가정용 소형 퇴비화용기에 의한 부엌쓰레기의 분산식 퇴비화;III. 실험실조건에서 이중벽 소형 용기에 의한 퇴비화 연구 (Decentralized Composting of Garbage in a Small Composter for Dwelling House;III. Laboratory Composting of the Household Garbase in a Small Bin with Double Layer Walls)

  • 주택용 소형 퇴비화용기에 의한 부엌쓰레기의 퇴비화 가능성을 알아보기 위하여 두가지 용기 (Type 3 과 Type 4)를 제작하여 실험실 실험을 통하여 조사하였다. 두용기의 구조는 다 같이 용기의 벽을 이중으로 하였으며 Type 4는 보온을 하였고 Type 3은 보온을 하지 않았다. 겨울철 실험을 통하여 type 3은 우리나라의 기후여건에 사용이 불가능한 것으로 판단되어 봄철 및 여름철 실험에서 제외하였다. 따라서 Type 4에 대한 3 계절 실험결과를 요 약하면 다음과 같다. 1) 퇴비화기간 중 상승한 최고온도는 겨울 $43^{\circ}C$, 봄철 $55^{\circ}C$ 그리고 여릅철 $56^{\circ}C$ 였다. 2) 퇴비화원료물질의 무게는 8주 후 평균 63.3% 그리고 부피는 78% 감소하였다. 3) 퇴비층의 밀도는 겨울철 1.5kg/l, 봄철 0.8kg/1 그리고 여름철 0.8kg/1였다. 4) 수분함량은 전 퇴비화기간 동안 큰 변동이 없었으며 8주 후 겨울철 79.3%, 봄철 75.0% 그리고 여름철 70.0%였다. 5) pH 는 모두 첫주에 증가한 후 2주째에 감소하였다. 그 이후는 계절별 차이는 있었으나 계속 증가하였다. 8주 후 pH는 겨울철 6.19, 봄철 7.59 그리고 여름철에 8.69였다. 6) 퇴비화시간의 경과에 따른 회분 및 유기물함량은 분해속도가 빠를수록 증감현상이 뚜렷하였고 cellulose 및 lignin은 증가, hemicellulose 함량은 감소하였다. 7) 질소함량은 3.3-6.8%로 높았으며, 특히 여름철에 대단히 높았다. 암모늄태 질소함량은 전반에 증가했다가 감소하였으며 겨울철 8주째 2.404mg/kg, 봄철 3주째 12,400mg/kg 그리고 여름철 3주째 20,718mg/kg으로 가장 높았다. C/N율은 퇴비화기간의 경과에 따라 감소하였으나 차이는 크지 않았다. 여름철에 질산화가 가장 활발하였다. 8) 휘발성 및 고급유기산함량은 초기에 증가했다가 계절별로 시기적 차이는 있으나 감소하였다. 총 유기산의 최고농도는 겨울철 6주째 9.7%, 봄철 6주째 14.8% 그리고 여름철에는 2주째 15.8%였다. 9) 퇴비의 부숙도와는 무관하게 각 무기성분함량은 $P_2O_5$ 0.9-4.4%, $K_2O$ 1.6-2.4%, CaO 2.2-5.4% 그리고 MgO 0.30-0.61%였다. 10) CN 및 각종 중금속함량도 퇴비화시간의 경과에 따라 큰 변화가 없었으며 각 함량범위는 계절 구분없이 CN 0.21-14.55 mg/kg, Zn 11-166 mg/kg, Cu 5-65 mg/kg, Cd 0.5-10.8 mg/kg, Pb 6-35 mg/kg, Cr ND-33 mg/kg 그리고 Hg ND-302.04 g/kg 이였으며 중금속 역시 다른 무기성분들과 마찬가지로 퇴비화가 진행되면서 축적되지는 않았다.

해조의 식용분말화에 관한 연구 (A Study on the Nutritive Value and Utilization of Powdered Seaweeds)

  • I. Subject of the study A study on the nutritive value and utilization of powdered seaweeds. II. Purpose and Importance of the study A. In Korea the shortage of food will be inevitable by the rapidly growing population. It will be very important study to develop a new food from the seaweeds which were not used hitherto for human consumption. B. The several kinds of seaweeds have been used by man in Korea mainly as side-dishes. However, a properly powdered seaweed will enable itself to be a good supplement or mixture to certain cereal flours. C. By adding the powdered seaweed to any cereals which have long been staple foods in this country the two fold benefits; saving of cereals and change of dietary pattern, will be secured. III. Objects and scope of the study A. Objects of the study The objects will come under four items. 1. To develop a powdered seaweed as a new food from the seaweeds which have been not used for human consumption. 2. To evaluate the nutritional quality of the products the analysis for chemical composition and animal feeding experiment will be conducted. 3. Experimental cocking and accepability test will be conducted for the powdered products to evaluate the value as food stuff. 4. Sanitary test and also economical analysis will be conducted for the powdered products. B. Scope of the study 1. Production of seaweed powders Sargassum fulvellum growing in eastern coast and Sargassum patens C.A. in southern coast were used as the material for the powders. These algae, which have been not used for human consumption, were pulverized through the processes of washing, drying, pulverization, etc. 2. Nutritional experiments a. Chemical composition Proximate components (water, protein, fat, cellulose, sugar, ash, salt), minerals (calcium, phosphorus, iron, iodine), vitamins (A, $B_1,\;B_2$ niacin, C) and amino acids were analyzed for the seaweed powders. b. Animal feeding experiment Weaning 160 rats (80 male and 80 female rats) were used as experimental animals, dividing them into 16 groups, 10 rats each group. Each group was fed for 12 weeks on cereal diet (Wheat flour, rice powder, barley powder, potato powder, corn flour) with the supplementary levels of 5%, 10%, 15%, 20% and 30% of the seaweed powder. After the feeding the growth, feed efficiency ratio, protain efficiency ratio and ,organs weights were checked and urine analysis, feces analysis and serum analysis were also conducted. 3. Experimental cooking and acceptability test a. Several basic studies were conducted to find the characteristics of the seaweed powder. b. 17 kinds of Korean dishes and 9 kinds of foreign dishes were prepared with cereal flours (wheat, rice, barley, potato, corn) with the supplementary levels of 5%, 10%, 15%, 20% and 30% of the seaweed powder. c. Acceptability test for the dishes was conducted according to plank's Form. 4. Sanitary test The heavy metals (Cd, Pb, As, Hg) in the seaweed powders were determined. 5. Economical analysis The retail price of the seaweed powder was compared with those of other cereals in the market. And also economical analysis was made from the nutritional point of view, calculating the body weight gained in grams per unit price of each feeding diet. IV. Results of the study and the suggestion for application A. Chemical composition 1. There is no any big difference in proximate components between powders of Sargassum fulvellum in eastern coast and Sargassum patens C.A. in southern coast. Seasonal difference is also not significant. Higher levels of protein, cellulose, ash and salt were found in the powders compared with common cereal foods. 2. The levels of calcium (Ca) and iron (Fe) in the powders were significantly higher than common cereal foods and also rich in iodine (I). Existence of vitamin A and vitamin C in the Powders is different point from cereal foods. Vitamin $B_1\;and\;B_2$ are also relatively rich in the powders.'Vitamin A in ·Sargassum fulvellum is high and the levels of some minerals and vitamins are seemed4 to be some influenced by seasons. 3. In the amino acid composition methionine, isoleucine, Iysine and valine are limiting amino acids. The protein qualities of Sargassum fulvellum and Sargassum patens C.A. are seemed to be .almost same and generally ·good. Seasonal difference in amino acid composition was found. B. Animal feeding experiment 1. The best growth was found at.10% supplemental level of the seaweed Powder and lower growth rate was shown at 30% level. 2. It was shown that 15% supplemental level of the Seaweed powder seems to fulfil, to some extent the mineral requirement of the animals. 3. No any changes were found in organs development except that, in kidney, there found decreasing in weight by increasing the supplemental level of the seaweed powder. 4. There is no any significant changes in nitrogen retention, serum cholesterol, serum calcium and urinary calcium in each supplemental level of the seaweed powder. 5. In animal feeding experiment it was concluded that $5%{\sim}15%$ levels supplementation of the seaweed powder are possible. C. Experimental cooking and acceptability test 1. The seaweed powder showed to be utilized more excellently in foreign cookings than in Korean cookings. Higher supplemental level of seaweed was passible in foreign cookings. 2. Hae-Jo-Kang and Jeon-Byung were more excellent than Song-Pyun, wheat cake, Soo-Je-Bee and wheat noodle. Hae-Je-Kang was excellent in its quality even as high as 5% supplemental level. 3. The higher levels of supplementation were used the more sticky cooking products were obtained. Song-Pyun and wheat cake were palatable and lustrous in 2% supplementation level. 4. In drop cookie the higher levels of supplementation, the more crisp product was obtained, compared with other cookies. 5. Corn cake, thin rice gruel, rice gruel and potato Jeon-Byung were more excellent in their quality than potato Man-Doo and potato noodle. Corn cake, thin rice gruel and rice gruel were excellent even as high as 5% supplementation level. 6. In several cooking Porducts some seaweed-oder was perceived in case of 3% or more levels of supplementation. This may be much diminished by the use of proper condiments. D. Sanitary test It seems that there is no any heavy metals (Cd, Pb, As, Hg) problem in these seaweed Powders in case these Powders are used as supplements to any cereal flours E. Economical analysis The price of the seaweed powder is lower than those of other cereals and that may be more lowered when mass production of the seaweed powder is made in future. The supplement of the seaweed powder to any cereals is also economical with the criterion of animal growth rate. F. It is recommended that these seaweed powders should be developed and used as supplement to any cereal flours or used as other food material. By doing so, both saving of cereals and improvement of individual's nutrition will greatly be achieved. It is also recommended that the feeding experiment for men would be conducted in future.

대기오염집중측정소별 2013~2015년 사이의 PM2.5 화학적 특성 차이 및 유발인자 조사 (Difference in Chemical Composition of PM2.5 and Investigation of its Causing Factors between 2013 and 2015 in Air Pollution Intensive Monitoring Stations)

  • In this study, difference in chemical composition of $PM_{2.5}$ observed between the year 2013 and 2015 at six air quality intensive monitoring stations (Bangryenogdo (BR), Seoul (SL), Daejeon (DJ), Gwangju (GJ), Ulsan (US), and Jeju (JJ)) was investigated and the possible factors causing their difference were also discussed. $PM_{2.5}$, organic and elemental carbon (OC and EC), and water-soluble ionic species concentrations were observed on a hourly basis in the six stations. The difference in chemical composition by regions was examined based on emissions of gaseous criteria pollutants (CO, $SO_2$, and $NO_2$), meteorological parameters (wind speed, temperature, and relative humidity), and origins and transport pathways of air masses. For the years 2013 and 2014, annual average $PM_{2.5}$ was in the order of SL ($${\sim_=}DJ$$)>GJ>BR>US>JJ, but the highest concentration in 2015 was found at DJ, following by GJ ($${\sim_=}SJ$$)>BR>US>JJ. Similar patterns were found in $SO{_4}^{2-}$, $NO_3{^-}$, and $NH_4{^+}$. Lower $PM_{2.5}$ at SL than at DJ and GJ was resulted from low concentrations of secondary ionic species. Annual average concentrations of OC and EC by regions had no big difference among the years, but their patterns were distinct from the $PM_{2.5}$, $SO{_4}^{2-}$, $NO_3{^-}$, and $NH_4{^+}$ concentrations by regions. 4-day air mass backward trajectory calculations indicated that in the event of daily average $PM_{2.5}$ exceeding the monthly average values, >70% of the air masses reaching the all stations were coming from northeastern Chinese polluted regions, indicating the long-range transportation (LTP) was an important contributor to $PM_{2.5}$ and its chemical composition at the stations. Lower concentrations of secondary ionic species and $PM_{2.5}$ at SL in 2015 than those at DJ and GJ sites were due to the decrease in impact by LTP from polluted Chinese regions, rather than the difference in local emissions of criteria gas pollutants ($SO_2$, $NO_2$, and $NH_3$) among the SL, DJ, and GJ sites. The difference in annual average $SO{_4}^{2-}$ by regions was resulted from combination of the difference in local $SO_2$ emissions and chemical conversion of $SO_2$ to $SO{_4}^{2-}$, and LTP from China. However, the $SO{_4}^{2-}$ at the sites were more influenced by LTP than the formation by chemical transformation of locally emitted $SO_2$. The $NO_3{^-}$ increase was closely associated with the increase in local emissions of nitrogen oxides at four urban sites except for the BR and JJ, as well as the LTP with a small contribution. Among the meterological parameters (wind speed, temperature, and relative humidity), the ambient temperature was most important factor to control the variation of $PM_{2.5}$ and its major chemical components concentrations. In other words, as the average temperature increases, the $PM_{2.5}$, OC, EC, and $NO_3{^-}$ concentrations showed a decreasing tendency, especially with a prominent feature in $NO_3{^-}$. Results from a case study that examined the $PM_{2.5}$ and its major chemical data observed between February 19 and March 2, 2014 at the all stations suggest that ambient $SO{_4}^{2-}$ and $NO_3{^-}$ concentrations are not necessarily proportional to the concentrations of their precursor emissions because the rates at which they form and their gas/particle partitioning may be controlled by factors (e.g., long range transportation) other than the concentration of the precursor gases.

LC-MS/MS를 이용한 농산물 중 살균제 Validamycin A의 시험법 개발 (Development of Analytical Method for Detection of Fungicide Validamycin A Residues in Agricultural Products Using LC-MS/MS)

  • 발리다마이신 에이는 아미노글리코사이드계 항생물질로 이탄당인 균당을 2개의 포도당으로 분해하는 효소(trehalase)의 작용을 저해하여 균 증식을 억제한다. 우리나라에서는 벼, 고추, 복숭아, 마늘, 양파, 참다래 등에 세균성 병해를 방제하기 위해 농약으로 등록되어 있다. 국외의 경우 일본에 잔류허용기준(MRLs)이 감자 포함 43품목에서 0.05-0.06 mg/kg으로 기준이 설정되어 있고, 유럽은 0.01 mg/kg 일률 기준을 적용하고 있으며 국내의 경우 기준이 설정될 예정이다. 축산물의 경우 잔류물의 정의를 설정하고 있지 않으나 농산물의 경우 잔류물의 정의는 일본에서 모화합물로 설정되어 있으며 국내에서도 모화합물로 규정할 예정이다. 따라서 발리다마이신 에이의 잔류허용기준 신설에 따른 안전관리를 위하여 공정시험법을 개발하였다. 분석기기는 발리다마이신 에이의 물리 화학적 특성을 고려하여 선택성과 감도가 우수한 LC-MS/MS를 분석기기로 선정하였고 메탄올/물(50/50, v/v)을 이용하여 진탕 추출 후 HLB 카트리지를 이용한 정제 조건을 확립하여 시험법을 개발하였다. 발리다마이신 에이의 직선성은 결정계수($r^2$)가 0.99이상으로 우수하였으며, 검출한계 및 정량한계는 각각 0.005, 0.01 mg/kg으로 높은 감도를 나타내었다. 대표 농산물 5종(현미, 감자, 대두, 감귤, 고추)에 대하여 정량한계, 정량한계 10배, 정량한계 50배 수준으로 회수율 실험한 결과 평균 회수율(5반복)은 72.5-118.3%이었으며 분석오차는 10.3% 이하로 정확성 및 재현성이 우수함을 확인할 수 있었다. 검증 결과 각 농도별 발리다마이신 에이의 평균 회수율은 70.9~117.7%이었고, 상대표준편차(RSD)는 7.9% 이하로 조사되었다. 두 실험실간 회수율 결과에 따른 평균값은 75.9~112.7%이며 RSD는 18%미만으로 본 연구는 국제식품규격위원회 가이드라인(Codex Alimentarius Commission, CAC/GL 40)의 잔류농약 분석기준 및 식품의약품안전평가원의 '식품등 시험법 마련 표준절차에 관한 가이드라인(2016)'에 적합한 수준임을 검증하였다. 따라서 본 연구에서 개발한 시험법은 농산물 중 발리다마이신 에이의 잔류검사를 위한 공정시험법으로 활용할 수 있으며 유사 농산물에 대한 적용도 가능하리라 판단되어 향후 국내 농산물 중 농약의 잔류허용기준 신설 및 잔류농약 검사의 기초자료로 활용 가능할 것이다.

QuEChERS법 및 LC-MS/MS를 이용한 농산물 중 Oxytetracycline의 잔류시험법 개발 및 검증 (Development and Validation of the Analytical Method for Oxytetracycline in Agricultural Products using QuEChERS and LC-MS/MS)

  • 옥시테트라사이클린은 침투이행성 살균제로 배추, 고추 등의 농산물에 무름병, 줄기속마름병에 효과가 있다. 본 연구는 농산물 중 기준신설 예정농약 옥시테트라사이클린이 농산물 대하여 등록되지 않아 이에 대한 공정시험법을 개발하기 위하여 수행되었다. 옥시테트라사이클린의 잔류물의 정의는 미국, 일본의 경우 농산물 대상으로 모화합물로 설정되어 있다. 현재, 국내에서는 농산물 중 옥시테트라사이클린의 잔류물의 정의 및 잔류허용기준이 설정되어있지 않고, 국내 농산물(고추 등)에 대한 잔류허용기준 신설이 최초 요청되었으며 국내 유통 농산물 중 잔류량에 대한 안전관리 확보를 위해 잔류물의 정의를 모화합물로 규정하고 적부판정을 위한 공정시험법을 개발하고자 하였다. 옥시테트라사이클린의 물리 화학적 특성을 고려하여 QuEChERS법을 이용한 추출 및 정제법을 최적화하여 LC-MS/MS에 의한 분석법을 확립하였다. 수용성 유기용매인 메탄올을 추출 용매로 사용하여 pH 조절 및 염화나트륨을 첨가하여 추출법을 최적화하고, d-SPE 흡착제를 이용하여 간섭물질을 효과적으로 제거하여 정제법을 확립하였다. 옥시테트라사이클린의 결정계수($r^2$)는 0.99 이상으로 높은 직선성을 보여주었고, 옥시테트라사이클린의 시험법 정량한계(LOQ)는 0.01 mg/kg이며, 대표 농산물 5종(현미, 감자, 대두, 감귤, 고추)에 대하여 LOQ (0.01 mg/kg), $10{\times}LOQ$ (0.1 mg/kg), $50{\times}LOQ$ (0.5 mg/kg) 수준으로 회수율 실험한 결과 평균 회수율(n=5)은 80.0~108.2%이었으며 상대표준편차는 11.4%이하로 확인되었다. 또한 실험실간 검증 결과 두 실험실간 회수율 결과에 따른 평균값은 83.5~103.2%이며 변이계수는 14.1% 이하로 조사되어, 본 연구는 국제식품규격위원회 가이드라인(Codex Alimentarius Commission, CAC/GL 40)의 잔류농약 분석 기준 및 식품의약품안전평가원의 '식품등 시험법 마련 표준절차에 관한 가이드라인(2016)'에 적합한 수준임을 확인하였다. 따라서 본 연구에서 개발한 시험법은 농산물 중 잔류할 수 있는 옥시테트라사이클린의 안전관리를 위한 공정시험법으로 활용 가능할 것이다.
