Cucumber mosaic cucumovirus (CMV) is an isometric plant virus with functionally divided genomic RNAs and a broad host range. RNA 1 and RNA 2 each encode one protein, both of which are essential for replication. RNA 3 encodes the viral coat protein and an additional protein thought to be involved in potentiating the cell-to-cell movement of the virus. Functions of the RNAs have been confirmed using a pseudorecombinant virus constructed with infectious cDNA-derived transcripts of the RNAs. Generally, CMV produces different symptoms in various host plants depending on the virus strains. In this mini-review, we describe the potential role of the genes associated with symptom expression of CMV RNAs.
Molecular analysis has been done for characterization of the interactions between three beet curly top virus (BCTV) strains and two Arabidopsis ecotypes in terms of virus inducible disease symptoms and infectivities. The total DNA was isolated from three tissues (shoot tips, infection origins and roots) of virus infected plants and this DNA was analyzed by quantitatively and qualitatively to elucidate virus movement and symptom development. CTV-Worland infected Col-O and Sei-O showed only symptom shown in hypersusceptible ecotype Sei-O by BCTV-worland was shoot tip stunting. Kinetics of virus DNA accumulation of three different viruses indicated that roots contained more virus DNA than shoot tips or infection origins, and that disease symptom severity was strongly correlated with virus DNA accumulation. These results suggest that the mild and Worland-specific symptoms shown in Sei-O by BCTV-worland are caused by the interactions of host factors provided by hypersusceptible ecotype and viral factors of mild strain.
Beet curly top virus is the DNA virus that is providing useful for basic studies of the infection of Arabidopsis thaliana with viral host and provides a system for studying both resistance and the molecular basis of symptom development. An importnat aspect of symptom development observed in BCTV-infected A. thaliana (ecotype Sei-O) was the induction of cell division on phloem and surrounding cortex cells. Analysis of the expression of GUS reporter gene activity in transgenic plants containing constructs with promoter of the auxin-inducible saur gene showed that saur promoter activity was induced concomitantly in symptomatic tissues at the inflorescence shoot tips of the transgenic lines. The auxin sensitivity tests showed that hypersusceptible ecotype, Sei-O produced more amounts of callus than susceptible ecotype, Col-O. These studies indicated that changes in auxin concentration were involved in the induction of cell division in BCTV-infected plants and clearly demonstrated that there was a strong correlation between auxin-induced gene expression and the activation of cell division.
Recently, some global cases of 2019 novel coronavirus (COVID-19) pneumonia have been caused by second- or third-generation transmission of the viral infection, resulting in no traceable epidemiological history. Owing to the complications of COVID-19 pneumonia, the first symptom and imaging features of patients can be very atypical and early diagnosis of COVID-19 infections remains a challenge. It would aid radiologists and clinicians to be aware of the early atypical symptom and imaging features of the disease and contribute to the prevention of infected patients being missed.
$Potato$$virus$$X$ (PVX) replication is precisely regulated by regulatory viral sequences and by viral and/or host proteins. In a previous study, we identified a 54-kDa cellular tobacco protein that bound to a region within the first 46 nucleotides (nt) of the 5' non-translated region (NTR) of the viral genome. Optimal binding was dependent upon the presence of an ACCA sequence at nt 10-13. To identify host factors that bind to 5' NTR elements including AC-rich sequences as well as stemloop 1 (SL1), we used northwestern blotting and matrixassisted laser desorption/ionization time-of-flight mass spectrometry for peptide mass fingerprinting. We screened several host factors that might affect PVX replication and selected a candidate protein, $Nicotiana$$tabacum$ WRKY transcription factor 1 (NtWRKY1). We used a $Tobacco$$rattle$$virus$ (TRV)-based virus-induced gene silencing (VIGS) system to investigate the role of NtWRKY1 in PVX replication. Silencing of $WRKY1$ in $Nicotiana$$benthamiana$ caused lethal apical necrosis and allowed an increase in PVX RNA accumulation. This result could reflect the balancing of PVX accumulation in a systemic $N.$$benthamiana$ host to maintain PVX survival and still produce a suitable appearance of mosaic and mottle symptoms. Our results suggest that PVX may recruit the WRKY transcription factor, which binds to the 5' NTR of viral genomic RNA and acts as a key regulator of viral infection.
Tobacco plants resistant (cvs. Xanthi-nc and Samsun-NN) and susceptible (cv. NC 82) to tobacco mosaic virus (TMV) were inoculated with TW to obtain basic information about the characteristics of resistance expression in tobacco plants by examining the viral populations, symptom development and gene expression of pathogenesis-related proteins (PR-proteins) such as PR-1 and $\beta$-1, 3-glucanase in different temperature conditions. TMV populations in resistant plants increased more at 37$^{\circ}C$ than at 27$^{\circ}C$, while the viral populations increased continuously and were not significantly influenced by the temperature conditions in the susceptible tobacco plants. Infection sites of resistant tobacco leaves were remarkably expanded in proportion with increased time at the high temperature.
Viral coat protein (CP) encoding cDNA with artificial start and stop codons was synthesized by reverse-transcriptase polymerase chain reaction (RT-PCR) from the Korean isolate of potato virus Y-vein nectrosis strain (pVY-VN). To make PVY CP cDNA to untranslatable form, three stop codons were inserted near the start codon by "megaprimer-PCR" method. The untranslatable CP cDNA was subcloned to plant expression vector and transferred to N. tabacum cv. NC82 by Agrobacterium-mediated transformation. Highly resistant plants to PVY infection were screened, based on symptom development after mechanical virus inoculation. By genomic PCR and Southern blot analysis, one or more copies of the untranslatable CP gene were found in all transformants. From northern blot analysis, highly resistant transgenic lines had very low level of CP transcript but susceptible lines had high level, suggesting resistance to PVY infection should be related to RNA-mediated mechanism.mechanism.
Mutants of the monopartite geminivirus beet curly top virus (BCTV) have been screened for infectivity, systemic movement, replication and symptom development in Arabidopsis thaliana. As known by coding for coat protein, R1 mutant was not infectious and did not move systemically. R2, R3 and L2/L3 mutants produced milder symptoms compared to wild type BCTV but the infectivity was reduced by 40% to 60%. R2 ORF is thought to be involved in the regulation of ssDNA and dsDNA accumulation because only dsDNA was accumulated on R2-infected organs. Disruption of ORF L4 resulted in reduced infections, but the viral DNA was accumulated in infected organs from roots to shoot tips as much as wild type BCTV on Sei-O. In addition, 4 mutants did not produce callus-like tissues on infected organs, suggesting that L4 ORF may play a role in the induction of host cell divisions by virus infection. This result was supported by the patterns of mRNA expression and promoter analysis of the cell cycle marker gene, cycl, on Arabidopsis. cycl mRNA was accumulated on symptomatic organs by wild type BCTV infections but not by L4 mutant. We conclude that the BCTV L4 ORF is essential for symptom developments, specially callus-like formation on infected organs.

We conducted this study to report the clinical manifestations of 5 acute viral hepatitis type A treated by oriental medical treatment. Five hepatitis A patients who visited OO oriental hospital from May 2008 to July 2009 were treated by oriental medical treatment, including herbal acupuncture, acupuncture and herbal medicine and western medical treatment, including fluid therapy and peroral medicine. Prodromes were similar to influenza and duration from symptom onset to first visit were $5.0\;{\pm}\;1.6$ days. Chief symptoms included jaundice, itching, dark urine, anorexia, fatigue, nausea, vomiting, abdominal discomfort. The mean values of the initial laboratory test were serum total bilirubin(TB) of $3.62\;{\pm}\;2.77\;mg/dL$, aspartate aminotransferase(AST) of $729\;{\pm}\;422\;IU/L$, alanine aminotransferase(ALT) $774\;{\pm}\;754\;IU/L$. The peak mean values of those were $6.30\;{\pm}\;2.13\;mg/dL$, $2177\;{\pm}\;2573\;IU/L$, $2238\;{\pm}\;1682\;IU/L$ respectively. The mean duration of hospitalization were $15\;{\pm}\;3$ days. Follow-up at 3 weeks after discharge showed that all patients recovered without complications. Treatment of oriental medicine has effectiveness on acute viral hepatitis type A. Further study is needed.

The current market size of insect industry in Korea is estimated at 300 million dollars and more than 500 local farms are related to many insect industry. One of the strong candidates for insect industry is Korean horn beetle, Allomyrina dichotoma. Early this year, we reported a viral disease extremely fatal to A. dichotoma larvae. While we were proceeding a nationwide investigation of this disease, it was informed that similar disease symptom has been occurred occasionally during past over 10 years. The symptom can be easily confused with early stage of bacterial infection or physiological damage such as low temperature and high humidity. A peroral infection with the purified virus to healthy larvae produced a result that only 21% of larvae survived and became pupae. Although some of the survived adult beetle was deformational, many of them had no abnormal appearance and even succeeded in mating. Later, these beetles were examined if they were carrying the virus, and all except one were confirmed as live virus carrier. This implies that these beetles may fly out and spread the disease to the nature. We found the evidence for this possibility by collecting a few wild A. dichotoma larvae which were virus infected, near two local farms rearing A. dichotoma larvae. So far, transovarial transmission of this virus to the eggs, or horizontal transmission to other commercially reared insects is not known yet.
