  • This study was carried out to obtain basic data on mycelial growth characteristics for an artificial cultivation of Volvariella volvacea. Twelve URP primers were used to assess genetic relationship of V. volvacea and its result was divided into two groups. But there was nothing different in morphological characteristics in V. volvacea. Among five kinds of mushroom media, MCM medium was selected as the favorable culture medium. The optimal range of temperature and pH for mycelial growth on V. volvacea were 35oC and pH 6~8, respectively. Carbon sources had not an effect on 10 strains of V. volvacea, and nitrogen source for the optimum mycelial growth was yeast extract. Also, we selected GMVV 79004 as a superior strain of 10 strains V. volvacea based on the mycelial growth and yield.

  • The edible paddy straw mushroom Volvariella volvacea (Bull. ex F.) Singer has high nutritional and medicinal values. They are grown on a wide variety of agricultural by-products using different several methods. The result of this present study showed that type of bed and cultivation condition (outdoor/indoor) had effects on the yield of paddy straw mushrooms grown on the spent oyster mushroom sawdust. The treatment of circular compact bed under indoor cultivation condition (CYIC) had the best yield and biological efficiency (B.E.) with 2,119.2 g/bed and 14.5%, respectively, followed by the treatment of conventional bed under outdoor cultivation condition (COOC) with the yield and B.E. of 1,935.5 g/bed and 13.2%, respectively, but the lowest yield and B.E. were observed in the treatment of conventional bed under indoor cultivation condition (COIC) with 1,226.1 g/bed and 8.4%, respectively. Paddy straw mushrooms cultivated on spent oyster mushroom sawdust should be proper in both outdoor and indoor conditions by conventional and circular compact methods as well. However, in indoor conditions, paddy straw mushrooms should be grown in a circular compact bed to ensure the temperature of the bed is suitable for the mushroom growth.

  • This experiment was undertaken to investigate proper conditions for protoplast isolation from strains of Agaricus bisporus, Flammulina velutipes, Lentinus edodes, Pleurotus florida, Pleurotus ostreatus, Pleurotus sajor-caju and Volvariella volvacea. Combination of ${\beta}-D-glucanase$, novozym 234 and snail enzyme with 0.6M $MgSO_4$ in 0.05M Na-maleate buffer, pH 5.8 was the most effective for isolation of protoplasts. High yields of protoplasts were obtained from $4{\sim}5$ days old mycelia of P. florida and P. ostreatus on mushroom complete agar medium. Protoplasts were also obtained in good yield from other fungi but A. bisporus and V. volvacea. In the latter two cases protoplast release was observed; however, the yield was much lower than those of the other fungi.

  • Diversities of Korean species belonging to the genus Volvariella have been studied through forty five dried specimens collected throughout the Korean peninsula for 28 years, from 1972 to 2000, and preserved in NIAST's herbarium. We have found one unrecorded species of the genus Volvariella in Korea: V. villosavolva(Lloyd) Sing, Stirp Taylori Korean common names were designated. Ten species of the genus Volvariella including the recorded Korean species of Volvariella have been identified and the keys to the species were constructed.

  • This study was carried out investigate optimal additive content of spent medium from oyster mushroom in Volvariella volvacea cultivation. The formation of spent mushroom medium from oyster mushroom consisted of 50% pine sawdust, 30% beet pulp and 20% cotton seed hull pellet. The higher amount of spent medium from oyster mushroom resulted in higher total nitrogen content and C/N ratio of medium decreased according to increasing mixing ratio of spent medium from oyster mushroom. The fruit body yield and biological efficiency of supplemented with 50% spent medium from oyster mushroom was same level of non additive medium.

  • Present experiments were conducted to determine the possibility of cultivation of tropical and subtropical saprophytic fungus, Volvariella volvacea (Bull. ex Fr.) Sing. under Korean climatic conditions and the-possibility of applying the method of indoor cultivation which was modified from the cultivation method of the common mushroom, Agaricus bisporus (Lange) Sing.. The results obtained were as follows: 1. In conventional outdoor cultivation, the adequate cultivation period of this fungus was from around the end of June to the end of August. 2. When rice straws submerged in water for 5 days were peak-heated for 6 days in a mush room house, the yield of the fungus was higher than that obtained from the rice straws peak-heated immediately after outdoor composting. 3. The highest yield, 20.49kg per 100kg rice straws, was obtained from the water-soaked and peak-heated rice straws which were cased with a 2 cm layer of loam at 7 days after spawning.

  • The hot-water extracts of four strains of Volvariella volvacea [Vv (KMCC04386), Vv-Chi (KMCC04382), V9-21 (KMCC04380), and VG-19 (KMCC05115)] were prepared to determine their antioxidant activities, -glucan content, and nutritional content. Among the four V. volvacea strains, Vv strain showed the highest DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity (23.7%). The highest total polyphenol and total flavonoid contents (8.17 mg y/g and 3.46 mg QE/g, respectively) were observed in the Vv-Chi strain. The ferric reducing antioxidant power (FRAP) and reducing power were significantly higher in the Vv-Chi strain compared to those in the other V. volvacea strains (p<0.05). There were no significant differences in the nitrite scavenging activity among the four different strains (p<0.05). The -glucan content in the four V. volvacea strains ranged from 15.13-16.07%, and the VG-19 strain had the highest -glucan content (15.73%). The VG-19 strain also had the highest total amino acid (986.8 mg/kg) and essential amino acid (369.3 mg/kg) contents among the four V. volvacea strains. The results of this study showed that the Vv-Chi strain exhibited the highest antioxidant activity, while the -glucan and nutritional contents were higher in the VG-19 strain compared to those in the other strains of V. volvacea.
