea and at Hongdo island and Heucksan island on the western sea of Korea from May to September in 1986. Authors investigated for the isolated strains to bacteriological identification, hemolysis about various erythrocytes and antibiotic susceptibilities. The results obtained were as follows: 1. V. damsela was isolated 14 strains from total 383 specimens; 233 cases of sea water, 40 cases of fish and 110 cases of shellfish, respectively. Eight strains were isolated from sea water and 6 strains were isolated from shellfish. 2. The biochemical characteristics which differentiate it from other Vibrio species were indole negative, ornithine negative, Voges-Proskauer positive, arginine positive, galactose positive, glucose positive, maltose positive, mannose positive, trehalose positive, and growth in nutrient broth with 1% to 6% NaCl. 3. On hemolysis reaction on blood agar media using human, rabbit and guinea pig erythrocytes, human erythrocytes were 11 strain positive, rabbit erythrocytes were 12 strain positive and guinea pig erythrocytes were 13 strain positive. 4. Senistivity test using with chemotherapeutic agents of "BioLab" Microbial Sensitivity Test Discs were generally sensitived to amikacin, ampicillin, cephalothin, chloramphenicol, clindamycin, erythromycin, gentamycin, kanamycin, methicillin, penicillin, streptomycin, tetracycline and tobramycin, respectively, but were resistant to lincomycin.

Preservation of Marine Heterotrophic Bacteria by Using a Deep-freezing Method

  • The effect of cryoprotectants and suspending solutions on the preservation of marine heterotophic bacteria was investigated. Six halotolerant and four halophilic bacterial isolates suspended in either distilled water or artificial seawater were preserved in glycerol and dimethylsulfoxide at -70$\^{C}$, respectively. After one year of preservation, the recovery rates on the appropriate agar plates were estimated. The survival rate was found to be dependent on the strain tested, regardless of the preservation conditions tested.
