실시간 정량 PCR을 통한 김치 유래 Weissella spp.에 의한 Listeria monocytogenes 생육 억제 (Growth Inhibition of Listeria monocytogenes by Weissella spp. from Kimchi Through Real-time PCR)

  • 본 연구에서는 김치로부터 W. cibaria 0D17와 W. confusa 0D23를 생화학적 특성 분석과 16s rRNA 염기서열 분석을 통해 분리, 동정하였으며, W. cibaria 0D17와 W. confusa 0D23 배양 상등액이 L. monocytogenes에 대한 항균 효과가 있는 것으로 나타났다. 또한, 실시간 정량 PCR을 통해 W. cibaria 0D17 및 W. confusa 0D23가 L. monocytogenes를 공동 배양했을 때의 생육억제 효과를 분석한 결과, $37^{\circ}C$에서는 생육 억제 효과가 있는 것으로 확인되었다. 따라서, 김치 유래 Weissella spp.가 갖는 L. monocytogenes에 대한 항균 활성에 대한 기초 자료로 활용가능 할 것으로 판단되며, W. cibaria 0D17 및 W. confusa 0D23가 생산하는 bacteriocin 등의 항균 물질 특성에 대한 연구가 추가적으로 진행될 필요가 있을 것이다.

김치에서 분리된 젖산균의 β-glucosidase 활성 탐색 (Exploration of β-Glucosidase Activity of Lactic Acid Bacteria Isolated from Kimchi)

  • ${\beta}$-Glucosidase 효소활성이 높은 균주를 선발하기 위하여 다양한 김치에서 분리된 젖산균의 ${\beta}$-glucosidase 활성을 탐색하였다. 김치에서 분리된 156개의 젖산균 중 134개의 균주만이 cellobiose를 탄소원으로 대사하였으며, 세포내 ${\beta}$-glucosidase 활성이 세포외 활성보다 현저히 높았다. 배추김치에서 분리된 W. cibaria KFRI88010 균주가 3.7${\pm}$0.5 unit/mg protein으로서 가장 높은 세포내 ${\beta}$-glucosidase 효소활성을 나타내었으며, 효소활성은 pH 5, ${37^{\circ}C}$ 반응조건에서 가장 높게 나타났다. $Mn^{2+}$를 비롯한 금속이온은 효소활성을 크게 저해하였다. W. cibaria KFRI88010 균주를 배양할 때 사용한 탄소원 중, fructose는 cellobiose나 glucose와 비교하여 약 2.5배 이상의 높은 세포내 ${\beta}$-glucosidase 효소활성을 나타내었다.

RecN 유전자 특이적 PCR을 이용한 Weissella 속 유산균의 검출법 개발 및 적용 (Development and Application of PCR-Based Weissella Species Detection Method with recN Gene Targeted Species-Specific Primers)

  • Weissella 속 유산균 검출의 차이를 이용한 한국산 및 중국산 김치 판별의 가능성 검토를 위하여 Weissella 속 9종 균주의 PCR 검출법을 개발하였다. 종(species) 수준에서의 Weissella 속 균주의 특이적 PCR 검출을 위한 primer는 recN 유전자의 염기서열을 이용하여 선정하였으며, 김치로부터 W. cibaria, W. confusa, W. koreensis, W. soli를 모두 검출하기 위해서는 20 ng template DNA가 필요한 것으로 나타났다. 한국산 김치시료로부터는 W. cibaria, W. confusa, W. koreensis가 높은 빈도로 검출되었지만, W. soli는 검출되지 않았다. 한편 중국산 김치시료로부터는 이들 4종의 Weissella 속 균주들이 모두 검출되었다. 본 연구자들이 개발한 W. soli 특이적 PCR 검출은 현시점에서 중국산 김치의 원산지 판별법으로 적용되기에는 한계점을 가지고 있지만, 미생물 군집의 차이를 이용한 새로운 과학적 검증법이 제시되어 그 가능성이 검토되었다는 점에서 의의를 가지고 있다.

Lipoteichoic Acid Isolated from Weissella cibaria Increases Cytokine Production in Human Monocyte-Like THP-1 Cells and Mouse Splenocytes

  • Lactic acid bacteria (LAB) have beneficial effects on intestinal health and skin diseases. Lipoteichoic acid (LTA), a cell wall component of gram-positive bacteria, is known to induce the production of several cytokines such as TNF-α, IL-1β, and IL-8 and affect the intestinal microflora, anti-aging, sepsis, and cholesterol level. In this study, Weissella cibaria was isolated from Indian dairy products, and we examined its immune-enhancing effects. Live and heat-killed W. cibaria did not induce the secretion of immune-related cytokines, whereas LTA isolated from W. cibaria (cLTA) significantly increased the secretion of TNF-α, IL-1β, and IL-6 in a dose-dependent manner. cLTA increased the phosphorylation of nuclear factor kappa-light-chain-enhancer of activated B cells, p38 mitogen-activated protein kinases, and c-Jun N-terminal kinases in THP-1 cells. The secretion of TNF-α and IL-6 was also increased in the cLTA-treated mouse splenocytes. These results suggest that cLTA, but not W. cibaria whole cells, has immune-boosting potential and can be used to treat immunosuppression diseases.

Weissella cibaria CMU 경구투여가 비글의 구취 저하에 미치는 효과 (Oral malodor-reducing effects by oral feeding of Weissella cibaria CMU in Beagle dogs)

  • This study assessed the effects of Weissella cibaria (W. cibaria) CMU on oral health in male and female beagles (n = 18) by measuring oral malodor and periodontal disease-related parameters (calculus, plaque, and gingivitis indices). Oral malodor and indicators of periodontal disease were assessed in five treatment groups: negative control (scaling and 0.24 mg of maltodextrin, n = 3), positive control (0.24 mg of maltodextrin, n = 3), and W. cibaria CMU groups (each n = 4) at low (CMU-L, $2{\times}10^7$ colony forming unit [CFU]), medium (CMU-M, $2{\times}10^8CFU$), and high (CMU-H, $2{\times}10^9CFU$) concentrations. After feeding with W. cibaria CMU for 6 weeks, total volatile sulfur compound concentrations in the CMU-L ($2.0{\pm}1.04ng/10mL$), CMU-M ($2.4{\pm}1.05ng/10mL$), and CMU-H ($2.6{\pm}1.33ng/10mL$) groups were significantly lower than in the positive control group ($3.2{\pm}1.65ng/10mL$). Also, CMU-L ($1.4{\pm}0.83ng/10mL$) and CMU-H ($1.9{\pm}1.14ng/10mL$) groups had methyl mercaptan levels lower than that in the positive control group ($2.4{\pm}1.21ng/10mL$) at week 2. The plaque index was significantly lower in the CMU-H group ($4.5{\pm}0.28$) than in the positive control group ($5.9{\pm}1.08$) at week 6. W. cibaria CMU could be useful as a novel oral hygiene probiotics for reducing volatile sulfur compounds production and inhibiting plaque growth in companion animals.

유세포분석기, 형광현미경, 용균반검사 분석을 이용한 Weissella cibaria 박테리오파지 정량분석 및 상관관계분석 (Enumeration of Weissella cibaria phage with cytometry, epifluorescence microscopy, and plaque assay)

  • 유세포분석기, 형광현미경 계수법과 현재 주로 활용하는 숙주감염에 의한 용균반검사법에 의한 박테리오파지를 계수하는 방법을 Weissella cibaria 박테리오파지로 비교 연구하였다. 상관관계 분석은 피어슨 상관 통계분석, 회귀분석, 차이검정등으로 하였다. 12개의 시료로 유세포분석기와 용균반검사에 의한 계수법을 비교한 결과는 피어슨 분석으로는 96.7%의 상관관계(상관값 r=0.967, $R^2=0.9352$)와 차이검정은 1.063을 보였다. 형광현미경과 용균반검사에 의한 계수법 비교결과는 피어슨 분석으로는 99.0% (상관값 r=0.990, $R^2=0.9811$) 상관관계와 차이검정은 1.605이였다. 그러므로 유세포분석기와 형광현미경 계수법은 용균반검사 계수법과 같이 W. cibaria 박테리오파지 계수에 적합할 것으로 사료된다.

Effect of Bacteriophages on Viability and Growth of Co-cultivated Weissella and Leuconostoc in Kimchi Fermentation

  • This study aimed to understand the survival and growth patterns of bacteriophage-sensitive Weissella and Leuconostoc strains involved in kimchi fermentation. Dongchimi kimchi was prepared, and Weissella and Leuconostoc were co-cultivated in the dongchimi broth. Weissella cibaria KCTC 3807 growth was accompanied by rapid lysis with an increase in the bacteriophage quantity. Leuconostoc citreum KCCM 12030 followed the same pattern. The bacteriophage-insensitive strains W. cibaria KCTC 3499 and Leuconostoc mesenteroides KCCM 11325 survived longer under low pH as their growth was not accompanied by bacteriophages. The bacteriophage lysate of W. cibaria KCTC 3807 accelerated and promoted the growth of Leuconostoc. Overall, our results show that bacteriophages might affect the viability and population dynamics of lactic acid bacteria during kimchi fermentation.

프로바이오틱스 섭취가 장기요양시설 노인의 구강환경 변화에 미치는 효과 (Effect of probiotics intake on oral environment changes of the elderly in long-term care facilities)

  • Objectives: This study was aimed at evaluating the effects of changes in the oral environment of the elderly in long-term care facilities after the intake of Weissella cibaria. Methods: The test group ingested the W. cibaria strain Chonnam Medical University (CMU), and the control group ingested the control food. Of all randomized trial subjects, 62 were analyzed (32 in the experimental group and 30 in the control group). In this 8-week demographic study, we evaluated self-perceived halitosis, changes in halitosis, sensory test results, tongue plaque index scores, saliva buffering capacity, and the salivary flow rate. Results: The W. cibaria CMU intake in the elderly in long-term care facilities during the experimental period did not demonstrate statistically significant changes in the salivary flow rate. However, self-perceived halitosis, organoleptic test results, tongue plaque index scores, and salivary buffering capacity demonstrated statistically significant differences between the experimental and control groups. These findings partially confirmed the beneficial effects of the W. cibaria CMU on the oral environment in the elderly. Conclusions: Research results on the role of probiotics in the oral cavity should be summarized, and utilization plans should be sought to obtain a clearer understanding of the clinical efficacy and related factors. The value of probiotic use may be high in improving the oral health of people by enabling treatment and prevention.

Isolation of Weissella strains as potent probiotics to improve antioxidant activity of salted squid by fermentation

  • The aim of this study was to enhance the antioxidant activity of salted squid by inoculation of two Weissella spp. strains (W. cibaria FB-069 and W. viridescens FB-077) isolated from traditional Korea salted squid. The safety and probiotic potential characteristics of these two strains were evaluated. The safety of these strains was analyzed based on hemolytic activity, mucin degradation, biogenic amino production, and resistance to antibiotics. These lactic acid bacteria showed probiotic potential, including resistance to gastrointestinal tract conditions, adhesion to Caco-2 cells, and aggregation. The low-salted squid fermented with Weissella strains had consistently higher antioxidant activity through changing their amino acid profiles. Therefore, W. cibaria FB-069 and W. viridescens FB-077 might be good candidates for fermentation of salted squid to develop functional food with enhanced health benefits.

Genomic analysis of WCP30 Phage of Weissella cibaria for Dairy Fermented Foods

  • In this study, we report the morphogenetic analysis and genome sequence of a new WCP30 phage of Weissella cibaria, isolated from a fermented food. Based on its morphology, as observed by transmission electron microscopy, WCP30 phage belongs to the family Siphoviridae. Genomic analysis of WCP30 phage showed that it had a 33,697-bp double-stranded DNA genome with 41.2% G+C content. Bioinformatics analysis of the genome revealed 35 open reading frames. A BLASTN search showed that WCP30 phage had low sequence similarity compared to other phages infecting lactic acid bacteria. This is the first report of the morphological features and complete genome sequence of WCP30 phage, which may be useful for controlling the fermentation of dairy foods.
