가정용 냉장고의 제상 주기와 온도 변화가 저장 식품의 품질에 미치는 영향 (Influence of Thermal Oscillation on Quality of Frozen Foods Stored in Domestic Refrigerator)

  • 가정용 냉장고의 제상 횟수와 제상에 따른 온도 상승이 냉동식품의 품질에 미치는 영향을 조사하기 위하여, 육류, 생선, 과일 및 아이스크림을 냉동실에 저장하면서 품질변화를 측정하였다. 냉동실의 제상 주기가 16시간 정도이고, 제상시 온도는 $-5^{\circ}C$로 상승하는 기존의 일반 냉장고와, 제상주기가 30시간 정도이고 제상시 온도는 $-15^{\circ}C$로 조절되어 제조된 냉동실에서의 품질변화를 비교하였다. 냉동저장 중에 일어난 육류와 생선의 단백질 변성 정도와 육즙의 생성율 및 육류의 색깔 변화 정도는, 제상 주기가 짧고 온도 상승폭이 보다 큰 기존의 냉동실에서 변화가 훨씬 심한 것으로 나타났다. 냉동저장 중 얼음의 재결정화로 인한 조직의 파괴 정도와 얼음 결정의 크기를 광학현미경과 전자현미경으로 각각 관찰한 결과, 온도 변동이 심할 경우 육류의 조직이 더욱 많이 파괴되었고, 아이스크림에서는 얼음 결정이 훨씬 커진 것을 알 수 있었다. 시간-온도 지시계에 의한 냉동실의 저장 효과를 비교한 결과 위에 서 측정한 품질변화 속도와 잘 일치하였다. 따라서 냉동실의 제상 주기를 길게 하고 제상시 온도 상승폭을 줄인다면 가정용 냉장고에서 유발되는 냉동식품의 2차적인 품질 저하를 크게 감소시킬 수 있음을 알았다.

대전광역시 대전천과 갑천의 물과 주변 토양들로부터 분리한 국내 미기록 야생효모들의 특성 (Characterization of the Unrecorded Wild Yeasts from the Water and Riverside Soils of Daejeoncheon and Gapcheon in Daejeon Metropolitan City, Korea)

  • 대전광역시의 대전천과 갑천의 물과 주변토양들로부터 분리한 야생효모 중 대전천에서 분리한 Debaryomyces udenii JSF601, Kazachstania telluris JSF602와 Tricosporon faecale JSF614, Candida infanticola WJSL0039, Candida palmioleophila WJSL0048, Pichia spartinae WJSL0087, Trichosporon coprophilum WJSL0093 등 7균주들과 갑천에서 분리한 Leucosporidium golubevii WJSL0108과 Ustilentyloma graminis WJSL0118의 야생효모들이 국내 미기록 효모 균주들로 최종 선별 하여 이들의 형태학적, 배양학적 특성 등을 조사하였다. 대전천에서 분리, 선별한 국내 미기록 균주들은 구형~타원형으로 Pichia spartinae WJSL0087, Kazachstania telluris JSF602, Tricosporon faecale JSF614와 Trichosporon coprophilum WJSL0093들은 자낭포자와 의균사를 형성하였다. Candida infanticola WJSL0039는 10% NaCl을 함유한 YPD 배지에서 생육하는 호염성 효모이었고 Pichia spartinae WJSL0087는 $37^{\circ}C$에서 생육하는 고온성 효모이었다. 갑천에서 분리, 선별한 국내 미기록 효모들은 모두 구형으로 자낭포자를 형성하였다. Ustilentyloma graminis WJSL0118 만이 의균사를 형성하였고 비타민을 첨가하지 않은 YPD 배지에서도 생육하였으며 $37^{\circ}C$에서 생육하는 고온성 효모이었다.

김치액의 색상에 의한 배추 김치의 품질 평가 (Quality Evaluation of Korean Cabbage Kimchi by Instrumentally Measured Color Values of Kimchi Juice)

  • 김치의 숙성도를 평가하기 위한 한 방법으로 김치의 숙성과정 중 자연적으로 유출되는 김치액의 색상 변화를 색차계를 이용해서 측정하고, 또한 이들 결과를 성분분석과 관능적인 평가와 비교 조사하였다. 성분분석과 관능검사 결과는, 16$^{\circ}C$에서 숙성 3일째 김치의 숙성 적기 pH인 4.3에 달하였으며 이때 비타민 C 및 carotenoids 함량도 거의 최고치에 달하고 신맛과 조직감도 적당하였다. 숙성온도 4$^{\circ}C$에서는 숙성 6일후에도 미숙단계에 있었다. 숙성온도 16$^{\circ}C$에서 김치액의 $L^{*}$ 값은 숙성기간 중 뚜렷한 변화를 나타내지 않았다. 반면 $a^{*}$ 값은 숙성 4일까지는 상승하다가 이후 급속히 감소하였으며 $b^{*}$ 값은 숙성 3일까지는 증가하다가 그후 거의 변화가 없었다. 김치액량, pH 및 산도는 김치액의 $L^{*}$ 혹은 $b^{*}$ 값과, carotenoids 함량은 다만 $a^{*}$ 값과 높은 상관관계를 나타냈다. 김치액의 색상으로 김치의 숙성 정도를 평가하기 위해서는 $a^{*}$ 혹은 $b^{*}$ 간을 지표로 삼는 것이 바람직하였다. 즉, $a^{*}$ 값이 담금 초기와 비교해서 동등 혹은 낮은 값을 나타내거나 $b^{*}$ 값이 거의 변화가 없을 때는 과숙기에 이르렀다고 할 수 있었다. 김치의 적숙기를 예측하는 한 방법으로 $a^{*}$$b^{*}$ 의 비( $a^{*}$ / $b^{*}$ )를 이용할 수 있었다. 즉, $a^{*}$ / $b^{*}$ 의 비값이 1이상 흑은 1에 가까울 때는 미숙기, 0.8 부근 일때는 적숙기, 0.8보다 작아 질수록 적숙기에서 과숙기로 점차 옮아감을 알 수 있었다.

Porphyromonas gingivalis가 일부 구강미생물의 형광 발현에 미치는 영향 (Red fluorescence of oral bacteria interacting with Porphyromonas gingivalis)

  • Objectives: Dental plaque is composed of 700 bacterial species. It is known that some oral microorganisms produce porphyrin, and thus, they emit red fluorescence when illuminated with blue light at a specific wavelength of <410 nm. Porphyromonas gingivalis belongs to the genus Porphyromonas, which is characterized by the production of porphyrin. The aim of this study was to evaluate red fluorescence emission of some oral microorganisms interacting with P. gingivalis. Methods: Five bacterial strains (P. gingivalis, Streptococcus mutans, Lactobacillus casei, Actinomyces naeslundii, and Fusobacterium nucleatum) were used for this study. Tryptic soy agar medium supplemented with hemin, vitamin K3, and sheep blood was used as a growth medium. The fluorescence emission of bacterial colonies was evaluated under 405 nm-wavelength blue light using a Quantitative Light-induced Fluorescence Digital (QLF-D) camera system. Each bacterium was cultured alone and co-cultured in close proximity with P. gingivalis. The red/green (R/G) ratio of fluorescence image was calculated and the differences of R/G ratio according to each growth condition were compared using the Mann-Whitney test (P<0.05). Results: Single cultured S. mutans, L. casei and A. naeslundii colonies emitted red fluorescence (R/G ratio=$2.15{\pm}0.06$, $4.31{\pm}0.17$, $5.52{\pm}1.29$, respectively). Fusobacterium nucleatum colonies emitted green fluorescence (R/G ratio=$1.36{\pm}0.06$). The R/G ratios of A. naeslundii and F. nucleatum were increased when P. gingivalis was co-cultured with each bacterium (P<0.05). In contrast, the R/G ratios of S. mutans and L. casei were decreased when P. gingivalis was co-cultured with each bacterium (P=0.002, 0.003). Conclusions: This study confirmed that P. gingivalis could affect the red fluorescence of other oral bacteria under 405 nm-wavelength blue light. Our findings concluded that P. gingivalis has an important role for red fluorescence emission of dental biofilm.

Korean Guideline for the Prevention and Treatment of Glucocorticoid-induced Osteoporosis

  • Background: To develop guidelines and recommendations to prevent and treat glucocorticoid (GC)-induced osteoporosis (GIOP) in Korea. Methods: The Korean Society for Bone and Mineral Research and the Korean College of Rheumatology have developed this guideline based on Guidance for the Development of Clinical Practice Guidelines ver. 1.0 established by the National Evidence-Based Healthcare Collaborating Agency. This guideline was developed by adapting previously published guidelines, and a systematic review and quality assessment were performed. Results: This guideline applies to adults aged ${\geq}19years$ who are using or plan to use GCs. It does not include children and adolescents. An initial assessment of fracture risk should be performed within 6 months of initial GC use. Fracture risk should be estimated using the fracture-risk assessment tool (FRAX) after adjustments for GC dose, history of osteoporotic fractures, and bone mineral density (BMD) results. All patients administered with prednisolone or an equivalent medication at a dose ${\geq}2.5mg/day$ for ${\geq}3months$ are recommended to use adequate calcium and vitamin D during treatment. Patients showing a moderate-to-high fracture risk should be treated with additional medication for osteoporosis. All patients continuing GC therapy should undergo annual BMD testing, vertebral X-ray, and fracture risk assessment using FRAX. When treatment failure is suspected, switching to another drug should be considered. Conclusions: This guideline is intended to guide clinicians in the prevention and treatment of GIOP.

저병원성 조류인플루엔자 감염에 따른 육계의 면역인자 변화 (Changes in Immunological Factors Induced by H9N2 Avian Influenza Challenge in Broilers)

  • 가금산업에서 전염성 질병은 농장의 생산성에 악영향을 미치는 중요한 요인이다. 그 중 저병원성 조류인플루엔자는 산란율과 증체율에 영향을 준다. 본 연구에서는 육계에서 저병원성 조류인플루엔자 감염시 혈청 단백질 및 사이토카인의 변화 양상을 조사하였다. 1일령의 육계를 1주령부터 4주령까지 각 주령마다 저병원성 조류인플루엔자를 공격 접종 후 혈중 chDBP, Chicken Ovotransferrin, chIL-1β, chIL-6, chIFN-γ의 양을 비교하였다. 어린 주령에 저병원성 조류인플루엔자를 공격접종한 그룹에서는 4주령에 음성대조군과 비교해 chIL-1β, chIL-6가 유의성 있는 낮은 값을 보이는 것을 확인하였다. 추가적인 연구를 통하여 chIL-1β, chIL-6가 질병의 경과를 확인할 수 있는 보조적인 감염 지표로 활용될 수 있을 것으로 판단된다.

유제품의 제2형 당뇨병 예방 효과: 총설 (Preventive Effect of Milk Products against Diabetes Mellitus Type 2: A Review)

  • 최근에 유제품의 섭취가 제2형 당뇨병의 위험성을 낮춘다는 연구결과가 점점 증가하고 있는 추세이다. 지금까지 발표된 유제품 섭취와 제2형 당뇨병 위험성의 관계에 대한 증거들을 검토해 보면, 유제품의 섭취와 제2형 당뇨병의 위험성 사이, 그리고 유제품의 섭취량에 따라 제2형 당뇨병의 발병률에 다양한 영향을 미치는 것으로 밝혀졌다. 특히 저지방 유제품의 섭취와 제2형 당뇨병의 위험성 감소는 일정한 관계를 보였으며, 일반적인 지방을 함유한 유제품은 인체에 좋은 영향을 주는 것으로 밝혀졌다. 하지만 몇몇 특정 유제품이 인체에 미치는 역할을 명확하게 확인할 향후 연구가 요구된다. 유제품이 제2형 당뇨병의 위험성을 낮추는 기전은 아직까지 명확하지 않지만, 유제품이 비만과 신진대사장애에 작용하는 기전과 유사할 것으로 사료되고 있다. 시간이 지날수록 유제품의 섭취가 제2형 당뇨병의 위험성을 낮춘다는 연구 결과와 일관성 있는 결과들이 지속적으로 제공되고 있다. 일반적인 지방과 특정 유제품을 더 잘 이해하기 위해서는 더 많은 연구가 진행되어야 하며, 지금까지의 연구 결과를 뒷받침하기 위해서는 잘 설계된 무작위통제실험과 기계적 연구를 통해 임상실험으로 반드시 검증하여야 하며, 이러한 노력들이 임상실습 및 공중보건지침으로 이행될 수 있도록 많은 노력이 절실히 요구 되어지고 있다.

효소고정화 방법에 따른 콜레스테롤 검출용 바이오센서의 전류 감응도 분석 (Analysis of Amperometric Response to Cholesterol according to Enzyme-Immobilization Methods)

  • 콜레스테롤의 신속하고 정확한 새로운 분석방법을 모색하기 위하여 본 연구에서는 전기적 전도성이 우수한 MWCNT를 이용하여 전극을 제작하였고, 여러 가지 효소고정화 방법을 통해 전기화학적 감응도 분석을 실시하였다. MWCNT의 전도성을 향상시키기 위해 아민기를 도입한 MWCNT-$NH_2$를 제조하였고, MWCNT-$NH_2$/GCE에 PB를 점착하여 작업전극을 제조하였다. 제조한 작업전극은 0.5~500 ${\mu}M$ $H_2O_2$ 농도 범위에서 농도가 증가함에 따라 전류가 비례적으로 증가하였고, 검출한계는 0.1 ${\mu}M$로 나타나 전극이 높은 감도를 가지고 있음을 확인하였다. 또한 콜레스테롤 검출을 위해 적합한 효소 반응기를 제작하기 위해 담체인 aminopropyl glass beads, CNBr-activated sepharose, Na-alginate, toyopearl beads에 cholesterol oxidase를 고정화시켜 바이오센서의 콜레스테롤 표준용액에 대한 감응도를 측정한 결과, aminopropyl glass beads과 CNBr-activated sepharose는 1~100 ${\mu}M$ 범위에서 선형관계를 보였으며, Na-alginate는 5~50 ${\mu}M$의 범위에서, toyopearl beads는 1~50 ${\mu}M$ 범위에서 선형관계를 나타내었다. 검출한계는 제작된 효소반응기 모두 1 ${\mu}M$로 나타나 콜레스테롤에 대한 높은 검출력을 보여주었으나, 특히 CNBr-activated sepharose와 Na-alginate를 이용한 효소반응기가 높은 coupling efficiency와 감응도를 보여 콜레스테롤 검출을 위한 본 바이오 센서 시스템에 적합한 것으로 나타났다.

Insulin-like growth factor가 소장 점막 세포 증식에 미치는 영향

  • Growth hormone (GH) plays a key role in regulating postnatal growth and can stimulate growth of animals by acting directly on specific receptors on the plasma membrane of tissues or indirectly through stimulating insulin-like growth factor (IGF)-I synthesis and secretion by the liver and other tissues. IGF-I and IGF-Ⅱ are polypeptides with structural similarity with proinsulin that stimulate cell proliferation by endocrine, paracrine and autocrine mechanisms. The initial event in the metabolic action of IGFs on target cells appears to be their binding to specific receptors on the plasma membrane. Current evidence indicates that the mitogenic actions of both IGFs are mediated primarily by binding to the type I IGF receptors, and that IGF action is also mediated by interactions with IGF-binding proteins (IGFBPs). Six distinct IGFBPs have been identified that are characterized by cell-specific interaction, transcriptional and post-translational regulation by many different effectors, and the ability to either potentiate or inhibit IGF actions. Nutritional deficiencies can have their devastating consequence during growth. Although IGF-I is the major mediator of GH's action on somatic growth, nutritional status of an organism is a critical regulator of IGF-I and IGFBPs. Various nutrient deficiencies result in decreased serum IGF-I levels and altered IGFBP levels, but the blood levels of GH are generally unchanged or elevated in malnutrition. Effects of protein, energy, vitamin C and D, and zinc on serum IGF and IGFBP levels and tissue mRNA levels were reviewed in the text. Multiple factors are involved in the regulation of intestinal epithelial cell growth and differentiation. Among these factors the nutritional status of individuals is the most important. The intestinal epithelium is an important site for mitogenic action of the IGFs in vivo, with exogenous IGF-I stimulating mucosal hyperplasia. Therefore, the IGF system appears to provide and important mechanism linking nutrition and the proliferation of intestinal epithelial cells. In order to study the detailed mechanisms by which intestinal mucosa is regulated, we have utilized IEC-6 cells, an intestinal epithelial cell line and Caco-2 cells, a human colon adenocarcinoma cell line. Like intestinal crypt cells analyzed in vivo or freshly isolated intestinal epithelial cells, IEC-6 cells and Caco-2 cells possess abundant quatities of both type Ⅰ and type Ⅱ IGF receptors. Exogenous IGFs stimulate, whereas addition of IGFBP-2 inhibits IEC-6 cell proliferation. To investigate whether endogenously secreted IGFBP-2 inhibit proliferation, IEC-6 cells were transfected with a full-length rat IGFBP-2 cDNA anti-sense expression construct. IEC-6 cells transfected with anti-sense IGFBP-2 protein in medium. These cells grew at a rate faster than the control cells indicating that endogenous IGFBP-2 inhibits proliferation of IEC-6 cells, probably by sequestering IGFs. IEC-6 cells express many characteristics of enterocyte, but do not undergo differentiation. On the other hand, Caco-2 cells undergo a spontaneous enterocyte differentiation. On the other hand, Caco-2 cells undergo a spontaneous enterocyte differentiation after reaching confluency. We have demonstrated that Caco-2 cells produce IGF-Ⅱ, IGFBP-2, IGFBP-3, and an as yet unidentified 31,000 Mr IGFBP, and that both mRNA and peptide secretion of IGFBP-2 and IGFBP-3 increased, but IGFBP-4 mRNA and protein secretion decreased after the cells reached confluency. These changes occurred in parallel to and were coincident with differentiation of the cells, as measured by expression of sucrase-isomaltase. In addition, Caco-2 cell clones forced to overexpress IGFBP-4 by transfection with a rat IGFBP-4 cDNA construct exhibited a significantly slower growth rate under serum-free conditions and had increased expression of sucrase-isomaltase compared with vector control cells. These results indicate that IGFBP-4 inhibits proliferation and stimulates differentiation of Caco-2 cells, probably by inhibiting the mitogenic actions of IGFs.

임산부의 산전관리와 산욕기 영양실태에 관한 연구 (Clinical Study on Prenatal care, and Dietary Intakes for Pregnant Women and new Mothers)

  • This study was projected to provide basic data on prenatal care for future direction in maternity and child care, and also to investigate the diet of women during pregnancy and the period directly afterwards in order to offer to mothers appropriate advice for the improvement of nutritional standards. A clinical study on prenatal care was based on 1054 delivery cases. A nutritional survey was performed on 174 mothers admitted to the department of obstetrics at St. Mary's Hospital during the period of March, 1975 to February, 1976. The results obtained are summarized as follows; I. Clinical study on prenatal care 1) The age distribution showed 59.4% of the mothers were between the ages of 25 to 29 years old. 2) The gestational period was highest between the 37th and 40th gestational weeks. 33.7% of the mothers were primigravidae and 31.8% of them primiparae. 3) 41.3% of the mothers had not received prenatal care or had only received it once before. 4) Induced deliveries were 61.8% and spontantaneous deliveries 38.2%. 61.9% of the mothers had received prenatal care, while those without prenatal care accounted for 61.6% of the total induced deliveries. 5) Low birth weights were 7.7% and 5.0% of the mothers had received prenatal care, while 11.5% had no prenatal care. 6) There were 1.13% of still births, 0.32% of the mothers had prenatal care and the remainder did not have prenatal care. 7) Of those receiving prenatal care, 2.1% showed in the $0{\sim}3$ Apgar score group, 6.3% in the $4{\sim}6$ Apgar score group, and 91.6% in the $7{\sim}10$ Apgar score group. Among the non-prenatally cared for group 5.0% of the newborns were in the $0{\sim}3$ Apgar score group, 9.7% were in $4{\sim}6$ Apgar score group and 85.3% were in the $7{\sim}10$ Apgar score group. 8) Obstetrical complications were developed in 11.86% of the pregnant women when they were hospitalized. Among the group receiving the prenatal care 8.1% of the mothers had obstetrical complications. In the group without prenatal care 17.16% of the mothers had obstetrical complications. The most common obstetrical complication was malpresentation. 9) The first prenatal care was received between the 37th and 40th gestationl weeks. II. Food intake during pregnancy The following are the results from the questionnaires of the mothers concerning diets during pregnancy; 1) Main meals and snacks In 32.2% of the cases, their main meals during the diet amounted to more than was usually eaten at other times. In 67.8% of the cases, their main meals during the diet were the same as that usually eaten. In 22.4% of the cases, snacks during the diet amounted to more than usually eaten at other times. In 77.6% of the cases, snacks during the diet were the same as usually eaten. 2) Itemized list The mothers made a special effort to include certain items in their diets, the following is a breakdown of those items; a. egg, meat, fish 33.3% b. fruit, vegetables 32.2%. c. milk, fruit juice 18.4% d. cake, bread 2.9% e. nothing special 13.2% 3) Milk 44.8% of the mothers had at least one cup of milk everyday. 33.4% of the mothers had at least one cup of milk on occasion. 15.5% of the mothers did not have any milk. 4) Vitamins 39.7% of the mothers had vitamins everyday. 24.7% of the mothers had vitamins occasionally. 35.6% of the mothers did not have any vitamins. 5) Anemic symptoms 9.2% of the mothers very often had anemic symptoms during pregnancy. 39.1% of the mothers often had anemic symptoms during pregnancy. 51.7% of the mothers did not have anemic symptoms at all. 6) Taboos on food 23% of the mothers recognized 'taboos' on food during pregnancy 27% of the mothers displayed on uncertainty about the 'taboos' on food during pregnancy 50% of the mothers displayed indifference toward the taboos. III. Nutritional survey on the new mothers diet. 1) The diets for new mothers can be divided into four categories, such as general diet, low sodium diet, soft diet and liquid diet. 2) Cooked rice and seaweed soup were the main foods for the new mothers as has been the traditional diet for Korean mothers. 3) The average diet contained 1,783g.
