  • We investigated whether three flavonoids including daidzein, baicalein, hesperidin and ursolic acid, a triterpene acid, affect proliferation of MCF-7 human breast cancer cells stimulated by estrogenic compounds. Ursolic acid and baicalein inhibited proliferation of MCF-7 cells induced by PhIP, a food-derived carcinogen with estrogenic activity. Daidzein and hesperidin inhibited estradiol-induced proliferation of MCF-7 cells. These compounds should be further investigated for the possible involvement in signaling pathway after estrogen receptor binding in breast cancer cells.

  • Triterpenoid saponins 유도체인 OA와 UA가 치아우식증 원인균종의 하나인 Streptococcus downei의 표준균주(ATCC $33748^T$) 및 임상균주(4주)에 대하여, 액체배지희석법을 이용한 MIC값 측정 및 time-kill 분석법을 통한 살균작용 유무를 측정하여 다음과 같은 결과를 얻었다. 1. S. downei ATCC $33748^T$ 및 S. downei 임상균주(4주)들의 OA 대한 MIC 값은 $8{\mu}g/ml$로 동일하였고, UA의 대한 MIC 값은 S. downei 표준균주와 임상균주 간에 차이가 있었다. 2. Time-kill 분석법을 실시한 결과 OA와 UA는 S. downei $33748^T$에 대하여 $2{\times}MIC$ (각각 $16{\mu}g/ml$$8{\mu}g/ml$) 이상의 농도에서 살균작용을 갖는다는 것을 알 수 있었다. 이상의 연구결과를 종합할 때, triterpenoid saponins 유도체인 OA와 UA는 뮤탄스 연쇄상구균 중 최근에 사람의 구강 내 치면세균막에 서식하는 것으로 밝혀진 S. downei,에 대해 살균제로 작용할 수 있음을 알 수 있었다.

  • Angiogenesis refers to the formation of new capillary blood vessels, or neovascularization occurring under various physical conditions, such as development of the embryo, formation of corpus luteum, wound healing and pathological conditions including tumor growth and metastases, hemangiomas, diabetic retinopathy, rheumatoid arthritis. There are many evidences that angiogenesis is important for the progressive growth of solid tumors and also permits the shedding of metastatic tumors from the primary site. Thus, treatment of angiogenesis inhibitors might be a novel strategy for tumor growth inhibition. Normal vascular endothelial cells are in a state of differentiation and angiogenic endothelial cells migrate and proliferate, and they subsequently differentiate into vessel-forming quiescent phenotype cells, Therfore, it was speculated that a modifier of cell differentiation could also affect angiogenesis. In order to identify new antiangiogenic factors, the research was conducted to estimate the inhibitory activities of cell differentiation agents by means of chick embryo chorioallantoic membrane(CAM) assay. Hence, we have established the CAM assay for the screening of antiangiogenic agents. Using the CAM assay, we found that ursolic acid, a tumor cell differentiation-inducing agent, showed a markedly inhibitory effect on chick embryonic angiogenesis.

조구등의 성분 (The Constituents of Uncaria Hooks)

  • Chemical constituents of Uncaria hooks were studied. Four compounds were isolated from the ether soluble fraction by chromatographic purification process. They were ident ified as isocorynoxeine-N-oxide, anti-isorhynchophylline-N-oxide, physcion and ursolic acid by spectral evidences. Their contents analyzed by HPLC or GC method were physcion 0.04%, ursolic acid 0.26%, isocorynoxeine-N-oxide 0.04% and anti-isorhynchophylline-N-oxide 0.03% respectively. Isocorynoxeine-N-oxide was not reported in nature so far.

  • The diethyl ether extract of isodon excisus var. coreanus exhibited significant inhibitory activity in aromatase assay. Bioactivity-guided fractionation of the extract led to the isolation of three active compounds: inflexin(ent-1${\alpha}$-hydroxy-3${\beta}$,6a-diacetoxykau r-16-en-11,15-dione) (1), ursolic acid (2), and ursolic acid 3-O-acetate (3).

Antimicrobial Activity of Oleanolic Acid, Ursolic Acid, and Sophoraflavanone G against Periodontopathogens

  • In general, oleanolic acid (OA) and ursolic acid (UA) have antimicrobial effect against Gram-positive bacteria but not Gram-negative bacteria whereas sophoraflavanone G has antimicrobial activity against both bacterial types. However, the antimicrobial effects of OA, UA, and sophoraflavanone G against periodontopathogens have not been studied to any great extent. The aim of this study was to investigate antimicrobial effect of OA, UA, and sophoraflavanone G against 15 strains (5 species) of oral Gram-negative bacteria, which are the major causative bacteria of periodontal disease. The antimicrobial activity was evaluated by minimal inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) determinations. OA and UA showed antimicrobial effects against all of the Porphyromonas gingivalis strains tested and also Prevotella intermedia ATCC $25611^T$. Interestingly, P. intermedia ATCC 49046 showed greater resistance to OA and UA than P. intermedia ATCC $25611^T$. In contrast, sophoraflavanone G had antimicrobial activity against all strains, with MIC and MBC values below $32{\mu}g/ml$, except Aggregatibacter actinomycetemcomitans. These results indicate that sophoraflavanone G may have potential for use in future oral hygiene products such as dentifrices and gargling solution to prevent periodontitis.

Enhancement of Platelet Aggregation by Ursolic Acid and Oleanolic Acid

  • The pentacyclic triterpenoid ursolic acid (UA) and its isomer oleanolic acid (OA) are ubiquitous in food and plant medicine, and thus are easily exposed to the population through natural contact or intentional use. Although they have diverse health benefits, reported cardiovascular protective activity is contentious. In this study, the effect of UA and OA on platelet aggregation was examined on the basis that alteration of platelet activity is a potential process contributing to cardiovascular events. Treatment of UA enhanced platelet aggregation induced by thrombin or ADP, which was concentration-dependent in a range of $5-50{\mu}M$. Quite comparable results were obtained with OA, in which OA-treated platelets also exhibited an exaggerated response to either thrombin or ADP. UA treatment potentiated aggregation of whole blood, while OA failed to increase aggregation by thrombin. UA and OA did not affect plasma coagulation assessed by measuring prothrombin time and activated partial thromboplastin time. These results indicate that both UA and OA are capable of making platelets susceptible to aggregatory stimuli, and platelets rather than clotting factors are the primary target of them in proaggregatory activity. These compounds need to be used with caution, especially in the population with a predisposition to cardiovascular events.
