Recolonization of Transfected Blastodermal Cells in Developing Embryos after Transferring into UV-irradiated Fertilized Hen′s Egg (UV-조사 수정란 내로 이식한 유전자 변화 배반엽 세포의 재구성)

  • Unfortunately, there is no technique which is stable and repetitive to produce transgenic chicken, although various ways of gene transfer including PGC-and embryonic cell-mediated gene transfer, DNA microinjection, virus inoculation and sperm cells have been employed. The aims of this study were 세 develop and establish such a stable, repetitive and efficient way of gene transfer giving a faithful gene expression during development after the reconstruction of embryo in an UV-irradiated egg. A dual reporter plasmid (pJJ9), a fusion gene containing lacZ and GFP driven by a CMV promoter was used to exploit either merits of both reporting markers. lacZ with strong signal or GFP with vital marking. Electroporated embryonic blastodermal cells (EBCs) in the presence of the pJJ9 DNA faithfully showed 377 bp PCR product and lacZ or GFP expressions in the identical cells in vitro of in vivo. Furthermore, analyses of expression pattern of the foreign DNA demonstrated that microinjected EBCs cells into the UV-irradiated recipient egg should participate in normal developmental process, for example, proliferation and differentiation into various tissues. Thirty percentages of the manipulated eggs showed lacZ expression in their tissues. These results together with the specific procedures used in this study should facilitate avian transgenesis.

  • Recently, researches related to automative mechanism have been widely studied to increase the driver's safety by continuously monitoring the driver's health condition to prevent driver's drowsiness. This paper describes the design of wearable chest belt for ECG and reflectance pulse oximetry for $SpO_2$ sensors based on wireless sensor network to monitor the driver's healthcare status. ECG, $SpO_2$ and heart rate signals can be transmitted via wireless sensor node to base station connected to the server. Intelligent monitoring system is designed at the server to analyze the $SpO_2$ and ECG signals. HRV(Heart Rate Variability) signals can be obtained by processing the ECG and PPG signals. HRV signals are further analyzed based on time and frequency domain to determine the driver's drowsiness status.

Antineuroinflammatory Effects of 7,3',4'-Trihydroxyisoflavone in Lipopolysaccharide-Stimulated BV2 Microglial Cells through MAPK and NF-κB Signaling Suppression

  • Neuroinflammation―a common pathological feature of neurodegenerative disorders such as Alzheimer's disease―is mediated by microglial activation. Thus, inhibiting microglial activation is vital for treating various neurological disorders. 7,3',4'-Trihydroxyisoflavone (THIF)―a secondary metabolite of the soybean compound daidzein―possesses antioxidant and anticancer properties. However, the effects of 7,3',4'-THIF on microglial activation have not been explored. In this study, antineuroinflammatory effects of 7,3',4'-THIF in lipopolysaccharide (LPS)-stimulated BV2 microglial cells were examined. 7,3',4'-THIF significantly suppressed the production of the proinflammatory mediators nitric oxide (NO), inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) as well as of the proinflammatory cytokine interleukin-6 (IL-6) in LPS-stimulated BV2 microglial cells. Moreover, 7,3',4'-THIF markedly inhibited reactive oxygen species (ROS) generation. Western blotting revealed that 7,3',4'-THIF diminished LPS-induced phosphorylation of extracellular signal-regulated kinase (ERK), c-Jun N-terminal kinase (JNK), glycogen synthase kinase-3β (GSK-3β), and nuclear factor kappa B (NF-κB). Overall, 7,3',4'-THIF exerts antineuroinflammatory effects against LPS-induced microglial activation by suppressing mitogen-activated protein kinase (MAPK) and NF-κB signaling, ultimately reducing proinflammatory responses. Therefore, these antineuroinflammatory effects of 7,3',4'-THIF suggest its potential as a therapeutic agent for neurodegenerative disorders.

A Performance Comparison Study of Lesion Detection Model according to Gastroscopy Image Quality (위 내시경 이미지 품질에 따른 병변 검출 모델의 성능 비교 연구)

  • Many recent studies have reported that the quality of input learning data was vital to the detection of regions of interest. However, due to a lack of research on the quality of learning data on lesion detetcting using gastroscopy, we aimed to quantify the impact of quality difference in endoscopic images to lesion detection models using Image Quality Assessment (IQA) algorithms. Through IQA methods such as BRISQUE (Blind/Referenceless Image Spatial Quality Evaluation), Laplacian Score, and PSNR (Peak Signal-To-Noise) algorithm on 430 sheets of high quality data (HQD) and 430 sheets of low quality data (PQD), we showed that there were significant differences between high and low quality images in lesion detecting through BRISQUE and Laplacian scores (p<0.05). The PSNR value showed 10.62±1.76 dB on average, illustrating the lower lesion detection performance of PQD than HQD. In addition, F1-Score of HQD showed higher detection performance at 77.42±3.36% while F1-Score of PQD showed 66.82±9.07%. Through this study, we hope to contribute to future gastroscopy lesion detection assistance systems that involve IQA algorithms by emphasizing the importance of using high quality data over lower quality data.

  • As it is changing into aging society, rehabilitation, welfare and sports industry markets are being expanded fast. Especially, the field of vital signals interface to control welfare instruments like wheelchair, rehabilitation ones like an artificial arm and leg and general electronic ones is a new technology field in the future. Also, this technology can help not only the handicapped, the old and the weak and the rehabilitation patients but also the general public in various application field. The commercial bio-signal measurement instruments and interface systems are complicated, expensive and large-scaled. So, there are a lot of limitations for using in real life with ease. this thesis proposes a wireless transmission interface system that uses EMG(electromyogram) signals and a control module to manipulate hardware systems with portable size. We have designed a hardware module that receives the EMG signals occurring at the time of wrist movement and eliminated noises with filter and amplified the signals effectively. DSP(Digital Signal Processor) chip of TMS320F2808 which was supplied from TI company was used for converting into digital signals from measured EMG signals and digital filtering. We also have used PCA(Principal Component Analysis) technique and classified into four motions which have right, left, up and down direction. This data was transmitted by wireless module in order to display at PC monitor. As a result, the developed system obtains recognition success ratio above 85% for four different motions. If the recognition ratio will be increased with more experiments. this implemented system using EMG wrist direction signals could be used to control various hardware systems.

  • Blood supply rather than nerve supply implies pulp vitality. To evaluate pulp vitality clinically, electric pulp test and thermal test which are based on sensory nerve response have been used in addition to many auxiliary data such as past dental history, visual inspection, radiographic examination, percussion, palpation and transillumination test. However, reactivity of the nerves to the stimulation is not synonymous with normalcy. Therefore measurement of pulpal blood flow using a laser Doppler flowmeter became a new trial to test the pulp vitality. The purpose of the present study was to evaluate normal pulpal blood flow level of maxillary teeth in adult to provide a guideline in determining the vitality of dental pulp. Pulpal blood flow was measured in maxillary central and lateral incisors, canines, first and second premolars and first molars of seventy nine adults of 22 - 30 years old using a laser Doppler flowmeter (PeriFlux 4001, Perimed Co., Stockholm, Sweden, 780 nm infrared laser, 1mW). For directly-made splints, silicone rubber impressions were taken directly from the mouth. For indirectly-made splints, alginate impressions were taken from the mouth and stone cast were made. After making depressions on the buccal surfaces of the cast teeth to indicate the hole positions, second impressions with vinyl polysyloxane putty were taken from the cast. Holes for the laser probes were made at the putty impressions 4mm above the gingival level. Laser probe (PF416 dental probe, 1.5mm) was inserted in the prepared hole and the splint was set in the mouth. After 10 minutes of patient relaxing, pulpal blood flow was recorded for 5 minutes on each tooth. The recorded flow was saved in the computer and calculated with a software 'Perisoft' version 5.1. Pulpal blood flow was also recorded in six teeth of five individuals with no response to electric pulp test and cold test, with periapical radiolucency, or with history of root canal treatment to compare with nonvital teeth. The difference between the mean flow values of each group of teeth were analyzed using one-way ANOVA and Duncan's Multiple Range test. The results were as follows: 1. The average pulpal blood flow values of all the tested teeth of each location were between 9 - 16 Perfusion Unit. Pulpal blood flow value was highest in maxillary lateral incisors, followed by first premolars, second premolars, canines, central incisors, and then first molars (p<0.01). 2. In six anterior teeth, indirectly-made splint group showed higher pulpal blood flow values than directly-made splint group (p<0.01). In posterior teeth, however, there was no significant flow value difference between directly-made splint group and indirectly-made splint one (p>0.05). 3. Teeth with vital pulps showed higher signal values than teeth with nonvital pulps (p<0.01), and the flow photographs showed heartbeat-synchronous fluctuations and vasomotions, while those were absent in non vital tooth.

  • Objectives : This study is to evaluate Effect of Inhibition Macrophage Migration Inhibitory Factor(MIF) activation by Hominis Placenta Herbal Acupuncture(HPA) on Rheumatic Arthritis(RA). Hominis Placenta is the placenta of healthy human, which is vital-strengthening medical stuff. In recent years, Hominis Placenta applied to chronic diseases because it makes us more resistance to diseases. Therefore it is supposed that HPA is effective on RA, a kind of autoimmune disease. When RA is induced, MIF is activated, too. MIF affects the process of inflammatory disease including RA. Methods : In order to investigate the effect of Hominis Placenta extraction on MIF(early RA inducing cytokine) and MMP(Matrix Metallo Proteinase)-9 mRNA expression by means of Reverse Transcriptase- Polymerase Chain Reaction(RT-PCR). In this study, we investigated the effect of Hominis Placenta extraction on MIF(early RA inducing cytokine) and MMP-9 mRNA expression by means of RT-PCR. Besides we investigated changing of MIF in synovial membrane and, Interleukin-6 receptor(IL-6R)-$\alpha$(pro-inflammatory cytokine), Signal transducers and activators of transcription(STAT)-3, MMP-9 after treating mouse, which is artificially attacked with RA, with HPA on its $ST_{35}$, LE201 in vivo. Results : 1. As a result of treating Lipopolysaccharide(LPS)-stimulated Raw246.7cell with HPA, MIF(RA related cytokine) and MMP-9 mRNA expression is reduced in vitro. And this reaction is concentration-dependatant. 2. In synovial membrane of the mice treated with HPA, inhibition of MIF, IL-6R-$\alpha$, STAT3 & MMP-9 activation is observed in vivo. Conclusions : From the above results, it might be suggested that HPA mitigate tissue damage originated from RA, because it intercepts the early process of by inhibition MIF activity.

  • Cognitive radio (CR) technology based on geo-location database access approach and/or wideband spectrum sensing approach is absolutely vital in order to recognize available frequency bands in white spaces (WSs), and efficiently utilize shared spectrums. This paper presents a new structure for the TVWS database access protocol implementation based on Internet Engineering Task Force (IETF) Protocol to Access WS database (PAWS). A wideband compressive spectrum sensing (WCSS) scheme using a modulated wideband converter is also proposed for the TVWS utilization. The developed database access protocol technology which is adopted in both the TV band device (TVBD) and the TVWS database operates well in the TV frequency bands. The proposed WCSS shows a stable performance in false alarm probability irrespective of noise variance estimation error as well as provides signal detection probabilities greater than 95%. This paper also investigates Federal Communications Commision (FCC) regulatory requirements of TVWS database as well as European Telecommunications Standards Institute (ETSI) policy related to TVWS database. A standardized protocol to achieve interoperability among multiple TVBDs and TVWS databases, which is currently prepared in the IETF, is discussed.

Effect of EC Level of Nutrient Solution on the Yield and Quality of Cut Rose Based on Mixed Coir and Perlite (코이어와 펄라이트 혼합배지를 이용한 절화장미 수경재배 시급액농도가 수량 및 품질에 미치는 영향)

  • Organic materials reveal the remarkable absorption and high buffer capacity for nutrient. Hence, organic materials need some different nutrient management skill from inorganic one. The objective of this study was to evaluate the effect of EC level of nutrient solution on the yield and quality of cut rose grown in the mixed substrate of coir and perlite. 3 EC levels of nutrient solution was treated, which were 0.7, 1.0, 1.3 times of standard solution (Aichiken solution, Japan) for cut rose hydroponics. EC of the standard solution was changed by season following as 1.4 (Apr.~June), 1.0 (July~Aug.), 1.4 (Sep.~Oct.), and $1.6dS{\cdot}m^{-1}$ (Nov.~Mar.) subsequently. The supply of nutrient solution was controlled by the signal of water potential at -5kPa using frequency domain reflectometry (FDR) sensor. As the results, marketable yield was similar for all treatments until 3rd harvest, but was decreased in high EC level from 4th harvest to 7th harvest as final. 0.7 times of standard solution decreased the ratio of unmarketable rose having short stem below 70cm and increased the ratio of high quality rose having long stem above 91cm. The flower weight and stem diameter of cut rose was higher in the low EC treatment than the others.

FISH Karyotype Analysis of Seven Rose Cultivars (주요 장미 7품종의 FISH 핵형분석)

  • Determination of ploidy level for the mother plant is prerequisite for effective breeding. The study was carried out to determine the ploidy level in 7 different plant materials by FISH karyotype analysis. Among the seven plant varieties analyzed, all exhibit tetraploid (2n = 4x = 28) based on the results observed in chromosome analysis. Four signals of 45S rDNAs were detected on the terminal region of the short arm of chromosome 7. The length of somatic metaphase chromosomes ranges from 1.67 to $2.67{\mu}m$ in 'Alexandra', 1.40 to $2.04{\mu}m$ in 'Freud', 1.64 to 2.24 in 'Little Silver', 1.69 to $2.26{\mu}m$ in 'Teresa', 1.70 to $2.65{\mu}m$ in 'Tineke', 1.35 to $2.08{\mu}m$ in 'Vital', 1.39 to $2.04{\mu}m$ in 'Yellow Mimi'. Total length of the chromosome ranges from $11.23{\mu}m$ in 'Freud' as minimum to $15.05{\mu}m$ in 'Alexandra' as maximum. The karyotypes were composed of metacentric, submetacentric, and subtelocentric chromosome but there is no subtelocentric chromosome.
