Identification of Verticillium dahliae and V. albo-atrum Causing Wilt of Tomato in Korea

  • In 1997, 103 isolates of Verticillium were obtained from roots and stems of tomato plants showing wilt symptoms in greenhouses in eight areas of Korea. Out of these isolates, 75 were edintified as V. dahliae while 28 were identified as V. albo-atrum based on their morpho-logical and cultural characteristics. Both Verticillium species produced colonies with conidiophores, which were more or less erect, hyaline, with verticillate branches, and with 3-4 phialides at each node. V. dahliae produced microsclerotia, while V. albo-atrum produced resting dark mycelium. Optimum temperatures for mycelial growth of V. dahliae and V. albo-atrum on PDA were 22 and $26^{\circ}$, respectively. Mycelial growth of V. albo-atrum was slower than that of V. dahliae. Pathoge-nicity tests revealed that tomato cvs. Zuikoh No. 102, Kyoryokubeiju No. 2, Zuiken, Kagimuza, and Momotaro were susceptible to V. albo-atrum, while cvs. Zuikoh No. 102 and Kyoryokubeiju No.2 were susceptible to V. dahliae.

Verticillium Wilt of Potato Caused by Verticillium albo-atrum in Daegwallyong Area in Korea

  • Verticillium wilt was first observed in 2001 on potatoes (Solanum tuberosum) cv. Superior at Daegwallyong area, one of the major seed potato producing areas in Korea. The wilted potato plants showed typical symptoms including gradual yellowing and interveinal necrosis. There was discoloration in the vascular tissues of the infected stems which turned light brown. Fungal isolates from discolored vascular tissues were whitish to creamy with folding on potato dextrose agar medium, where they used to produce resting dark mycelia but no micro-sclerotia. Conidiophores were septate with side branches, swelled at the base, and arranged in a whorl. Conidia were 2.5-11.2$\times$2.0-4.5 $\mu\textrm{m}$ um in size and were borne in small clusters at the tips of phialides. Optimal temperature range for mycelial growth was $25-30^{\circ}C$. Based on these cultural and morphological characteristics, the fungus was identified as Verticillium albo-atrum Reink & Berth. Pathogenicity tests by root dipping method revealed that the fungus caused the same symptoms as observed in naturally infected potato plants. This is the first report of Verticillium wilt on potato caused by Verticillium albo-atrum in Korea.

일본산(日本産) 수입(輸入) 채소(菜蔬) 종자(種子)에서 검출(檢出)된 진균(眞菌) (Fungi Detected in the Seeds of Vegetable Crops Imported from Japan)

  • Sixteen seed samples of nine vegetable crops imported from Japan were tested for plant quarantine by routine methods of seed health testing. Nineteen species of fungi including Alternara alternata were identified from the seed samples. Some species were very frequently detected in the seeds. In this experiment, Verticillium albo-atrum, V. dahliae and Macrophomina sp. from the seeds of spinach, and Cercospora capsici, Drechslera hawaiiensis and V. albo-atrum from the seeds of sweet pepper were detected for the first time, respectively. It has been reported that they had a pathogenicity on some vegetable crops in Japan and other countries. The data suggest that an epidemic damage may be occurred if the infected seeds are sown and cultivated in Korea.

Ribosomal DNA의 Internal Transcribed Spacer(ITS) 부위의 염기서열 분석에 의한 Gliocladium 속과 근연속에 관한 계통 분류학적 연구 (Phylogenetic Analysis of the Genus Gliocladium and its Related Taxa by Comparing the Sequences of Internal Transcribed Spacers and 5.8S r-DNA)

  • The phylogenetic position of Gliocladium and its related taxa were investigated, using the neighbor-joining method of the sequences from internal transcribed spacers (ITS1 and ITS2) and 5.8S ribosomal DNA (rDNA). It was focused especially on the generic concept by comparing with the related genera such as Trichoderma, Hypocrea, Verticillium, Penicillium and Talaromyces. Gliocladium species and its related genus were divided into three groups by the phylogenetic analysis using the neighbor-joining method. The first group includes Penicillium-like strains such as Penicillium, Tararomyces, Verticillium and one species of Gliocladium (G. cibotii JCM 9203 and JCM 9206). Especially, Gliocladium cibotii JCM 9203 is thought to be the similar species with Verticillium bulbillosum JCM 9214. Between these two species, Gliocladium cibotii and Verticillium bulbillosum, the intraspecies concept needs to examined with culture condition. and morphological properties. The second group includes two species Verticillium, Verticillium tricorpus and Verticillium albo-atrum which extracted from the GenBank database in NCBI (National Center for Biotechnology Information). Trichoderma-like strains, such as Trichoderma, Hypocrea and several species of Gliocladium are included in the third group. Also, Gliocladium penicillioides IFO 5869 and Gliocladium catenulatum ATCC 10523 formed the subgroup of Trichoderma-like strains. The species of Gliocladium were dispersed in Trichoderma-like and Penicillinum-like group, and only one species of Gliocladium cihotii used in our study was located in Penicillium-like genus group. The species of Verticillium appeared in all three groups and the species of Trichoderma formed the monophylogeny with Hypocrea (telemorph). Also, Gliocladium virens was grouped with Trichoderma harzianum with a high bootstrap value, supporting that Gliocladium virens is to be placed in Trichoderma. The results suggest that Gliocladium is polyphyletic, and is more Trichoderma-like than Penicillium-like.

광 반사방식을 이용한 감염 씨감자 비파괴 선별 기술 개발 (Development of non-destructive measurement method for discriminating disease-infected seed potato using visible/near-Infrared reflectance technique)

  • Pathogenic fungi and bacteria such as Pectobacterium atrosepticum, Clavibacter michiganensis subsp. sepedonicus, Verticillium albo-atrum, and Rhizoctonia solani were the major microorganism which causes diseases in seed potato during postharvest process. Current detection method for disease-infected seed potato relies on human inspection, which is subjective, inaccurate and labor-intensive method. In this study, a reflectance spectroscopy was used to classify sound and disease-infected seed potatoes with the spectral range from 400 to 1100 nm. Partial least square discriminant analysis (PLS-DA) with various preprocessing methods was used to investigate the feasibility of classification between sound and disease-infected seed potatoes. The classification accuracy was above 97 % for discriminating disease seed potatoes from sound ones. The results show that Vis/NIR reflectance method has good potential for non-destructive sorting for disease-infected seed potatoes.
