Microbe Hunting: A Curious Case of Cryptococcus

  • C. neoformans-associated cryptococcosis is primarily a disease of immunocompromised persons, has a world-wide distribution, and is often spread by pigeons in the urban environment. In contrast, C. gattii causes infection in normal hosts, has only been described in tropical and semi-tropical areas of the world, and has a unique niche in river gum Eucalyptus trees. Cryptococcosis is acquired through inhalation of the yeast propagules from the environment. C. gattii has been identified as the cause of an emerging infectious disease centered on Vancouver Island, British Columbia, Canada. No cases of C. gattii-disease were diagnosed prior to 1999; the current incidence rate is 36 cases per million population. A search was initiated in 2001 to find the ecological niche of this basidiomycetous yeast. C. gattii was found in the environment in treed areas of Vancouver Island. The highest percentage of colonized-tree clusters were found around central Vancouver Island, with decreasing rates of colonization to the north and south. Climate, soil and vegetation cover of this area, called the Coastal Douglas fir biogeoclimatic zone, is unique to British Columbia and Canada. The concentration of airborne C. gattii was highest in the dry summer months, and lowest during late fall, winter, and early spring, months which have heavy rainfall. The study of the emerging colonization of this organism and subsequent cases of environmentally acquired disease will be informative in planning public health management of new routes of exposure to exotic agents in areas impacted by changing climate and land use patterns.

Cryptococcosis is an infection associated with an encapsulated, basidiomycetous yeast Cryptococcus neoformans. The route of entry for this organism is through the lungs, with possible systemic spread via the circulatory system to the brain and meninges. There are four cryptococcal serogroups associated with disease in humans and animals, distinguished by capsular polysaccharide antigens. Cryptococcus neoformans: variety grubii (serotype A), variety neoformans (serotype D), and variety gattii (serotypes B and C) (Franzot et at. 1999). C. neoformans variety gattii has recently been elevated to species status, C. gattii. C. neoformans val. grubii and var. neoformans have a world-wide distribution, and are particularly associated with soil and weathered bird droppings. In contrast, C. gattii (CG) is not associated with bird excrement, is primarily found in tropical and subtropical climates, and has a restricted environmental niche associated with specific tree species. (Ellis & Pfiffer 1990) Ellis and Pfeiffer theorize that, as a basidiomycete, CG requires an association with a tree in order to become pathogenic to mammals. In Australia, CG has been found to be associated with five species of Eucalypts, Eucalyptus camaldulensis, E. tereticornis, E. blakelyi, E. gomphocephala, and E. rudis. Eucalypts, although originally native to Australia, now have a world-wide distribution. CG has been found associated with imported eucalypts in India, California, Brazil, and Egypt. In addition, in Brazil and Columbia, where eucalypts have been naturalized, native trees have been shown to harbour CG (Callejas et al. 1998; Montenegro et al. 2000). In British Columbia, Canada, since the beginning of 1999, there have been 120 confirmed cases of cryptococcal mycoses associated with CG in humans, including 4 fatalities (data from British Columbia Centre for Disease Control), and over 200 cases in animal pets in BC (data from Central Laboratory for Veterinarians). What is remarkable about the BC outbreak of C. gattii-cryptococcosis is that all of the cases have been residents of, or visitors to, a narrow area along the eastern coast of Vancouver Island, BC, from the tip of the island in the south (Victoria) to Courtenay on the north-central island as illustrated in Figure 1. Of the first 38 human cases, 58% were male with a mean age of 59.7 years (range 20 - 82): 36 cases (95%) were Caucasian. Ten cases (26%) presented with meningitis, the remainder presented with respiratory symptoms. Cultures recovered from cases of cryptococcosis associated with the outbreak were typed as serogroup B, which is specific to CG (Bartlett et al. 2003). This was the first reported outbreak of CVG in Canada, or indeed, the world. Where infection with CG is endemic, for example, Australia, the incidence of cryptococcosis ranges from 1.8 - 4.7 per million between the southern and northern states (Sorrell 2001). However, the overall incidence of cryptococcosis in immunocompenent individuals has been estimated at 0.2 per million population per year (Kwon-Chung et al. 1984). The population of Vancouver Island is approximately 720,000,consequently, even if the organism were endemic, one would expect a maximum of 0.15 cases of cryptococcal disease annually.

Effects of 3,5,3'-triiodo-L-thyronine ($T^3$) on Growth of GR-transgenic Coho Salmon, Oncorhynchus Kitsutch

  • GH-transgenic coho salmon (Oncorhynchus kitsutch) juveniles in tGH*T$_3$and tGH*PTU were fed with the diets containing 1 ug/g fish of 3,5,3'-triiodo-L-thyronine (T$_3$) and 30 ug/g fish of 6-n-propyl-2- thiouracil (PTU), respectively, to assess the effect of these drugs on the change of physiological activity, growth and survival rate in comparison with normal transgenic (tGH*C) and nontransgenic coho salmon (Wild) for 90 days. Although the daily food intakes of all transgenic (tGH)-groups were higher than Wild, the amount was reduced by exogenous PTU supply. The fred efficiencies of tGH-groups were lower than Wild, but the efficiency was reduced both by T$_3$and PTU. The survival rate of tGH-group was significantly higher than that of Wild, but there was no significant difference among tGH-groups. Although the growth of tGH-coho salmon was faster than Wild. the growth rate of transgenic salmon was increased by exogenous T$_3$, but was reduced by PTU Plasma TT$_4$levels of tGH-groups was approximately 2-fold higher relative to Wild, but there were no difference of plasma TT$_4$levels among tGH-groups. plasma TT$_3$level or tGH-coho salmon was increased by exogenous T$_3$administration, but was reduced by exogenous PTU. In addition, although plasma GH levels of all tGH-groups were higher than that of Wild, the GH level in plasma of transgenic coho salmon was increased by exogenous T$_3$and reduced by exogenous PTU. In the meantime, the transgenic fishes also displayed head, jaw and opercular abnormalities typical of the offsets of this gene construct in coho salmon, indicating that some imbalance in growth processes has been induced. However, the abnormalities of transgenic coho salmon was reduced following exogenous PTU administration.

  • The current size, characteristics. and channels of distribution of the offshore export (non North American) market for ginseng products are described. The apparent size of the worlds export market for ginseng products in 1987 is 3,363 tonnes. The major export markets for ginseng are Hong Kong (1,972.4 tonnes) and Japan (540.9 tonnes). The volume of ginseng imported into Hong Kong has doubled over the past five years from 976.3 tonnes in 1983 to 1,972.4 tonnes in 1987. This increase is reflective of an expanding Chinese market. In terms of products forms, the toast majority of ginseng imported into China is in root form, while Japan imports both red and white ginseng as well as ginseng beverages and medical products. Hong Kong is the destination of over 86f; of the volume of North American ginseng exported between 1983 and 1987. North American ginseng products are distributed by ginseng brokers or importers/exporters.

  • It is estimated that world ginseng production has increased from 3,200 tonnes In 1983 to 5,132 tonnes in 1987. China produced approximately hart of world ginseng production and South Korea's production amounts to 31.9% of world ginseng production. Canadian ginseng production of 114 tonnes in 1987 represents only 2.2% of estimated world production. World ginseng production is projected to increase to 6,856 tonnes by 1992. Canadian production as a proportion of total world production is expected to increase to 3.3% however, North American production overall is projected to decrease 12.0% of world production by 1992. Assuming that the value of each country's production is equal to average export price, the value of world ginseng production is estimated to be $ 443 million The estimated values of Korean, North Anerican and Canadian ginseng production represent $ 198.3million, 977.1 million, and $ 12.5 million, redpectively.

Marine macroalgae of the Aleutian Islands: I. Bangiales

  • We sequenced the rbcL gene in more than 100 collections of foliose Bangiales made in the Aleutian Islands and western Alaska Peninsula during the past 25 years. This work allows us to recognize four previously undescribed species, two in the genus Boreophyllum and two in Pyropia. Boreophyllum aleuticum appears to be endemic to the Aleutian Islands, whereas B. ambiguum is known to occur from the Yakutat area to the tip of the Alaska Peninsula. The two previously undescribed species of Pyropia are more broadly distributed. Pyropia taeniata, which was previously identified under the name Py. pseudolinearis, occurs from northern Southeast Alaska through the Aleutian Islands. Pyropia unabbottiae, which is sister to Py. abbottiae, occurs from southern Vancouver Island to Attu Island. Collections throughout the Aleutian Islands allow us to document the distribution of another dozen species of foliose Bangiales in this region, including Boreophyllum aestivale, Fuscifolium tasa, Pyropia fallax, Py. fucicola, Py. gardneri, Py. kurogii, Py. nereocystis, Py. pseudolanceolata, Py. torta, Wildemania amplissima, W. norrisii, and W. variegata. We were unable to confirm the occurrence of the following species previously recorded from the Aleutian Islands: Porphya ochotensis, Pyropia abbottiae, Py. perforata, Py. pseudolinearis, P. purpurea, P. umbilicalis, Py. yezoensis and Wildemania schizophylla. At least two undescribed filamentous Bangiales also occur in the Aleutian Islands.

  • 본 연구는 PvuII PCR-RFLP를 이용하여 Yorkshire종 돼지에서 Estrogen Receptor의 유전적 다형과 산자수간의 관련성을 분석하기 위하여 수행되었다. 무창 돈사에서 사육중인 242두의 종빈돈으로부터 혈액을 채취하여 PvuII PCR- RFLP로 ER 유전자형을 결정하였다. ER 유전자 좌위에서 유전자 빈도는 각각 0.39(A)와 0.61(B)였다. ER 유전자형별 산자수에 대한 효과는 최소제곱평균을 설명하는 고정모형을 설정하여 추정하였다. 분석결과 복당 총산자수와 실산자수에서 특정 ER 대립유전자(B)에서 산자수 증진효과가 관찰되었다. 따라서 돼지 ER 좌위의 유전적 변이는 번식돈의 산자수 증대와 관련된 marker-assisted selection(MAS)에 응용될 수 있을 것으로 사료된다.

Suitable Dietary Protein/Lipid Ratio for Hybrid, Female Red Sea Bream Pagrus major and Male Black Sea Bream Acanthopagrus schlegeli in the Juvenile Stage, Compared with Red Sea Bream

  • To determine a suitable dietary protein/lipid (CP/CL) ratio in the early juvenile stages of hybrid porgy ($F_1$), female red sea bream (RSB) ${\times}$ male black sea bream, five diets with various CP/CL ratios-60/7, 55/12, 51/17, 46/23, and 41/28-were prepared and provided to juveniles in triplicate. At the smaller juvenile stage, $F_1$, weighing 0.32 g, a significantly higher specific growth rate (SGR) and feed efficiency (FE) were seen with 60/7 and 55/12 diets. However, in RSB weighing 0.26 g, SGR and FE were higher with the 60/7 diet than the other diets at $21^{\circ}C$. At the larger juvenile stage, $F_1$, weighing 3.7 g, there was no significant difference in SGR or FE among the diets, but RSB weighing 4.0 g fed 60/7, 55/12, and 51/17 diets had higher SGR and FE than 46/23 and 41/28 diets at $24^{\circ}C$. Moreover, survival and apparent nutrient retention of $F_1$ at both stages were significantly higher than those in RSB. These results indicate that both $F_1$ and RSB weighing ca. 0.3 g require a higher dietary CP/CL than those weighing ca. 4 g. Additionally, $F_1$ in both trials showed the suitability of a lower dietary CP/CL than RSB, indicating that mass production of $F_1$ juveniles will be more economical than RSB.
