Isolation of Angiotensin Converting enzyme inhibitors from Ripe Cucurbita moschata Duch

  • Angiotensin converting enzyme (ACE) inhibitor acts on the inhibition of ACE and causes a decrease in blood pressure. There have been several reports on screening of ACE inhibitors from natural food products and protein hydrolysates of various food sources. Ripe Cucurbita moschata Duch has been used as an oriental medicine in Korea. To isolate ACE inhibitors, crude water extracts of the edible portion of ripe Cucurbita moschata Duch were obtained after heating in water at 95$^{\circ}C$ for 2 h. Crude extracts were then filtered using PM-10 and YM-1 membranes. The membrane-filtered solution was loaded onto Sephadex G-15 column equlibrated with a phosphate buffer. Among the four major fractions of gel permeation chromatography, the second fraction had the highest inhibitory activity of 65%. Further purification of the fraction using reversed-phase HPLC with a $C_{18}$ column produced ACE inhibitors, which were identified as a mixture having molecular mass of 222 and 273 by Tandem mass spectrometry.

Effect of Internal Fluid Resonance on the Performance of a Floating OWC Device

  • In the present study, the performance of a floating oscillating water column (OWC) device has been studied in regular waves. The OWC model has the shape of a hollow cylinder. The linear potential theory is assumed, and a matched eigenfunction expansion method(MEEM) is applied for solving the diffraction and radiation problems. The radiation problem involves the radiation of waves by the heaving motion of a floating OWC device and the oscillating pressure in the air chamber. The characteristics of the exciting forces, hydrodynamic forces, flow rate, air pressure in the chamber, and heave motion response are investigated with various system parameters, such as the inner radius, draft of an OWC, and turbine constant. The efficiency of a floating OWC device is estimated in connection with the extracted wave power and capture width. Specifically, the piston-mode resonance in an internal fluid region plays an important role in the performance of a floating OWC device, along with the heave motion resonance. The developed prediction tool will help determine the various design parameters affecting the performance of a floating OWC device in waves.

괭생이 모자반에 의한 수중 중금속 Cd(II), Pb(II) 이온의 제거 (Removal of Heavy Metals, Cd(II) and Pb(II) Ions in water by Sargassum Herneri)

  • 갈조류인 Sargassum horneri를 중금속 흡착제로 사용하여 중금속 Cd(II) 및 Pb(II)이온을 제거하였다. Sargassum horneri는 동해안에서 채취하였고, 풍건 건조시켜 40~60 mesh의 입자 크기로 흡착제를 만들어 사용하였다. batch법은 진탕 시간에 따른 흡착량을 측정함으로써 흡착속도를 조사하였고, column법은 해조 분말을 충진 시킨후 일정농도의 중금속용액을 1 mL/min의 속도로 흘려보내면서 흡착시키고 흡착량을 알아 보았다. 중금석 흡착에 미치는 pH의 영향은 batch법과 column법 모두, pH 10.5>7.0>3.5순으로 나타났다. Pb(II)이온이 Cd(II)이온보다 높은 흡착량을 보였다. 특히 batch법에서는 모든 pH의 조건하에서 5분이내에 최대 흡착량에 도달하였다. 회수율은 batch법에 의해 흡착된 중금속의 회수율이 column법에 의해 흡착된 중금속의 회수율 보다 조금 높게 나타났다.

강변여과에서의 은나노입자의 영향 : 실험실규모 컬럼 실험 (Effect of silver nanoparticles on the performance of riverbank filtration: Column study)

  • Soil column experiments were evaluated effects of silver nanoparticles (i.e., 0, 2.5, 5, and 10 mg/L) on the microbial viability which is strongly associated with the degradation of organic matter, pharmaceutically active compounds(PhACs) and biological oxidation of nitrogenous compounds during river bank filtration. The addition of silver nanoparticles resulted in almost no change in the aqueous matrix. However, the intact cell concentration decreased with addition of silver nanoparticles from 2.5 to 10 mg/L, which accounted for 76% to 82% reduction compared to that of control (silver nanoparticles free surface water). The decrease in adenosine triphosphate was more pronounced; thus, the number and active cells in aqueous phase were concurrently decreased with added silver nanoparticles. Based on the florescence excitation-emission matrix and liquid chromatograph - organic carbon detection analyses, it shows that the removal of protein-like substances was relatively higher than that of humic-like substances, and polysaccharide was substantially reduced. But the extent of those substances removed during soil passage was decreased with the increasing concentration of silver nanoparticles. The attenuation of ionic PhACs ranged from 55% to 80%, depending on the concentration of silver nanoparticles. The attenuation of neutral PhACs ranged between 72% and 77%, which was relatively lower than that observed for the ionic PhACs. The microbial viability was affected by silver nanoparticles, which also resulted in inhibition of nitrifiers.

막증류 공정의 전처리 공정으로서 천연 제올라이트 컬럼 적용 (Scalants removal from synthetic RO brine using natural zeolite)

  • Membrane distillation (MD) is the thermally driven water separation process based on the vapor pressure difference across the membrane. In order to increase the water recovery of the conventional RO process, the additional MD-PRO pocess was suggested. In this study, the syntheric RO brine was used as a feed solution of the MD process. Due to the high salinity of the RO brine, the MD membrane could be fouled by the scalants. In order to mitigate the scaling on the MD membrane surface, the pre-treatment process using the column filled by natural zeolite was applied. The roughing filter was installed between the pre-treatment process and MD system in order to prevent possible particulate fouling by the debries of the natural zeolite. Moreover, in order to enhance the CEC of the natural zeolite, the NaCl soaking was conducted. The flux and electronic conductivity were monitored under given experimental conditions. And the membrane morphology and the chemical compositions were analyzed by using the SEM-EDX.

Theory and Practices of Water Pollution Control by Wetland - a Case Study of Reed Wetland in Baiyangdian Lake

  • Wetland is an important eco-system on the earth and can effectively control agricultural non-point source pollution. Reed is a typical wetland plant for land/inland water ecotone in north China. The studies indicated that reed had a underground rooted-stem, which formed a 'high speeded-connecting vessels' i.e. reed root channel (RRC) in Baiyangdian lake of Hebei province. It spread predominantly along horizontal direction underground and are mainly distributed at 18-75 cm. The results of field work from healthy reed-wetland (HRW) and regarded reed-wetland (RRW) showed that the reed, averagely, in HRW is 4.2 m height, 1 cm diameter, 65/m2 density; in RRW is 2.4 m height, 3 mm diameter, 86/m2 density. These results indicated the regradation of the function of RRC in RRW. The results of laboratory work of sewage purification from reed soil column (RSC) $(0\~100cm)$ and wheat soil column (WSC) showed that the efficiency of purification to sewage, in RSC, is high than in WSC, especially for phosphorus. The efficiency of purification, in RSC, is $92.6\%$ for total phosphorus, $43.5\%$ for total nitrogen, $54.1\%$ fur COD, respectively; in WSC, is $86.0\%$ for total phosphorus, $241.3\%$ for total nitrogen, $29.8\%$ for COD, respectively.

Nutrient dynamics study of overlying water affected by peroxide-treated sediment

  • Background: Loading of excess nutrient via bioremediation of polluted sediment to overlying water could trigger anoxia and eutrophication in coastal area. The aim of this research was to understand the changes of overlying water features such as dissolved oxygen (DO); pH; oxidation reduction potential (ORP); $chlorophyll-{\alpha}$ ($Chl-{\alpha}$); and nitrogen nutrients ammonia ($N-NH_4{^+}$), nitrate ($N-NO_3{^-}$), and nitrite ($N-NO_2^-$) when the sediment was not treated (control) and treated by calcium peroxide for 5 weeks. Methods: The water samples were analyzed for measuring physical and chemical properties along with the sediment analyzed by polymerase chain reaction (PCR) including denaturing gradient gel electrophoresis (DGGE) for identifying the phylogenetic affiliation of microbial communities. Results: Results showed that due to the addition of calcium peroxide in sediment, the overlying water exposed the rise of dissolve oxygen, pH, and ORP than control. Among the nitrogen nutrients, ammonia inhibition was higher in calcium peroxide treatment than control but in case of nitrate inhibition, it was reversed than control. $Chlorophyll-{\alpha}$ was declined in treatment column water by 30% where it was 20% in control column water. Actibacter and Salegentibacter group were detectable in the calcium-peroxide-treated sediment; in contrary, no detectable community ware found in control sediment. Both phylogenetic groups are closely related to marine microflora. Conclusions: This study emphasizes the importance of calcium peroxide as an oxygen release material. Interaction with peroxide proved to be enhancing the formation of microbial community that are beneficial for biodegradation and spontaneity of nutrient attenuation into overlying water.

토양증기추출(Soil Vapor Extraction)을 이용한 토양 내 Trichloroethylene (TCE)과 Toluene정화 실험 (Laboratory Tests for Trichloroethylene (TCE) and Toluene Remediation in Soil Using Soil Vapor Extraction)

  • 토양증기추출(Soil Vapor Extraction)법을 이용하며 대표적 휘발성 NAPL(Non-aqueous phase liquid)인 TCE (trichloroethylene)와 toluene을 토양으로부터 제거하는 칼럼 실험을 실시하였다. 균질한 Ottawa sand와 실제 오염지역의 토앙들이 사용되었으며, 토양특성 및 증기추출 조건들이 정화효율에 미치는 영향을 규명하였다. 직경 2.5cm, 길이 30cm인 유리 칼럼이 사용되었고, 붉은색으로 염색된 TCE 또는 toluene 4 gram이 주입되었다 주입 공기가 일정한 속도로 칼림 내에 주입될 수 있도록 공기 유량계를 설치하여 0.03 L/min의 일정한 속도로 공기가 주입되도록 하고, 퍼지 장치를 설치하여 주입 공기의 습윤도를 99% 이상으로 유지하였다. 칼럼으로부터 유출되는 가스는 자동 가스시료주입 밸브장치를 이용하여 가스크로마토그래피 칼럼 내로 주입되게 하여, FID 검출기로 유출 가스 농도를 분석하였다. Ottawa sand로 충진된 칼럼실험에서는 매질의 입자크기, 함수율, 토양 내 오염물 체류시간 등을 변화시켜 실험을 반복하였다. TCE로 오염된 세립질 Ottawa sand 칼럼실험에서 유출 공기의 최대 농도는 조립질 Ottawa sand 칼럼의 유출 농도보다 약 20% 정도 감소하였고, 오염지역의 실제토양 칼럼실험에서는 최대유출농도가 조립질 Ottawa sand칼럼의 농도보다 약 50% 감소하였으나, 20 liter 공기 주입 후부터는 모두 비슷한 농도감소 현상을 나타내었으며 초기 주입량의 90 % 이상이 제거되었다. 함수율증가에 따른 유출공기의 농도 감소는 거의 나타나지 않았으며, TCE 주입 후 7일 동안 방치하였다가 SVE를 실시한 칼럼 실험에서도 잔류하는 TCE의 양이 약간 증가하였지만 20liter 공기 추출 후에는 초기 주입량의 90% 가, 40 liter 공기 추출 후에는 98% 이상이 제거되었다. Toluene 으로 오염된 칼럼 실험에서도 TCE와 비슷한 제거 경향을 나타냈으며 200 liter 공기 추출 후에는 오염물 초기 주입량의 98% 이상이 제거되었다. 본 실험 결과로부터 증기추출법을 이용한 TCE, toluene 정화 효율성이 규명되었으며, 휘발성 NAPL로 오염된 실제 토양을 복원하기위한 SVE법의 적용가능성을 확인할 수 있었다.
