Cdc25B is a mitotic regulator that might act as a starter phosphatase to initiate the positive feedback loop at the entry into mitotic (M) phase. In the present study, distribution of Cdc25B mRNA in duodenal mucosa of the chicken was demonstrated by means of in situ hybridization histochemistry (ISHH) using sense and antisense digoxigenin (DIG)-labeled RNA probes. The results showed that there were many labeled cells distributing in the duodenal mucosa of the adult chicken. Of these labeled cells, 81.60${\pm}$9.63% of Cdc25B mRNA positive cells was distributed in the basilar part and mid-portion of the intestinal gland and 36.21${\pm}$8.81% in the middle and basilar portion of villi of the small intestine of the chicken, respectively. Most of these labeled cells were positive in the regions of the stem cell and proliferation. The signals of ISHH decreased from basilar to upper part in the crypt of Lieberkuhn and weakened in the inferior villi of the duodenum. Moreover, the positive signals were both in the cytoplasm and cell nucleus. However, the labeled cells were negative in both the lamina muscularis mucosae and muscular layer. The results of ISHH suggested the existence of Cdc25B mRNA and vigorous proliferation activities in the duodenal mucosa of adult chicken, replenishing the cells which had sloughed off from the superior part of the villus. Our results provide some molecular evidence for a regular pattern of avian intestinal epitheliosis and functional partition and provide an approach to further study of the locations of Cdc25B in the chicken.
Facioscapulohumeral muscular dystrophy (FSHD) is an autosomal dominant neuromuscular disorder which has been clinically shown to cause progressive weakness and result in atrophy of the facial muscles, shoulder girdle and upper arm muscles. The responsible gene for the FSHD has been located on chromosome 4q35-qter. The probes p13E-11 and pFR-1 detect DNA rearrangements associated with FSHD as under 28 kb DNA fragment in genomic southern analysis digested with EcoRI and the fragment contains 3.3 kb Kpn I tandem repeats. In this study, 4 fetuses with a family history of FSHD were analysed by genomic southern hybridization analysis with probes to determine whether they carried the deleted region. Of the 4 fetuses, three of them had mothers who were FSHD patients and the other one had a father affected with FSHD. After 10-11 weeks of gestation, we performed chorionic villi sampling and extracted DNA from uncultured and cultured tissue cells for the direct DNA analysis. The result of the southern analysis showed two fetuses having received about 15-18 kb of deleted genes from the father and the mother respectively, and found to be FSHD patients. The other two fetuses were shown to have two normal alleles from the parents and found to be normal. Two pregnancies which were determined to be normal were carried to term delivering two healthy babies.

This study was examined the effects of Daesenggi-Tang on intestinal mucosa and gastrointestinal transit time and plasma lipids in rats. Adult male rats were fed four weeks on diets containing no addition(basal diet group), 5% cellulose(cellulose group) and Daesenggi-Tang group(Daesenggi-Tang group). The results were as follows; 1. The gastrointestinal transit times was significantly decreased in Daesenggi-Tang administered group compare to basal diet and cellulose groups. Carmine red mixed with Daesenggi-Tang, as a marker, was administered through a gastric tube for stomach or intracecally by a chronically implanted catheter for colon transit. Small intestinal transit and large intestinal transit time were significantly decreased in Daesenggi-Tang administered group compare to basal diet. 3. The height of jejunal villi was developed in Daesenggi-Tang administered group compare to basal diet. The thickness of mucosa and muscle layer of colonic mucosa were significantly developed in Daesenggi-Tang administered group compare to basal diet group. 4. The numerical change of goblet cell in colonic mucosa was increased acid mucin stained alcian blue in Daesenggi-Tang administered group compare to basal diet and cellulose group. 6. HDL-cholesterol of plasma lipid was increased in Daesenggi-Tang administered group compare to basal diet and cellulose groups. Theses results suggests that Daesenggi-Tang may be used in prevention and treatment of constipation resulting in increase of fecal weight, decrease of gastrointestinal transit time, development of intestinal villi, intensify of stainability of acid mucin in colon.
연구목적: 본 연구에서는 정상 환자와 습관성 유산 질환 환자에서 유래된 융모막 조직 내에서의 면역억제유전자들의 발현 양상에 대해 알아보고자 하였다. 연구재료 및 방법: 임신 6주와 8주의 습관성 유산 질환 환자와 정상 환자로부터 융모막 조직을 채취하였다 (Normal N=6; RSA N=6). 면역조직화학적 분석을 통해서 조직을 관찰하고 세포가 살아있음을 확인한 후에, 역전사 중합효소연쇄반응을 통해서 면역억제유전자인 placenta protein 14 (PP14), indoleamine 2, 3-dioxygenase (IDO) 그리고 mucin1 (MUC1) 유전자의 발현 정도를 비교하였다. GAPDH 발현에 기준한 면역억제유전자 발현을 정량 분석하여 Student's t-test를 시행하였고, p<0.05를 유의성이 있는 것으로 판정하였다. 결 과: 습관성 유산 질환 환자의 경우, 임신 6주와 8주의 융모막 조직에서의 면역억제유전자 (PP14, IDO, MUC1) 발현이 현저하게 낮은 양상을 보이고 있었다. 습관성 유산과 면역억제유전자의 발현이 통계학적으로 유의성 있는 연관성을 가지고 있다는 것이 확인되었다. 결 론: 면역억제유전자 (PP14, IDO, MUC1)의 발현이 습관성 유산 질환 환자에서 특이적으로 낮게 나타나는 것으로 보아 습관성 유산 질환의 진단과 치료 연구 방안에 이 유전자들의 발현이 이용될 수 있을 것으로 사료된다.
A 27-year-old pregnant woman who had one son with mental and growh retardation and dysmorphic features, was referred for genetic counselling. Cytogenetic investigations revealed 4/22 translocation in the mother(46, XX, t(4;22)(p14;P11)), partial trisomy 4p in son(46, XY, -22, +der(22), t(4;22)(p14;p11)mat). The father had normal karyotype. Amniocentesis and chorionic villi sampling were performed in 3 successive pregnancies. The karyotypes of fetus in 3rd, 4th pregnancies by amniocentesis were 46, XX, t(4;22)(p14;p11) and 46, XX, t(4;22) (p14;p11), and the karyotype of fetus in 5th pregnancy by chorionic villi sampling was found to be 46, XX, -22, +der(22) t(4;22)(p14;p11)mat. We report 3 succesive prenatally diagnosed familial partial trisomy 4p and 4/22 translocation of maternal origin with review of literature.
Eosinophils play an essential role in allergy reaction after parasite infection. To examine the immune reaction induced by eosinophils, we investigated the allergy reaction in BALB/c mice infected with Echinostoma hortense's metacercariae, as well as the effect of ketotifen, an anti-allergy drug, on eosinophil immune reaction in the villi of host intestine. The worm recovery rate was higher in ketotifen-treated mice than in untreated mice and the worms in ketotifen-treated mice survived longer than those in untreated mice. The antibody titer in the serum of ketotifen-treated mice was very low. Especially, Echinostoma hortense infection strongly increased serum IgE level and eosinophil infiltration into the villi of the mouse intestine. Ketotifen treatment suppressed eosinophil infiltration into the infected areas and inhibited IL-4 production. The reduced IL-4 production may be related with the reduction of IgE, IgG1 and IgG2 production. In conclusion, ketotifen inhibited eosinophil infiltration functioning in the allergy reaction induced by parasite infection and the expression of immunoglobulins and cytokines.
This Study was conducted to assess the single dose toxicity of DA-125, a new anthracycline anti-cancer agent, in rats and mice. The Drug was administered once intravenously to both sexes of rats and mice. Then followed a 14-day period of observation. The $LD_{50}$ Values (95% confidence limit) were estimated to be 60.9 mg/kg (57.5~64.3 mg/kg) for male rats and 60.2 mg/kg (56.2~64.5 mg/kg) for female rats, and 85.8 mg/kg (81.0~90.9 mg/kg) for male mice and 84.5 mg/kg (78.2~91.9 mg/kg) for female mice. Both sexes of rats and mice given the drug revealed the clinical sign of decreased locomotor activity, emaciation, hair loss, red-dish brown urine, salivation, and watery diarrhea. In addition, body weight from the next day to the 7th day tended to be decreased slightly in rats and mice treated with DA-125. Death occurred from the next day after administration to the 12th day. Macroscopically, congestion of gastrointestinal organ, lung, and adrenal glands were found in both sexes on the dead rats and mice. Histopathological examination of dead rats manifested atrophy of spleen, hypoplasia of bone marrow, hypcplasia and necrosis of lymphocyte in thymus, atrophy of villi in small intestine (duodenum, jejunum, and ileum), hyperplasia of granular epithelium in small intestine, degeneration of germinal epithelium in testis, defer oration of tubular epithelium in kidney, and vacuolation and myolysis of myocardium in heart. Histopathological examination of dead mice revealed hypoplasia of spleen and mesenteric lymph node, local necrosis of liver, atrophy of villi in small intestine, hyperplasia of glandular epithelium in small and large intestine, degeneration of tubular in kidney, degeneration of germinal cells in testis, and slight vacuolar degeneration of myocardium in heart.

Constipation is a common clinical problem that comprises of symptoms that include excessive straining, hard feces, feeling of incomplete evacuation and infrequent defecation. Although many conditions, such as metabolic problems, fiber deficiency, anorectal problem, an drug, can cause constipation. This study was examined the effects of Chilsun-Whan on intestinal mucosa and gastrointestinal transit time and plasma lipids in rats. Adult male rats were fed for weeks on diets containing no addition(basal diet group), 5% cellulose(cellulose group) and 2.5% . Chilsun-Whan group(Chilsun-Whan group). The results were as follows; 1. The fecal weght was significantly increased 2 times in Chilsun-Whan administrated group compare to basal diet group. 2. The gastrointestinal transit times was significantly decreased in Chilsun-Whan administrated group compare to basal diet. 3. To evaluate the antitumor activity, antimetastatic and radioprotective effects of Kamisasammaekmundongtang(KSMT), studies were done experimentally. The results were obtained as follows: 1. In cytotoxicity against P388, A549 and B16-F10, KSMT was not showed satisfiable cytotoxicity as compared with control. 2. In Inhibitory effect on activity of DNA topoisomerase I, KSMT has strong inhibitory effect. 3. The inhibitory effect on adhesion of A549 to complex extracellular matrix was significantly increased at 0.5mg/ml, 1mg/ml of KSMT. 4. The T/C% was 122 in KSMT treated group in S-180 bearing ICR mice. 5. In antiangiogenetic effect on CAM assay, inhibitory rate was 33% in KSMT treated group. 6. In pulmonary colonization assay, a number of colonies in the lungs were decreased significantly in KSMT treated group as compared with control group. 7. By FACS analysis of splenic leukocyte after exposure to radiation by linear accelerator, T-helper cell, B cell and macrophage in KSMT treated group were significantly increased while splenocytes were decreased in control group. 8. In histological changes of jejunum of $Bald{\setminus}C$ mice after exposure to radiation by linear accelerator, exclusion and fusion of villi were decreased as compared with control group. But in duodenum and ileum, exclusion and fusion of villi were not decreased as compared with control group. 9. WBC, PLT were increased in KSMT treated group as compared with control group after exposure to radiation by linear accelerator, but the increasing effect was not significant. Above results suggest that KSMT may be useful in prevention of cancer metastasis and protection from damage by radiotherapy. But the further study of KSMT would be demanded.
Sprague-Dawley rats (n=32) were fed a diet containing basal (control), cellulose (5%), or alcohol insoluble residue (AIR) (5%) extracted from the stem and root barks of elm (Ulmus davidiana var. japonica Nakai) for 4 weeks. The effects of the diets, on gastrointestinal functions and morphology were evaluated. The weight gains, food intake, and food efficiencies for the cellulose and AIR diet-fed groups were not significantly different from those of the AIR-free (basal) diet. The gastrointestinal transit times of the stem and root bark AIR diets were significantly reduced (p<0.01) compared to the basal diet, and were slower than those of the cellulose diet. The fecal weights of the stem and root bark AIR diets were significantly increased (p<0.01) up to 4-fold compared to those of the basal diet. The height of the mucosal villi, and mucosal and muscle layer thicknesses of the colon were greater and more developed in the stem and root bark AIR diets (p<0.01) than in the basal diet. The villus heights in the jejunum and the colon mucosal goblet cells were more developed in the order of cellulose > stem bark AIR > root bark AIR diets.
