Cytochemical and Ultrastructural Studies on Tracheal Epithelium in the Aging Rat (노화에 따른 Rat 기관상피의 세포화학적 및 전자현미경적 연구)

  • The present studies were designed to determine the feasibility of using the rat tracheal epithelium as models for induction of aging. The ultrastructural and cytochemical changes of tracheal epithelium were investigated in rats at ages of five, twelve and twenty four months. Some major changes in the tracheal epithelium with advancing age were observed by electron microscopy. The results were summarized as fellow: 1. With the advance of age, lysosome, vacuole and multivesicular bodies were increased in number and numerous myelinoid bodies were observed in cytoplasm of ciliated cells. 2. In goblet cell, serous cell and brush cell lysosome and myelinoid bodies were increased in number with the advance of age, and an myelinoid bodies was often found within the secretory granule. 3. Cytochemical studies showed that acid phosphatase activities was observed in multivesicular bodies and lysosome, strong activities with the advance of age. And alkaline phosphatase activity are observed in microvilli, granule and lateral membrane of secretory granule cells, and strong activities with age. Consequently suggest that with the advance of age, tracheal epithelium show ultrastructural and cytochemical alteration of some kind of cell organelles in all kind of cell.

Ultrastructure and Activity Pattern of Peroxidase in Secretory Trichomes of Drosera capensis (장대끈끈이주걱 분비모의 미세구조와 peroxidase 활성)

  • Glandular trichomes present on the leaf surface of Drosera capensis were examined using transmission electron microscopy. A large number of stalked glands exist on the adaxial surfaces of the leaf blade. The secretory head is composed of two layers of secretory cells, one layer of middle cells, and the inner tracheids. The secretory cells contain rough endoplasmic reticulum, mitochondria, plastids, Golgi apparatus, and vacuoles. The secretory cells show prominent cell wall ingrowth, and thick cuticle restricted on the subcuticular wall. Frequently, the cuticle has some pores, canal-like structures, showing electron -dense granules being penetrated through them. Ultrastructural localization using diaminobenzidine showed the electron-dense deposits in the vacuole. No peroxidase activity was seen in the cell wall and cytolasm. The activity of peroxidase (POX) isozymes in Drosera which isoelectric point (pI) is 3.6 and some anionic POX isozymes which pIs are laid between 3.6 and 4.6 were especially increased according to the development and the formation of glandular trichomes. Also, the activity of some POX isozymes which isoelectric points are laid between 4.6 and 5.1 were increased in the regions of leaves which has trichomes.

Toxoplasmosis in piglets (자돈의 톡소플라즈마증 발생)

  • Suckling piglets and weaned pigs showed anorexia, dehydration, severe abdominal breathing, emaciation and paresis from Oct. 1993. to Nov. 1993. Five 2-week-old piglets were submitted for diagnosis in Kangwon National University. At necropsy, the pin-point well demarcated yellowish white foci were scattered on the surface of the lung, heart, liver, spleen and kidney. Histologically, multifocal areas of necrosis with mononuclear cells infiltration were found in the lung, heart, liver, lymph node, spleen, kidney and small intestine. These lesions tended to be associated with blood vessels. Variable round to ovoid tachyzoites were located at the periphery of the lesions. The organisms were demonstrated as Toxoplasma gondii by immunohistochemical staining method. Ultrastructurally, this parasite was surrounded with parasitophorous vacuole in alveolar macrophage. The parasite was crescent-shaped and $6{\sim}8{\times}1{\sim}2{\mu}m$ in size. It was enclosed by an thick outer membrane and an underlying thin inner membrane. Several club-shaped paired organelles and conoids lay in the cytoplasm at the anterior. Numerous round body and one to several mitochondria were presented in the cytoplasm. Based on the gross findings, histopathology, immunohistochemical and electron microscopic findings, this case was diagnosed as toxoplasmosis in piglets.

Histological Studies of Gonad in the Hybrid Species Cobitis. sinensis-longicorpus Complex(Pisces, Cobitidae) (잡종기원의 Cobitis sinensis-longicorpus complex(Pisces, Cobitidae)에 대한 생식소의 조직학적 연구)

  • Cobitis sinensis-longicorpus complex considered as hybrid origin between C. sinensis and C. longicorpus coccurred ommonly in the upper streams of the Nakdong River, Korea. Histological examinations of their gonad were accompanied with 272 individuals of C. sinensis-longicorpus complex collected. Most of fishes collected were females, however, only 6 individuals were found males. The ovarian tissues of females are completely fertile undergoing normal oogenensis. In the male gonads, testicular lobule structure with abnormal vacuolar tissues were observed. Spermatogonia and spermatocytes were also observed of their testis however spermatids or sperms were not shown in their developmental stages. From these facts, we infer that female population of C. sinensis-longicorpus complex may be unique reproductive hierarchy accomplishing their reproduction with participation of males of their closely related bisexual species.

Redescription of Two Urostylid Ciliates (Ciliophora: Urostylida), Anteholosticha pulchra and Metaurostylopsis struederkypkeae from Korea

  • Two urostylid ciliates, Anteholosticha pulchra (Kahl, 1932) Berger, 2003 and Metaurostylopsis struederkypkeae Shao et al., 2008, new to Korea, were collected from the Yellow Sea and the East Sea, Korea, respectively. They were identified based on live observation and protargol impregnation. Taxonomical characters of A. pulchra are as follows: $190-300{\times}30-55\;{\mu}m$ size in vivo; contractile vacuole located on the left side of the posterior 1/4 of the cell; spherical-reddish granules at cirral bases and around dorsal bristles, somewhat sparsely distributed throughout the cell surface; four frontal and two frontoterminal cirri; four dorsal kineties; caudal cirri absent. Metaurostylopsis struederkypkeae is characterized as follows: $80-110{\times}40-50\;{\mu}m$ size in vivo; caudal cirri absent; two types of cortical granules: type 1, yellow-green arranged along the ventral cirral rows and dorsal kineties; type 2, small and reddish, with an irregular arrangement; four frontal, four to eight frontoterminal, and two to six transverse cirri; five to seven left and three to five right marginal rows. Sequences of small subunit ribosomal DNA were determined from both species, and pairwise distances with their relatives were analyzed.

  • A strategy for development of a functional rice in proved with human lactoferrin and enhancement of nutrient compounds was planned. For the purposes we have cloned and characterized a human lactoferrin cDNA from human mammary gland cDNA library A endosperm storage vacuole targeting sequence and the cDNA fragment was linked to endosperm specific glutelin promoter. The fusion gene fragment was inserted into a binary vector containing MAR gene. In addition a new ${\beta}$-galactosidase gene from Bifidobacterium of human was used as a reporter gene in the vector system, Rice plants showing a high concentration of amino acids in the endosperm cells were developed by using a biochemical mutation and bred for the transformation with the binary vector system Finally we have established a transformation method for the rice endosperm cells.

  • The origin and the function of cytolysomes were studied in the mesophyll cells and the root-tip cells of Glycine max Merr. and Zea mays L. fixed by paraformaldehyde-glutaraldehyde-$OsO_4$. The cytolysome-like structures were found of three main types of configurations: multivesicular, myelin like (multilamllar) and multitubular. More complex and mixed ones were also observed. The origin of these structures seems to be initiated by invaginations or in holdings of the plasmalemma into the cell interior, and that by aggregation and convolution of endoplasmic reticulum in the cytoplasm. Invagination of the plasmalemma were found of two main types of configurations: concentric whorls of lamellar and multivesicular. The structures were also observed within vacuoles and cytoplasm. Since the structurers are widely distributed in the cells and are greatly varied in sizes and shapes. These structures originate from the plasmalemma and the cytoplasm subsequently protrudes into the vacuole, and that seem to play an important role on the formation of the autophagic vacuoles. The possible function and fate of these structures are discussed.

Effect of SOD on Ultrastructural Changes of Gastric Parietal Cells in the Cisplatin Treated Rats (흰쥐에서 cisplatin에 의한 위벽세포의 미세구조변화에 미치는 SOD의 영향)

  • This study aims to demonstrate the effect of SOD (superoxide dismutase), one of the antioxidant enzymes, on the ultrastructural changes in the parietal cells caused by the administration of cisplatin in the rat. A total of 60 healthy Sprague-Dawley rats weighing about 200 gm were used as experimental animals. Cisplatin (6 mg/kg) was administered intraperitoneally to rats pretreated with 15,000 unit/kg of SOD or rats without the pretreatment. The experimental animals were sacrificed at 6 hours, 12 hours, 24 hours and 3 days after the administration of cisplatin. The results were as follows: 1. SOD alone did not affect the ultrastructural changes in the gastric parietal cells in the rat. 2. Irregular shaped mitochondria, mitochondria with dim cristae, dilated cristae, ruptured outer membrane, electron lucent matrix and degenerative mitochondria were seen in cisplatin treated rat. Whorled membranous body, many lysosomes and large vacuole were observed in the gastric parietal cells in cisplatin treated rat. 3. Mitochondria with dilated cristae and electron lucent matrix and irregular shaped mitochondria were observed in the gastric parietal cells of the cisplatin treated rat with pretreatment of SOD. These results suggest that SOD attenuates the toxic effect of the cisplatin in the gastric parietal cells of the rat.

Pathogenetic Impact of Vacuolar Degeneration by Accelerated Transport of Helicobacter pylori VacA

  • Vacuolar degeneration of the gastric epithelial cells is a characteristic feature of the derangement of mucosa where Helicobacter pylori colonizes, and H. pylori vacuolating cytotoxin (VacA) has been suggested to playa key role in it. To elucidate the VacA-involved degenerative mechanism, VacA was purified, and its impact on degeneration of HeLa cells was determined. In the presence of ammonium chloride, cell vacuolation by VacA was dose-and time-dependent, however, no detectable degeneration of the cells was observed with the VacA concentration tested. A further increase of vacuolation was shown in cells pre-treated with diethyl pyrocarbonate (DEPC) , and this resulted in a change of the cell morphology to become spherical. Similar phenomena were also observed when HeLa cells were co-cultivated with intact H. pylori cells. It was remarkable to note that the degree of growth inhibition was proportional to the increase in vacuole formation, suggesting that the vacuolation rate would be critical for cell degeneration. Surprisingly, although VacA was itself inhibited by DEPC, its uptake was markedly increased by this agent, similar to that found in cells with Nabutyrate. These data indicate that the cell's tolerance of VacA transport may be critical for vacuolar degeneration and may be changeable during H. pylori inhabitation.

Phagocytic osteoclasts in the alveolar bone of diabetic rats with periodontitis

  • Periodontitis is a bacteria-induced inflammatory disease associated with alveolar bone loss. Osteoclast is a macrophage-lineage cell that exhibits phagocytic activity; however, osteoclast phagocytic activity has not been demonstrated under pathological conditions. Diabetes is a pathological condition that exacerbates alveolar bone loss via periodontitis; therefore, we examined phagocytic osteoclasts in diabetic rats that had periodontitis. The rats were divided into the control (C), periodontitis (P), and diabetes with periodontitis (DP) groups. Diabetes and periodontitis were induced by streptozotocin injection and ligature of the mandibular first molars, respectively. On days 3 and 20 after the ligature, the rats were sacrificed, and osteoclasts containing inclusions were quantified by tartrate-resistant acid phosphatase staining. On day 3, there were more osteoclasts containing inclusions in the DP group than in the C group. Among inclusions, osteocyte-like cells and dense bodies were more frequently observed in the DP group than in the C group. Cytoplasm-like structures were elevated more in the DP group than in the C and P groups. However, no differences were observed on day 20. Interestingly, some osteoclasts were in contact with the osteocytes within the exposed lacunae and contained several inclusions within a large vacuole. Thus, the elevation of phagocytic osteoclasts in rats with diabetes and periodontitis provides insight into the role of osteoclast phagocytic activity under pathological conditions.
