  • Biosorption of lead by marine algae, Undaria pinnatifida, was examined. The biosorption capacity of lead by U. pinnatifida was above 30% of its own weight and proportional to the initial lead concentration. However, the opposite result was shown in different initial weight of biomass. The mechanism of biosorption was accorded to the ion exchange process.

Effects of supplemental undaria powder, herb and wasabi in the diets on growth, body composition, blood chemistry and non-specific immune response of juvenile flounder, Paralichthys olivaceus

  • This study was conducted to investigate the effects of several additives(macroalgae, wasabi, and herb) in formulated diets on the growth, body composition, blood chemistry and non-specific immune response of juvenile flounder. Three replicates of juveniles (average weight 8.4 g) in flow-through aquarium system were fed one of six isonitrogenous (45%) and isolipidic (8%) diets containing 5 and 10% Undaria powder, 2% wasabi leaf, 2% wasabi stem, and 0.5% herb (Obosan) for 8 weeks. Survival was not affected by the different dietary additives (P>0.05). The highest weight gain and feed efficiency offish fed the diet containing 0.5% herb were significantly higher than those of fish fed the diets containing 10% Undaria powder (P<0.05). No significant differences were found in contents of moisture, crude protein, lipid and ash of whole body (P>0.05). Fish fed the diet containing 10% Undaria powder showed the highest moisture and the lowest crude lipid contents in the liver. Although hematological parameters (red blood cell, hematocrit and hemoglobin) and serum constituents (glucose, total cholesterol and glutamic-oxaloacetic transaminase) contents of fish varied between treatments, no specific trend was observed throughout feeding periods. Lysozyme activity in the serum and nitroblue tetrazolium reduction of macrophage in the head kidney from fish fed the diets containing herb was significantly higher than those of fish fed the control diet. The results of this study suggest that herb as an additive in this formulated diet may improve growth and non-specific immune response of juvenile flounder.

  • 원유 가격의 상승과 지구온난화로 인하여 재생 가능한 바이오매스를 이용하여 산업적으로 중요한 화합물을 생산하는 연구가 주목받고 있다. 특히, 3세대 바이오매스인 해조류는 비식량 자원, 높은 생산 수율, 온실가스 저감 등 장점을 가지고 있기에 연구 되어야 할 필요가 있다. 본 연구에서는 고체분석 방법을 이용하여 4종의 해조류 중에 미역이 가장 많은 당류를 함유하는 것을 확인하였다. 미역의 효과적인 전처리를 위해 반응표면분석법을 이용하였으며 이를 통해 고체의 부하 및 촉매의 농도 증가가 총 당의 추출률과 관계 있음을 확인하였다. 4종의 락토바실러스 균주에서 미역의 전처리물을 이용하여 젖산 생산 수행하였으며 L. alimentarius와 L. brevis가 해조류 이용한 젖산생산에 적합한 균주임을 확인하였다.

미역 포자엽 fucoidan의 중금속 흡착 특성 (Biosorption of Lead and Cadmium by Fucoidan from Undariafinnatifida)

  • 국내에서 다량 생산되는 갈조류인 미역포자엽, 다시마, 톳, 모자반으로부터 fucoidan을 추출, 분획하여 Pb와 Cd의 흡착 특성을 조사하였다. 4종의 갈조류에서 추출한 fucoidan의 분획물 모두 Pb의 흡착량이 Cd보다 높았으며, 시료간에는 미역포자엽 분획물 (Fr-3.0)이 Pb와 Cd 모두 흡착력이 상대적으로 우수하였다. pH와 농도 변화에 따른 홉착력의 변화는 $C_f$ (잔류농도)가 50mg/L까지는 pH 간 차이가 없었으나, 농로가 증가함에 따라 pH 5.5 처리구가 다른 처리구에 비하여 흡착량이 증가하였다. 실험을 통하여 얻은 Pb 와 Cd의 최대 흡착량은 pH 5.5에서 각각 94mg/g ($C_f$164mg/L), 64mg/g ($C_f$197mg/L)였으며 Langmuir sorption model을 통해 구한 최대 흡착량은 pH 5.5에서 각각 178mg/g, 122mg/g이었다. Cd 공존시 Pb 흘착량은 $C_f$가 낮을 범위에서는 변화가 없었으나 $C_f$가 증가함에 따라 공존 이온의 흡착 방해로 인하여 감소하였다.

갈조류 미역(Undaria pinnatifida)의 분리당화발효와 다양한 효모를 이용한 바이오에탄올의 생산 (Bioethanol Production from Seaweed Undaria pinnatifida Using Various Yeasts by Separate Hydrolysis and Fermentation (SHF))

  • 해조류 중 갈조류인 미역으로부터 분리당화발효(SHF)를 위한 전처리 및 효소당화를 검토하고, 기존의 분리당화발효(SHF)를 개선하기 위해 공배양발효(co-culture)를 수행하였다. 비순치 효모와 고농도 mannitol에 순치(adaptive evolution)한 효모를 이용한 공배양발효를 실시한 결과 발효 72시간에 12.2 g/l의 에탄올과 에탄올 수율($Y_{EtOH}$) 0.41을 나타내었다. 이러한 기존의 분리당화발효(SHF)를 개선한 공배양발효를 통해 에탄올 생산 수율이 0.23에서 0.41로 35.2% 증가하였으며, 에탄올 발효시간도 108시간에서 72시간으로 33.3% 감소하였다. 이러한 연구결과는 해양 바이오매스인 해조류로부터 바이오연료 생산과정에 있어 유용한 정보를 제공하는 것으로 판단된다.

LPS로 유도된 RAW 264.7 대식세포에 대한 미역(Undaria pinnatifida) Ethyl Acetate 분획물의 항염증 효과 (Anti-Inflammatory Effect of Ethyl Acetate Fraction Isolated from Undaria pinnatifida on Lipopolysaccharides-Stimulated RAW 264.7 Cells)

  • An ethanolic extract of Undaria pinnatifida was fractionated using several solvents. Of the fractions, the ethyl acetate fraction had the greatest inhibitory effect on lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 macrophage cells. Using this fraction (U. pinnatifida ethyl acetate extract, UPE), we investigated the molecular mechanism underlying its inhibitory effect on LPS-stimulated RAW 264.7 cells. Pretreatment of the cells with up to $100{\mu}g/mL$ UPE significantly inhibited NO production and inducible nitric oxide synthase (iNOS) expression, in a dose-dependent manner. Similarly, UPE treatment markedly reduced the production of pro-inflammatory cytokines, such as interleukin (IL)-1, IL-6 and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), while it strongly suppressed the nuclear translocation of nuclear factor-kappa B (NF-${\kappa}B$) by preventing proteolytic degradation of inhibitor of nuclear factor ${\kappa}B$ $(I{\kappa}B)-{\alpha}$. Moreover, UPE treatment significantly reduced the phosphorylation of phosphatidylinositol 3-kinase (PI3K)/Akt and mitogen-activated protein kinase (MAPK) in LPS-stimulated cells. These results indicate that UPE contains anti-inflammatory compounds and suggest that it might be used as a functional food material that assists in prevention of inflammatory diseases.

Purification and Anticoagulant Activity of a Fucoidan from Korean Undaria pinnatifida Sporophyll

  • Crude fucoidan was extracted from the sporophyll of Korean Undaria pinnatifida collected at a coastal area ofWando, Korea, mainly by dilute acid extraction, ethanol precipitation, CaCU Precipitation, with an yield of approxi-mately 3.9% in mass. It was further purified by DEAE-cellulose column chromatography and its chemical composi-don and in vitro anticoagulant activity was determined. The average molecular mass of the purified fucoidan wasestimated about 2.1 x 103 kDa by size-fractionation HPLC and it consisted of neutral sugar (52.34% in mass), uronicacid (26.2%), and sulfate esters (7.4%). From the HPAEC-PAD analysis, the monosaccharide composition of thepurified fucoidan was shown to be fucose, galactose, xylose, and mannose, with a molar ratio of 1, 0.2, 0.02, 0.15,respectively, demonstrating that major monosacd-iande was fucose (72.3% in mol percentage) and other sugars,xylose (1.5%), galactose (14.6%), and mannose (10.9%) were present as minor component. The results suggested thatthis fucoidan is a sulfated, U-type fucoidan. The activated partial thrombloplastin time (APTT) assay of the purifiedfucoidan showed that the purified fucoidan elicited anticoagulant activity in a dose-dependent manner. Five jUg ofsporophyll fucoidan delayed the blood clotting time up to 5 times than untreated control and also up to 1.5 timesthan the same amount of the commercial fucoidan, respectively. Although it is preliminary, these results suggestthat the fucoidan of Korean Undaria vinnatifida sporophyll would be promising candidates for the development ofan anticoaeulant.

RAW 264.7 세포에서의 미역(Undaria pinnatifida) 뿌리 에탄올 추출물의 항염증활성 (Anti-inflammatory Activity of Ethanol Extract of Undaria pinnatifida Root in RAW 264.7 Cells)

  • The anti-inflammatory effects of the ethanol extract of Undaria pinnatifida root (UPREE) were investigated using the lipopolysaccharide (LPS)-induced inflammatory response in RAW 264.7 cells by measuring the production of nitric oxide (NO), interleukin (IL)-6, tumor necrosis factor (TNF)-${\alpha}$, and IL-$1{\beta}$, and cell proliferation. We found that NO levels were reduced by 34% at $100{\mu}g/mL$. Moreover, the production of IL-6 and TNF-${\alpha}$ was suppressed by the UPREE treatment. In particular, the IL-6 production was inhibited by more than 30% at $100{\mu}g/mL$ UPREE. The proliferation of RAW 264.7 cells was measured by MTT assay, and we found no cytotoxicity in those cells treated with UPREE compared to the control. Our results suggest that UPREE shows promise as a therapeutic anti-inflammatory treatment.
