Cloning of the $Glu-tRNA^{Gln}$ Amidotransferase (gatCAB) Gene from Staphylococcus aureus

  • In Gram(+) bacteria and organelles in higher eukarotes, $Gln-tRNA^{Gln}$ utilized for protein biosynthesis is formed by a tRNA-dependent amino acid transformation using mischarged $Gln-tRNA^{Gln}$ as the intermediate. In this study, the gatCAB gene encoding $Gln-tRNA^{Gln}$ amidotransferase (Glu-AdT) of Staphylococcus aureus was cloned and its nucleotide sequence wa determined. The S. aureus gatCAB gene was organized in an operon structure consisting of three open reading frames (gatC, gatA, and gatB), similar to that of Bacillus subtilis. The gene sequences for the A and B subunits of$Gln-tRNA^{Gln}$ amidotransferase showed significant homology (77 and 87% homology with amino acid sequence) with the gatA and gatB genes of B. subtilis, yet the C subunit (gatC) showed a relatively lowe homology with the B. subtilis gatC gene and other orthologues. The cloned S. aureus <$Gln-tRNA^{Gln}$ amidotransferase gene was highly expressed in Escherichia coli, and the resulting crude enzyme could convert misacylated <$Gln-tRNA^{Gln}$ into $Gln-tRNA^{Gln}$ in vitro.

Development of smart transducer with embedded sensor for automatic process control of ultrasonic wire bonding

  • A ring-shaped lead zirconate titanate (PZT) piezoceramic sensor has been integrated with the Langevin-type piezoceramic driver of an ultrasonic wire-bonding transducer to form a smart transducer for in-situ measurement of three essential bonding parameters: namely, impact force, ultrasonic amplitude and bond time. This sensor has an inner diameter, an outer diameter and a thickness of 12.7 mm, 5.1 mm and 0.6 mm, respectively. It has a specifically designed electrode pattern on the two major surfaces perpendicular to its thickness along which polarization is induced. The process-test results have indicated that the sensor not only is sensitive to excessive impact forces exerted on the devices to be bonded but also can track changes in the ultrasonic amplitude proficiently during bonding. Good correlation between the sensor outputs and the bond quality has been established. This smart transducer has good potential to be used in automatic process-control systems for ultrasonic wire bonding.

Traditional Unani perspective of perceived insufficient milk (Qillatul Laban) and Galactogogues: A literary research with recent studies

  • The most important reason mentioned for the early discontinuation of breast feeding and introduction of supplementary bottles is Perceived Insufficient Milk (PIM), which is relatively common in women. This is of public health concern because the use of breast milk substitutes increases the risk of morbidity and mortality among infants in developing countries and shortens birth intervals. Thus, a literary search in classical text for aetiopathogenesis, symptoms and treatment of PIM were appraised to implement in contemporary era. The classical Unani texts viz., Al Qanon fit Tib (Canon of Medicine), Al Hawi (Continens Liber), Zakheera Kharzam Shahi, Tarjuma Kamilus Sana, Tibbe Akbar, Akseer Azam, and Kitabul Kulliyat were reviewed. Further certain galactogogue herbs which are in use since antiquity such as fenugreek, cotton seeds, cumin, asparagus, black cumin etc were explored in different search engines on website for proven galactogogue activity. The causes of PIM are abnormal temperament of body or breast, anaemia, anxiety, depression, malnutrition etc. The principle treatment is treating the cause viz., the temperament is corrected by diet and drugs in abnormal temperament, elimination of humour is required in dominance of humour etc. The aforementioned drugs are proven scientifically for their galactogogue activity. The classical texts are having valuable information regarding PIM, which can be implemented in present era. Aforementioned Unani drugs are proven scientifically for with their galactogogue effect, however, clinical trials are scarce. Therefore, further randomized controlled clinical trials are recommended.

  • Ginger (Zingiber officinale) belonging to the family Zingiberaceae is a perennial herb. It is widely distributed in tropical Asia. In India, it is cultivated mainly in Kerala, Andhia Pradesh, Uttar Pradesh, West Bengal and Maharashtra. It is one of the most common spices, which is in use since centuries for its versatile medicinal actions like antiemetic, stomachic, expectorant, anti-inflammatory, aphrodisiac etc in traditional system of medicine (Unani, Ayurveda, and Chinese medicine). It is useful for the treatment of various gastrointestinal, pulmonary, cardiovascular and sexual disorders. The phytochemical study of ginger showed the presence of many volatile oils and oleo-resins like gingerol, zinger one, zingiberol etc. Numerous experimental and clinical trials have proven ginger for its range of therapeutic activities such as antibacterial, antidiabetic, antiemetic, hypolipidaemic, hepatoprotective etc properties. The present article aims to explore traditional Unani and pharmacological activities of this herb reported till date.

Identification of Streptomyces sp. AMLK-335 Producing Antibiotic Substance Inhibitory to Vancomycin-Resistant Enterococci

  • The actinomycete strain AMLK-335 was antagonistic to vancomycin-resistant enterococci (VRE). Based on the diaminopimelic acid (DAP) type, and morphological and physiological characteristics revealed by scanning electron microscopy (SEM), AMLK-335 was confirmed to belong to the genus Streptomyces. Analysis of the 16S rDNA nucleotide sequences found AMLK-335 to have a relationship with Streptomyces platensis. The production of antibiotic from this strain was most favorable when cultured on glucose, polypeptone, yeast extract (PY) medium for 6 days at $27^{\circ}$. The antibiotic was identified as cyclo(L-phenylalanyl-L-prolyl) by comparing ti with the reported MS and NMR spectral data. Cyclo(phe-pro) from the PY cultures of AMLK-335 was most effective (K-98-258). Futhermore, cyclo(phe-pro) had antimicrobial activity against Bacillus subtilis, Microcuccs luteus, Staphylococcus aureus, and Saccharomyces cerevisiae, but it wa ineffective against Candida albicans, Streptomyces murinus, and Aspergillus niger.

Measurement of Cell Death Constant in Anabaena flos-aquae (Cyanophyceae) by the Molecular Probe (Anabaena flos-aquae 에서의 세포사멸계수(Cell Death Constant)의 측정)

  • The measurement of cell death constant in Anabaena flos-aquae was tested by the Live/Dead BacLight Viability kit(Molecular Probes Co., Seatle, WA). When the Live/Dead BacLight Viability kit was applied to Anabaena flos-aquae, the cells with intact cell membranes(live cells) stained fluorescent green, while the cell with damaged membranes(dead cells) stained fluorescent red and the background remained virtually nonfluorescent. The rations of live : dead cells in the cell suspension were controlled artifically and Live/Dead BacLight Viability kit was applied to them. The ratios of green:red fluorescent cells in the cell suspension were the same as those of live : dead cells controlled artifically. It was also approved by the fluorescence emission. The cell death constant was measured in the P-limited Anabaena flos-aquae chemostal culture in the N-fixing and $KNO_3-supplied$ conditions. The culture in N-fixing chemostat had a dead cell proportion of 1.2% at the growth rate of 0.7/day and increased to 2.6% at the growth rate of 0.3/day. The cell death constant of N-fixing culture was 0.008/day.There was a same trend in the $KNO_3-supplied$ chemostat culture. The proportion of dead cell was 1.6% of dead cell proportion at the growth rate of 0.7/day and increased to 4.3% at the growth rate of 0.3/day.

Relationships between Invertebrate Availability and the Abundance of Three Species of Shrews and the Shrew-mole in Managed Forests (산림 내에서 곤충의 현존도와 식충목 4 종의 풍부도간의 상관 관계)

  • The abundance of coarse woody debris (CWD) has been hypothesized to increase niche for forest dwelling insectivores, concentration of nutrients, and the diversity of invertebrates. However, no abjective assessment based on replication in the field has yet been done. this study was carried out to test the relationships between invertebrate availability and the abundance of four insectivores (Sorex trowbridgii, S. vagrans, S. monticolus and Neurotrichus gibbsii) in western Washington, USA. Pitfall traps were used to determine abundance and diversity of invertebrates. Abundance of insectivores was not different between habitats except for S. vagrans which was more abundant in habitats with low amounts of CWD than in habitats with high amounts of CWD. Simpson’s diversity index computed for invertebrates did not differ between habitats. There were no significant relationships between the diversity indices of invertebrates and the abundance of shrews. Shrew abundance and the Coleoperan beetles also did not show any significant relationship. The results suggested insectivores might select a wide range of prey items in addition to surface-active invertebrates.

  • The movement of in-situ walls has become very important as construction in large cities moves upward, instead of outward. Tall structures typically have deep excavations not on1y to provide extra space for parking, but also to reduce the potential settlement of the building. These large excavations require a robust bracing system to resist the lateral earth pressures as the depth increases. Methods to predict deflections of the retaining systems are of utmost importance because wall movements allow potentia1 settlement of adjacent structures. Case studies will be analyzed and measured waI1 def1ections will be compared with predictions from empirica1ly derived charts.

  • In this paper, we present a polarizer that consists of three step rotary detents which can selectively convert linear polarzation into circular polarization and vice versa. For the design of the polarizer, the transmission line theory is applied to design the wa- veguide mode transducer for the modes to be smothly converted in waveguides, and a dielectric plate is inserted in circular waveguide for the conversion of a polarized wave with the angle of an inserted dielectric plate. Also, we simulated to obtain the optimum values of the transmission and the reflection coefficient characteristics at input and output port, and proved the propriety of the theory from the knowledge of measuring the constructed polarizer with the designed data.

Localization of Acoustic Sources on Wind Turbine by Using Beam-forming Techniques (빔-형성 기법을 이용한 풍력 터빈 음원의 국부화)

  • The previous work (Cheong et al., 2006) where the characteristics of acoustic emissions of wind turbines has been investigated according to the methods of power regulation, has showed that the acoustic power of wind turbine using the stall control for power regulation is more correlated with the wind speed than that using the pitch control. In this paper, basically extending this work, the noise generation characteristics of large modern upwind wind turbines are experimentally indentified according to the power regulation methods. To investigate the noise generation mechanisms, the distribution of noise sources in the rotor plane is measured by using the Beam-forming measurement system (B&K 7768, 7752, WA0890) consisting of 48 microphones. The array results for the 660 kW wind turbine show that all noise is produced during the downward movement of the blades. This result show good agreement with the theoretical result using the empirical formula with the parameters: the convective amplification; trailing edge noise directivity; flow-speed dependence. This agreement implies that the trailing edge noise is dominant over the whole frequency range of the noise from the 660 kW wind turbine using the pitch control for power regulation.
