Isolation of Infectious Pancreatic Necrosis Virus from Goldfish (Carassius auratus) and Chum Salmon (Oncorhynchus keta) in Korea. (금붕어(Carassius auratus)와 연어(Oncorhynchus keta)에서 췌장괴저 바이러스(IPNV)의 분리에 대하여)

  • Two viruses were isolated from kidney and spleen tissues of goldfish(Carasius auratus) and the ovarian fluid of chum salmon(Oncorhynchus keta). Both viruses replicated and produced cytopathic effect in EPC, CHSE-214, and CHH-1 cell lines at $15^{\circ}C$. The isolates were resistant to pH 3 and choloroform. Antiserum to infections pancreatic necrosis virus(IPNV) serotype VR 299 neutralized the infectivity of both of the isolates. Electron microscopy showed that the particles had typical IPNV particle morphology with average diameters of 55nm, This paper describes the first isolation of viruses infecting cultured fish in Korea.

  • The occurrence of potato(Sotanum tuberosum) viral diseases caused by Potato virus X(PVX), Potato virus Y (PVY), Potato leafroll virus(PLRV), Potato vims S(PVS), Potato virus M(PVM), Potato virus A(PVA), Potato virus T(PVT), Alfalfa mosic virus(AIMV), Tobacco mosic virus(TMV), Potato mop top virus(PMTV) Tobacco rattle virus(TRV) and Potato spindle tuber viroid(PSTVd), potato witches' broom phytoplasma, have been identified so far in Korea. Major viral diseases such as PVX, PVY and PLRV had been studied more deeply, however, the others are just identified and only partially characterized since the first study on the relation between PVX nucleic acid and virus protein by Kim in 1961. The most studies on potato viral diseases are mainly focused on the problems of seed potato production. The National Alpine Agricultural Experiment Station(NAAES), since it began its activities in 1961, has given special attention to this problem by doing studies to identify, characterize and control potato virus diseases. This effort resulted in the development of new potato virus detection methods as a basis for elaborating new method of control, such as the production of seed potato free of virus and the selection of new virus-resistant transgenic potatoes. The further studies of potato viral diseases required would be fallowings: the continuous monitoring for the occurrence of identified or not identified potato viruses in Korea, the isolation of resistant viral genes, the development of control method for the non-persistently transmitted viruses like PVY, special vectors such as nematode and fungus transmitted viruses, TRV and PMTV and the development of control methods against potato viral diseases by viral cross protection, therapy, transgenic plant, and the use of the agents or molecules, such as virus inhibitors and antiviral proteins, etc., blocking viral replication.

  • The severe acute respiratory syndrome -SARS virus spreaded rapidly in Asia in 2003 caused a lethal and serious epidemic. Establishing special isolation wards of high-level epidemic prevention has becomes one of the most important tasks of epidemic prevention. Taking the biosafety level of US CDC biology laboratory as an example, the laboratory of the highest level, BL4 and that of the second high level, BL3 are only allowed to handle and research this kind of highly dangerous viruses safely, to ensure the safety of researchers and the surrounding environment.This research aims to study the functions and the design requirements of "special isolation wards", which is equivalent to the high-level biotechnology laboratory, and have an integrated discussion about the design and construction techniques of this kind of wards, expecting to provide a reference for constructing special isolation wards and maintaining operation safely in Taiwan.

Isolation and Properties of Cucumber Mosaic Virus Inducing Mosaic Symptoms in Hippeastrum hybridum Hort (아마리리스에 모자익병을 일으키는 CMV에 관한 연구)

  • Cucumber Mosaic Virus (CMV) was isolated from naturally infected Hippenstrum hybridum. The virus caused mosaic symptoms on Nicotiana glutinosa and local lesions on Vignaunguiculata. The thermal inactivation point was 56C, dilution end point $10^{-3}$ and longevity in vitro was 2 days for CMV from Hippeastrum. Purified virus was obtained using citrate chloroform extraction procedure and polyethylene glycol precipitation followed by sucrose density gradient centrifugation. Purified virus had a typical absorption at 245nm. Electron micrographs of the purified virus from Hippeastrum showed spherical particles with 30nm in diameter. The purified virus reacted with CMV antiserum in agar gel double diffusion test.

Development of a novel reverse transcription PCR and its application to field sample testing for feline calicivirus prevalence in healthy stray cats in Korea

  • Background: Feline calicivirus (FCV) is a major and highly infectious pathogen in cats worldwide. However, there have been limited studies about the status of FCV infections in Korea. Objectives: To investigate the current status of FCV infections in stray cats in Korea. Methods: A novel reverse transcription polymerase chain reaction (RT-PCR) assay was developed based on the conserved nucleotide sequences of reported FCV strains. Field swab samples were collected from 122 cats (2 hospital admitted cats and 120 stray cats) in 2016 and 2017. All the samples were tested by virus isolation and 2 different RT-PCRs, including the novel RT-PCR, for the detection of FCV. Results: The novel RT-PCR assay showed no cross-reactivity to the nucleic acids of the other feline pathogens tested, and the limit of detection was calculated as 100 TCID50/mL based on an in vitro assessment. The novel RT-PCR assay detected 5 positive samples from the 122 field samples, which showed perfect agreement with the results of the virus isolation method. In contrast, another RT-PCR assay used in a previous study in Korea detected no positive samples. The prevalence of FCV infection in stray cats was 2.5% (3/120) based on the results of virus isolation and the novel RT-PCR assays. Conclusions: The current study is the first report of the detection and prevalence of FCV in stray cats in Korea. The novel RT-PCR assay developed in this study showed high sensitivity and specificity, which indicates a useful diagnostic assay to identify FCV infection in cats.

  • Recent studies have shown the importance of viruses as potential sources of plankton mortality, which affect primary production and biogeochemical functions of their hosts. Here, we report basic characteristics of a novel virus (Stephanopyxis palmeriana virus: SpalV) that causes lysis of a culture of the diatom S. palmeriana, which was isolated in Jaran Bay, Korea, in August 2008. SpalV is a round-shaped viral particle ~25-30 nm in diameter that propagates in its host's cytoplasm. In addition, it shows species-specific infectivity among the tested diatom species. The burst size and latent period are estimated to be roughly 92 infectious units $cell^{-1}$ and <80 h, respectively.

  • Diatoms are a major component of the biological community, serving as the principal primary producers in the food web and sustaining oxygen levels in aquatic environments. Among marine planktonic diatoms, the cosmopolitan Skeletonema costatum is one of the most abundant and widespread species in the world's oceans. Here, we report the basic characteristics of a new diatom-infecting S. costatum virus (ScosV) isolated from Jaran Bay, Korea, in June 2008. ScosV is a polyhedral virus (45-50 nm in diameter) that propagates in the cytoplasm of host cells and causes lysis of S. costatum cultures. The infectivity of ScosV was determined to be strain- rather than species-specific, similar to other algal viruses. The burst size and latent period were roughly estimated at 90-250 infectious units/cell and <48 h, respectively.

  • Investigate the epidemics for influenza outbreaks. The outbreak pattern of the internal patients housed in the 10 designated hospitals was monitered to investigate and the characteristics of the virus isolates are as follows. 232 strains of influenza virus was isolated from the oral specimen of 1,320 respiratory disease patients in Pusan from Oct. 1998 to Jun. 1999. Among these isolates, 222 strains were A-type and the rest were B-type. The outbreak pattern for sex-and age-groups is as follows. The male outbreak was similar to the female outbreak: male outbreak, 47.4% and female outbreak, 52.5%. Most of the patients were less than 10 years old. The monthly influenza outbreak was consistent from Dec. 1998 to Apr. 1999. and The 113 strains from the A-type isolates were A/ Sydney/05/97(H3N2)-like, the 109 strains were A/Beijing/262/95(H1N1)-like, and all of the 10 B-type isolates were B/Harbin/07/94-like.

  • The outbreak patterns of the internal and external and external patients in the 20 designated hospitals and in 16 health centers were monitored to investigate and the characteristics of the virus isolates were as follows. Two hundreds and thirteen strains of influenza virus were isolated from the oral specimens of 1,686 patients with respiratory disease in Pusan. 1999.
