UV Sensitivity of Korean Skin and The Effects of Factors affecting SPF Determination (한국인 피부의 자외선 감수성과 SPF 측정치에 미치는 인자의 영향)

  • Multiport-600 Solar SimulatorR is one of the most recent and convenient in-strument for evaluation of sun protection factor(SPF). In this study, we examined the practicability of the SPF determining system using Multiport -600 and the effects of several factors-light sources, seasons and experimental animals-on the minimal erythema dose(MED) and SPF. We also tested the UV sensitivity according to the sites of Korean people, And the ultraviolet radiation reaching the earth's surface In Seoul have been observed for one year. As a result of this study, the determinig system for SPF using Multiport-600 was proved to be a good system in accuracy and time-saving. The biological activity of fluorescence UV lamp of PUVA-800R was significantly higher than natural light or solar simulator with Xe arc lamp, and the determined MED became lower in inverse proportion to room temperature rise. Skin sensitivity by ultraviolet adiation was hights. in order \circled1 back \circled2 inns, upper arm \circled3 outer upper arm \circled4 foream. We also observed that UV radiation intensity was highest at noon in july and 1 sun burn unit(MED) was 28 minutes at that time.

  • Recombinant interleukin-2 (IL-2) has demonstrated as an antineoplastic agent in mice and human, but the relatively low response rates observed in clinical trials. Therefore, the present study was undertaken in order to evaluate therapeutic activities of IL-2 for the establishment of therapeutic applications. At the onset of the experiment, normal C3H/HeN mice were injected with $3{\times}10^6$ RD-995 tumor cells, murine ultraviolet radiation-induced fibrosarcoma, subcutaneously. Beginning on day 25, experimental groups were treated with a 5-day course of IL-2 (subcutaneous injection of 30,000 IU every 12 hours for 5 days). The result of this experiment revealed that RD-995 tumor grew progressively in control mice. Subcutaneous IL-2 therapy decreased tumor growth until day 23, then the tumor grew progressively. No significant difference in the survival of IL-2 therapy decreased tumor growth until day 23, then the tumor grew progressively. No significant difference in the survival of IL-2 treated mice were observed compared with the control mice.

Effects of Inhibitors on Cross-Adaptive Response to Ultraviolet Radiation or Ethyl methanesulfonate in Chinese Hamster Ovary Cells

  • This study was performed by the sister chromatid exchanges (SCEs) to investigate the effects of Aphidicolin (APC) or 2, 4-dinitrophenoi (DNP) on cross-adaptive response to ultraviolet radiation (UV) or ethyl methanesulfonate (EMS) in Chinese hamster ovary (CHO) cells. The pretreatment with 1 J/m$^2$ UV decreased the yield of SCEs induced by subsequent treatment with 8 mM EMS in CHO cells. And the treatment with 10 $\mu$g/ml APC or 50 $\mu$M DNP during incubation after pretreatment with 1 J/m$^2$ UV increased the yield of SCEs induced by 8 mM EMS. The pretreatment with 2 mM EMS decreased the yield of SCEs induced by subsequent treatment with 5 J/m$^2$ UV. The treatment with 10 $\mu$g/ml APC during incubation after 2 mM EMS increased the yield of SCEs induced by 5 J/m$^2$ UV. These results suggest that APC and DNP inhibit cross-adaptive response to pretreatment with UV and subsequent treatment with EMS, and also cross-adaptive response to pretreatment with EMS and subsequent treatment with UV is inhibited by APC in CHO cells.

  • The adaptive response and cross-adaptive response to sister chromatid exchanges (SCEs) and DNA single-strand breaks (SSBs) in Chinese hamster ovary (CHO)-K$_1$ cells treated with ultraviolet radiation (UV), ethyl methanesuffonate (EMS), or bleomycin (BLM) were investigated. Two assays were used in this study; SCEs and alkaline elution. The pretreatment with low conditioning dose of 2 mM EMS or 1 J/m$^2$ UV decreased the yield of SCEs induced by subsequent treatment with 8 mM EMS, 5 J/m$^2$ UV or 5 $\mu$g/ml BLM. And the pretreatment with low conditioning dose of 1 $\mu$g/ml BLM decreased the yield of SCEs induced by subsequent treatment with 5 $\mu$g/ml BLM or 5 J/m$^2$ UV. The rejoining of DNA SSBs in cells subsequently treated with 2 J/m$^2$ UV, 50 mM EMS or 400 $\mu$g/ml BLM is higher than that only treated with 2 J/m$^2$ UV, 50 mM EMS or 400 $\mu$g/ml BLM. These results suggest that there are the adaptive response and cross-adaptive response to SCEs, and is the adaptive response to the rejoining of DNA SSBs in CHO cells.

Expression of Poly (ADP-ribose) Polymerase During Apoptosis Induced by Ultraviolet Radiation in HeLa $S_3$ Cells

  • Induction of apoptosis allows the organism to get rid of abnormal cells and also of tumor cells. Understanding the mechanism involved in Ultraviolet radiation (UV) induced apoptosis may improve its therapeutic efficacy. In this study, we present expression of poly (ADP-ribose) polymerase (PARP) during apoptosis induced by UV in HeLa $S_3$ cells. Four different assays were performed in this study: morphological assessment of apoptotic cells and cell viability, DNA fragmentation analysis by agarose gel electrophoresis, quantitative assay of fragmented DNA, and expression of PARP by the western blot analysis. The percentages of apoptotic HeLa $S_3$ cells irradiated with $75J/m^2$ UV was increased continuously from 3 hrs incubation. DNA ladder pattern was appeared at 6 hrs. The amount of nucleosomal DNA fragments in cells treated UV increased from 3 to 12 hrs incubation and gradually decreased. The cleavage of PARP in HeLa $S_3$ cells irradiated with UV was induced, and the cleavage of PARP was more delayed in the cells pretreated with $5J/m^2$ UV and subsequently irradiated with $75J/m^2$ UV. than that in the cells only irradiated with $75J/m^2$ UV. Thus these data suggest that the cleavage of PARP relates with DNA fragmentation associated with apoptosis.

Effect of Electrical Stimulation Level on Quantitative Sensory Test Induced Erythema by UV Radiation (전기자극수준이 자외선에 의한 홍반의 정량적 감각검사에 미치는 효과)

  • Purpose : This study is to investigate the modulatory effects to the ultraviolet induced erythema of pain processing system. Methods : Thirty six healthy volunteers were divided into none treatment group (n=6), indomethacine group (n=6), subsensory level electrical stimulation group (n=6), sensory level electrical stimulation group (n=6), motor level electrical stimulation group (n=6), noxious level electrical stimulation group (n=6). Subjects were induced erythema for three times minimal erythema dose (MED) at upper arm of dermatome C6 level. Each experimental group had mechanical pain threshold (MPT), electrical pain threshold (EPT), thermal pain threshold (TPT). Results : This study revealed that we observed that pain thresholds were significantly correlated with each other in pain processing system. The effect of electrical stimulation levels evaluates were shown to be significant differences pain control effect in electrical stimulation group (sensory, motor level electrical stimulation groups) more than indomethacine group, subsensory level and control group. Conclusion : In this study, it was found that the effect of ultraviolet induced erythema of pain control by modulatory electrical stimulation.

Inhibition of melanogenesis by tyrosinase siRNA in human melanocytes

  • Tyrosinase (TYR) plays a critical role in cellular melanogenesis and, thus, has been the major target of pharmacological approaches for the control of skin pigmentation. This study examined an alternative molecular approach using TYR-small interfering RNA (siRNA) to control melanogenesis in the human melanocytes. Both the mRNA and protein levels of TYR were significantly lowered by TYR-siRNA treatment, whereas TYR-related protein 1 and TYR-related protein 2 displayed no such changes. TYR-siRNA treatment inhibited the cellular melanin synthesis from the externally supplied TYR substrate L-tyrosine. TYR-siRNA also suppressed melanin synthesis and decreased the viability of cells exposed to ultraviolet radiation, supporting a critical role of melanin in protection against ultraviolet radiation. These results suggest that molecular approaches using siRNA targeted to the enzymes of melanogenic pathway may provide a novel strategy for the control of cell pigmentation.

Gamma radiation shielding properties of poly (methyl methacrylate) / Bi2O3 composites

  • This work investigated the gamma-ray shielding performance, and the physical and mechanical properties of poly (methyl methacrylate) (PMMA) composites embedded with 0-44.0 wt% bismuth trioxide (Bi2O3) fabricated by the fast ultraviolet (UV) curing method. The results showed that the addition of Bi2O3 had significantly improved the gamma shielding ability of PMMA composites. Mass attenuation coefficient and half-value layer were examined using five gamma sources (Cs-137, Ba-133, Cd-109, Co-57, and Co-60). The high loading of Bi2O3 in the PMMA samples improved the micro-hardness to nearly seven times that of the pure PMMA. With these enhancements, it was demonstrated that PMMA/Bi2O3 composites are promising gamma shielding materials. Furthermore, the fast UV curing exerts its great potential in significantly shortening the production cycle of shielding material to enable rapid manufacturing.

Modeling Grain Rotational Disruption by Radiative Torques and Extinction of Active Galactic Nuclei

  • Extinction curves observed toward individual Active Galactic Nuclei (AGN) usually show a steep rise toward Far-Ultraviolet (FUV) wavelengths and can be described by the Small Magellanic Cloud (SMC)-like dust model. This feature suggests the dominance of small dust grains of size a < 0.1 ㎛ in the local environment of AGN, but the origin of such small grains is unclear. In this paper, we aim to explain this observed feature by applying the RAdiative Torque Disruption (RATD) to model the extinction of AGN radiation from FUV to Mid-Infrared (MIR) wavelengths. We find that in the intense radiation field of AGN, large composite grains of size a > 0.1 ㎛ are significantly disrupted to smaller sizes by RATD up to dRATD > 100 pc in the polar direction and dRATD ~ 10 pc in the torus region. Consequently, optical-MIR extinction decreases, whereas FUV-near-Ultraviolet extinction increases, producing a steep far-UV rise extinction curve. The resulting total-to selective visual extinction ratio thus significantly drops to RV < 3.1 with decreasing distances to AGN center due to the enhancement of small grains. The dependence of RV with the efficiency of RATD will help us to study the dust properties in the AGN environment via photometric observations. In addition, we suggest that the combination of the strength between RATD and other dust destruction mechanisms that are responsible for destroying very small grains of a <0.05 ㎛ is the key for explaining the dichotomy observed "SMC" and "gray" extinction curve toward many AGN.

Protective effect of Oxya chinensis sinuosa methanol extract on UVB-induced damage in human retinal pigment epithelial cells

  • The human eye, constantly exposed to solar radiation, can be damaged by UV radiation. In particular, ultraviolet B (UVB)-induced damage plays an important role in retinal degeneration and cell aging. In this study, we investigated the protective effects of the methanol extract of Oxya chinensis sinuosa (OCM), an edible insect known for its high protein content (64.2%), and various pharmacological effects, on human retinal pigment epithelial cells. ARPE-19 cells were treated with OCM and subsequently UVB irradiated. Our results showed that OCM effectively attenuates UVB-induced cell damage by reducing MAPK phosphorylation (JNK and p38 MAPK). Additionally, OCM increased the phosphorylation of Akt, and cell cycle regulators, including p21 and p27, in a dose-dependent manner. Moreover, OCM treatment increased ARPE-19 cell proliferation by activating the S6K1/S6 pathway. This study suggests that OCM prevents UVB-induced retinal cell damage by increasing cell proliferation via ROS reduction, suggesting its potential as a functional therapeutic superfood against retinal cell damage.
