Characterization of Vibrio harveyi, the Causal Agent of Vibriosis in Cultured Marine Fishes in Korea

  • An epizootic causing mortality among cultured marine finfishes occurred in 1999 in the province of Kyoungsang, Korea. The disease was characterized by the presence of enterocele, abdominal swelling, and gastroenteritis. The causative bacteria were isolated from olive flounder (Paralichthys olivaceus), black rockfish (Sebastes schlegeli), turbot (Scophthalmus maximus) and the rearing water. These bacteria showed swarming activity on agar plates and yellowish or greenish colonies on thiosulfate-citrate-bile salts-sucrose (TCBS) agar plates, but no luminescence. The pathogen was identified as Vibrio harveyi based on morphological and biochemical characteristics and the sequence of l6S rDNA. The lethal doses (LD$_{50}$) of olive flounder and black rockfish were estimated to be $1.24\times10^6-1.36\times10^8$ and $3.24\times10^5-5.8\times10^7$ CFU/fish respectively following intraperitoneal injection.

A detection method for vibrio vulnificus using monoclonal antibodies

  • Monoclonal antibodies were prepared in order to an assay method for Vibrio vulnificus. Sixteen mouse ybridoma cell lines were established by immunization of whole cell antigen to BALB/c mice, fusion with SP2/O myeloma cells, and cloning. Most of them secreted IgM.lambda. antibodies. A sandwich enzyme-linked immunosorbent assay was developed with rabbit anti-V. vulnificus polyclonal antibodies as capture antibody, an IgM monoclonal antibody as detector antibody, and goat anti-mouse IgM-alkaline phosphatase conjugate as developer antibody. The range of detection was 10$\^$4/ to 10 V. vulnificus cells per microplate well. When four related Vibrio species were tested for cross-reactions, V. parahaemolyticus showed 3.5% reactively and V. carchariae, V. fluvialis, and V. furnisii showed negligibal (<1%) cross-reactivity.

조피볼락에 대한 Vibrio harveyi의 병원성 (Pathogenicity of Vibrio harveyi to black rockfish, Sebastes schlegeli)

  • 본 연구는 V. harveyi의 luminescence 유무와 해수 어류에 대한 병원성의 관계를 알아보고자 non-luminous V. harveyi 와 luminous V. harveyi 분리 균주로 조피볼락에 대한 병원성 시험을 시행하였다. 시험 균주 V. harveyi 모두 haemolytic activity를 나타내었으며, 어체 내 생존능은 non-luminous V. harveyi 분리 균주 FR 2가 강한 생존능을 나타내었다. 또한 조피볼락에 인위 감염시, non-luminous V. harveyi 분리 균주 FR 2가 강한 병원성을 나타내었으며 luminous V. harveyi 분리 균주 VIB 391은 낮은 병원성을 나타내었다. 결론적으로 조피볼락에서는 non-luminous V. harveyi 분리 균주 FR 2가 강한 병원성을 나타내었으며 luminous V. harveyi 분리 균주 VIB 391은 매우 약한 병원성을 나타내어 luminescence의 유무에 따라 해수 어종에 대한 병원성의 차이가 있을 것으로 생각된다
