중국 황도화(黃島話)의 'NP1+VP+기(起)+NP2'구문에 관한 초보적 고찰

  • In Huangdao Dialect, the "NP1+VP+起+NP2" sentence structure can not only be used with monosyllabic and bisyllabic adjectives and active adjectives, but also with passive adjectives. The usage of the VP is broader in Huangdao Dialect than in Qingdao dialect in case it is a verb phrase. The negative of "NP1+VP+起+NP2" sentence structure uses "不" or "沒有." If common comparative element can be found between NP1 and NP2, the negative particles are used at the beginning of the sentence in general to generate topic sentences. NP1 is often a common noun, modifying phrase, and quantifying phrase, while it can also be a verb-object phrase or modifying phrase. Huangdao Dialect may exhibit specific quantifying phrases that express comparative differences. The inquisitive style of "NP1+VP+起+NP2" sentence structure include appending a question mark at the end of the sentence, combining the positive and negative forms of the adjective, and appending "'是沒" or "是不是" in the front of the adjective.

Anti-HIV agent VP-0501의 생체이용성 향상을 위한 아미노산 프로드럭 개발 및 약물동태연구 (Pharmacokinetics of a new anti-HIV agent VP-0501 and development of its amino acid prodrug for improving oral bioavailability)

  • We have studied pharmacokinetics of a new anti-human immunodeficiency virus (HIV) agent VP-0501 and its amino acid prodrug VP-0501AL which is designed to improve oral bioavailability. After oral administration at 100 mg/kg dose in rats (n = 4), VP-0501 was not detectable in plasma (<50 ng/ml), while after the administration of VP-0501AL, VP-0501 was quantitatively detected, at least for 8 hrs, with Cmax of ca. $2.5{\mu}g/ml$ and AUC of $8hr^{\ast}{\mu}g/ml$. When VP-0501 was intravenously administered at 50mg/kg, this compound appeared at a marginal level in plasma with AUC of $2hr^{\ast}{\mu}g/ml$, $t_{1/2}$ of 2 hr, $C_0$ of $0.7{\mu}g/ml$, and MRT of 3 hr. On the other hand, with intravenous VP-0501AL at the same dose, both the prodrug VP-0501AL and its metabolite VP-0501 appeared comparatively at higher level in the plasma: pharmacokinetic parameters of VP-0501AL including $Vd_{\beta}$, AUC, $t_{1/2,{\beta}}$, $C_0$, $CL_{tot}$, and MRT were ca. 2 L/kg, $70hr^{\ast}{\mu}g/ml$, 2 hr, $180{\mu}g/ml$, 0.7 L/hr/kg, and 1 hr, respectively. These results demonstrate that attachment of amino acid alanine to VP-0501 is an effective approach for improvement of its oral bioavailability. Therefore, VP-0501AL is expected to become a new highly bioavailable and potent anti-AIDS drug candidate/lead compound.

일본어 유악센트 방언과 무악센트 방언의 통사적 애매성의 해소와 운율적 특징 (Syntactic Ambiguities and their Resolution in Prosody in Japanese)

  • The prosody can play a crucial role in differentiating ambiguous sentences to correctly reflect their intended syntactic structures. In what way do the speakers in Tokyo and Sendai dialects of Japanese use prosodic elements to differentiate syntactic ambiguities? Acoustic measurement was made of utterances of ambiguous sentences in Japanese to observe prosodic strategies for disambiguation. Materials were sentences of the type ADV-VP1-NP-VP2, ADV-NP1-NP2-VP2, where the ambiguity lies in locative adverbial modification, ADV modifying either VP1 or VP2. For this construction the Japanese create the same ambiguities. After defining the depth of a syntactic boundary, F0 of the phrase before and after the boundary, and duration of the syllable and pause before the boundary were measured. The results show that Tokyo dialects speakers use F0 after syntactic boundary, and Sendai dialects speakers use of the syllable and/or pause before the boundary.

  • 4-비닐피리딘과 N,N'-1,6-헥사메틸렌비스아크릴아미드를 라디칼중합하여 여러 가지 가교도를 가지는 가교 폴리(4-비닐피리딘) (CHP4VP)을 합성하였고, 이들 가교고분자와 구리(II)와의 착물을 평형흡착법으로 제조하였다. 제조한 착물들을 ascorbic acid (AA)의 산화반응 촉매로 사용하여 촉매활성을 조사하였다. CHP4VP-Cu(II) 착물에 의한 AA 산화반응은 Michaelis-Menten형 동력학적 거동을 나타내었다. CHP4VP-Cu(II) 촉매계의 촉매활성은 CHP4VP의 가교도가 증가할수록 증가하였고, 또 CHP4VP-Cu(II) 착물은 재사용 후에도 촉매활성이 거의 감소하지 않았다. 그러나 CHP4VP-Cu(II) 착물은 전보의 N,N'-메틸렌비스아크릴아미드가 가교제로 포함된 가교 폴리(4-비닐피리딘)-구리(II) 착물보다 AA 산화반응에 대한 촉매활성이 감소하는 경향을 나타내었다. 이들 결과로 부터 촉매계에 포함된 CHP4VP의 가교도와 가교제의 소수성이 AA 산화반응에 중요한 역할을 함을 알 수 있었다.

cDNA Cloning and Expression of Human Rotavirus Outer Capsid Protein VP7 in Insect Cells

  • Rotavirus is a major cause of severe gastroenteritis in young children and animals throughout the world. The VP7 of rotavirus is thought to induce the synthesis of neutralizing antibodies and to be responsible for determining viral serotypes. The cDNA coding for the VP7 capsid protein of human rotavirus, obtained from Korean patients (HRV-Y14), was cloned and its nucleotide sequence was determined. Comparative analysis of the nucleotide sequences between VP7 of Y14 and that of other foreign isolates showed $92.7~95.2\%$ homology to G1 serotypes (RV-4, KU, K8, WA), $74.2\%$ homolgy to G2 serotype HU-5, $76.4\%$ homology to G3 serotype SA-11, and $77.6\%$ homology to G4 serotype A01321. These data suggest that HRV-Y14 can be classified as a G1 serotype. cDNA coding for VP7 of HRV-YI4 was subcloned into the baculovirus vector and the VP7 glycoprotein was expressed in insect cells. The expressed proteins in Sf9 cell extract and tissue culture fluid were separated on SDS-PAGE, and Western blot analysis with monoclonal antibody raised against the synthetic peptide containing 21 amino acids within the VP7 conserved region was performed. The molecular weight of recombinant VP7 was estimated to be 36 kDa which is about the same size as the native VP7. Addition of tunicamycin in the culture media caused a reduction of the molecular weight of the recombinant VP7 indicating that the expressed protein was glycosylated.

고에너지 엑스선을 조사한 갑상선의 세포막모델에서 요오드이온의 선택적 투과성 전달 특성 (Selective Permeate Transport Characteristics of Iodine ion at Cell Membrane Model of Thyroid which Irradiated by High Energy X-Ray)

  • 본 논문에서는 고에너지 엑스선(6 MV X선)을 조사한 갑상선의 소포세포막 모델에서 막의 두께차이, 고정전달자[VP-AN% (w/ w)]의 농도차이, OH-이온의 농도차이가 세포내부와 세포외부사이로 갑상선호르몬의 구성성분인 요오드이온의 농도차이로 발생하고 갑상선호르몬의 생성량에 영향을 미치는데 이러한 영향을 구명하기위해서 소포의 세포막을 통한 요오드의 선택적 투과전달 특성에 대하여 연구하였다. 이 실험에 사용한 세포막 모델은 피리딘 양이온을 고정전달자로 한 poly(4-vinylpyridine- co-acrylonitrile :VP-AN)분말을 인장강도의 강화용인 polysulfone과 결합시킨 불균질의 복합막을 사용하였다. 고정전달자[VP-AN% (w/ w)]의 농도차이(0~62 %)에서 36.5 ℃로 방사선을 조사한 소포의 고분자 막(세포막모델)은 형태학적 변화인 인장강도가 비 조사된 세포막모델보다 약 1.2 ~ 1.8배 정도 감소되었다. 방사선을 조사한 갑상선의 소포세포막모델 [2mole AN%(w/w)]에서 막의 두께차이, 고정전달자[VP-AN% (w/ w)]의 농도차이, OH-이온의 농도 차이로 발생하는 요오드이온의 선택적 투과플럭스는 비 조사된 세포막모델보다 막의 두께가 변화 할 때 각각 약 2.1 ~ 4.5배, 약 2.2 ~ 2.5배, 약 2.1 ~ 2.67배 정도 감소되었다. 결국 방사선조사로 인해서 갑상선의 소포세포막에서 요오드이온의 선택적 투과 전달기능이 비정상적으로 되어 세포장해가 발생하고 갑상선호르몬의 생성량 감소도 동반하게 되었다고 사료됐다.

Characterization of Carotenoid Biosynthetic Pathway Using Viviparous Mutant Embryos in Maize ( Zea mays L. )

  • Carotenoid compounds in embryos of wild-type(WT) and viviparous mutants of maize(Zea mays L.) were analyzed using high performance liquid ehromatography (HPLC) with a photodiode array detector. Zeaxanthin accumulates in WT embryos as the major carotenoid. Phytoene accumulates in vp2 and vp5. Phytofluene in w3 and ${\xi}$-carotene in the vp9 mutant embryos. This indicates that the vp2 and vp5 mutants impair phytoene desaturase from 15-cis-phytoene to 15-cis-phytofluene. The w3 mutant has neither an isomerase from 15-cis-phytofluene to all-trans-phytofuene nor phytofluene desaturase from phytofluene to ${\xi}$-carotene. The vp9 mutant does not have the ${\xi}$-carotene desaturase from ${\xi}$-carotene to lycopene. Our analysis shows that the terminal carotenoid. ${\gamma}$-carotene(${\beta},{\Psi}$-carotene), accumulates in the vp7 mutant embryos. The ${\varepsilon}$-carotene(${\varepsilon},{\varepsilon}$-carotene), a product of ${\delta}$-carotene(${\varepsilon},{\Psi}$-carotene) in some plants, however, has not been found in maize embryos. The vp7 mutant impairs a cyclization step from ${\gamma}$-carotene to both ${\beta}$-carotene and ${\alpha}$-carotene. We suggest that monocyclic ${\gamma}$-carotene is the sole precursor of both bicyclic ${\beta}$-carotene(${\beta},{\beta}$-carotene) and ${\alpha}$-carotene(${\beta},{\varepsilon}$-carotene) in maize.

Abscisic Acid Binding to Extracts from Normal and Viviparous-1 Mutant Aleurone Layers of Zea mays L.

  • Aleurone layers of normal and vp1 mutant maize kernels were extracted and centrifuged at 100,000g to yield a cytosol fraction. Binding of [3H]ABA cis, trans (+)ABA to a soluble macromolecular components present in the cytosol was demonstrated by Sephadex chromatography and non-denaturing PAGE. The binding component was of high molecular weight and seems to be an aggregate of proteins. A rapid DEAE-cellulose filter method for assaying bound [3H]ABA to a soluble protein was adapted. Binding assays were performed with cytosol that had been preheated or incubated with several enzymes, indicating that heat and protease treatments disrupted the binding. This suggested that binding occurred to proteins. Some properties of the ABA binding proteins were described. The [3H]ABA binding were reduced dramatically when unlabeled ABA was added as a competitor, suggesting a specific binding of [3H]ABA. Gel filtration profiles and autoradiogram of [3H]ABA binding showed no difference in the binding components of Vp1 and vp1/vp1 mutant cytosol, indicating that Vp1 protein is not a sole ABA binding protein.

Surface Micelle Formation of Polystyrene-b-Poly(2-vinyl pyridine) Diblock Copolymer at Air-Water Interface

  • We have studied the surface micelle formation of polystyrene-b-poly(2-vinyl pyridine) (PS-b-P2VP) at the air-water interface. A series of four PS-b-P2VPs were synthesized by anionic polymerization, keeping the PS block length constant (28 kg/㏖) and varying the P2VP block length (1, 11, 28, or 59 kg/㏖). The surface pressure-area ($\pi$-A) isotherms were measured and the surface morphology was studied by atomic force microscopy (AFM) after Langmuir-Blodgett film deposition onto silicon wafers. At low surface pressure, the hydrophobic PS blocks aggregate to form pancake-like micelle cores and the hydrophilic P2VP block chains spread on the water surface to form a corona-like monolayer. The surface area occupied by a block copolymer is proportional to the molecular weight of the P2VP block and identical to the surface area occupied by a homo-P2VP. It indicates that the entire surface is covered by the P2VP monolayer and the PS micelle cores lie on the P2VP monolayer. As the surface pressure is increased, the $\pi$-A isotherm shows a transition region where the surface pressure does not change much with the film compression. In this transition region, which displays high compressibility, the P2VP blocks restructure from the monolayer and spread at the air-water interface. After the transition, the Langmuir film becomes much less compressible. In this high-surface-pressure regime, the PS cores cover practically the entire surface area, as observed by AFM and the limiting area of the film. All the diblock copolymers formed circular micelles, except for the block copolymer having a very short P2VP block (1 kg/㏖), which formed large, non-uniform PS aggregates. By mixing with the block copolymer having a longer P2VP block (11 kg/㏖), we observed rod-shaped micelles, which indicates that the morphology of the surfaces micelles can be controlled by adjusting the average composition of block copolymers.

Carboxylated Polysulfon/MeVpl-DVB 막의 제조와 특성에 관한 연구 (A Study on the Preparation of Carboxylated Polysulfone/MeVpI-DVB Membranes and Its Characteristics)

  • 본 연구에서는 고분자 주사술에 카르복실기를 도입하는 방법으로서 직접 리튬화 반응을 이용하여 polysulfone을 카르복실화(CPSf)한 후, 음이온에 대해 선택성이 있는 것으로 알려진 pyridinium 양이온을 고정전달자로 한 poly(1-alkyl-4-vinylpyridinium iodide-codivinylbenzene) (MeVpI-DVB)을 합성하고, 이들을 혼합한 고분자 분리막을 제조하여 특성화 한 후, 막의 열적특성 및 투과특성을 고찰하였다. CPSf와 MeVpI-DVB의 합성으로 혼성 반응 생성물이 아닌 blend가 형성되었으며, MeVpI-DVB에 대해 CPSf의 함량이 많을수록 열적안정도가 우수함을 알 수 있었고, 합성막은 종류에 따라 이온교환용량은 1.0~1.8(meq/g dry mem.), 함수율은 0.16~0.26(g $H_{2}O$/g dry mem.), 고정이온농도는 6.4~7.3(meq/g $H_2{O}$)의 범위를 보였다. $Cl^-$의 플럭스 변화는 CPSf 함량이 많을수록 증가하는 경향을 나타내었다.
