The nucleotide sequence of the xylanase gene (xynY) from alkalophilic Bacillus sp. YC-335 was determined and analyzed. An open reading frame of 1, 062 base pairs for xynY gene was observed and encoded for a protein of 354 amino acids with a molecular weight of 38, 915. S1 nuclease mapping showed that the transcription initiation sites of the xynY gene were different in Bacillus sp. YC-335 and Escherichia coli HB101 (pYS55). S1 mapping also showed that -10 region of the xynY gene recognized by RNA polymerases of E. coli and Bacillus sp. YC-335 were TACAGT and TATGAT , respectively. A ribosome binding site sequence with the free energy of -17.0 Kcal/mol was observed 9 base pairs upstream from the unusual initiation codon, TTG. The proposed signal sequence consisted of 27 amino acids, 2 of which were basic amino acid residues and 21 were hydrophobic amino acid residues. When the amino acid sequences of xylanases were compared, Bacillus sp. YC-335 xylanase showed more than 50% homology with xylanases from B. pumilus, B. subtilis, and B. circulans.
This study was conducted to examine how a low-head dam removal (partial removal) could affect benthic macroinvertebrate communities in a stream. Benthic macroinvertebrates and substrates were seasonally sampled before and after dam removal (March 2006-April 2007). Benthic macroinvertebrates and substrates were quantitatively sampled from immediately upstream (upper: pool) and downstream (lower: riffle) sites, the location of the dam itself (middle), and immediately above the impoundment (control: riffle). After the removal, species richness and density of benthic macroinvertebrates as well as the EPT group (Ephemeroptera, Plecoptera and Trichoptera) increased to higher levels than those before the removal, while functional feeding groups and habitat orientation groups changed more heterogeneously at the upper site. At the lower site, species richness and density decreased somewhat immediately after dam removal, which was associated with an increase of silt and sand, but recovered after monsoon floods which helped to enhance substrate diversity at the upper site. Decreased dominance index and increased diversity index in both the upper and lower sites are evidence of positive effects from the dam removal. In conclusion, we suggest that even a partial removal of a dam, resulting in increased substrate diversity in the upper site, could sufficiently help rehabilitate lost ecological integrity of streams without major habitat changes.
본 연구에서는 동천의 하류와 연결된 호계천의 수질환경현황 및 문제점을 분석하여 하천의 실태를 파악하고, 수질 및 악취를 비롯하여 병원미생물제거를 위한 친환경적이며 가장 경제적인 방안을 제시하고자 하였다. 조사 정점은 부산시 동구 범일동 호계천 수질에 대하여 장마철을 포함하여 지난 수개월 동안에 걸쳐 선정된 3개 정점에 대하여 미생물제제의 투입전후의 수질과 악취변화를 분석하였다. 활성화된 미생물제제는 $0.042m^3$/시간의 속도로 4일간 지속적으로 유지하여 처리하였고 약 2개월에 걸쳐 7-10일 간격으로 실시하였다. 제제처리 전후의 DO 및 오염물질별 단위시간당 평균총량을 계산하여, 제제처리 전후의 DO 및 오염물질별 제거효율을 분석한 결과 용존산소는 중류 및 하류의 경우 상류(대조군)에 비해서 1.7배 정도의 높은 농도를 나타내었다. 이는 하천의 정화작용으로 인해 회복이 됨을 의미한다. COD의 경우는 중류 및 하류에 있어서 가장 높은 제거효율을 보여 대조군에 비해 약 2배의 제거효율을 보였다(약 60%에 육박). 그리고 제제처리 전에 비해 제제처리 후 상류지점의 악취에 비해 중류지점의 악취감소가 평균 약 65% 정도 감소가 나타났으며, 하류지점의 악취감소는 평균 약 19% 정도 감소하였다. 따라서 미생물제제처리에 의한 악취저감이 현저함을 알 수 있었다. 미생물군집의 종(Species) 수준에서는 전반적으로 Hydrogenophaga caeni, Sphaerotilus natans, Acidovorax radicis, Acidovorax delafieldii 및 Cloacibacterium rupense이상위우점종으로 나타났으며 병원균인 Arcobacter cryaerophilus는 제제처리 후 중류에서는 제거되는 효과를 보였다.
This study was to analyze fish composition and ecological indicator characteristics from eight sampling sites of Geum River, October 2007. Total number of family and species sampled were 9 and 40. The most dominant family was Cyprinidae (27 species, 85%), and then followed by Cobitidae, Odomtobutidae, and Gobiidae. Constancy values of Zacco platypus and Zacco temminckii were 1.00 and 0.30, respectively, and the relative abundance of Acheilognathus koreensis and Pseudogobio esocinus were greater than 5% of the total. The number of Korean endemic species sampled in this Geum River study was 7 family 19 species, which is 47.5% of total 40 species, and endangered species of Pseudopungtungia nigra and Gobiobotia brevibarba were only 0.5% of the total and these species were only distributed within the upstream regions. Exotic species, Micropterus salmoides, which is known as large-mouth bass, were observed in two sites of G3 and G7. Analytical results of fish community showed that community dominance index was 0.19, which is low, and the species evenness index (0.74), diversity index (2.03), and richness index (3.00) appeared high. These results indicate that structure of fish community is stable in the Geum River. According to various guilds analysis, the relative abundance of tolerant and omnivores at all sites were 40% and 47% of the total, respectively. This monitoring data may contribute changes of fish fauna and compositions in relation to habitat modifications and chemical water quality degradations in the future.
In order to use fundamental data for conservation of species diversity and stream health with dam development, fish investigation in up- and downstream of Pyeonglim Dam was seasonally conducted in 2007~2008. In addition, data of water qualities, investigated for environmental impact assessment, were used to determine a water quality standard level in Pyeonglim Stream, and it revealed that water environment condition was good in the stream. During the study period, a total number of fish caught from the 4 study sites was 5,449 representing 9 families 25 species, and there were 8 Korean endemic species (32.0%) including Rhodeus uyekii, Squalidus gracilis majimae, and Microphysogobio yaluensis. Two species of Zacco temminckii (37.5%) and Zacco platypus (26.3%) were dominant and subdominant in all sites. There were few individuals of the $2^{nd}$-class endangered species (Sand lamprey, Lampetra reissneri) and the introduced species (Bluegill, Lepomis macrochirus), found in the upstream of the dam. and further investigation for conservation of the endangered species and for prevention of the introduced species is needed in future. With fish fauna and multi-metric health assessment model in each sampling attempt, index of biotic integrity(IBI) was evaluated and it resulted mostly in good(26~35) and excellent(36~40) condition in all sites. The results indicate that it is very important to study not only environmental impact assessment with fish composition but also stream health assessment in order to conserve healthy aquatic ecosystem.
CBOD(carbonaceous BOD) decay rate was investigated for the execution of water quality model in Nakdong-Rive basin. Estimation of laboratory-derived CBOD decay rate, $k_l$ and CBOD decay rate in natural waters, $k_d$ were carried out. Hydraulic factors were applied for the calculation of $k_d$. Values of biochemical oxygen demand were investigated in Weagwan, Koreong, Jeokpo, Namgi and Mulgeom sites for the four times. The ranges of $k_l$ value were $0.04{\pm}0.01{\sim}0.14{\pm}0.03$. The values of $k_l$ in upstream sites were much larger than those in the downstream sites. The values of $k_d$ were 0.025, 0.036, 0.005 and 0.001 at Weagwan, Jeokpo, Namgi and Mulgeom, respectively, indicating that values of $k_d$ were generally larger than those of $k_l$.

Mammalian urothelium undergoes unique membrane specialization by making the asymmetric unit membrane (AUM) that is covered with the apical cell surface during terminal differentiation. The AUM contains several major integral membrane proteins including uroplakin Ia, Ib, II and III. The genes for uroplakins have been cloned from humans and mice, but not from porcine. In this study, we report the cloning of the UPII genomic DNA, which codes for the full length open reading frame for the uroplakin II protein. The deduced amino acid sequence encodes of a hydrophobic NH$_2$-terminal peptide, a prosequence, and a mature protein. The prosequence contains three potential N-glycosylation sites and a RGRR cleavage site that may be involved in uroplakin II processing and maturation. Northern and immunohistochemistry analyses showed that the porcine UPII gene is only expressed in urothelium and that the protein was specifically localized in urothelial superficial cells. A 2kb of upstream in the promoter sequence contains multiple transcription factor binding sites, including GC-box, SPI, AP2, and GATA-box sites, but not for TATA or CAAT-box sequences. Comparison of the porcine UPII promoter sequence with that of the murine by MEME system presented two conserved motifs, suggesting a cis-acting regulatory role for the conserved sequences. Sequence homology between two species in motif A and B was 79% and 80% respectively, although their relative locations were different. During the gestation, mouse bladder at estrus stages and day 10 after parturition showed higher UPII expression, while showed lower expression at peri-implantation stage. Taken together, our results showed that the porcine UPII gene was expressed highly and specifically in the bladder urothelium and that steroid hormones for implantation changed the expression of UPII in the bladder, although the biological significance of UPII remains to be not determined.
To investigate environmental impacts of inland fish- farms on the water quality of a stream, environmental parameters including physical, chemical and biological factorswere estimated at trount- farm in 1994. Influences( F-1) were polluted as passing through fish tank( F-2) and settle pond( F-3), so organic and inorganic nutrient concentrations became very high in effluents( F-4). BOD$_{5}$, Chl. a, Nitrite and heterotrophic bacterial numbers in the junction site(5-2) of F-4 and S-1( upstream) were highest among the stream sites. BOD$_{5}$ was proved to a good parameter of organic pollution, and nitrite of Inorganic nutrients in the fishery and it's affected stream, The high concentration of BOD$_{5}$ and Chl. a in S-2 were decreased in downstream(5-3, 5-4). It seems that this results are due to dilution or sedimentation effect. This supposition requires further testing about the sediment of stream. The results of this paper demonstrate that fish farming can deteriorate water quality of a stream by changing environmental factors.
This paper presents the evaluation of the impact of climate change on water resources and yield capacity of Asa and Kampe reservoirs. Trend analysis of mean temperature, runoff, rainfall and evapotranspiration was carried out using Mann Kendall and Sen's slope, while runoff was modeled as a function of temperature, rainfall and evapotranspiration using Artificial Neural Networks (ANN). Rainfall and runoff exhibited positive trends at the two dam sites and their upstream while forecasted ten-year runoff displayed increasing positive trend which indicates high reservoir inflow. The reservoir yield capacity estimated with the ANN forecasted runoff was higher by about 38% and 17% compared to that obtained with historical runoff at Asa and Kampe respectively. This is an indication that there is tendency for water resources of the reservoir to increase and thus more water will be available for water supply and irrigation to ensure food security.
Traditionally, small-molecule or antibody-based therapies against human diseases have been designed to inhibit the enzymatic activity or compete for the ligand binding sites of pathological target proteins. Despite its demonstrated effectiveness, such as in cancer treatment, this approach is often limited by recurring drug resistance. More importantly, not all molecular targets are enzymes or receptors with druggable 'hot spots' that can be directly occupied by active site-directed inhibitors. Recently, a promising new paradigm has been created, in which small-molecule chemicals harness the naturally occurring protein quality control machinery of the ubiquitin-proteasome system to specifically eradicate disease-causing proteins in cells. Such 'chemically induced protein degradation' may provide unprecedented opportunities for targeting proteins that are inherently undruggable, such as structural scaffolds and other non-enzymatic molecules, for therapeutic purposes. This review focuses on surveying recent progress in developing E3-guided proteolysis-targeting chimeras (PROTACs) and small-molecule chemical modulators of deubiquitinating enzymes upstream of or on the proteasome.
