Hepatitis B virus X protein enhances NFκB activity through cooperating with VBP1

  • Hepatitis B virus X protein (HBx) is essential for hepatitis B virus infection and exerts a pleiotropic effect on various cellular machineries. HBx has been also demonstrated as an indirect transcriptional transactivator of various different viral and cellular promoters. In addition, HBx is involved in the development of various liver diseases including hepatocellular carcinoma. However the mechanism of HBx in hepatocellular carcinogenesis remains largely unknown. In this study, to identify possible new cellular proteins interacting with HBx, we carried out yeast two-hybrid assay. We obtained several possible cellular partners including VBP1, a binding factor for VHL tumor suppressor protein. The direct physical interaction between HBx and VBP1 in vitro and in vivo was confirmed by immunoprecipitation assay. In addition, we found that VBP1 facilitates HBx-induced $NF{\kappa}B$ activation and cell proliferation. These results implicate the important role of HBx in the development of hepatocellular carcinoma through its interaction with VBP1.

Effects of far infrared radiological materials on immune response and changes of fecal microorganism in pigs (돼지에 있어 원적외선 방사물질의 첨가가 면역반응 및 분중 미생물의 변화에 미치는 영향)

  • This research was conducted to investigate the effects of far infrared radiological materials on growth performance, immune response and changes of microbial flora in feces for growing pigs. Twelve growing pigs($Duroc{\times}Yorkshire{\times}Landrace$, average initial body weight of $15.6{\pm}0.5kg$) were used in a 40 day growth assay. Treatments were control(saline injection with diet: S); saline injection with Bio-Plus in diet(SBP); and vaccination with Bio-Plus in diet(VBP) in a randomized complete block design with initial BW as the blocking criterion. Serum concentrations of IgG of SBP and VBP were higher than those of S at day 10(p<0.05), 20(p<0.05), 30(p<0.05) and 40(p<0.05). Pigs fed with treatment diets had increased lymphocyte level compared to S at day 20(p<0.05) and 40(p<0.05). Cortisol was lower in treatments than in S at day 30(p<0.05). At day 20, there was no significant difference in E. coli among the treatments. However, it was observed that E, coli of the treatments was decreased compared to S at day 40(p<0.01). Lactobacillus of SBP was significantly higher(p<0.05) than that of S at day 40. In conclusion, the results of the experiment suggest that far infrared radiological materials could be a very beneficial immune response for growing pigs in health aspects.

Experimental study of strength characteristics of reinforced broken rock mass

  • As the structure of broken rock mass is complex, with obvious discontinuity and anisotropy, it is generally necessary to reinforce broken rock mass using grouting in underground construction. The purpose of this study is to experimentally investigate the mechanical properties of broken rock mass after grouting reinforcement with consideration of the characteristics of broken rock mass (i.e., degree of fragmentation and shape) and a range of reinforcement methods such as relative strength ratio between the broken rock mass and cement-based grout stone body (λ), and volumetric block proportion (VBP) representing the volumetric ratio of broken rock mass and the overall cement grout-broken rock mass mixture after the reinforcement. The experimental results show that the strength and deformation of the reinforced broken rock mass is largely determined by relative strength ratio (λ) and VBP. In addition, the enhancement in compressive strength by grouting is more obvious for broken rock mass with spherical shape under a relatively high strength ratio (e.g., λ=2.0), whereas the shape of rock mass has little influence when the strength ratio is low (e.g., λ=0.1). Importantly, the results indicate that columnar splitting failure and inclined shear failure are two typical failure modes of broken rock mass with grouting reinforcement.

The Results of Radiation Therapy Alone vs Radiation Plus Chemotherapy of Uterine Cervix Cancer (자궁경부암에서 방사선 단독요법 및 항암화학과 방사선 병용요법의 치료성적)

  • Pupose: Radiation therapy(RT) is conventionally standard treatment for locally advanced stage for uterine cervix cancer. Recently to improve treatment results, combined chemotherapy and radiation therapy was tried We retrospectively analysed our experience of 122 patients. Comparision of the results in 45 patients treated with RT alone and 77 patients treated with RT plus chemotherapy was made Materials and Mathods: From January 1985 to December 1991 122 patients with cervix cancer were treated with whole pelvic external RT and ICR(34 1 ICR, 77 2 ICR, 11 high dose rate ICR) in our department. Forty five patients were treated with RT alone, and 77 patients were treated with combined RT plus chemotherapy Mean age was 58 years(range:29-81). Histologic types were 111 squamous cell carcinoma, 5 large cell carcinoma, 3 adenocarcinoma, and 2 adenosquamous cell carcinoma. According to the FIGO stage 6 had stage $IA(4.9\%),$ 11 had $IIA(9.0\%),$ 37 had $IIB(30.3\%),$ 3 had $IIIA(2.5\%),$ 63 had $IIIB(51.6\%).$ and 2 had stage $IV(1.6\%).$ In 77 patients with RT Plus chemotherapy, 36 patients were treated with VBP(vinblastin, bleomycin, cisplatinum) , 39 patients with cisplatinum plus 5-FU and 2 patients with 5-FU. Results: Complete response after external RT (3960cGy-5500cGy) was achieved in 61 patients$(50\%).$ The actuarial 5 year and 9 rear survival rate was $57.8\%\;and\;53.9\%,$ respectively. Five rear actuarial survival rate was $63.1\%$with RT alone(n=45) and $55.9\%$ with RT plus chemotherapy(n=77). The 5 rear survival rate was $35.5\%$ for 1 course of ICR and $67\%$ for 2 courses of ICR. There was statistically significant advantage of survival with RT alone group who were treated with 2 courses of ICR and dose to the A Point)=8000cGy (4/25 died). In RT plus chemotherapy group, dose response was not seen and there was no difference in 5 year survival between 1 course and 2 course of ICR $(50\%\;vs\;56.8\%),$ and dose to point A less than 8000 cGy and more than 8000 $cGy(55.6\%\;vs\;55.7\%).$ There was no significant difference in survival between RT alone and RT plus chemotherapy for patients with tumor size greater than 3cm in size. Five year survival rate for early stage (Stage IB and IIA) with RT alone group and with RT Plus chemotherapy group was $60\%\;and\;77.0\%,$ respectively In advanced stage (stage IIB, IIIA, IIIB, IVA) the 5 year actuarial survival rate were $62.6\%,$ for RT alone group vs $53.6\%$ for RT plus chemotherapy group. Conclusion: Present study demonstrates that there is no survival advantage with adding chemotherapy in advanced stage of uterine cervix cancer. RT alone is considered as treatment of choice for patients with locally advanced cervix cancer. There was increased survival in RT alone group treated with RT dose above 8000 cGy to point A and 2 course of ICR. but 2 course of ICR and RT dose above 8000 cGy to point A did not affect survival advantage in RT plus chemotherapy group.
