Liquid Chromatography Quadrupole Time-Of-Flight Tandem Mass Spectrometry for Selective Determination of Usnic Acid and Application in Pharmacokinetic Study

  • A rapid and sensitive method for determining usnic acid of Lethariella cladonioides in rat was established using high performance liquid chromatography (HPLC) quadrupole time-of-flight (QTOF) tandem mass (MS/MS). Rat plasma was pretreated by mixture of acetonitrile and chloroform to precipitate plasma proteins. Chromatographic separation was achieved on a column ($50{\times}2.1$ mm, $5{\mu}m$) with a mobile phase consisting of water (containing $5{\times}10^{-3}$ M ammonium formate, pH was adjusted to 3.0 with formic acid) and acetonitrile (20:80, v/v) at a flow rate of 0.3 mL/min. A tandem mass spectrometric detection with an electrospray ionization (ESI) interface was conducted via collision induced dissociation (CID) under negative ionization mode. The MS/MS transitions monitored were m/z 343.0448 ${\rightarrow}$ m/z 313.2017 for usnic acid and m/z 153.1024 ${\rightarrow}$ m/z 136.2136 for protocatechuic acid (internal standard). The linear range was calculated to be 2.0-160.0 ng/mL with a detection limit of 3.0 pg/mL. The inter- and intra-day accuracy and precision were within ${\pm}7.0%$. Pharmacokinetic study showed that the apartment of usnic acid in vivo confirmed to be a two compartment open model. The method was fully valid and will probably be an alternative for pharmacokinetic study of usnic acid.

Case Study on the Instability of the Slopes in Unsaturated Residual Soils Considering the Rainfall Characteristics (강우특성을 고려한 불포화 잔적토 비탈면의 붕괴사례 연구)

  • This study has obtained Soil Water Retention Curve (SWRC) of the unsaturated soil from the volumetric pressure plate extractor test and the triaxial compression tests was also conducted. By using the rainfall data measured in the site the seepage analysis of unsteady flow was performed with the program of SEEP/W in Geostudio 2007 and stability of the slope was analyzed with SLOPE/W program. Results of analyses show that shear strength of the unsaturated soil increases with the increase of matric suction. And it was also found that the net volumetric stress and the apparent cohesion increased with the matric suction. The seepage analysis of rainfall represents that the increasing rate of negative pore pressure at the zone of large negative pore pressure is appeared to be high even though lower rainfall intensity, but this tendency declines with ground depth. The stability analysis of slope was carried out for the actual plane of failure with the data representing the field condition. The factor of safety thus calculated was about unity (1.0) or just below, which means that the adopted method of analysis is in good agreement with the field condition.

The Effect of Skewness of Nonlinear Waves on the Transmission Rate through a Porous Wave Breaker (파형의 왜도가 투과성 방파제 투과율에 미치는 영향)

  • It has been presumed that highly nonlinear skewed waves frequently observed in a surf zone could significantly influence the transmission behaviour via a porous wave breaker due to its larger inertia force than its nonlinear counterparts of zero skewness [Cnoidal waves]. In this study, in order to confirm this perception, a numerical simulation has been implemented for 6 waves the skewness of that range from 1.02 to 1.032. A numerical simulation are based on the Tool Box called as the ihFoam that has its roots on the OpenFoam. Skewed waves are guided by the shoal of 1:30 slope, and the flow in the porous media are analyzed by adding the additional damping term into the RANS (Reynolds Averaged Navier-Stokes equation). Numerical results show that the highly nonlinear skewed waves are of higher transmitted ratio than its counterparts due to its stronger inertia force. In this study, in order to see whether or not the damping at the porous structure has an effect on the wave celerity, we also derived the dispersive relationships of Nonlinear Shallow Water Eq. [NSW] with damping at the porous structure being accounted. The newly derived dispersive relationships shows that the phase lag between the damping friction and the free surface elevation due to waves significantly influence the wave celerity.

Method for Evaluating Radionuclide Transport in Biosphere by Calculating Elapsed Transport Time (이동 경과 시간 계산을 이용한 생물권에서의 방사성 핵종 이동 평가 방법)

  • For geological disposal of radioactive wastes, a method was proposed to evaluate the radionuclide transport in the biosphere by calculating the elapsed time of nuclide migration. The radionuclides were supposed to be introduced from a natural barrier and reached a large surface water body following a groundwater flow in a shallow subsurface. The biosphere was defined as a shallow subsurface environment that included aquifers on a host rock. Using the proposed method, a calculation algorithm was established, and a computer code that implemented the algorithm was developed. The developed code was verified by comparing the simulation results of the simple cases with the results of the analytical solution and a public program, which has been widely used to evaluate the radiation dose using the radionuclide transport near the surface. A case study was constructed using the previous research for radionuclide transport from the hypothetical geological disposal repository. In the case study, the code calculated the mass discharge rate of radionuclide to a stream in the biosphere. Because the previous research only demonstrated the transport of radionuclides from the hypothetical repository to the host rock, the developed code in the present study could help identify the total transport of radionuclide along the complete pathway.

Quantitative analysis of cholesterol in infant formula by isotope dilution liquid chromatography-tandem mass spectrometry (동위원소희석 액체크로마토그래피 질량분석법에 의한 분유 내 콜레스테롤의 정량)

  • An isotope dilution liquid chromatography tandem mass spectrometry was developed as a primary method for the quantitative analysis of cholesterol in infant formula. Cholesterol-d4 was used as an internal standard and spiked into the infant formula sample. In order to release cholesterol out of cholesteryl ester, which is cholesterol bound to fatty acids in infant formula, saponification was carried out. Saponification conditions were optimized with heating temperature, reaction time and the concentration of KOH. The optimum conditions were as follows; heating temperature was 70 ℃, reaction time was 180 min and the concentration of KOH was 0.8 mL of 8 M KOH for about 0.1 g infant formula sample. Extraction of cholesterol out of sample solution was carried out with hexane uisng liquid-liquid extraction. Chromatographic analysis was carried out using Phenomenex Kinetex C18 column. Mobile phase was 0.1% acetic acid in methanol/water (v/v, 99/1) and flow rate was 0.3 mL/min. Cholesterol and cholesterol-d4 were monitored at mass transfer m/z 369/259 and 373/263 respectively. Reproducibility of the method was evaluated to be 0.23% of the measurement result. The expanded uncertainty of the measurement result of cholesterol in infant formula was approximately 1.9% at a 95% confidence level. NIST standard reference material having certified values of cholesterol in infant formula, was analyzed in order to verify this method. The ID-LC/MS/MS results were well agreed with the certified values of NIST SRM within the uncertainty.

Studies on the Positive Inotropic Mechanism of Aconiti Tuber (부자 강심성분의 작용기전에 관한 연구)

  • Aconiti tuber butanol fraction, which is isolated from the chloroform insoluble and water soluble extract of Aconitum volubile, has been recently known to have a potent positive inotropic effect in the isolated cardiac muscle preparations of various animals. The positive inotropic mechanism of Aconiti tuber butanol fraction, in relation with the external calcium, was studied using the isolated cat papillary muscle. The positive inotropic effect was dependent on the calcium concentration in the nutrient medium, and a synergistic relation could be demonstrated between Aconiti tuber butanol fraction and the external calcium. The inotropic effect of $10^{-4}g/ml$ of Aconiti tuber butanol fraction was equivalent to that of 0.06mM of calcium in the medium. After the treatment with a calcium influx inhibitor, Verapamil$(2{\pm}10^{-7}-10^{-6}M)$, the contractile force of the papillary muscle was markedly inhibited. In these preparations, Aconiti tuber butanol fraction restored the decreased contractility in a dose-dependent manner. It was suggested that the positive inotropic effect of Aconiti tuber butanol fraction might be related with the stimulating action on the calcium influx through the slow inward calcium channels in the cardiac cell membrane. In contrast with digitalis cardiac glycoside, Aconiti tuber butanol fraction infused intravenously into the anesthetized rabbit decreased the systemic arterial blood pressure and increased the carotid blood flow, but produced no prominent changes in the heart rate.

The Acid Rock Drainage and Hydraulic Characteristics of the Waste Rock Dump (폐석적치장의 산성배수발생 및 수리특성 분석)

  • This study was carried out to plan the prevention of the generation and discharge of Acid Rock Drainage (ARD). The Acid Base Accounting(ABA) test was performed for geological materials such as pit wall, waste rock and stream sediments near the Imgi abandoned pyrophyllite mine in Busan, Korea. In addition, hydraulic characteristics were tested with the disk tension infiltrometer around the waste rock dump. Maximum Potential Acidity(MPA) of geological materials near the Imgi mine was 246.942kg $H_2SO_4/t$, and maximum Acid Neutralising Capacity(ANC) was 8.7kg $H_2SO_4/t$. These results indicate the pit wall and waste rock, except most of stream sediments are acid generating geological materials. These have salt and free hydrogen ion which resulted from oxidation of sulfides. Hence they could be convert rain water to acid rock drainage. Although the waste rock dump of the Imgi mine have very low infiltration rate, slopes of the waste rock dump have many "V" type erosion gullies and multi-layers. These gullies and multi-layers have coarse clastic particle layers which have very large hydraulic conductivity. Through this coarse clastic particle layer a large part of rain flow into ground. And also this layer could function as aeration path which induced oxidation of sulfide minerals and generation of ARD continuously.

Heat Transfer Enhancement by Fins in a Latent Heat Storage System Using Phase Change Material (상변화물질을 이용한 축열시스템에서 핀에 의한 열전달 촉진 연구)

  • Heat transfer characteristics of low temperature latent heat storage systems have been examined for the circular finned and unfinned tubes using Na$_2$B$_4$O$\_$7/10H$_2$O as a phase change material. In order to reduce the supercooling of PCM, 3 wt% of Na$_2$B$_4$O$\_$7/10H$_2$O was added as the nucleating agent and 2.2 wt% of acrylic acid sodium sulfate was used as the thickener. The heat storage vessel has dimension of 530 mm height, 74 mm 1.D. and inner heat transfer tube is 480 mm height and 13.5 mm O.D. Water was employed as the heat transfer fluid. During the heat recovery experiment, the heat recovery rate was affected by the flow rates and inlet temperature of heat transfer fluid. The enhancement of heat transfer by fins over the unfinned tube system was found to be negligible in the thin finned tube systems, whereas the heat transfer coefficient in the thick finned tube system is approximately 60% higher than that in the unfinned lobe system. The experimentally determined heat transfer coefficient for the unfinned tube and thick finned tube systems are 150-260 W/㎡$^{\circ}C$ and 230-530 W/㎡$^{\circ}C$, respectively. The fin efficiency based on the heat transfer coefficient and area increased by fins was found to be 0.05 and 0.26 for the thin and the thick finned tube systems.

  • A method was developed for the determination of fructooligosaccharides and raffinose contents in infant formula. The samples were extracted and analyzed by liquid chromatography equipped with carbohydrate column and evaporative light scattering detector. The mobile phase used for the gradient mode was water-acetonitrile, at a flow rate of 1.0mL/min. The method showed a mean recovery of 95-99%, the relative standard deviation obtained in the precision study was 0.774-8.982%, the quantification and detection limits were 25-50mg/L.

Manufacture of the Hydrophobic HY-type Zeolite-honeycomb and Its Adsorption/Desorption Characteristics for the Benzene, o-xylene, and MEK (소수성 HY-형 제올라이트제 하니컴의 제조 및 그 하니컴의 벤젠, o-xylene, MEK에 대한 흡.탈착특성)

  • We performed the experiments to manufacture the hydrophobic $200cells/in^2$-zeolite honeycomb using HY-type zeolite of Si/Al ratio of 80 for separating and removing the VOCs emitted from small and medium size-plants by adsorption and to determine the drying method for the honeycomb at $105^{\circ}C$ without cracking, then measured performances of the honeycomb to adsorb the benzene, o-xylene, and MEK and to desorb the benzene and MEK saturated on the honeycomb by the nitrogen gas as the desorption gas. As a results, the good honeycomb was formed and the honeycomb was not cracked when the mixing ratio of the zeolite to bentonite to methyl cellulose to polyvinyl alcohol to glycerine to water is 100 : 8.73 : 2.18 : 4.19 : 1.38 : 126 and dried the honeycomb at $105^{\circ}C$ for 24 hours in the drying oven. The shape of the dried honeycomb was not changed after calcination, and the compressive strengths of the honeycomb after drying and calcination were 6.7 and $0.69kg/cm^2$, respectively. The adsorption efficiencies of the honeycomb for benzene, o-xylene, and MEK were $92{\sim}96%$ at the room temperature. The desorption efficiency at $180^{\circ}C$ was higher than that at $150^{\circ}C\;by\;1.5{\sim}13.8%$ depending on the flow rate of the nitrogen gas, and it was found that desorption efficiency is higher than 85% at $180^{\circ}C$ and 1.0L/min of the nitrogen gas. At $180^{\circ}C$ and 0.2 L/min, the concentration of the benzene and MEK in the used desorption gas are higher than 40,000 and 50,000ppm, respectively, so it be used as the fuel for preheating the desorption gas fed into the column in desorption cycle.
