Outbreaks of foodborne diseases associated with Vibrio species such as V. parahaemolyticus, V. vulnificus, and V. cholerae frequently occur in countries having a dietary habit of raw seafood consumption. For rapid identification of different Vibrio species involved in foodborne diseases, whole-cell protein pattern analysis for 13 type strains of 12 Vibrio species was performed using SDS-PAGE analysis. Pathogenic Vibrio species such as V. parahaemolyticus, V. vulnificus, V. cholerae, V. alginolyticus, V. fluvialis, and V. mimicus were included in the 12 Vibrio species used in this study. Each of the 12 Vibrio species showed clearly specific band patterns of its own. Two different strains of V. parahaemolyticus showed two different SDS-PAGE whole-cell protein patterns, giving the possibility of categorizing isolated strains in the same V. parahaemolyticus species into two subgroups. The 36 Vibrio isolates collected from sushi restaurants in Busan were all identified as V. parahaemolyticus by comparing their protein patterns with those of Vibrio type strains. The identified isolates were categorized into two different subgroups of V. parahaemolyticus. The whole-cell protein pattern analysis by SDS-PAGE can be used as a specific, rapid, and simple identification method for Vibrio spp. involved in foodborne diseases at the subspecies level.
Thirty-eight Vibrio spp. were isolated from the sea waters harvested from the 22 stations located on the west coast of the Korean peninsula in September 2006. The isolates consisted of V. parahaemolyticus (n=21), V. alginolyticus (n= 16) and V. cholerae non-01 (n=1), among which 35 isolates displayed resistance against two of the tested antibiotics. Among the 38 isolates, 18 isolates exhibited multi-drug resistance against more than four 4 antibiotics. In particular, minimum inhibitory concentration $(MIC)_{50}$ and $MIC_{90}$ of ampicillin-resistant isolates were as high as $2,048{\mu}g{\cdot}mL^{-1}$ and $4,096{\mu}g{\cdot}mL^{-1}$ respectively. $\beta$-lactamase production was examined to analyze the ampicillin-resistance. Some Vibrio spp. isolates produced $\beta$-lactamase, however antibiotics resistance pattern and $\beta$-lactamase production were not clearly related to each other. A genetic relationship between resistance and gene expression was confirmed in the ampicillin-resistant isolates.

The enterobacterial repetitive intergenic consensus (ERIC)-PCR is a recently described DNA fingerprinting technique based on amplification of repetitive element distributed in bacteria. We applied of ERIC-PCR to clinical isolates of Vibrio parahaemolyticus and other bacteria associated diarrhea. Twenty isolates of V. parahaemolyticus were used for intragenic genotyping, which were isolated from 2001 to 2002 in Chungbuk National University hospital. For interspecies genotyping, V. vulnificus, V. alginolyticus, V. parahaemolyticus, Escherichia coli, Salmonella and Shigella spp. were used. The genotyping were analyzed by ERIC-PCR. The genotyping of V. parahaemolyticus were grouped two major pattern (A, B) and were subdivided into 10 subtypes (A1, A2, B1-B8) by ERIC-PCR. These method distinctly differentiated bacterial species associated diarrhea. Those results show that ERIC-PCR can be reliable and efficient method for genotyping of V. parahaemolyticus and bacteria associated diarrhea.

The rotation of the flagellar motor is powered by the electrochemical gradient of specific ions across the cytoplasmic membrane. Recently, the gents of the Na'-driven motor have been cloned from marine bacterium of Vibrio sp. and some of the motor proteins have been purified and characterized. Also, motx gene encoding a channel component of the sodium type flagellar motor was identified from Vibrio Huuiaiis (KTCC 2473). The amino acid sequence of MotX protein from V, Huvialis shared 90, 85, $85\%$ identity with V, cholerae, V. alginolyticus, V parahaemolyticus, respectively. We have studied the localization of the expressed MotX protein in Escherichia coli by immune-gold labeling of ultra-thin frozen section. Our observation of the expressed protein indicated that MotX protein could be existed as attachment to inner membrane in E. coli.
To determine the physiological, biochemical characteristics and toxicity of hemolysin produced by a novel sucrose positive Vibrio (Vibrio sp. D5) isolated from estuary of Kum river, it was compared with already known sucrose positive Vibrio. Salinity, pH, temperature and conductivity of place where Vibrio sp. D5 was isolated were $4.7\%_{\circ},\;7.6,\;24^{\circ}C$ and $7800{\mu}MHOS$, respectively. Physiological and biochemical characteristics distingiushed Vibrio sp. D5 from other sucrose positive Vibrio: V. alginoipicus, V. cholerae, V. cincinnatiensis, V. fluvialis, V. furnissii and V. metschnikovii. The range of salinity and pH for growth of Vibrio sp. D5 were $0.5\%\~7.5\%$ and $4.5\~9.5$, respectively. Vibrio sp. D5 exhibited typical yellow colony on TCBS agar plate and curved rod type upon transmission electron microscopy (TEM). Vibrio sp. D5 had lethal toxicity against mouse in case of intraperitoneal injection with its culture and showed hemolysin activity on human blood agar and sheep blood agar. Ubrio sp. D5 also demonstrated vascular permeability activity toward rat. From the above results, Vibrio sp. D5 was ascertained to be a novel pathogenic Vibrio.
Fifty four strains of pathogenic vibrios were isolated from the Gwangan Beach from May to October, 2002. The isolated vibrios were composed of 7 different species: Vibrio parahaemolyticus, V. cholerae non-O1, V. alginolyticus, V. vulnificus, V. hollisae, V. fluvialis, ane V. mimicus. In the detection rate, V. parahaemolyticus was most predominant as $46\%$(25/54). From the isolated strains, only 25 strains have hemolytic activity or 25 strains only proteolytic activity on agar plates. Eleven strains showed both hemolytic and proteolytic activity. No strains showed urease activity. All strains of V parahaemolyticus did not show hemolytic activity, while V. cholerae non-O1 strains showed $\beta$ hemolytic activity. Kanagawa phenomena of pathogenic vibrios did not accord with hemolytic activity of the culture supernatant at the late log phase. Some strains showed high hemolytic activity despite having proteolytic activity, but some weak hemolytic activities despite having no proteolytic activity.

A novel bacteriophage, designated as VPP97, that infects the strains of Vibiro parahaemolyticus (hallophilic, Gram-negative bacterium) isolated most commonly from marine environments, has been discovered, and several of its properties have been determined. The plaques were clear and sized $0.6{\sim}1.0\;mm$ in diameter. The virion forms a single band on 70% sucrose gradient and ${\rho}1.50$ CsCl gradient by sucrose gradient centrifugation and CsCl gradient centrifugation respectively. It has a hexagonal head and a relatively long tail, as shown by electron microscopy. Vibrio alginolyticus, Vibrio fluvialis and Vibrio furnissii were also sensitive to this phage. It was almost totally inactivated at $70^{\circ}C$ and at pH below 5 or over 10. The nucleic acid of VPP97 is composed of DNA. The VPP97 had 9 specific structural proteins sized between 21.5 kDa and 97.4 kDa on SDS-PAGE. When V. parahaemolyticus cultures were treated with either phage VPP97 or one of the several antibiotics for 2 hours, the viable number of V. parahaemolyticus treated with the phage VPP97 is lower than that treated with chloramphenicol, erythromycin or penicillin, but not lower than that treated with tetracycline. Mice that have responded to the phage treatment revealed the lower numbers of V. parahaemolyticus in small intestine and less damage on small intestine compared to the untreated mice. Therefore, we suggest that the phage treatment appears effective to the infection by V. parahaemolyticus.

In this study, the initial number of bacteria before ozone sterilizing shows 290 per $1m{\ell}$ in RUN 1 and RUN 2 equally, but the removal rate shows more than 50% in RUN 1, and 100% in RUN 2 respectively when ozone injection amount is $0.28mg/{\ell}$. It is regarded as a satisfactory result that E-coli concentration without ozone contact is 890, rapid removal effect of E-coli is observed in $0.28mg/{\ell}$ of ozone, and E-coli is removed perfectly in $0.84mg/{\ell}$ of ozone. It is thought that an excellent efficiency is obtained for vibrio alginolyticus because the initial number of bacteria before ozone contact is positive, but it is altered to negative after ozone contact. CODcr shows the tendency which is somewhat reduced as the ozone injection is increased, but the general effect is appeared not so much, and it is thought that the tendency is caused by the reason that sea water contains much salt which is estimated as a component of CODcr, therefore it is regarded that ozone contact has not an important effect on salinity. It is thought that the frequency of changing salt water in the fish preserve of a sliced raw fish restaurant can be reduced to under the standard because NTU of 7 days after sea water injection is 0.70 in the experiment of turbidity, hut more than 50% of turbidity removal efficiency is appeared at $0.28mg/{\ell}$ of ozone injection, and it shows 70% at $0.84mg/{\ell}$ of ozone injection in RUN 1 and RUN 2 commonly.
A bacterial strain inhibiting the growth of Vibrio anguillarum, the causative agent of vibriosis, was isolated from fish intestines. The isolated strain HS36 was identified as Aerococcus urinaeequi based on the characteristics of the genus according to Bergey's Manual of Systematic Bacteriology and by 16S rRNA sequencing. The growth rate and antibacterial activity of strain HS36 in shaking culture were higher than those in static culture, while the optimal pH and temperature for antibacterial activity were 7.0 and 30℃, respectively. The active antibacterial substance was purified from a culture broth of A. urinaeequi HS36 by Sephadex G-75 gel chromatography, Sephadex G-25 gel chromatography, and reverse-phase high-performance liquid chromatography. Its molecular weight, as estimated by Tricine SDS-polyacrylamide gel electrophoresis, was approximately 1,000 Da. The antibacterial substance produced by strain HS36 was stable after incubation for 1 h at 100℃. Although its antibacterial activity was optimal at pH 6-8, activity was retained at a pH range from 2 to 11. The purified antibacterial substance was inactivated by proteinase K, papain, and β-amylase treatment. The newly purified antibacterial substance, classified as a class II bacteriocin, inhibited the growth of Klebsiella pneumoniae, Salmonella enterica, and Vibrio alginolyticus.
The monthly distribution of marine bacteria in the coastal waters of Chung-Mu was investigated from April, 1976 to March, 1977. The aim of this study was to obtain basic data for the prevention of food poisoning and for the efficient aquaculture of the area. Samples of sea water, mud, fish and shell fish were taken every month. The results are as follows: 1. One thousand four hundred and twenty-six strains were isolated from 732 samples of sea water, mud, fish and shell fish. They were 450 strains of Pseudomonas fluorescens, 422 strains of Achromobacter liquefacience, 72 strains of Vibrio parahaemolyticus, 234 strain of Vibrio alginolyticus, and 248 strains of Proteus vulgaris. 2. V. parahaemolyticus occupied $9.84\%$ of the total samples and $52\%$ of them were found in the sea water and mud. 3. The muds sampled districtly beneath the aquaculture raft contained much gas. They seemed to originate from the deposition of excretion of shell fish. The fatness of the shell fish was low where the gas was abundant. 4. It was .found that the shell fish with low fatness contained much Vibrio sp. 5. Regional distribution shows that marine bacteria were abundant in the order of station 10, 9, 8, 11, 1. The area around station 9 was polluted by reclamation of the area, and station 8 showed the influence of the excrement treat tank located nearly.
