Ultrastructural Changes of the Hermaphrodite Duct Epithelium by Season in a Korean Slug Incilaria fruhstorieri (한국산 산민달팽이(Incilaria fruhstorferi) 장웅동체관 상피의 계절에 따른 미세구조의 변화)

  • A studv on the ultrastructural changes of the hermaprodite duct epithelium in spring and summer specimens of a Korean slug Incirovia fruhstorferi was conducted. In spring specimen, the hermaphrodite duct was 0.3 H 0.2 mm in diameter and seemed to be a little bit abnormal feature, due to shrinkage. The epithelium of the duct was composed of cells containing various-sized vacuoles and their cytoplasm showed high electron density. In summer specimen, the hermaphrodite duct was 0.4 $\times$ 0.23 mm in diameter and seemed to be more voluminous than that of the spring specimen. The lumen of the duct in the summer specimen was lined by a epithelium composed of several cell types such as ciliated cuboidal, pseudostratified cuboidal, ciliated columnar and irregular cells. Phasocvtotic activities of the duct epithelium are observed only in the summer specimen. Both of the duct epithelia in the spring and summer specimens were surrounded by connective tissues at their bases. The results obtained suggest that the duct epithelia of the slugs experience seasonal variation in relation to their reproductive cycles.

  • This study was investigated to elucidate the effect of chlorambucil on the heart in aging rats. Male rat ranging in age from 3 to 36 months were used. Each age groups(3, 6, 12, 18, 24, 36 months) included control and chlorambucil treated rats. As a part of the study, the ultrastructural changes in the left ventricular myocardial cells were described by using the qualitative and morphometric techniques. The results are summarized as follows. Age associated ultrastructural changes included: an increase in vacuoles, protrusion of plasma membranes, lipid droplets, and lipofuscins in myocardium of control groups. These changes which begin to occur at 12 months and continue through 36 months. At the 36 months some instance of unusual formation of contraction band and separation of intercalated discs were encountered. Morever, these changes and contents with chlorambucil treatment were remarkably increased in comparison with control groups. Age-dependent changes of control group measured with morphometry were not observed in the volume densities of mitochondria and myofibrils. But there was increase in interstitium. On the other hand decrease in sarcoplasmic reticulum and T-tubule system. In chlorambucil treated groups, volume densities of mitochondria and interstitium were increased in comparison with those of the control groups. But sarcoplasmic reticulum and T-tubule system were remarkably decreased.

  • The present studies were designed to determine the feasibility of using the rat tracheal epithelium as models for induction of aging. The ultrastructural and cytochemical changes of tracheal epithelium were investigated in rats at ages of five, twelve and twenty four months. Some major changes in the tracheal epithelium with advancing age were observed by electron microscopy. The results were summarized as fellow: 1. With the advance of age, lysosome, vacuole and multivesicular bodies were increased in number and numerous myelinoid bodies were observed in cytoplasm of ciliated cells. 2. In goblet cell, serous cell and brush cell lysosome and myelinoid bodies were increased in number with the advance of age, and an myelinoid bodies was often found within the secretory granule. 3. Cytochemical studies showed that acid phosphatase activities was observed in multivesicular bodies and lysosome, strong activities with the advance of age. And alkaline phosphatase activity are observed in microvilli, granule and lateral membrane of secretory granule cells, and strong activities with age. Consequently suggest that with the advance of age, tracheal epithelium show ultrastructural and cytochemical alteration of some kind of cell organelles in all kind of cell.

  • The ultrastructural changes and differentiation mechanism of chromoplasts and leucoplasts from Proplastids in root tip cells of Panax ginseng seedlings were studied with transmission electron microscope. Initial cells have so many proplastids with a few osmiophilic droplets and a lot of mitochon dria at early stage of germination, therefore electron density of cytoplasm is generally higher than that of the other cells just like periblem, plerome and root cap. Proplastids are observed in the initial cells, but only leucoplasts appeared in the central root cap cells. Because root cap cells are derived ultimately from initial cells, the cell organelles in the root cap cells are directly related by those of initial cells. This result postulates that leucoplast is diferentiated from proplastid, and this is the same with other's concepts. On the contrary, the precise observations of chromoplast with crystalline inclusions in the peripheral root cap cells can conclude the direct pathway of chromoplast development from proplastid. Because of the differences of these result from those of other experiments, new scheme of plastid development, direct differentiation of chromoplast from proplastid, can be postulated. And this is the originality of this research on the differentiation of plastids.

  • This study was undertaken to investigate ultrastructural changes according to the radiosensitivity in the spermatogenic cells and Sertoli cell of the seminiferous tubules in Korean native pheasants. During spermatogenetic period, testes were collected from male adult Korean native pheasant and they were used as experimental and control birds. The experimental group was divided into a single-dose whole body irradiation group (400, 600, 800 and 1,000 rads) and a split-dose whole body irradiation groups(400/2, 600/2, 800/2 and 1,000/2 rads). The experimental birds were sacrificed at 24 and 72 hrs after irradiation and the control pheasants were sacrificed at the same time. Ultrastructural changes of Sertoli cells and spermatogonia were investigated by ultrathin section with electron microscope. The results obtained are summarized as follows; 1. The apoptosis was observed after 72 hrs group of the single-dose irradiation of 400 rads. 2. The cytoplasmic organelles of spermatogonia were severely damaged more than that of sertoli cell in 72 hours group of split-dose irradiation of 800 rads. 3. The cytoplasmic organelles of Sertoli cell were severely damaged except the nuclear membrane of Sertoli cells in 72 hrs group of split-dose irradiation of 1,000 rads.

Effects on Ultrastructural Changes and Several Phosphatase Activities by Actinomycin D in Migrating Primordial Germ Cells of Developing Rat (발생중인 흰쥐 원생식세포의 이동과 미세구조 및 몇몇 Phosphatase의 활성에 미치는 Actinomycin D의 영향)

  • In this study, the pathway and date of migrating Primordial germ cells (PGCs) were observed light microscopically and ultrastructural changes of them during migration were observed by electron microscopic examination. For these purpose, alkaline phosphatase reactions were used for identifying the PGCs and acid phosphatase reactions were used for observing their degenerating activities. Also, effects of actinomycin D on the migration of PGCs were examined. According to these results, at the 9th gestation day, PGCs were observed in the endodermal cells of yolk sac, at the 11th gestation day, they were seen in the hindgut and then entered into the dorsal mesentery by the 13th gestation day. At the 14th gestation day, they were located in the genital ridges. When PGCs were located in the hindgut and genital ridges, the positive reactions of alkaline phosphatase were dominated, but acid phosphatase reactions were limited in all stage except they were in dorsal mesentery. However, these reactions were lessened in case of actinomycin D treatment. By electron microscopic examination, PGCs had pseudopodia, tail process, trailing cytoplasm and nuage as the ultrastructural characteristics. In addition, these morphological features were damaged by actinomycin D treatment.

Electron Microscopic Changes in the Epithelial Damage Of the Maxillary Mucosa Induced by Platelet Activating Factor (혈소판 활성인자에 의한 상악동 점막상피의 손상에 대한 전자현미경적 변화)

  • Platelet activating factor (PAP) has been known as implicating as one of potent inflammatory mediators and reported 0 be involved in inflammation and allergy. PAF induces ciliary dysfunction and epithelial damage of human paranasal sinus mucosa in vitro. However, several recent papers have reported that PAF may not readily damage the airway epithelium. The aim of this study was to investigate the ultrastructural evidence to elucidate the pathogenesis of epithelial damage induced by PAF. Sixteen $\mu\textrm{g}$ g of PAF was applied into the maxillary sinuses of 6 rabbits. Rabbits were divided into 2 subgroups along with time interval at 1st and 3rd experimental day, and sinus mucosae were taken for the histopathologic study using electron microscopy. At 1st day, epithelial cells showed no ultrastructural change. Ultrastructures of the cilia were well preserved. Subepithelial space showed no evidence of the infiltration of inflammatory cells. Intravascular platelet aggregation and swelling of endothelial cells were evident. At 3rd day, epithelial cells showed vacuolar degeneration. Fusion of cilia forming giant cilia and focal loss of cilia were evident. Eosinophils were infiltrated in subepithelial and intraepithelial space. Swelling of endothelial cells, and migration of inflammatory cells into the connective tissue were evident. This study implies that epithelial damage induced by PAF may be secondary to the cytotoxicity of mobilized eosinophils rather than direct cytotoxicity of PAF.

Ultrastructural Changes of Chinese Cabbage Root Tissues Associated with Pathogenesis of Plasmodiophora brassicae

  • Roots of Chinese cabbage (Brassica campestris var. chinensis) seedlings infected with Plasmodiophora brassicae were examined by light and electron microscopy to reveal histopathological changes related to pathogenesis in the susceptible host. The pathogen colonized the cortex and partly the stele as well, invading up to the xylem. Gall tissues could be differentiated from the initially infected tissues, involving less compact organization and new vascular development. The infected cells were much hypertrophied, and contained one to several plasmodia. Except cellular hypertrophy, no pathological ultrastructural modification was noted in the infected calls. Infected cytoplasm became dense with ground cytoplasm, inconspicuous central vacuole, and increased cellular organelles such as mitochondria and dictyosomes. There were two types of nuclear states of plasmodium, uninucleate and multinucleate. Both plasmodia were structurally similar, filled with lipid droplets, bounded with envelope, and containing mitochondria, endo-plasmic reticulum, and sometimes small vacuoles. Plasmodial fragmentation, which may be regarded as a way to discharge plasmodial materials into host cytoplasm, commonly occurred, forming plasmodial fragments by outgrowth of plasmodial cytoplasm and regional compartmentalization. Plasmodial fragments were degenerated sometimes followed by forming chains of spherical vesicles especially in the uninucleate plasmodial state. These ultrastructural features indicate the biotrophic nature of the pathogen associated with its pathogenesis in the susceptible host.

Ultrastructural Changes in the Ganglion and Granule-Containing Cells in the Heart of Vacor-Induced Diabetic Mongolian Gerbil (Vacor 유발 당뇨 모래쥐의 심장신경절과 과립함유세포의 미세구조)

  • The ultrastructural changes of the cardiac ganglion and granule-containing cells in the heart of vacor-induced diabetic Mongolian gerbils were studied by electron microscopy. After one month of vacor-induced diabetes the ganglion cells showed increase in numbers of dense bodies and mitochondria compared with the normal cardiac ganglion. Most of the satellite cells were filled with numerous phagosomes containing digested debris. Both electron-dense and lucent types of degenerating axon terminals were observed. The former was characterized by clusters of agranular vesicles and numerous mitochondria. The electron lucent type of degenerating axon terminal contained a few agranular vesicles and swollen mitochondria. Degenerating unmyelinated and myelinated axons contained large numbers of dense bodies, lamellar bodies, and mitochondria. Numerous macrophages containing phagosomes were reveled in the interstitial spaces. Some of the granule-containing cells in the heart showed a variety of degenerative changes and a decreased number of dense-cored vesicles. After three months of vacor-induced diabetes the unmyelinated and myelinated axons showed degenerative changes, whereas no structure changes could be demonstrated in intraatrial ganglion and granule containing cells. The satellite cells containing engulfed debris were observed in the cardiac ganglion cells. These results suggest that the degenerative changes occur in the cardiac ganglion cells of vacor-induced diabetic Mongolian gerbils as well as atrial granule-containing cells.

Effects of DMTU and SOD on Ultrastructural Changes of Gastric Chief Cells in Adriamycin Treated Rats (Superoxide dismutase 및 Dimethyl thiourea가 흰쥐 위샘 으뜸세포에서 Adriamycin 투여 후 나타나는 미세구조의 변화에 미치는 영향)

  • Adriamycin is a one of anthracyclin antibiotics isolated from the culture media of Streptomyces peucetius var casius. The formation of reactive oxygen metabolite by redox cycling during the metabolism and the inhibition of DNA synthesis results in antineoplastic effects of adriamycin. The authors have demonstrated the effects of SOD(superoxide dismutase) or DMTU (dimethyl thiourea), which are used as an antioxidant, on the ultrastructural changes of the gastric chief cells after the administration of adriamycin in the rat. Adriamycin (30 mg/kg) was administered intraperitoneally to the Sprague-Dawley rats weighing about 220 gm and SOD (15000 unit/kg) or DMTU (500 mg/kg) were administered intraperitoneally to the rats 30 minutes after the administration of adriamycin. The gastric chief cells 24, 48 and 72 hours after the administration of adriamycin were observed with Hitachi-600 electron microscope. The results were as follows. 1. SOD or DMTU alone did not affect the ultra structures of the gastric chief cells in the rat. 2. Dilation, sacculation and segmentation of the cisternae of rough endoplasmic reticulum, dilation of the saccules of Golgi complex and dilated mitochondria with electron lucent matrix were seen in the adriamycin treated rats. In the course of time, the ultrastructures of the chief cell changed markedly. 72 hours after drug administration, severely dilated cisternae of rough endoplasmic reticulum, with clumping of chromatin around the nuclear envelope and mitochondria with electron lucent matrix and dilated cristae were seen in the chief cell. 3. The treatment of SOD is more effective than DMTU to attenuated the ultrastructural changes of the chief cells in the adriamycin administered rat. Consequently it is suggested that adriamycin would induce the degenerative changes of the organelles of the chief cell. The treatment of SOD is more effective than DMTU to attenuate the adriamycin induced damage.
