The disease known as cerebral cavernous malformations mostly occurs in the central nervous system, and their typical histological presentations are multiple lumen formation and vascular leakage at the brain capillary level, resulting in disruption of the blood-brain barrier. These abnormalities result in severe neurological symptoms such as seizures, focal neurological deficits and hemorrhagic strokes. CCM research has identified 'loss of function' mutations of three ccm genes responsible for the disease and also complex regulation of multiple signaling pathways including the WNT/β-catenin pathway, TGF-β and Notch signaling by the ccm genes. Although CCM research is a relatively new and small scientific field, as CCM research has the potential to regulate systemic blood vessel permeability and angiogenesis including that of the blood-brain barrier, this field is growing rapidly. In this review, I will provide a brief overview of CCM pathogenesis and function of ccm genes based on recent progress in CCM research.

Betulae Cortex of Betula platyphylla Suk. var. japonica Hara (Betulaceae) has long been used for treatment of various inflammation, fever and cough in Eastern Asia. In order to investigate antibacterial activity of the Betulae Cortex against Streptococcus mutans or Porphyromonas gingivalis, MIC (minimal inhibitory concentration) and pH were checked, and for anti-inflammation activity, the experiments about trypsin-induced paw edema, vascular permeability and myeloperoxidase activity in rat's hind-paw tissue, were carried out with various extracts of Betulae Cortex (BCXs) respectively. The BCXs showed significant antibacterial activity, and at the dose of over 50 mg/kg, BCX showed significant inhibition on the paw edema, vascular permeability and myeloperoxidase activity. These results indicate that BCXs have antibacterial activity against oral cariogenic bacteria and anti-inflammatory effect.

The anti-inflammatory activity of the aqueous extract from Gleditsiae spina was investigated utilizing carrageenin-induced edema, granuloma pouch and adjuvant arthritis in rats. The effects of this agent on vascular permeability and acetic acid-induced writhing in mice were also examined. Its anti-inflammatory activity on carrageenin edema was observed with oral administration. The aqueous extract from Gleditsiae spina(400 mg/kg, 7 days) showed significant inhibitory effects on granuloma and exudate formation in rats. In the method of adjuvant arthritis, the aqueous extract(400 mg/kg), orally administered for 21 days, inhibited the development of hind paw edema in rats. The aqueous extract was also inhibited the increase in vascular permeability and the number of writhings induced by acetic acid in mice. In the present study, the aqueous extract of Gleditsiae spina was indicated to have the anti-inflammatory action.

Object : We investigated the effect of Gamiokyaek-tang(GOYT) on immediate type hypersensitivity. Methods : We investigated anti-dinitrophenyl(DNP) IgE-mediated passive cutaneous anaphylaxis(PCA) and acetic acid-induced vascular permeability in rodents. Also we measured MTT assay, ${\beta}-hexosminidase$ activity and IL-4 from RBL-2H3 and nitric oxide from Raw264.7. Results : GOYT inhibited passive cutaneous anaphylaxis and acetic acid-induced vascular permeability by oral administration. All the concentrations of GOYT from 0.1 to 5mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosminidase$ release and IL-4 production were significantly reduced by 1, 2 and 5mg/ml of GOYT. In Raw264.7, nitric oxide level was decreased by 5mg/ml of GOYT. Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity.

Experimental studies were done to research effects of Leejungtang and Leejungtanggamibang on the anti-allergic effects. We studied the vacular permeability response induced by histamine and serotonin, homologous PCA provoked by IgE-like antibody against EWA, contact dermatitis induced by picryl chloride and delayed type hypersensitivity response to SRBC. The results were obtained as follows: 1. In effects of the Leejungtang and Leejungtanggamibang on the vascular permeability responses to intradermal serotonin, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 2, In effects of the Leejungtang and Leejungtanggamibang on the vascular permeability responses to intradermal histamine, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 3. In the homologous PCA provoked by IgE-like antibody against white albumin, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 4. In the delayed type hypersensitivity responses to picryl chloride, the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect. 5. In the delayed type hypersensitivity responses to sheep red blood cell the Leejungtang revealed insignificant effect and Leejungtanggamibang revealed significantly inhibitory effect.
He impregnated a highly porous, knitted polyester (Dacron) graft with a biodegradable nonproteinaceous material, algin. This new vascular graft is blood tight but still retains high porosity in the body. It does not need to be preclotted with blood before implantation and has good tissue ingrowth and biological healing properties due to the high porosity. The algin impregnated graft was investigated by "in vitro" examinations in this study. It was characterized by ESCA analysis, SEM observation, and measurements of water permeability, algin coating weight, mechanical properties and whole blood clotting time. The water permeability of the graft was reduced more than 99% by the algin impregnation treatment without changing any mechanical properties. "In vivo" examinations of the algin impregnated vascular graft are on progress.
Cheonggisan(CGS) is well known for its effect on such allergic disease as urticaria and atopic dermatitis. Gagamcheonggisan(GCGS) was formulated by subtracting several herbs from CGS and adding several herbs to CGS. Even though it is being used frequently in the clinicai medicine for the treatment of above hypersensitivity diseases, basic study to make sure the mechanism of its action is rare. In this study the author tried to know the effect of CGS and GCGS on the vascular permeability, contact dermatitis, granular secretion from mast cells and function of macrophages. The results obtained in this study are as follows : 1. Administration of CGS and GCGS decreased the vascular permeability induced by serotonin and histamine. The decrease by serotonin is more typical and dose-dependent. 2. Administration of CGS and GCGS inhibited foot-pad and ear swelling responses induced by sheep red blood cells and picryl chloride respectively, the inhibition of foot-pad swelling responses is bigger than that of ear swelling responses and both of them are not dependent on the dose3. Treatment of peritoneal mast cells with CGS and GCGS water extract decreased the histamine release triggered by compound 48/80 in a dose dependent fashion 4. Administration of CGS and GCGS increased the phagocvtic activity of peritoneal macrophages and treatment of peritoneal macrophages with CGS activated phagocytic function in a dose dependent fashion. 5. Administration of CGS and GCGS enhanced such reactive oxygen intermediates(ROIs) as superoxide and hydrogen peroxide production from peritoneal macrophages. 6. Treatment of CGS and GCGS activated peritoneal macrophages for the production of ROIs. The above results show that CGS and GCGS decreased the hypersensitivity reactions by inhibiting non-specific inflammatory mediator release and vascular permeability without affecting general immune responsiveness.
