Intrasporangium속 방선균의 Phenylalanine 분지대사 경로의 조절 (Regulation of Phenylalanine Specific Pathway in a Species of Intrasporangium)

  • 토양에서 분리 동정한 phenylalanine 생산균인 Intrasporangium속 방선균의 phenylalanine 생합성 분지경로의 대사조절 특성을 조사하기 위해 chorismate mutase와 prephenate dehydratase를 부분정제한 결과 chorismate mutase는 prephenate dehydratase와 전혀 별개의 단백질로서 두 종의 isoenzyme, chorismate mutase I (CM I)과 chorismate mutase II (CMII)로 구성되어 있음을 알았다. CMI은 최종대사산물인 L-phenylalanine, L-tyrosine 및 L-tryptophan에 대해 완전내성을 보였으나 CMII는 1.5mM tyrosine에 의해서 약 50% 활성 저해를 나타내었다. 이에 비해 prephenate dehydratase는 0.02 mM phenylalanine 존재하에서 95% 이상의 활성 저해를 나타냈으나 이와 같은 L-phenylalanine 활성 저해효과는 positive effector인 L-tyrosine과 L-methionine에 의해 크게 감소되었다. 또한 chorismate mutase 생합성은 feedback repression을 받지 않는데 비해 prephenate dehydratase는 1mM phenylalanine 존재에 의해 약 94%의 효소합성 저해를 나타냈다. 그러나 5mM tyrosine을 동시에 첨가했을 때는 전혀 저해효과를 인식할 수 없었다. 따라서 Intrasporangium속 방선균 역시 대다수의 다른 미생물 균종과 마찬가지로 phenylalanine 분지 경로에서 prephenate dehydratase에 의해 촉매되는 두번째 반응이 가장 중요한 대사조절 단계가 되고 있다는 것을 알 수 있었다.

The Effect of Reducing Dietary Phytate Intake and Zinc Supplementation on the Iron Status of Elderly Korean Women

  • To investigate the effects of dietary phytate reduction and zinc supplementation on biochemical iron parameters in elderly Korean women consuming inadequate iron, fifteen healthy women aged 64-75 years were recruited for a feeding study. A high-phytate diet (27.8 phytate:zinc molar ratio) was provided for 9 days, followed by a nine-day low-phytate diet(12.3) and a subsequent 28-day period of unregulated meals with zinc supplementation (22 mg/d as zinc gluconate). Serum iron increased significantly with the low-phytate diet (130.4 $\mu g$/L) but returned to the level observed during the high-phytate diet (113.0 $\mu g$ /L) period when subjects were taking zinc supplements (105.8 $\mu g$ /L). However, serum ferritin in the subjects decreased significantly with the low-phytate diet (73.8 $\mu g$ /L) as well as with zinc supplementation (57.2 $\mu g$ /L), compared to levels following consumption of the high-phytate diet (89.6 $\mu g$ /L). Transferrin receptor and transferrin saturation were unchanged with the treatments. In summary, zinc supplementation might result in deteriorated iron status in elderly Korean women who consume inadequate iron, while there was no significant effect from reducing dietary phytate.

Single-Phase Self-Excited Induction Generator with Static VAR Compensator Voltage Regulation for Simple and Low Cost Stand-Alone Renewable Energy Utilizations Part II : Simulation and Experimental Results

  • In this paper, the power conditioner composed of the stand-alone single-phase squirrel cage rotor type self-excited induction generator (SEIG) driven by prime movers such as a wind turbine and a micro gas turbine (MGT) is presented by using the steady-state circuit analysis based on the two nodal admittance approaches using the per-unit frequency in addition to a new state variable defined by the per-unit slip frequency along with its performance evaluations for the stand-alone energy utilizations. The stande-alone single-phase SEIG operating performances in unregulated voltage control loop are then evaluated on line under the conditions of the speed change transients of the prime mover and the stand-alone electrical passive load power variations with the simple theoretical analysis and the efficient computation processing procedures described in the part I of this paper. In addition, a feasuible PI controlled feedback closed-loop voltage regulation scheme of the stande-alone single-phase SEIG is designed on the basis of the static VAR compensate. (SVC) and discussed in experiment for the promising stand-alone power conditioner. The experimental operating performance results are illustrated and give good agreements with the simulation ones. The simulation and experimental results of the stand-alone single-phase SEIG with the simple SVC controller for its stabilized voltage regulation prove the practical effectiveness of the additional SVC control loop scheme including the PI controller with fast response characteristics and steady-sate performance improvements.

댐 운영을 고려한 금강의 생태.수문학적 변화 평가 : I. 유황변화 분석 (Evaluation of Eco-Hydrological Changes in the Geum River Considering Dam Operations: I. Flow Regime Change Analysis)

  • 본 연구에서는 금강의 생태계에 영향을 끼쳤을 가능성이 있는 주요 개발 및 관리 활동들을 기초로 하여 생태수 문학적인 하천평가 시나리오 구성을 위한 개념모델을 제시하였다. 또한 생태수문학적 하천평가를 위하여 여러 지원 프로그램과 함께 유역물수지 모델인 KModSim과 하천생태분석 모델인 RAP을 이용한 분석방법을 개발하였다. 용담댐과 대청댐의 건설 및 운영에 따라 예측되는 유황변화를 분석하기 위하여 다양한 시나리오에 따른 수문분석을 실시한 결과에 의하면, 수통 및 공주지점에서 초과시간 10%에 해당하는 "Percentile 10" 값은 각각 20.5%, 8.0% 감소한 반면, "Percentile 90" 값은 각각 156.3%, 340.8%나 증가하여 저유량의 변화가 크게 나타났으며, 이는 한국의 비조절하천의 특징인 큰 유황변동율을 감소시킨 것으로 나타났다. 후속 논문에서는 생태-수문학적 분석결과를 제시 할 것이다.

Tree Diversity, Distribution and Population Structure of a Riparian Forest from Certain Zones along the Dikhu River in Nagaland, India

  • At present the biodiversity of the riparian forest is under threat due to various anthropogenic pressures. Hence study was conducted along the three unprotected zones of riparian forest in Dikhu river, Nagaland, north-east India in order to compared the diversity, distribution and population structure of riparian trees. In each zone $100{\times}100m^2$ plot were marked and subdivided into 20 plots of $10{\times}10m^2$. 10 plots on each side of the river were taken randomly covering 0.02 ha. Only tree with dbh ${\geq}10cm$ and dbh above 1.5 m above ground level were recorded as individual species. A total of 29 tree species belonging to 18 families were recorded from the three zones of the river. Tree species richness was highest at the middle zone (19) followed by upper (14) and lower zone (7). The most abundant species and family recorded at upper, middle and lower zones were Melia azaderach of Euphorbiaceae ($380\;stems\;ha^{-1}$), Terminalia chebula of Euphorbiaceae ($432.5\;stems\;ha^{-1}$) and Duabanga grandiflora of Lythraceae ($365\;stems\;ha^{-1}$) respectively. The ranges of diversity indices observed in the three zones were: Shannon-Wiener index (1.25-0.73), Simpson diversity index (0.42-0.93), Evenness index (0.47-0.37) and Index of co-dominance (0.75-0.94). Rest of other indices were also estimated and compared. Distribution of trees shows the contagious pattern common in the upper and middle zones and regular in the lower zone. The girth size class analysis demonstrated that the riparian forest is in less mature succession stage. This study emphasize the need for management and conservation of riparian forest by developing policy to declare the riparian zone as protected area to prevent further degradation and loss of biodiversity from these unregulated zones along the river.

  • The CKLF-like MARVEL transmembrane domain-containing family (CMTM) is a novel family of genes first reported at international level by Peking University Human Disease Gene Research Center. The gene products act between chemokines and the transmembrane-4 superfamily. Located in several human chromosomes, the CMTMs CKLF and CMTM1 to CMTM8 may be unregulated in tumors and act as potential tumor suppressor genes with important roles in the immune, male reproductive and hematopoietic systems. In-depth studies in recent years established a close relation between CMTMs and tumorigenesis and metastasis. The CMTM family has a significant clinical value in diagnosis and treatment of diseases linked to tumors and the immune system.

미계측 유역 평균갈수량 산정을 위한 지역회귀모형의 개발 (Development of Regional Regression Model for Estimating Mean Low Flow in Ungauged Basins)

  • 본 연구에서는 미계측 유역의 평균갈수량 추정을 위한 지역회귀모형을 개발하고자 하였다. 12개 다목적댐과 4개의 용수댐에서 관측된 조절되지 않은 유입량 자료로부터 평균갈수량을 산정하였고, 이를 유역면적, 유역경사, 유역밀도, 연평균강수량, 유출곡선지수 등의 유역특성인자와의 상관분석을 통해 다양한 형태의 지역회귀모형을 개발하였다. 평균갈수량의 관측값과 추정값의 비교를 통해 각 회귀모형의 성능을 평가하였고, 유역면적, 연평균강수량, 유출곡선지수를 설명변량으로 하는 회귀모형이 가장 우수한 성능을 보였다. 또한 비유량법과 기존에 개발된 기존회귀모형과의 비교를 통해서 본 연구에서 개발한 모형의 적용성이 가장 우수한 것으로 분석되었다.

환경시료 중 폴리브롬화비페닐에테르(Polybrominated Biphenyl Ethers) 분석법 (Analytical Method of Polybrominated Biphenyl Ethers(PBDEs) in Environmental Samples)

  • 최근 새로운 환경오염물질로 부각되고 있는 미규제 오염물질 중 브롬화 난연제로 많이 사용되고 있는 PBDEs에 대한 환경시료 중의 분석법을 확립하기 위해 표준물질이 준비된 PBDEs 40종을 대상으로 전처리 회수율 및 검출한계를 산정하였으며, 이때 사용한 환경시료는 PBDE 및 TBBPA의 사용량이 많은 지역의 수질 및 토양시료를 대상으로 하였다. GC/MS을 이용한 PBDEs의 검출한계는 수질 시료가 5.0~10.0 pg/L로 나타났으며, 토양시료는 0.5~2.0 ng/g 이었다. 수질시료의 용매별 추출효율은 디에틸에테르를 제외한 단일용매의 경우에는 에틸아세테이트, n-헥산 및 디클로로메탄 등의 평균회수율이 89~98%로 나타났으며, 이들 용매의 20% 아세톤 혼합용매의 평균회수율이 89~103%로 나타내났다. 속실렛 추출법에서는 브롬화 정도 및 브롬의 치환 위치에 따라 회수율이 다르게 나타났으며, 아세톤:n-헥산(1:1)이 아세톤:디클로 로메탄(1:1)의 경우보다 추출 효율이 높게 나타났다.

국내 주요 정수장 수돗물의 퍼클로레이트 검출현황 (Occurrence of Perchlorate in Drinking Water in Korea)

  • Perchlorate is an unregulated contaminant but recently many cases of perchlorate contamination have been reported in the US. In Japan, also, a couple of contamination incidents caused by perchlorate occurred. Against this backdrop, investigation on perchlorate was urgent for Korea. Accordingly, perchlorate investigation in tap water and river water was conducted one time a week for seven consecutive weeks at major water sources of Korea including Han River, Guem River, Yeong-san River, and Nak-dong River. Perchlorate was not found at Han River, Guem River, and Yeong-san River. However, all the 4 tap water samples in Nak-dong River recorded high level of perchlorate. Among others, NT1 located at Daegu posted the highest value of $22.3{\mu}g/L$. This level is lower than $24.5{\mu}g/L$, the threshold recommended by US EPA. Still, perchlorate contamination in drinking water can deal a heavy blow to nerve development of infants and children by causing iodine deficiency. At the 1st and 2nd investigations, perchlorate concentration reached $18.7{\sim}95.6{\mu}g/L$ and $4.0{\sim}25.6{\mu}g/L$ respectively, both of which are relatively higher. The high perchlorate concentration in Nak-dong River was possible because of the waste water discharged from LCD manufacturing factory which was located at NS3 in Gumi, Korea. Perchlorate concentration of waste water from the factory was $730{\sim}1,858{\mu}g/L$.

Sesquiterpene Derivatives Isolated from Cyperus rotundus L. Inhibit Inflammatory Signaling Mediated by NF-${\kappa}B$

  • The immune system is finely balanced by the activities of pro-inflammatory and anti-inflammatory mediators or cytokines. Unregulated activities of these mediators can lead to the development of various inflammatory diseases. A variety of safe and effective anti-inflammatory agents are available with many more drugs under development. Of the natural compounds, the sesquiterpenes (nootkatone, ${\alpha}$-cyperone, valencene and ${\alpha}$-selinene) isolated from C. rotundus L. have received much attention because of their potential antiinflammatory effects. However, limited studies have been reported regarding the influence of sesquiterpene structure on anti-inflammatory activity. In the present study, the anti-inflammatory potential of four structurally divergent sesquiterpenes was evaluated in lipopolysaccaride (LPS)-stimulated RAW 264.7 cells, murine macrophages. Among the four sesquiterpenes, ${\alpha}$-cyperone and nootkatone, showed stronger anti-inflammatory and a potent NF-${\kappa}B$ inhibitory effect on LPS-stimulated RAW 264.7 cells. Molecular analysis revealed that various inflammatory enzymes (iNOS and COX-2) were reduced significantly and this correlated with downregulation of the NF-${\kappa}B$ signaling pathway. Additionally, electrophoretic mobility shift assays (EMSA) elucidated that nootkatone and ${\alpha}$-cyperone dramatically suppressed LPS-induced NF-${\kappa}B$-DNA binding activity using 32Plabeled NF-${\kappa}B$ probe. Hence, our data suggest that ${\alpha}$-cyperone and nootkatone are potential therapeutic agents for inflammatory diseases.
