  • We have investigated the relative roles of α1-adrenoceptors and purinoceptors in contractions to low and high frequency stimulation of the mouse vas deferens, in terms of the time course of responses. In separate experiments, isometric contractile responses were obtained to 10 pulses at 1 Hz and 40 pulses at 10 Hz. Responses to 1 Hz stimulation consisted of a series of discrete peaks. The α1A-adrenoceptor antagonist RS100329 (10-9M-10-7M) significantly reduced the response to the first pulse, the α1D-adrenoceptor antagonist BMY7378 (10-7M-10-6M) significantly reduced the response to the first two pulses, and the non-selective α1-adrenoceptor antagonist prazosin (10-8M) reduced the response to the first 4 pulses at 1 Hz. Responses to 10 Hz stimulation consisted of an early peak response and a maintained plateau response. RS100329 significantly reduced the peak response but did not significantly affect the plateau response. Prazosin, significantly reduced both the peak and plateau responses. The α1A-adrenoceptor antagonist RS17053 in high concentrations reduced mainly the plateau response leaving a clear early peak response. The plateau response of contraction was almost abolished by the purinoceptor antagonist suramin. These results suggest that there is a relatively minor early α1D-adrenoceptor and a larger early α1A-adrenoceptor component to stimulationevoked contractions of mouse vas deferens, but the major α1-adrenoceptor component is revealed by prazosin to be α1B-adrenoceptor mediated. α1B-Adrenoceptor activation probably facilitates contractions mediated by other α1-adrenoceptors and by purinoceptors. These results suggest that combined non-selective α1-adrenoceptor blockade, particularly α1B-adrenoceptor blockade, in addition to P2X1-purinoceptor blockade is useful in reducing male fertility.

  • Fine structures of the testis and vas deferens in the fall-web worm, Hyphantria cunea Drury, are studied with electron microscope. Adult worms have single testis close to the midlines of the abdomen. Testis is composed of 4 follicles which are incompletly separated from each other and bounded together by a peritoneal sheath. The peritoneal sheath consisted of outer cuticular layer and two kinds of inner layers, in which glycogen particles are dispersed commonly. These two layers are divided by the morphology of cytoplasmic granules. Follicular epithelium forming the wall of the follicles have melanin pigment granules, and trachea or tracheoles are extended through this epithelium. In the cysts of adult testis, matured spermatozoa are grouped together in bundles and after releasing the sperm bundles to the vas deferens, lamellar shaped lysosomes appeared in the cytoplasms of the cyst cells. The number of spermatozoa per cyst is exactly 256 ($2^8$), this number is characteristics of the Lepidoperan species. Vas deferens is a tube with a fairly thick bounding epithelium, a basement membrane and a layer of circular muscle outside it. At the apical portion of the epithelial cells, microvilli are well developed. And in the cytoplasms of these cells, numerous excretory granules are observed.

  • The sesarmid crab Muradium tetragonum, inhabiting the mangrove, are considered as a key consumer of litter and thereby play an important role in the detritus food chain and energy flow in the mangrove ecosystem. The present investigation was carried out with objectives to enlighten the reproductive system of Muradium tetragonum through histological and histochemical studies. Histological organization of the testis of M. tetragonum revealed that each testis has a lobular structure consisting of several testicular lobules arranged around the collecting duct. Histology of the deferens of M. tetragonum revealed it to be composed of three-layer of tissues along the entire length:the outer connective tissue, the middle muscular and the inner epithelial layer. Based on the histological architecture these three regions are recognized as proximal vas deferens (PVD), middle vas deferens (MVD) and distal vas deferens (DVD). Histological characteristics of the ovary of M. tetragonum during different phases of ovarian development were studied. Based on the colour changes of the ovary and diameter of the oocytes five stages of ovarian development can be pronounced. Histochemical analysis of the male reproductive tissues of M. tetragonum signifies the secretion of a different biomolecule by specifying their origin in the reproductive tissue and their possible transformation into spermatophores. In the female reproductive tissues, histochemical evaluation envisaged the secretory products during different stages of ovarian development The secretory substances of the spermatheca expound on the significance of its secretion in dehiscing the spermatophore wall and in nourishing as well as protecting the spermatozoa.

  • The effects of ginseng total saponins (GTS) on the action of U-50,488H, a $textsc{k}$-opioid receptor agonist, on the electrically induced twitch responses of mouse vats deferens were studied. U-50,488H ($10^9$~$10^{-5}$M) inhibited the twitch contractions in a dose-dependent manner, which were caused by adenosine 5'-triphosphate (ATP) released from the stimulated sympathetic nerve, and this effect was antagonized by naloxone ($10^6$ M). GTS, which itself induced the inhibition of the twitch contractions, acted additively to U-50,488H, GTS and U-50,488H had no effect on the tension of the unstimulated organs. The contractions elicited by ATP were not affected by U-50,488H, but inhibited by GTS. These results suggest that U-50,488H suppressed the twitch contractions by the inhibition of neurotransmitter release from presynaptic nerve terminals via action on opioid receptor, but G75, by inhibiting the action of the neurotransmitter on the smooth muscle.

  • To elucidate one of the effect of piperine on the peripheral sympathetic nervous system, influence of piperine upon the contractile action of norepinephrine, methoxamine and tyramine as well as uptake and release of $[^{3}H]-norepinephrine$ has been investigated in naive and chronic piperine-treated vas deferens of rats. $pA_2$ value for ${\alpha}_1-adrenoceptor$ of phentolamine was significantly increased. Chronic piperine-treated group was markedly shown increased efflux of $[^{3}H]-norepinephrine$ and muscular tension, but was not affected the neuronal up-take and release of $[^{3}H]-norepinephrine$. It can be concluded that potentiation of the effect of norepinephrine by acute and chronic piperine treated group may be due to the change of affinity of ${\alpha}_1-adrenoceptor$, and partly due to possible modification of storage mechanism.

  • The objective of this study was to examine the binding potential of sperm to the epithelium of the sperm storage tubules (SST) in vitro and in vivo to assess the functional maturation of fowl sperm. Sperm from the testis, epididymis, as well as the proximal, middle and distal vas deferens were incubated in vitro with either the uterovaginal junction (UVJ)- or infundibular tissue containing SST at $39^{\circ}C$ for 30 min. Aliquots of sperm were also artificially inseminated into the uteri of hens, and the UVJ and infundibulum were collected 24 h post artificial insemination (AI). After incubation and AI, tissues were washed to remove loosely adhered sperm and subjected to fluorescence staining with 4', 6-diamidino-2-phenylindole, dihydrochloride (DAPI) for counting the number of bound sperm per 0.25 mm2 of surface area. Sperm from the testis, epididymis, and the three segments of the vas deferens exhibited their differential (p<0.05) binding capacity, which increased gradually from the testicular to distal vas deferens sperm under both in vitro and in vivo conditions. Existing similar trend, sperm, regardless of their source had a lesser affinity to bind to the epithelium of the infundibular SST than to the UVJ-SST. These experimental results suggested that fowl sperm may undergo gradual changes in the process of functional maturation, whereby they gain the ability to bind to the epithelium of the SST during their passage through the male reproductive tract (MRT).

  • This study was performed to investigate the effect of octreotide on the contractility of rat vas deferens. The smooth muscle strips isolated from the prostatic portion were myographied in isolated organ bath, Electric field stimulation (monophasic square wave, duration: 1 mSec, voltage : 50 V, frequency : 5 Hz or 30 Hz, train: 10 Sec) produced reproducible contraction. The contraction was composed of two component, first phasic component (FPC) and second tonic component (STC). These contractions were abolished by tetrodotoxin ($1{\mu}M$). Octreotide inhibited the field stimulation induced contractions both FPC and STC concentration-dependently. The FPC was decreased by a desentization of purinergic receptor by pretreatment of mATP, and the STC was decreased by pretreatment of reserpine(3 mg/kg, IP) 24 hours before experiments. Octreotide reduced the field stimulation induced contraction in the presence of mATP and of reserpinized muscle strips. The inhibitory effect of octreotide was more potent at 5 Hz than at 30 Hz. Octreotide did not affect basal ton and exogenous norepinephrine- or ATP-induced contraction. These results suggest that octreotide inhibit the contractility of the isolated rat vas deferens by inhibition of the release of neurotransmitters, both ATP and norepinephrine from adrenergic nerve terminal.

  • Vas deferens is a long tube and could be divided into four regions as its morphological characters: a short and slender proximal region, a convoluted region, a straight and more thick distal region, and an ejaculatory duct, It is connected with the posterior outer horns of the testis and runs to the gonopores opened on the coxa of the fifth walking legs, The proximal region consists almost entirely of simple cuboidal epithelial cells, $12\~28{\mu}$m in height, surrounded by a thin basal membrane. A small aggregation of high cuboidal epithelium is obsened from one side of the proximal region. The convoluted and distal region is composed of two kinds of epithelial cells; high cuboidal epithelial cells, $40\~120{\mu}m$ in height located in dorsal portion and simple cuboidal epithelial cells of $12\~28{\mu}$m in height located in ventral and lateral portion. The ejaculatory duct is surrounded with two kinds of muscle layers, inner longitudinal and outer circular muscle fibers, The lumen is lined with high and simple cuboidal epithelium in almost equal proportions. The proximal region contains mature sperm and basophilic substances. The eosinophilic substances appeared newly going toward the convoluted region and are laid along simple cuboidal epithelial cell layer. Tube-like content (spermatophore) of the vas deferens is ejaculated from a pair of genital pores at mating. Two masses of the content fuse together side by side and are usually deposited on the female thorax between the second and fourth or fifth pereiopods, The spermatophore formed measures $2.7\~4.0$ mm in length and $1.5\~2.7$ mm in width.

  • We report the case of a 46-year-old Chinese male patient who visited our clinic complaining of infertility. Semen analysis revealed azoospermia, and azoospermia factor c region partial deletion (b1/b3) was detected using Y chromosome microdeletion analysis. Testicular sperm extraction was performed after genetic counseling. The bilateral ductus deferens and a portion of the epididymis were absent, whereas the remaining epididymis was expanded. Motile intratesticular spermatozoa were successfully extracted from the seminiferous tubule. On histopathology, nearly complete spermatogenesis was confirmed in almost every seminiferous tubule. To our knowledge, this is the first case report of b1/b3 deletion with a congenital bilateral absence of the vas deferens and almost normal spermatogenesis.
