Adaptation success of Zostera marina to a new transplant environment

  • Marked declines in seagrass meadows are increasingly being reported from coasts around Korea and many regions of the world. The transplantation of seagrasses has been attempted to mitigate and control their degradation from a range of anthropogenic factors. In this study, Zostera marina shoots, which were collected from a donor bed in Koje Bay, were transplanted in Jindong Bay in December 2004. In 2008, a comparative investigation of shoot morphology, growth, and reproduction strategy of Z. marina was carried out between the donor and transplant sites to assess their adaptation success. Shoot height, individual shoot weight, and leaf productivity at the transplant site were significantly greater than those at the donor site. However, below-/aboveground tissue ratio was significantly lower at the transplant site compared to the donor site. Z. marina survival was maintained through vegetative reproduction, while peak season for lateral shoot recruitment was late winter for both donor and transplant site populations. However, vegetative reproduction mainly occurred during late winter and spring at the transplant site, whereas lateral shoots were evident across all seasons except late spring in the donor site. More pronounced seasonal variations were found at the transplant site compared to the donor site. These results indicate that Z. marina populations at the two sites possess distinct phenotypic variations induced by different environmental conditions, and Z. marina transplants have adapted well to the new transplant environment.

인공 황산비 및 질산비가 애기장대의 생장과 종자발아에 미치는 영향 (Effects of Simulated Sulfuric and Nitric Acid Rain on Growth and Seed Germination of Arabidopsis thaliana)

  • The experiment was carried out to investigate the effects of sulfuric acid and nitric acid among the main components of simulated acid rain (SAR) on the growth of vegetative organs and seed germination of Arabidopsis thaliana. The Arabidopsis treated with SAR supplemented with sulfuric and nitric acids, respectively, showed 28% and 30% decrease of shoot and root growth compared to the control plants, and also many necrotic spots on leaf surfaces after SAR treatment were observed. The shoot and root length for plants grown with nitric acid rain was 14% and 17% lower, respectively, compared to the control, whereas those grown with sulfuric acid rain was 24% and 25% lower than control plants. When Arabidopsis seeds were sown in distilled water, germination rate was 100% after 7 days. However, 80% in SAR medium supplemented with sulfuric and nitric acids, 88% in sulfuric acid rain medium and 93% in nitric acid rain medium. The germination abilities of seeds harvested from SAR supplemented with sulfuric and nitric acids, sulfuric acid rain, and nitric acid rain were 73%, 73% and 94%, respectively. Consequently, sulfuric acids showed more inhibitory effects than nitric acids on the growth of vegetative organs as well as germination rates in Arabidopsis.

  • Plant biotechnology involving genetic modification has been rather controversial. However, the major issues related to safety are being addressed by continued improvements in technology. Some of the related facts will be highlighted to set the tone for a scientific discussion on the possibilities of using the technology for crop improvement. Our main research interest is to understand the molecular regulation of shoot bud regeneration in plant tissue culture, which is essential for crop improvement by biotechnology. We have isolated and characterized some genes that are associated with adventitious shoot regeneration. These include a MADS-box cDNA (PkMADS1) from paulownia kawakamii, which regulates vegetative shoot development and in vitro shoot regeneration from leaf explants. Another gene we have characterized from petunia codesfor a cytokinin binding protein (PETCBP). Preliminary functional analysis of this gene indicated that this also affects adventitious shoot bud initiation. Also, the antisense suppression of this gene in petunia causedexcessive branching. Results from our work and selected other publications will be used to highlight the possibilities of manipulation of such genes to improve crop species.

슬래쉬소나무의 화아원기(花芽原基) 형성(形成)의 생리학적(生理學的) 연구(硏究)(I) 당년지(當年枝)의 형태(形態)와 자화(雌花)의 착화(着花)와의 관계(關係) (Physiology of Strobilus Initiation in Slash Pine I. Ovulate Strobilus Production in Relation to Shoot Morphology)

  • 채종원(採種園)에서 18년생(年生) 접목묘(接木苗)인 슬래쉬 소나무를 대상(對象)으로 하여, 구과형성(毬果形成)과 가지의 형태학적(形態學的)인 상관관계(相關關係)를 조사했다. 춘기(春期)에 조사한 지당(枝當) 자화수(雌花數)와 상관관계가 가장 큰 것은 전년도(前年度) 신초지(新梢枝)의 지당(枝當) 엽중량(葉重量)이었다. ($R^2=+0.41$). 웅화수(雄花數)는 신초지(新梢枝)의 엽중량(葉重量)과 부(負)의 상관(相關)을 보여주었다. ($R^2=-0.21$). 웅화(雄花)에서 자화(雌花)로의 성전환(性轉換)은 그 가지의 생장력(生長力)(vigor)이 증가함에 따라서 촉진(促進)되었다. 자화(雌花)가 거의 착화(着花)되지 않는 개체목(個体木)의 토부수관(土部樹冠)에서 채취(採取)한 정아(頂芽)와, 자화(雌花)나 웅화(雄花)가 대량(大量)으로 착화(着花)한 개체(個体)의 정아(頂芽)로, 유관속 조직의 생장(生長)을 비교(比較)한 결과, 전자(前者)가 후자(後者)보다 훨씬 빨랐으며, 이는 자화(雌花)를 생산(生産)하지 않는 정아(頂芽)는 강(强)한 영양생장(營養生長)의 경향(傾向)을 나타냄을 뒷받침하는 것이다.

개화기에 저온 피해를 받은 '후지'/M.9 사과나무의 하계전정 시기가 신초생장 및 과실품질에 미치는 영향 (Influence of Summer Pruning Time on Shoot Growth and Fruit Quality of 'Fuji'/M.9 Apple Tree Damaged by the Low Air Temperature at Flowering Period)

  • BACKGROUND: The low temperature at flowering period break the balance between vegetative and reproductive growth of apple tree. Summer pruning has been used to control vegetative growth. So, this study was conducted to investigate the effect of summer pruning time on shoot growth and fruit quality of 'Fuji'/M.9 apple trees damaged by the low temperature at flowering period. METHODS AND RESULTS: The following treatments were applied to tree : a) control (no summer pruning), b) pruned 26 June, c) pruned 30 July, d) pruned 28 August, and e) pruned 26 September. The summer pruning significantly increased light penetration and fruit red color by reducing the total shoot growth compared with control. And the summer pruning control the outbreak of apple valsa canker. But the summer pruning at the end of June increased regrowth of shoot and pruning weight compared with the summer pruning at the end of August. The summer pruning at 30 July had the highest fruit weight, but return bloom was the highest in the summer pruning at 28 August. CONCLUSION(S): These results indicated the optimum summer pruning time of 'Fuji'/M.9 apple trees damaged by the low temperature at flowering period were the end of August.

동해 덕산항에 생육하는 포기거머리말(Zostera caespitosa Miki) 군집의 생물계절학과 형태 변이 (Phonology and Morphometrics Change of Zostera caespitosa Miki Populations at the Duksan Port in the Eastern Coast of Korea)

  • 동해 덕산항에 생육하는 포기거머리말(Zostera caespitosa Miki)군집의 생물계절학과 형태 변이를 1998년 3월부터 2000년 8월까지 조사하였다. 포기거머리말의 형태적인 형질(식물체 길이, 잎 길이, 엽초 길이, 잎 너비와 잎 수),생식 형질(불염포, 육수화서와 종자)의 수와 크기, 생육밀도, 생물량과 이화학적 특성 (수온, 영양염 농도)을 관측하였다. 형태적인 형질은 엽초를 제외하고 계절에 따라 유의한 차이 (p<0.05)를 나타내었다. 식물체길이는 수온에 따라 명확한 년 순환을 나타내었다. 영양지는 지속적으로 출현하였으나 생식지는 1월 중순부터 4월초까지 적은 수만이 출현하였다. 포기거머리말의 개화기는 2월 중순부터 시작되었으며, 4월초에 성숙한 종자가 출현하였다. 해수내의 질산염과 인산염의 농도는 계절에 따른 차이를 나타내었으나, 암모니아의 농도는 계절에 따라 다양하게 나타났다. 식물체의 형태적인 형질과 이화학적인 형질은 유의한 상관관계를 나타내지 않았으나, 포기거머리말 생식 단계와 생활사는 수온에 영향을 받는 것으로 추정된다. 덕산항에서 포기거머리말군집의 생육밀도와 지상부 생물량은 각각 $511.6\pm{25.6}\;shoots\;m^{-2}$$413.4\pm{19.8}\;g\;dry\;wt\;m^{-2}$로 추산되었다.

The Rice FON1 Gene Controls Vegetative and Reproductive Development by Regulating Shoot Apical Meristem Size

  • Most plant organs develop from meristems. Rice FON1, which is an ortholog of Clv1, regulates stem cell proliferation and organ initiation. The point mutations, fon1-1 and fon1-2, disrupt meristem balance, resulting in alteration of floral organ numbers and the architecture of primary rachis branches. In this study, we identified two knockout alleles, fon1-3 and fon1-4, generated by T-DNA and Tos17 insertion, respectively. Unlike the previously isolated point mutants, the null mutants have alterations not only of the reproductive organs but also of vegetative tissues, producing fewer tillers and secondary rachis branches. The mutant plants are semi-dwarfs due to delayed leaf emergence, and leaf senescence is delayed. SEM analysis showed that the shoot apical meristems of fon1-3 mutants are enlarged. These results indicate that FON1 controls vegetative as well as reproductive development by regulating meristem size.

음나무(Kalopanax pictus Nakai)의 종자번식 및 녹지삽목 번식기술 (Seed Germination and Softwood Cutting Technique of Kalopanax pictus Nakai)

  • 본 연구는 음나무의 대량증식을 위한 실용적인 실생번식과 녹지 삽목 기술을 개발하고자 실시 하였다. 실생 번식을 위한 노천매장과 1$0^{\circ}C$ 후숙처리는 1주이내에 배의 크기를 3mm이상 유도하였고 발아율은 대조구에 비해 노천매장과 1$0^{\circ}C$에서 후숙된 종자에서 70%이상 높게 나타났다. 녹지 삽목 실험에서 는 삽수의 모수령이 발근에 미치는 효과를 조사한 결과를 보면, 1년생 삽목묘 신초를 삽수로 사용한 것이 68.9%의 발근율로 가장 높고, 모수의 수령이 유령일수록 발근율이 높고, 반면 수령이 증가할수록 발근 능력이 감소하였다. 삽목 시기별 발근 실험에 있어서 1차 생장한 6월 13일 삽목한 삽수가 68.9%로 발근율이 가장 높아 6월중순이 삽목 적기였다. 식물생장조정제 종류 및 농도별 처리에서는 IBA 1000mg.L$^{-1}$ 농도에서 30분간 침적했을 때 발근율이 70%, NAA 1000mg.L$^{-1}$에서 66.7%의 순으로 2000mg.L$^{-1}$ 30초, 500mg.L$^{-1}$ 10시간 처리구 보다 발근율이 높게 나타났다.

Temperature Effects on Shoot Growth and Flowering of Kumquat Trees

  • This study investigated the effects of temperature on the shoot growth and flowering of potted kumquat [Fortunella margarita ( Lour.) Swingle] trees grown in subtropical conditions of I-Lan County in Taiwan. Temperature treatments included T 25-32, T 17-25, T 22, and T 18. The T 25-32 treatment trees were to the day/night temperatures of $25/18^{\circ}C$ for 2 weeks, followed by 28 weeks at $32/25^{\circ}C$. T 17-25 was exposed for 4 weeks to $17/10^{\circ}C$ followed by 26 weeks at $25/18^{\circ}C$. T 22 and T 18 were exposed at $22/18^{\circ}C$ and $18/13^{\circ}C$, respectively, for the entire duration of the experiment. Control trees were placed in a plastic greenhouse under conditions similar to the natural environment. The kumquat trees exposed to high-temperature environment of $32/25^{\circ}C$ showed more frequent and speedy sprouting of new buds, but induced the earlier termination of shoot elongation growth, resulting in decreased vegetative growth. The temperature treatments lower than $22^{\circ}C$ suppressed the new shoot production but increased the shoot growth period, resulting in increased shoot length and diameter. Temperatures higher than $25/18^{\circ}C$ readily induced flowering, with flowering being advanced under the higher temperature conditions such as $32/25^{\circ}C$. However, flowering was substantially inhibited under temperature conditions lower than $22/18^{\circ}C$, indicating the negative role of relatively lower temperatures on flowering of kumquat trees.

개화기 신초비틀기에 의한 '캠벨얼리' 포도의 착립 및 과실품질 (The Effect of Shoot Twist at Bloom on Fruitfulness and Fruit Quality of 'Campbell Early' Grapevine)

  • 본 연구는 수세가 강한 '캠벨얼리'를 시험재료로 착립률 증진을 위한 개화기 신초 비틀기의 효과를 규명하기 위하여 수행되었다. 수세가 강한 '캠벨얼리'의 신초 비틀기 처리에 따른 발아기 및 개화기는 처리별 차이가 없었으나 수확기는 3일 정도 지연되었다. 신초 비틀기 처리구에서 수확기가 지연된 것은 '캠벨얼리'의 과방중과 착립률이 개화전 7일과 개화시 모두 3절 비틀기 처리구에서 높게 나타났기 때문이었다. 신초 비틀기에 의한 '캠벨얼리'의 가용성 고형물 및 산도는 처리간 차이를 보이지 않았으나, 수량은 '개화 전 7일 3절 비틀기' 처리에서 12.1 kg/주으로 무처리에 비하여 1.6배 증가하였다. 안토시아닌 함량은 무처리에 비하여 수량이 많은 처리구에서 낮았으나 착색도인 Hunter 값은 처리간 유의적 차이를 보이지 않았다.
