Evaluating the Nutritional Composition of Unripe Citrus and Its Effect on Inhibiting Adipogenesis and Adipocyte Differentiation

  • Citrus fruits offer a range of health benefits due to their rich nutritional profile, including vitamin C, flavonoids, carotenoids, and fiber. It is known that unripe citrus has higher levels of vitamin C, dietary fiber, polyphenols, and flavonoids compared to mature fruits. In this study, we assessed the nutritional components of unripe citrus peel and pressed juices, as well as their anti-obesity potential through the modulation of adipocyte differentiation and the expression of adipogenesis-related genes, specifically PPARγ and C/EBPα, in 3T3-L1 preadipocytes. Our analysis revealed that unripe citrus peel exhibited elevated levels of fiber and protein compared to pressed juice, with markedly low levels of free sugar, particularly sucrose. The content of hesperidin, a representative flavonoid in citrus fruits, was 3,157.6 mg/kg in unripe citrus peel and 455.5 mg/kg in pressed juice, indicating that it was approximately seven times higher in unripe citrus peel compared to pressed juice. Moreover, we observed that the peel had a dose-dependently inhibitory effect on adipocyte differentiation, which was linked to a significant downregulation of adipogenesis-related gene expression. Thus, our findings suggest that unripe citrus possesses anti-obesity effects by impeding adipogenesis and adipocyte differentiation, with the peel demonstrating a more pronounced effect compared to pressed juice.

노지재배 풋귤 수확시기가 수량과 과실생육 및 다음해 착화에 미치는 영향 (Effect of the Harvest Season on the Yield and Growth of Unripe Fruit and Biennial Flowering of 'Miyagawa' Satsuma Mandarin in Open Field Cultivation)

  • BACKGROUND: As consumption of unripe mandarin increases, its cultivation has increased in open field cultivation areas. Because unripe mandarin must be harvested before ripening and color change, the optimum harvest time must be determined. This study investigated the effect of the harvest season on the yield of unripe fruit and biennial flowering of 'Miyagawa' satsuma mandarin. METHODS AND RESULTS: Two areas of unripe mandarin orchard were selected, and the yield, fruit growth, working time, and flowering of trees the following year were investigated. Fruit was harvested at 40, 60, 80, 100, and 120 days after full bloom and at general ripening. Fruit yield of unripe mandarin increased with later harvest time from 100th to 120th day except normal ripening. The next year, biennial occurred with normal ripening and harvesting, but not at the 120th day after full bloom. At the 40th day (earliest harvest time), summer and autumn shoots were present, but not after the 100th day. The 40th day required the most harvesting time; because the time gradually decreased with later harvest, the harvest time was shortest on the 120th day, and general ripening occurred shortly after the 120th day. CONCLUSION: Harvesting of unripe mandarin 100-120 days after full bloom was ideal to reduce harvesting time, enhance yield, and enable flowering the following year.

  • We investigated the anti-inflammatory activities of unripe fruit of Citrus grandis Osbeck growing at Jeju Island, through the evaluation of their inhibitory effect on the production of inflammatory markers (IL-6, iNOS, COX, TARC and MDC) in RAW264.7 murine macrophage cells and HaCaT human keratinocyte cells. Among the sequential solvent fractions obtained from crude extract, hexane and chloroform $(CHCI_3)$ fractions showed potential inhibitory activity on the mRNA expressions of IL-6, iNOS and COX-2 at the concentration of $100\;{\mu}g/ml$ in RAW264.7 cells. Also, EtOAc fraction showed inhibitory activity on the thymus and activation-regulated chemokine (TARC)/CCL17 and macrophage-derived chemokine (MDC)/CCL22 at the concentration of $50\;{\mu}g/ml$ in HaCaT cells. These results suggest that the unripe fruit of C. grandis may have anti-inflammatory activity through the suppression of inflammatory markers (IL-6, iNOS, COX, TARC and MDC).

제주 재래종 감귤 ‘홍귤’, ‘편귤’ 과피 분획물의 항산화, 항염증 효과 (Antioxidant and Anti-inflammatory Effects of Solvent Fractions from the Peel of the Native Jeju Citrus ‘Hongkyool’ and ‘Pyunkyool’)

  • 한약재로 사용되고 있는 제주 재래 감귤 중 홍귤, 편귤 품종 과피 추출물의 기능성 소재로서의 활용 가능성을 알아보기 위해 용매별 분획물이 보유한 항산화 및 항염증 활성을 조사하였다. 홍귤, 편귤 모두 부탄올 분획물에서 총 페놀성 화합물의 함량이 각각 534.4 mg/g, 342.9 mg/g으로 가장 높았으며, 그 다음으로 에틸아세테이트, 헥산, 그리고 물 분획물 순이었다. 총 플라보노이드 함량에 있어서도 부탄올 분획물에서 홍귤 431.8 mg/g, 편귤 415.7 mg/g으로 가장 높게 나타났다. DPPH 라디칼 소거활성은 부탄올 분획물 1 mg/ml 에서 홍귤(89.2%), 편귤(64.2%) 모두 가장 높았으며, ABTS 라디칼 소거활성에 있어서도 두 품종 모두 부탄올 분획물에서 낮은 농도에서도 매우 우수한 활성을 보였으며 그 뒤로 에틸아세테이트, 헥산, 물 분획물 순이었다. 항염증 효과 측정에서 홍귤, 편귤 에틸아세테이트 분획물 모두 NO, IL-6, iNOS 및 COX-2 단백질 생성 억제 효과를 확인할 수 있었다. 특히 홍귤 에틸아세테이트 분획물 100 μg/ml 농도에서 항염증 효과가 우수하였다.

미숙감귤의 항산화 및 항노화 활성에 대한 평가 (Evaluation on antioxidative and antiaging effect of immature fruits from Citrus unshiu Markovich)

  • Objective : This study was designed to investigate effect of immature fruits from Citrus unshiu Markovich to mature them on antioxidative and antiaging-related enzyme activities in vitro. Methods : Fruits from Open field-cultivated C. unshiu Markovich (ripe and unripe fruits of Citri) made a purchase in Jeju island. We measured total polyphenol and flavonoid contents in mature C. unshiu Markovich (MC) and immature C. unshiu Markovich (IMC) 70% ethanol extract. DPPH free radical, ABTS radical, and hydroxyl radical scavenging activities in both extracts were determined. Also, reducing power, SOD-like activities, elastase inhibition, and collagenase inhibition activities of these extract were assayed at 5, 1, 0.5, and 0.1 mg/㎖. Results : The total polyphenol contents in the ethanol extract of IMC were higher than those of MC (26.99 ± 0.89 mg/TAEg). And total flavonoid contents in them were very similar to both extract (MC; 7.25 ± 0.4 mg/RUEg and IMC; 7.44 ± 0.18 mg/RUEg). DPPH free radical scavenging activities, ABTS radical scavenging capacity, reducing power, and SOD-like activities of IMC-treated group showed significant increase compared to those of MC group in all treated concentrations. Particularly, DPPH free radical scavenging and elastase inhibition activities in IMC-treated group at 5 mg/㎖ showed similar with positive control group. Besides, IMC extract at 5 mg/㎖ concentration surpassed positive control (BHA or Vit. C) in SOD-like activities and ABTS radical scavenging capacity. Conclusion : These results suggest that IMC ethanol extract may be used as a natural antioxidant and a antiaging material for development of functional foods and cosmeceuticals.

Nutritional Evaluation of Some Tropical Crop Residues: In Vitro Organic Matter, Neutral Detergent Fibre, True Dry Matter Digestibility and Metabolizable Energy Using the Hohenheim Gas Test

  • The Hohenheim in vitro gas test was used to assess the nutritional value of some crop residues of known in vivo digestibility. The crop residues are groundnut shells (GNS) corn cobs (CC); cassava peels (CaP); unripe and ripe plantain peels (UPP, RPP) and citrus pulp/peels (CPP). Compared to other crop residues, crude protein (CP) content of CC was low. Except for CaP and CPP that had low neutral detergent fibre (NDF) and acid detergent fibre (ADF), other residues contained a high amount of cell wall constituents. Net gas production was significantly different among the crop residues (p<0.05). Gas production was highest in CPP followed by CaP. CC, UPP and RPP have the same volume of net gas production, while the least net gas production was in GNS. True dry matter (TDM) digestibility was significantly different (p<0.05) among the residues. GNS was the least in TDM digestibility. CaP, UPP and RPP had similar TDM digestibility values, while the highest TDM digestibility was obtained in CPP. OM digestibility was different among the residues (p<0.05). CaP and CPP had the same ME value while CC, UPP and RPP had close ME values and GNS the least in ME (p<0.05). The potential extent (b) and rate (c) of gas production were statistical different among the residues (p<0.05). The Hohenheim gas test gave high in vitro organic matter (OM) digestibility for CC, CaP, UPP and RPP and CPP. Fermentable carbohydrates and probably available nitrogen in the crop residues influenced net gas production. The results showed that crop residues besides, providing bulk are also a source of energy and fermentable products which could be used in ruminant livestock production in the tropics.
