Transport Properties of Polymer Blend Membranes of Sulfonated and Nonsulfonated Polysulfones for Direct Methanol Fuel Cell Application

  • The relation between the phase separated morphologies and their transport properties in the polymer blend membrane for direct methanol fuel cell application was studied. In order to enhance the proton conductivity and reduce the methanol crossover, sulfonated poly(arylene ether sulfone) copolymer, with a sulfonation of 60 mol% (sPAES-60), was blended with nonsulfonated poly(ether sulfone) copolymer (RH-2000, Solvay). Various morphologies were obtained by varying the drying condition and the concentration of the casting solution (10, 15, 20 wt%). The transport properties of proton and methanol molecule through the polymer blend membranes were studied according to the absorbed water. AC impedance spectroscopy was used to measure the proton conductivity and a liquid permeability measuring instrument was designed to measure the methanol permeability. The state of water in the blend membranes was confirmed by differential scanning calorimetry and was used to correlate the morphology of the membrane with the membrane transport properties.

  • An extensive experimental survey on the retention behaviors of phenols in the binary solvent system such as methanol-water, acetonitrile-water and tetrahydrofuran-water as well as the ternary solvent system such as methanol-acetonitrile-water and methanol-tetrahydrofuran-water is presented. A linear equation, which describes the capacity factor as a function of the solvent composition in the mobile phase and is able to predict the retention behaviors of phenols, was obtained. The iso-eluotropic lines for the binary and ternary solvent system are based on the equal strength of the methanol-water solvent which shows an optimum separation of the phenols used. The specific effect of each solute in the binary solvent system appeared to be larger than those in the ternary system.

Study on The Anti-HSV(Herpes Simplex Virus) Activity of Natural complex Products (한약 탕제를 이용한 항 Herpes virus 제제의 개발 연구)

  • In order to search for anti-HSV agents from natural complex products, we extended the number of specimens. Both methanol extract and boiling water extract of the natural complex products were screened to detect anti-HSV activity by MTT assay. Anti-HSV activities of thirteen natural complex products extracted by methanol and boiling water were screened. Three of 13 natural complex products extracted by methanol showed efficacy against HSV. Natural complex products showing anti-HSV activities as methanol extracts were No.3, 6, 11 and their Sl were 323.809, 2811.041 and 708.20. As water boiling extracts, No.8 and No.11 have displayed Sl of 16.45 and 60.39 respectively. Especially anti-HSV activities of natural complex products extracted by methanol No.6 was stronger than other ones.

A Study on the Anti-Oxidation Effects of Menthae Herba (I) (부하(溥荷)의 항산화효능(抗酸化效能)에 대한 연구 (I))

  • Objectives : In order to develop effective and safe anti-oxidation, the effects of Menthae Herba (M. spicata L. and M piperita L.) were examined. Methods : M. spicata L. and M. piperita L. of Menthae Herba were used to make water extracts and methanol extracts, and then its anti-oxidative effects were compared in vitro. Results : 1. M. spicata L. was stronger than M. piperita L. in anti-oxidative effects of methanol extracts, and water extracts showed similar anti-oxidation effects. 2. Water extracts of M. spicata L. was stronger than M. piperita L. showed a stronger superoxide anit-oxidation than the same-density ascorbic acid, so that water extracts were proved to be stronger than methanol extracts. 3. Methanol extracts' DPPH anti-oxidation was similar in M. spicata L. and M piperita L.. As for water extracts, M. spicata L. showed higher anti-oxidation and M. piperita L. had no anti-oxidation. 4. As for anti-oxidation of LDL and linoleic acid of methanol extracts, both M. spicata L. and M. piperita L. were stronger than a-tocopherol. As for the water extracts, these two showed similar level of anti-oxidative effect on linoleic acid, which is weaker that mehtanol extracts. 5. M. spicata L. includes more phenol than M. piperita L. Conclusions : Menthae Herba (M. piperita L. M. spicata L.) had anti-oxidation effect, so that it can be medically developed for anti-oxidation.

Neuroprotective Effect of Gagamchongmyung-tang on the Deficits of Learning and Memory by MCAO in the Rat (허혈유발 흰쥐에 있어서의 인지장애에 미치는 가감총명탕의 효과)

  • Objective : Gagamchongmyung-tang is clinically one of the most popular prescriptions as an herbal medicine for the treatment of amnesia. In order to evaluate its neuroprotective effects on the ischemia-induced cognitive deficits caused by middle cerebral artery occlusion (MCAO), we examined its ability to reduce impairments of learning and memory of rats in the Morris water maze. Method and Result : Focal cerebral ischemia produced a decrease in acetylcholine transmission in the hippocampus, and deficits of learning and memory in the Morris water maze task. Treatment with two types of Gagamchongmyung-tang, methanol and water extracts, produced a substantial increase in acquisition in the Morris water maze. Treatment with methanol extract of Gagamchongmyung-tang increased the performance of the retention test in the Morris water maze. Consistent with behavioral data, immunohistochemical data showed that treatment with methanol extract, but not water extract, of Gagamchongmyung-tang significantly recovered reduction of AchE and ChAT reactivity in the hippocampal CAl area. Conclusion : These results demonstrated that methanol extract of Gagamchongmyung-tang has protective effects against ischemia-induced learning and memory impairments, and provided evidence of methanol extract of Gagamchongmyung-tang as a putative treatment for amnesia, vascular dementia, and longer memory.

Influence of Roasting Time on Antibacterial and Antioxidative Effects of Coffee Extract (배전시간에 따른 커피 추출물의 항균 및 항산화 효과)

  • The influence of roasting time on antibacterial and antioxidative effects of methanol and water coffee extracts was investigated. Extract yield differed with roasting time. The maximum yield of methanol extract was 20.02% and 24.00% at respective roasting times of 12 and 20 min. The maximum yield of water extracts was 2.70% and 18.58% at 5 and 25 min roasting time, respectively. Antibacterial effects of each extract were determined by the classical minimal inhibitory concentration (MIC) paper disc diffusion method. Methanol extracts of different coffee samples inhibited growth of various strains except Escherichia coli. Extracts obtained following roasting times of 12, 14, 16, 20, and 25 min in particular displayed the most potent activity against Staphylococcus aureus. Among these extracts, that obtained from 12 min roasted coffee samples produced a MIC of $16.125{\mu}g$/mL against S. aureus. Water extracts applied at $1,000{\mu}g$/mL were growth inhibitory except against Salmonella choleraesuis and Prevotella intermedia. However, growth inhibition by water extracts was weak, with inhibitory zones of only 6-8 mm diameter produced. Determinations of free radical elimination for the different coffee extracts using 1,1-diphenyl-2-picrylhydrazyl were compared with ascorbic acid and butylated hydroxytoluene positive controls. Methanol and water extracts of different coffee samples ($100{\mu}g$/mL) showed $67.1{\sim}92.3%$ and $66.4{\sim}93.3%$ radical scavenging activity, respectively. However, longer roasting time (especially >20 min) tended to somewhat lower free radical elimination using both extracts. Total phenol in different coffee samples measured by the Folin-Denis method revealed the highest level of phenol contents with non-roasted coffee, whereas phenol content differed with different roasting time, ranging from $87.{\sim}126.5\;mg/g$ in methanol extracts. In water extracts, the phenol content was maximum at 8 min roasting time, whereas in other samples the content was varied from $95.0{\sim}199.1\;mg/g$.

  • Rate constants of solvolysis of 2-thiophenesulfonyl chloride were determined in aqueous binary mixtures with methanol, ethanol, acetone in water and in methanol. These data are interpreted using the equation of Grunwald-Winstein and Kivinen relationship. Also, kinetic solvent isotope effects in water and in methanol and product selectivities in alcohol-water mixtures were determined. Kinetic solvent isotope effect for hydrolysis of 2-thiopenesulfonyl chloride was 2.24 and 1.47 for methanol and water, respectively. Selectivity values for formation of ester relative to acid in ethanol-water mixtures show maximum S value. From kinetic solvent isotope effect in methanol and water, selectivity data in aqueous alcoholic solvents and solvent effects, it is proposed that the reaction channel favoured in low polarity solvents is general-base catalysis and/or is possibly addition elimination (S$_A$N) reaction pathway and in high polarity solvents iS S$_N$2 reaction mechanism.

Proton Conductivity and Methanol Permeability of Sulfonated Polysulfone/PPSQ Composite Polymer Electrolyte Membrane (설폰화된 폴리설폰/PPSQ 유-무기 복합 전해질막의 수소이온 전도도 및 메탄올 투과 특성)

  • Sulfonated polysulfone (SPSF) with poly(phenylmethyl silsesquioxane, PPSQ) composite polymer electrolyte membranes were prepared and their proton conductivity, water uptake and methanol permeability of membranes were characterized. By controlling the ratio of $(CH_3)_3SCI\;and\;CISO_3H$ and reaction time, SPSF with $37\~75\%$ degree of sulfonation were synthesized. The increase of sulfonate groups in the base polymer resulted in the increase of the water uptake in the membranes as well as methanol permeability. Composite membranes were prepared by casting of DMF solution of SPSF and PPSQ. The proton conductivity of the composite membrane at room temperature was $2.8\times10^{-3}\~4.9\times10^{-2}S/cm.$ The increase of PPSQ contents in composite membranes resulted in a decrease in water uptake and methanol permeability. Composite membranes containing $5\%$ PPSQ did not make a significant effect on the proton conductivity nO methanol permeability compared with that of pristine SPSF, but a significant decrease of water uptake was observed.

Antimicrobial and Antioxidative Activities of Solvent Fraction from Humulus japonicus (환삼덩굴의 용매분획별 항균성 및 항산화성)

  • The biological activities of Humulus japonicus were extracted by water and methanol. Methanol was better solvent than water in the extraction for antimicrobial activities against six different species of bacteria and two yeasts. The methanol extract was systematically fractionated with various organic solvents which have different polarities. From the result of antimicrobial activities against six species of bacteria and two species of yeasts, methanol extract was superior to water extract. The methanol extract of Humulus japonicus showed antimicrobial activity against the all species of microorganisms tested except Escherichia coli . The butanol fraction of methanol extract showed antimicrobial effect on the all species tested. The minimal inhibition concentration(MIC) of the butanol fraction on the growth of microorganisms was ranged between $0.1{\sim}0.4%$. The water extract of Humulus japonicus did not show inhibition of the activity of trypsin but methanol extract showed inhibitory activity. The chloroform fraction of methanol extract showed comparatively higher trypsin inhibitory activity than other fractions. The concentration of 50% inhibition$(IC_{50})$ by chloroform fraction was 1.0 mg/ml. Enzyme-inhibitor complex formation was above 90% of the while for 20 min. It was revealed that methanol extract of Humulus japonicus inhibited peroxide production of lard and soybean oil as substrate by antioxidative test. The chloroform fraction of methanol extract had the highest activity. When 0.2% of chloroform fraction was added, induction period of soybean oil and lard were extended 15, 9 days, respectively.

Antioxidant and tyrosinase inhibitory activity of white beech mushroom (Hypsizygus marmoreus) extracts (흰색 느티만가닥버섯 추출물의 항산화 활성 및 tyrosinase 저해 효과)

  • The objective of this study was to evaluate antioxidant effect and tyrosinase inhibitory activity of white beech mushroom (Hypsizygus marmoreus) extracts. The white beech mushroom was extracted into hot water and methanol. Total polyphenol content was highest in the hot water extract ($8.4{\pm}3.27mg\;GAE/g$) compared to the methanol extract ($7.3{\pm}2.85mg\;GAE/g$). The flavonoids contents in hot water and methanol extracts were $3.8{\pm}3.81ug/mg$ and $2.5{\pm}1.95ug/mg$, respectively. The tyrosinase inhibitory activity of extract was increased in a dose dependent manner and tyrosinase inhibitory activity of extract (hot water extract, 69.72%; methanol extract, 52.67% at 40 mg/ml) was lower than those of positive control 2% arbutin (96%). The DPPH radical scavenging activity of the hot water and methanol extract was 80% and 74%, respectively. Hot water extract ($63.34{\pm}1.00uM\;TE/g$) were more effective in ORAC (oxygen radical absorbance capacity) value than methanol extract ($46.33{\pm}0.48uM\;TE/g$). The toxicity of hot water and methanol extracts was investigated using WST-1 (4-[3-(4-iodophenyl)-2-(4-nitrophenyl)-2H-5-tetrazolio]-1,3-benzene disulphonate) assay on the B16BL6 melanoma cells.
