소라에서 분리한 Vibrio균으로 제조한 다시마 Single Cell Detritus(SCD)의 품질특성 (Quality Characteristics of Sea Tangle Single Cell Detritus (SCD) Manufactured by Vibrio sp. Isolated from Batillus cornutus)

  • 해조류를 식품으로 이용하기에는 분말화가 필수적이나 분쇄 등의 분말화 과정에서의 온도상승은 이들 생리활성성분의 열화를 가져올 수 있으며, 수십$\sim$수백 ${\mu}m$의 분말입자는 입자 상호간의 응집으로 인하여 유동성이 좋지 않다. 해조류의 기계적 분쇄의 단점을 해결하기 위하여, 다시마(Laminaria japonica)의 분해능이 강한 Vibrio sp.를 소라(Batillus cornutus)에서 찾아 다시마 SCD(Single Cell Detritus)를 제조하였으며 SCD가 가지는 여러 가지 성분 및 생리활성을 분석하였다. 다시마 SCD의 조단백 함량과 총아미노산의 함량은 다시마분말보다 높았으나 ash의 함량은 낮았다. 다시마 SCD와 분말시료의 항암활성은 각각 31.20과 29.27%로 차이가 없었다(p<0.05). ACE저해효과는 분말시료가 39.31%로 SCD의 26.07%보다 약간 높았다. 항혈액응고 측정 결과 다시마분말은 55.3초로 다시마 SCD의 34.5초보다 높았다. 또한 다시마 SCD와 분말은 항산화효과와 뇌허혈에 의한 신경세포사 억제효과는 없는 것으로 나타났다. 비록 다시마 SCD의 생리활성은 분말보다 약간 떨어지나 조단백질, 아미노산 및 지질의 함량이 높기 때문에 빵, 국수 및 수산 연제품 등의 첨가제로서 응용할 수 있는 큰 장점이 있다고 판단된다.

  • 대한민국 제주도 연안 해수로부터 agarase를 생산하는 균주 S4를 분리하였다. 균주 S4는 그람-음성의 막대형 세포로 부드러운 베이지색 원형 콜로니를 형성하며, 한 개의 극성 편모를 갖는다. S4 균주는 $15-42^{\circ}C$, 0.5-5%(w/v) NaCl, pH 6.0-9.0, 0.5-5%(w/v) NaCl 농도에서 안정된 성장을 보인다. S4 균주의 G+C content는 49.93 mol%, 세포내 주요 지방산 (>15%)은 $C_{18:1}{\omega}7c$, $C_{16:0}$, Summed feature 3(comprising $C_{16:1}{\omega}7c/iso-C_{15:0}$ 2-OH)이다. 16S rRNA 염기서열, 생화학적 및 분류학정 특징에 기초하여 S4 균주를 Vibrio sp. S4로 명명하였다. 0.1% agar를 첨가한 액체배지에서 S4 균주는 72시간에 세포농도와 agarase 활성이 최대치를 보였다. 반면, agar 를 첨가하지 않은 배양액에서의 agarase 활성은 무시할만한 수준이었으며, 이는 균주의 agarase 유전자 발현이 agar에 의해 유도됨을 시사하고 있다. 균주 S4가 세포외부로 분비하는 총 agarase는 $45^{\circ}C$와 pH 7.0에서 최상의 효소 활성을 보였으며, agarose를 분해하여 (neo)agarotetraose와 (neo)agarohexaose를 생산하였다.

Bioactive Cyclic Dipeptides from a Marine Sponge-Associated Bacterium, Psychrobacter sp.

  • A bacterial strain with good antibacterial activities against Staphylococus aureus and Escherichia coli was isolated from a marine sponge Stelleta sp., and it was identified as a Psychrobacter sp. by comparative 16S rDNA sequence analysis. In our search for bioactive secondary metabolites from this psychrophillic and halotolerent bacterium, sixteen cyclic dipeptides (1-16) were isolated and their structures were identified on the basis of NMR analysis. In the test of the compounds for the protective effect against Vibrio vulnificusinduced cytotoxicity in human intestinal epithelial cells, cyclo-(L-Pro-L-Phe) (5) exhibited significant protective effect. Compounds 2, 6, and 11, which contain D-amino acid, were first isolated from bacteria.

넙치 자어, 로티퍼와 알테미아의 세균총에 관한 분석 (Studies of Bacterial Flora of Rotifer sp., Artemia sp. and Olive Flounder larvae, Paralichthys olivaceus)

  • The purpose of this research was to investigate the bacterial flora of healthy olive flounder larvae (Paralichthys olivaceus) and live feeds (Rotifer spp. and Artemia spp.). The total bacteria counts were $9.2{\times}10^7$ and $1.2{\times}10^{10}cfu/g$ and Vibrio sp.(82.8%) was dominant in rotifers. The total bacteria counts were $3.8{\times}10^6$ and $9.2{\times}10^6cfu/g$ and Vibrio sp.(73.3%) was dominant in artemia. In olive flounder larvae, the total bacteria counts were $1.4{\times}10^6{\sim}8.3{\times}10^7cfu/g$ and V. harveyi (38.5%) was dominant. It might be potential marker of disease outbreak in olive flounder larvae.

Assessment of the ozonation against pathogenic bacteria in the effluent of the quarantine station

  • This study investigated how ozone treatment can successfully inactivate pathogenic bacteria in both artificial seawater and effluents discharged from the fishery quarantine station in Pyeongtaek Port, Korea. Vibrio sp. and Streptococcus sp. were initially inoculated into the artificial seawater. All microbes were almost completely inactivated within 10 min and 30 min by injecting 6.4 mg/min and 2.0 mg/min of ozone, respectively. It was discovered that the water storing Pleuronichthys, Pelteobagrus, and Cyprinus imported from China contained the indicator bacteria, Vibrio sp., Enterococcus sp., total coliforms, and heterotrophic microorganisms. Compared to the control, three indicator bacteria were detected at two to six times higher concentrations. The water samples displayed a diverse microbial community, comprising the following four phyla: Bacteroidetes, Proteobacteria, Firmicutes, and Actinobacteria. Almost all indicator bacteria were inactivated in 5 min at 2.0 mg/min of ozonation; comparatively, 92.9%-98.2% of the less heterotrophic microorganisms were deactivated within the same time period. By increasing the dosage to 6.4 mg/min, 100% deactivation was achieved after 10 min. Despite the almost complete inactivation of most indicator bacteria at high doses after 10 min, several bacterial strains belonging to the Proteobacteria have still been found to be resistant under the given operational conditions.

  • Monoclonal antibodies were prepared in order to an assay method for Vibrio vulnificus. Sixteen mouse ybridoma cell lines were established by immunization of whole cell antigen to BALB/c mice, fusion with SP2/O myeloma cells, and cloning. Most of them secreted IgM.lambda. antibodies. A sandwich enzyme-linked immunosorbent assay was developed with rabbit anti-V. vulnificus polyclonal antibodies as capture antibody, an IgM monoclonal antibody as detector antibody, and goat anti-mouse IgM-alkaline phosphatase conjugate as developer antibody. The range of detection was 10$\^$4/ to 10 V. vulnificus cells per microplate well. When four related Vibrio species were tested for cross-reactions, V. parahaemolyticus showed 3.5% reactively and V. carchariae, V. fluvialis, and V. furnisii showed negligibal (<1%) cross-reactivity.

Purification and Characterization of Chitinase from a Marine Bacterium, Vibrio sp. 98CJ11027

  • Chitin-degrading marine bacterial strain 98CJ11027 was isolated from bryozoa from the coastal area of Cheju Island, Korea, and identified as a member of the genus Vibrio. The molecular mass of the main extracellular chitinase (chitinase I), purified from strain 98CJ11027, was estimated to be 98 kDa. The optimal condition for chitinase I activity is pH 6.0 and 45℃. The activity was inhibited by $Fe^+2$ and$Cu^+2$. Chitinase I displayed the hydrolysis type of chitobiosidase and catalyzed reversed hydrolysis leading to the synthesis of tetraacetylchitotetraose.
