Bio-functions of Marine Carotenoids

  • Carotenoids being most important pigments among those occurring in nature, have received increased interest owing to their beneficial effects on human health. An effort is made to review marine carotenoids as important bioactive compounds with reference to their presence, chemical, and biofunctional benefits they afford. The potential beneficial effects of marine carotenoids were particularly focused on astaxanthin and fucoxanthin, major marine carotenoids found in marine animals and aquatic plants, respectively. Both carotenoids show strong antioxidant activity which is attributed to quenching singlet oxygen and scavenging free radicals. The potential role of the carotenoids as dietary antioxidants has been suggested as being one of the main mechanism by which they afford their beneficial health effects such as anticancer activity and anti-inflammatory effect. Only recently, antiobesity effect and antidiabetic effect have been noted as specific and novel bio-functions of fucoxanthin. Nutrigenomic study reveals that fucoxanthin induces uncoupling protein 1 (UCP1) expression in white adipose tissue (WAT) mitochondria to lead to oxidation of fatty acids and heat production in WAT. Fucoxanthin improves insulin resistance and decreases blood glucose level, at least in part, through the down-regulation of tumor necrosis factor $\alpha$ ($TNF{\alpha}$) in WAT of animals.

  • Obesity is a worldwide problem that is associated with metabolic disorders. Obesity is caused by the accumulation of an abnormal amount of body fat in adipose tissue. Adipose tissue is a major metabolic organ, and it has been classified as either white adipose tissue (WAT) or brown adipose tissue (BAT). WAT and BAT are characterized by different anatomical locations, morphological structures, functions, and gene expression patterns. WAT is mainly involved in the storage and mobilization of energy in the form of triglycerides. On the other hand, BAT specializes in dissipating energy as heat through uncoupling protein-1 (UCP-1)-mediated non-shivering thermogenesis. Novel type of brown-like adipocyte within WAT called beige/brite cells was recently discovered, and this transdifferentiation process is referred to as the "browning" or "britening" of WAT. Recently, Brown fat and/or browning of WAT have been highlights as a new therapeutic target for treatment of obesity and its related metabolic disorders. Here, we describe recent advances in the study of BAT and browning of WAT, focusing on proteomic approaches.

보중치습탕이 3T3-L1 지방전구세포의 분화 및 지방생성 억제에 미치는 영향 (Inhibitory Effects of Bojungchiseub-tang on Adipocyte Differentiation and Adipogenesis in 3T3-L1 Preadipocytes)

  • Bojungchiseub-tang (BJCST) has been used in symptoms and signs of edema, dampness-phlegm, kidney failure, and so on. BJCST is also expected to have strong anti-obesity activities. However, little is known about the mechanisms of its inhibitory effects on adipocyte differentiation and adipogenesis. In the present study, we examined the effects and mechanism of BJCST on transcription factors and adipogenic genes of 3T3-L1 preadipocytes to understand its inhibitory effects on adipocyte differentiation and adipogenesis. Our results showed that BJCST significantly inhibited differentiation and adipogenesis of 3T3-L1 preadipocytes in a dose-dependent manner. To elucidate the mechanism of the effects of BJCST on lowering lipid content in 3T3-L1 adipocytes, we examined whether BJCST modulate the expressions of transcription factors to induce adipogenesis and adipogenic genes related to regulate accumulation of lipids. As a result, the expression of steroid regulatory element-binding protein (SREBP)1, cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$, $C/EBP{\delta}$, and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) genes, which induce the adipose differentiation, liver X receptor $(LXR){\alpha}$ and fatty acid synthase (FAS) genes, which induce lipogenesis and adipose-specific aP2, Adipsin, lipoprotein lipase (LPL), CD36, TGF-${\beta}$, leptin and adiponectin genes, which compose fat formation were decreased. BJCST also reduced the expression of acyl CoA oxidase (ACO) and uncoupling protein (UCP) genes related to lipid oxidation. In conclusion, BJCST could regulate transcript factor related to induction of adipose differentiation and inhibited the accumulation of lipids and expression of adipogenic genes.

  • Nutrigenomics refers to research that investigates the interaction between nutrition and the human genome. Caffeine in tea and coffee is widely and routinely consumed by people. This study was performed to confirm the effect of caffeine treatment on the gene expression and cytokine profiling in 3T3-L1 adipocyte cells using microarray and protein array methodology. Treatment of caffeine in 3T3-L1 adipocyte cells increased expression of several genes related with obesity including adipocyte C1Q and collagen domain containing (ACDC), Adipsin (ADN), uncoupling protein 3(UCP3), while glyceraldehyde-3-phosphate dehydrogenase (GAPDH), which is known as lipid storage enzyme, was decreased by caffeine treatment. Furthermore, cytokines, such as interleukin-3 (IL-3), interleukin-12(IL-12), interleukin-13 (IL-13), granulocyte colony stimulating factor (GCSF), granulocyte macrophage colony stimulating factor (GM-CSF) and vascular endothelial growth factor (VEGF), were decreased in caffeine treated 3T3-L1 adipocyte cells. These results provided interesting information about the genes related with caffeine and cytokine expression profiling in obesity.

Effects of Panax ginseng extracts prepared at different steaming times on thermogenesis in rats

  • Background: Panax ginseng (PG) has a long history of use in Asian medicine because of its multiple pharmacological activities. It has been considered that PG in a type of white ginseng may induce undesirable thermogenic effects, but not in a type of red ginseng. However, there is a lack of evidence about the correlation between ginsenoside and thermogenesis. Methods: We investigated the effects of PG with different ginsenoside compositions on body temperature, blood pressure, and thermogenesis-related factors in rats. Results: With increasing steaming time (0 h, 3 h, 6 h, and 9 h), the production of protopanaxadiol ginsenosides increased, whereas protopanaxatriol ginsenosides decreased in white ginseng. In both short- and long-term studies, administration of four ginseng extracts prepared at different steaming times did not induce significant changes in body temperature (skin, tail, and rectum) and blood pressure of rats compared to saline control. In addition, there were no significant differences in the molecular markers related to thermogenesis (p > 0.05), mRNA expressions of peroxisome proliferator-activated receptor-gamma coactivator-$1{\alpha}$ and uncoupling protein 1 in brown adipose tissue, as well as the serum levels of interleukin-6, inducible nitric oxide synthase, and nitrite among the treatment groups. Conclusion: These observations indicate that the potential undesirable effects of PG on body temperature could not be explained by the difference in ginsenoside composition.

Genetic Variations of Eight Candidate Genes in Korean Obese Group

  • Obesity is a complex metabolic disorder with a strong genetic component. There are many candidate genes for obesity and its related phenotypes. We studied genetic variations between Korean obese and lean groups. Polymorphisms investigated were the Msp I polymorphism of the $\alpha$$_{2A}$-adrenergic receptor ($\alpha$$_{2A}$-AR) gene, the Mnl I polymorphism of the $\alpha$$_2$-adrenergic receptor ($\alpha$$_2$-AR) gene, the BstO I polymorphism of the $\beta$$_3$-adrenergic receptor ($\beta$$_3$-AR) gene, the Pml I polymorphism of the lamin A/C (LMNA) gene, the Hga I polymorphism of the clearance receptor (NPRC) gene, the Msp I polymorphism of the leptin gene, BclI polymorphism of the uncoupling protein 1 (UCPI) gene and the Hha I polymorphism of the fatty acid binding protein 2 (FABP2) gene. Among these genetic markers, Pml I polymorphism at the LMNA gene and Bcl I polymorphism at the UCP1 gene were significantly associated with obesity. However, further studies are required whether thease findings are reproduced in large population, although two polymorphisms might be useful as genetic markers in the ethiology of obesity in Korean population.ion.

Epac2a-knockout mice are resistant to dexamethasone-induced skeletal muscle atrophy and short-term cold stress

  • Exchange protein directly activated by cAMP (Epac) 2a-knockout (KO) mice exhibit accelerated diet-induced obesity and are resistant to leptin-mediated adipostatic signaling from the hypothalamus to adipose tissue, with sustained food intake. However, the impact of Epac2a deficiency on hypothalamic regulation of sympathetic nervous activity (SNA) has not been elucidated. This study was performed to elucidate the response of Epac2a-KO mice to dexamethasone-induced muscle atrophy and acute cold stress. Compared to age-matched wild-type mice, Epac2a-KO mice showed higher energy expenditures and expression of myogenin and uncoupling protein-1 in skeletal muscle (SM) and brown adipose tissue (BAT), respectively. Epac2a-KO mice exhibited greater endurance to dexamethasone and cold stress. In wild-type mice, exogenous leptin mimicked the responses observed in Epac2a-KO mice. This suggests that leptin-mediated hypothalamic signaling toward SNA appears to be intact in these mice. Hence, the potentiated responses of SM and BAT may be due to their high plasma leptin levels.

과일식과 걷기 운동이 UCP2 유전자 다형성 특성에 따른 여대생의 신체 조성 및 혈액 성분에 미치는 영향 (Effects of a Fruit Diet and Exercise on Body Composition and Hematological Parameters according to UCP2 Genotype in Female Collegians)

  • The objective of this study was to examine the effects of a fruit diet and exercise on body composition and hematological parameters in female collegians according to their uncoupling protein 2 (UCP2) genotypes. Nine subjects were recruited from the SahmYook University. 4 subjects carried the ID genotype, 5 the DD genotype, and none of the subjects carried the II genotype. After 10 days of the program, BMI were significantly decreased (p<0.05) in the ID genotype and the DD genotype. Mean total energy intake from fruit diet was 888.1 kcal. Fat mass and % body fat were significantly decreased (p<0.05) in subjects with the ID genotype. Hemoglobin level was significantly increased (p<0.05) in subjects with the DD genotype. This study provided evidence that fruit diet and exercise could decrease the BMI in subjects with the ID and DD genotypes and increase the hemoglobin level in those with the DD genotype.

고지방 식이로 유도된 비만쥐에서 식이 레반이 체지방 형성 및 혈청 렙틴과 UCP 발현에 미치는 영향 (Effects of Dietary Levan on Adiposity, Serum Leptin and UCP Expression in Obese Rats Fed High Fat Diet)

  • The effects of dietary levan, high-molecular-weight $\beta$-2,6-linked fructose polymer, on adiposity, serum leptin and UCP expression in rats fed high fat diet were studied. The adipose tissue hormone, leptin has been proposed to be involved in the regulation of food intake and energy expenditure. Uncoupling protein (UCP), a mitochondrial protein that uncouples the respiratory chain from oxidative phosphorylation, generates heat instead of ATP, thereby increase energy expenditure. To determine whether the dietary levan may have the anti-obesity effect, 4 wk old Sprague Dawley male rats fed high fat diet for 6 wks to induce obesity, and subsequently fed one of three diets for further 6 wks: 1) high fat (40% of calories) diet without levan 2) with 3% (w/w) levan 3) with 5% levan. For the comparison, control group fed AIN-76A diet. Visceral and peritoneal fat masses were lower in high fat diet with levan groups compared to high fat diet group. The effect of levan was dose-dependent. Adipocyte size was significantly reduced in the levan diet groups compared to the no levan diet group. Serum cholesterol level was not affected by levan containing diet, while the serum HDL cholesterol level was higher in leven diet groups. In addition, serum triglyceride level was markedly reduced by levan containing diet, thus lower than that of control group. Serum leptin was reduced by levan containing diet and lower in 5% levan group compared to 3% levan group (p < 0.001), as a result, serum leptin and insulin levels of 5% levan group were reduced to level of control group. Futhermore, the serum leptin level reflected the adiposity. The expression of UCP 1, and UCP 2 in brown adipose tissue was up-regulated by levan containing diet. In conclusions, levan containing diet reduced adiposity and serum triglyceride but increased UCP expression in the obese rats fed high fat diet. (Korean J Nutrition 35(9) : 903~911, 2002)

  • Bitter melon (Momordica charantia) is used in traditional herbal medicine in many Asian countries for the treatment of several diseases such as diabetes, eczema, night blindness, psoriasis, and rheumatism. Especially, most reports concerning the biological activities of bitter melon have focused on its effects on diabetes and hyperglycemia. Also, bitter melon is regarded as a longevity food, suggesting that it has several beneficial effects on anti-aging and the maintenance of a healthy state. Thus, we investigated whether bitter melon could increase the capacity of exercise in this study. Interestingly, bitter melon fruit extract activated AMP-activated protein kinase (AMPK), which is important for regulating glucose homeostasis, mitochondrial content and exercise capacity. In addition, bitter melon extract increased the expression of enzymes involved in fatty acid oxidation such as mitochondrial uncoupling protein 3 (UCP3), carnitine palmitoyl transferase 1b (CPT1b), and pyruvate dehydrogenase lipoamide kinase isozyme 4 (PDK4). Moreover, exercise tolerance was much more enhanced in bitter melon treated animals compared to the non-treated control group. These results suggest that bitter melon is a promising candidate for the development of functional foods beneficial for physical strength and the enhancement of exercise capacity.
