Effects of Nitrogen Sources and C/N Ratios on the Lipid-Producing Potential of Chlorella sp. HQ

  • Microalgae are being researched for their potential as attractive biofuel feedstock, particularly for their lipid production. For maximizing biofuel production, it is necessary to explore the effects of environmental factors on algal lipid-producing potential. In this study, the effects of nitrogen (N) sources (NO2-N, NO3-N, urea-N, NH4-N, and N-deficiency) and carbon-to-nitrogen ratios (C/N= 0, 1.0, 3.0, and 5.0) on algal lipid-producing potential of Chlorella sp. HQ were investigated. The results showed that for Chlorella growth and lipid accumulation potential, NO2-N was the best amongst the nitrogen sources, and NO3-N and urea-N also contributed to algal growth and lipid accumulation potential, but NH4-N and N-deficiency instead caused inhibitory effects. Moreover, the results indicated that algal lipid-producing potential was related to C/N ratios. With NO2-N treatment and carbon addition (C/N = 1.0, 3.0, and 5.0), total lipid yield was enhanced by 12.96-20.37%, but triacylglycerol (TAG) yields decreased by 25.52-94.31%. As for NO3-N treatment, carbon addition led to a 17.82-57.43%/25.86-82.67% reduction of total lipid/TAG yields. When NH4-N was used as the nitrogen source, total lipid/TAG yields were increased by 46.67-113.33%/28.99-74.76% with carbon addition. The total lipid/TAG yields of urea-N treatment varied with C/N ratios. Overall, the highest TAG yield (TAG yield: 38.75 ± 5.21 mg/l; TAG content: 44.16 ± 4.35%) was achieved under NO2-N treatment without carbon addition (C/N = 0), the condition that had merit for biofuel production.

A Study on the Behavior of Surface-Applied Urea with $^{15}N$ Isotope Dilution Technique in Paddy Soil (논토양에서 중질소(N-15)를 이용한 표면시용 요소로부터 유래하는 질소의 행동에 관한 연구)

  • The pot experiment using $^{15}N$ isotope dilution technique was carried out to calculate the balance of nitrogen of surface applied urea in the rice-soil system. The $^{15}N$ concentration was determined by stable isotope ratio mass spcetrometer (model: VG ISO-GAS MM622). In the pots with $^{15}N$ labeled urea application at the rates of 15 and 30 kg N/10a, the percentage of nitrogen derived from fertilizer (NDFF) in rice was higher at the rate of 30 kg N/10a (average 89%) than at the rate of 15 kg N/10a (average 64%). However, the recovery as percentage of fertilizer N by rice was higher at the rate of 15 kg N/10a (65.5%) than at the rate of 30 kg N/10a (54.2%). The percentage of the fertilizer N remained in extractable inorganic N form at the rates of 15 and 30 kg N/10a were $13.5%\;(NH_4-N\;5.53%,\;NO_3-N\;7.99%)$ and $16.5%\;(NH_4-N\;7.49%,\;NO_3-N\;8.98%)$ in unplanted soil, and $2.0%\;(NH_4-N\;0.63%,\;NO_3-N\;1.32%)$ and$2.3%\;(NH_4-N\;0.87%,\;NO_3-N\;1.40%)$ in soil planted to rice, respectively. The dominant form of inorganic-N in soil after harvest was $NO_3-N$ form rather than $NH_4-N$ form regardless of urea application rate or rice cultivation. The percentage of the fertilizer N remained in organic N form at the rates of 15 and 30 kg N/10a were 65.0 and 41.8% in unplanted soil, and 23.7 and 26.9% in soil planted to rice, respectively. In conclusion, the efficiency of surface-applied urea was greater at the rate 15 kg N/10a than at the rate of 30 kg N/10a.

  • The popularity of diesel engines is derived primarily from their higher thermal efficiency resulting from higher compression ratio. NOx removal from the diesel emissions is very important to meet stringent emission regulations. NOx emission from diesel engines is removed by an urea-SCR or an LNT system. The urea-SCR system needs the urea-solution supply system with concentration and level sensor. This study was carried out to develop a sensor for the measurement of urea-solution concentration by an electric conductivity method. Considering experimental parameters were the material of electrode, two kinds of electric power(AC or DC), the distance between two electrodes, and the length of electrode. It was found that the AC electric power was more useful to measure the urea-solution concentration compared to DC, because it prevented an ionization of the urea-solution. The silver rod coated with Pt is the most useful electrode, tendency of which is similar to Pt rod, and the cost is more economic. We could also find out the optimum distance between two electrodes and the length of electrode was 10mm and 3mm, respectively.

  • The surfaces of sheets added with N-chloro-polyacrylamide (N-Cl-PAM) are analyzed using X-ray photoelectron spectroscopy (XPS) to clarify the chemical bonding involved in the paper strength development induced by N-Cl-PAM. The comparison of the observed N1s chemical shift of the sheet with those of the paper strength additives and the model compound, 1-butyryl-3-propyl urea, illustrated the presence of covalent bonds of alkyl acyl urea and urethane on the fiber surfaces. Thus the formation of the covalent bonds by N-Cl-PAM themselves and by N-Cl-PAM with cellulose and hemicellulose may be an explanation for much higher effectiveness of N-Cl-PAM on the improvement of wet strength of paper than A-PAM.

Decomposition and N Release of Hairy Vetch Applied as a Green Manure and its Effects on Rice Yield in Paddy Field

  • Decomposition of green manure is necessary for the nutrient supply in farm soil. Hairy vetch as a green manure is superior to other winter legumes in terms of wintering ability and N accumulation. This experiment was carried out to investigate the decomposition and N release of hairy vetch and its effect on rice production as the following crop in paddy field. Decomposition of hairy vetch placed by soil depth of 0, 10 and 20cm at transplanting time was investigated by mesh bag method, which was enclosed chopped residue in mesh bags. The fate of $^{15}$ N derived from $^{15}$ N-labeled hairy vetch was investigated at harvest in three levels of N fertilization. Grain yield of the transplanted paddy rice cultured with hairy vetch as starter N were compared with that of applying urea as starter N in the field. Hairy vetch residue decomposed very rapidly both in transplanted and dry-seeded paddy field. In transplanted paddy field, hairy vetch residue lost 72-81 % and 86-90% of its weight after one and five month, respectively, as affected by incorporation depth. The C/N ratio of the decomposing vetch residue increased sharply during the early stages and after then, decreased slowly. The amounts of N and P released from the vetch were about 90% and 97% of initial content after one month, respectively. Recoveries of hairy vetch-$^{15}$ N by rice plant were 30.6, 34.6 and 35.7% in 0, 6 and 12 kg urea-N 10 $a^{-l}$ application, respectively, indicating that N fertilization increased the recovery of hairy vetch. $^{15}$ N. Hairy vetch residue incorporated as starter maintained significant N $H_4$$^{+}$-N concentration in soil water of plow layer until effective tillering stage. Grain yield in the plot applied with hairy vetch was not significantly different from that in the plot with urea. We concluded that hairy vetch incorporation could substitute starter N fertilization and showed possibility to reduce N amount of top-dressing.g.g.

Effects of Application of Slow-Released Nitrogen Fertilizer Using Waste Paper Slurry on the Growth and Yield of Rice and Chemical Properties of Soil (폐지섬유를 이용한 완효성 요소비료 시용이 벼의 생육, 수량 및 토양에 미치는 영향)

  • The aim of this study was to investigate the effects of slow-released nitrogen fertilizer(SRNF) on the growth and yield of rice. SRNF produced from wasted paper was applied to a clay loam paddy field comparing to urea fertilized field and only P-and K-fertilized field. Some agronomic components like as growth development and yield component were observed and physico-chemical properties of the soils were analyzed. Plant height and tiller numbers per hill showed higher in rice plant treated with SRNF than in one treated with urea at the early grow stage whereas they appeared to be all much the same at the end of growth stage. While the chlorophyll content in SRNF-treated rice shoot was higher than in urea-treated one, the photosynthetic activity in urea-treated rice shoot was slightly higher than in SRNF-treated rice. In harvested grain, the nitrogen content was higher than in SRNF treated rice than in urea treated rice, but in straws the content was less. At the harvesting stage, nitrogen uptake in grains was about 4% higher in SRNF-treated rice than in urea treated rice whereas in straws rather 20% lower. The N efficiency in SRNF treated rice was lower than in urea treated rice. In the soils treated with SRNF, pH, organic matter and phosphorus were higher than in the soils treated with urea. Total N content in SRNF treated soil was lower after experiment than in urea treated soil.

  • A series of waterbome poly(urethane-urea) anionomers were prepared from isophorone diisocyanate (IPDI), polycaprolactone diol (PCL), dimethylol propionic acid (DMPA), ethylene diamine (EDA), and triethylamine (TEA), NaOH, or Cu($(COOCH_3)_2$) as neutralizing agent. This study was performed to decide the effect of neutralizing agent type on the particle size viscosity, hydrogen bonding index, adhesive strength, antistaticity, antibacterial and mechanical properties. The particle size of the dispersions decreased in the following order: TEA based samples (T-sample), NaOH based samples (N-sample), and Cu($(COOCH_3)_2$) based sample (C-sample). The viscosity of the dispersions increased in the order of C-sample, N-sample, and T-sample. Metal salt based film samples Of and C-sample) had much higher antistaticity than TEA based sample. By infrared spectroscopy, it was found that the hydrogen bonding index (or fraction) of samples decreased in the order of T-sam-pie, N-sample, and C-sample. The adhesive strength and tensile modulus/strength decreased in the order of T-sample, N-sam-pie, and C-sample. The C-sample had strong antibacterial halo, however, T- and N-samples did not

A Study on Nitrogen Intake and Excretion in Young Korean Adult Women on Normal Diet (정상식이를 섭취하는 여대생들의 질소 섭취 및 배설에 관한 연구)

  • A study was conducted to measure nitrogen intake and excretion in young korean women on their normal diet and leading normal activity level. Dietary survey by food record, fecal and urinary samples were collected for 3 days in 19 healthy female college students in Korea. On one of the 3 days, duplicate of one-day diet was collected from each subjects. Mean daily intakes of energy and protein were calculated from food recorded. Duplicate diet samples and pooled fecal samples were analyzed for N content. Pooled urine samples were analyzed for total N, urea N, and creatinine content. Apparent N absorption, apparent N balance and urea N/creatinine N were calculated to evaluate protein nutritional status. The results obtained are summarized as following ; 1) Accordingly to food record, mean daily intakes of energy, protein, carbohydrate and far were 1535.2 $\pm$53.78Kcal, 55.95$\pm$2.97g(total nitrogen 8.95$\pm$0.45g), 254.13$\pm$10.31g and 39.24$\pm$2.76 g, providing 14.6%, 66.2%, 19.2% of total energy respectively. 2) Nitrogen intake by chemical analysis was 7.16$\pm$0.31g/day (protein 44.75$\pm$1.94kg/day) providing 82.39$\pm$4.58% of nitrogen intake by food record. The difference of total nitrogen intake between food record and chemical analysis in diets was significant(p<0.05). 3) Mean daily fecal nitrogen excretion was 1.38$\pm$0.10g and then mean apparent digesbility of protein was 80.53$\pm$5.21%. 4) Mean daily urinary nitrogen excretion, urea N excretion and creatinine excretion were 6.03$\pm$0.30g, 4.52$\pm$0.22g, and 0.88$\pm$0.04g respectively. Urinary urea nitrogen was 75.2$\pm$1.38% of total urinary nitrogen excretion and urinary nitrogen was 85.4$\pm$3.56% of total nitrogen intake by chemical analysis. 5) Mean urea N/creatinine N ration was 14.01$\pm$0.77. 6) Mean nitrogen balance was -0.244$\pm$0.33g/day. From the above results, it is concluded that the subjects in this study seem to be in marginal protein nutritional status and therefore should increase dietary protein intake.

Effects of Slow-Release Nitrogen Fertilizers, Latex coated urea and Meister 10, on Direct Seeded Rice in Flood Soil (벼 담수직파재배시 완효성질소비료의 시용 효과)

  • This study was conducted to find out the effect of slow-release nitrogen fertilizer, such as latex coated urea(LCU) and Meister10(MS10) on direct seeded rice in flooded soil(FS). Junghwabyeo, and early maturity rice cultivar grown on the plots which were treated with None-nitrogen, urea, LCU and MS10 plot. Growth characteristic, yield and yield components were investigated. Nitrogen uptake-efficiency and physico-chemical properties of soil before-after experiment were analyzed. Plant height and number of tillers $m^{-2}$ in LCU and MS10 plot at early grow stages were higher than those of Urea plot. MS10 and LCU plot showed similar tendency on the plant height but MS10 plot was higher than LCU plot on the number of tillers $m^{-2}$. The number of seedling $m^{-2}$ were no significant differences among None-N, urea, and MS10 plot. Heading date and leaf color were no significant differences among None-N, urea, and MS10 plot. Culm length in LCU and Ms10 plot were longer compared with urea plot, but panicle length were similar among with Urea, LCU and MS10 plot. Number of panicles $m^{-2}$ was highest in MS10 plot and it was similar between LCU and Urea plot. Yield were greater in order of Ms10 > LCU > Urea plot. Nitrogen uptake and nitrogen efficiency were greater in order of MS10 > LCU > urea plot. After the experiment, total content of nitrogen in soil was not changed at all treatments, but pH, P and K of soil were lower than that of before experiment at all treatments.
