Solubilization of Dairy Sludge using Ultrasonic Pretreatment (초음파를 이용한 유가공 슬러지의 가용화)

  • The effects of ultrasonic (1.2~1.7 kJ/g TS) pretreatment on the solubilization of dairy and livestock sludge were separately evaluated to investigate the possibility of recycling dairy sludge as a potential source of organic carbon. Compared to other industrial wastewater and sewage sludge, dairy sludge has higher organic matter content and no toxic materials. The solubilization rates of dairy and livestock sludge, at a specific energy input of 1.7 kJ/g TS, were 14.5% and 10.6%, respectively. After the 90-minute ultrasonic treatment, the soluble COD (chemical oxygen demand) increased about 7.1 times that of the initial SCOD, at an increase rate of $0.022m^{-1}$. In comparison, the increase in soluble nitrogen, which was ~3.4 times that of the initial soluble nitrogen concentration, was much smaller than the increase in SCOD; thus, the C/N ratio increased from 4.0 to 8.7.

  • The production process of cellulosic ethanol from rice straw using acidic saccharification was studied in this experimental work. The hydration by ultrasonic energy and the acidic saccharification using 10~30 wt% of $H_2SO_4$ were performed as pretreatment processes. Also, 10~50 wt% of yeast for 3~6 days was used for fermentation process. The yield of cellulosic ethanol was decided in the fermentation process. The optimum pretreatment condition was 375W of ultrasonic power and 30 min of hydration time using 20 wt% of $H_2SO_4$ and 2 h of the acidic saccharification time. Finally, the optimum fermentation condition was at the condition of 30 wt% of yeast and 3 days of fermentation time.

Studies of Pretreatment Mehtods for Additional Reduction of Sewage Sludge (최종 하수처리장 슬러지의 추가감량을 위한 슬러지 전처리 연구)

  • In this study, biochemical methane potential test was conducted to estimate ultimate methane and carbon dioxide yield for anaerobic digestion and pretreatment with sewage sludge cake. Two of 0.2 % TS of sewage sludge cakes were treated with 5M NaOH or sonication of 0.51 W/mL during 30 min respectively. Another sample was treated simultaneously with NaOH and sonication in same condition. Then, initial soluble COD increased from 33.1 mg/L to 494 mg/L. After BMP test, methane production ranged from 3.12 and 84.2 mL $CH_4$ per g of Volatile Solid (VS) and 9.2 and 13.5 mL $CO_2$ per g of Volatile Solid (VS) for carbon dioxide. In other tests, injection of nutrient media or sludge supernatant produced 73.1 and 73.8 mL $CH_4$ per g of Volatile Solid (VS) and 11.2 and 13.6 mL $CO_2$ per g of Volatile Solid (VS) respectively. When BMP test finished, 62 % of initial volatile solids decreased to 33.8~45.4 %. Simultaneous pretreatment increased soluble COD, reduction rate of volatile solids and digestion efficiency than those for alkaline and ultrasonic pretreatment.

Enhanced extraction and Antioxidant activity analysis of Flavone C-glycosides from Black bamboo using Ultrasonic wave (초음파를 이용한 오죽으로부터 Flavone C-glycosioes의 추출 및 항산화활성분석 향상)

  • In this work, the amounts of Flavone C-glycosides homoorientin, orientin extracted from Black bamboo by various ultrasonic waves frequency (35, 72, 170 KHz, 300 Watt $\pm$1) time (15, 30, 60 min) and temperature ($25^{\circ}C$) were compared using 50% aqueous ethanol solution. And describesanalysis of the antioxidant potential of Black bamboo using an high-performance liquid chromatography (HPLC) on-line $ABTS^+$antioxidant screening method. In conjunction with the analysis of their 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid ($ABTS^+$) radical scavenging ability. The optimum operating conditions were experimentally determined to analyze the Flavone C-glycosides homoorientin, orientin in the pretreatment ultrasonic extracts. From the results, the effect on extraction yield of variations in frequency, solvent composition and extraction time was investigated. The highest yield of Black bamboo was obtained by ultrasonic waves with a frequency of 72 KHz and an extraction time of 60 min, The extraction frequency 35 KHz ($25^{\circ}C$), time 30 min was selected as an optimal antioxidant activity condition.

Diamond Crystal Growth Behavior by Hot Filament Chemical Vapor Deposition According to Pretreatment Conditions

  • The change of the deposition behavior of diamond through a pretreatment process of the base metal prior to diamond deposition using HFCVD was investigated. To improve the specific surface area of the base material, sanding was performed using sandblasting first, and chemical etching treatment was performed to further improve the uniform specific surface area. Chemical etching was performed by immersing the base material in HCl solutions with various etching time. Thereafter, seeding was performed by immersing the sanded and etched base material in a diamond seeding solution. Diamond deposition according to all pretreatment conditions was performed under the same conditions. Methane was used as the carbon source and hydrogen was used as the reaction gas. The most optimal conditions were found by analyzing the improvement of the specific surface area and uniformity, and the optimal diamond seeding solution concentration and immersion time were also obtained for the diamond particle seeding method. As a result, the sandblasted base material was immersed in 20% HCl for 60 minutes at 100 ℃ and chemically etched, and then immersed in a diamond seeding solution of 5 g/L and seeded using ultrasonic waves for 30 minutes. It was possible to obtain optimized economical diamond film growth rates.

  • A type of titanium alloy, ferro-titanium, is the main material used to manufacture steel and stainless steel. Considering economic aspects, ferro-titanium ingots are intended to be manufactured using low-cost titanium scrap, and the best pretreatment process for removing impurities from recycled titanium scrap surfaces was studied here. Instead of ordinary acid or organic solvents, ecofriendly methods were researched and applied, and chip scrap materials were used. A high-quality ferro-titanium ingot was manufactured from titanium scrap after a pretreatment process was applied, and the impurities and properties were analyzed and compared with commercial material standards through a component analysis.

Extraction of Genistein and Formononetin from Sophoraflavescens Aiton using Ultrasonic wave (초음파를 이용한 고삼에 포함된 Genistein 및 Formononetin의 추출)

  • In this work, we the ettect on extraction amounts and general composition content of phytoestrogen genistein and formononetin extracted from Sophoraflavescens Aiton by various ultrasonic waves(35, 72, and 170 KHz) and extraction time(30, and 60 min) were compared using extraction solvent water 100%. The pretreatment step was composed of ultrasonic waves extraction, filtration, concentration, and membrane filtration. The extracted sample was analyzed by reversed-phase high performance liquid chromatography(RP-HPLC). And the mobile phase applied was linearly changed with A/B of 80/20~65/35 vol% for 60 min(A water/acetic acid, 99.9/0.1 vol%, B acetonitrile/acetic acid, 99.9/0.1 vol%). The experimental results, general composition carbohydrate(0.255 to 0.413%) excepts, other ingredients was confirmed almost similarly. Also, The highest yield of extraction amount 3.17g was obtained by ultrasonic waves with a frequency of 170 KHz and an extraction time of 60 min. This work offers would be useful for chemical and biological studies of natural plants and its products.

Optimum Conditions for Transformation of Synechocystis sp. PCC 6803

  • This study was conducted to determine the optimal conditions for introduction of exogenous DNA into Synechocystis sp. PCC 6803. Of the three transformation techniques studied, electroporation, ultrasonic transformation and natural transformation, natural transformation showed the highest efficiency. Additionally, this study demonstrated that the higher plasmid concentration and longer homologous recombining fragments resulted in a greater number of transformants. For successful transformation, the lowest concentration of plasmid was $0.02\;{\mu}g/ml$, and the shortest homologous recombining fragment was 0.2 kb. Use of Synechocystis sp. PCC 6803 in the logarithmic growth phase resulted in two-fold higher transformation rate than that of the same organism when cells in the latent phase or the plateau phase were used for transformation. Pretreatment of the host strain, Synechocystis sp. PCC 6803, with EDTA (2 mM) for two days prior to transformation increased the transformation efficiency by 23%. Additionally, incubation of the cells and DNA for 5 h under light conditions increased the transformation efficiency by two orders of magnitude. Moreover, recovery treatment of the cells before they were plated onto antibiotic medium also increased the transformation efficiency.

The Pretreatment Effects on Methane Fermentation of Microalgal Biomass (미세조류의 전처리에 따른 메탄발효 특성)

  • This study was conducted to identify the characteristics of methane fermentation and lysis effects of pre-treated microalgae. Chemical compositions of microalgae showed that the VS(volatile solid) was 86.1% of TS(total solid), and the protein was 63.5% of VS. These values were higher than those of activated sludge. The cell lysis test of raw microalgae biomass was conducted by many physicochemical methods. presenting that the degree of cell lysis was affected by following order: ultrasonic(100min.), alkali(pH 13), ultrasonic(10min), thermal($120^{\circ}C$), thermal($50^{\circ}C$), and acidic(pH 3) treatment. Methane fermentation with many pre-treated samples was performed, showing that the concentration of acetic acid was the highest. followed by propionic acid, butylic acid and valerie acid among all VFA(volatile fattic acid). In methane production. ultrasonic samples were only more effective than untreated one in total gas and methane productivity. but other samples were less effective. Especially. the alkalic sample had an inhibitation effect on methanogens.

The Effect of Pre-treatment on the Anaerobic Digestion of waste Activated Sludge (하수슬러지의 혐기적 소화효율 향상을 위한 전처리 효과)

  • The slow degradation rate of sewage sludge in anaerobic digesters is due to the rate limiting step of sludge hydrolysis. Therefore, the pre-treatment process had been carried out using acidic(pH 1.5, 3, 4, 5) and alkaline(pH9, 10, 13), thermal(50, 100, 150, $200^{\circ}C$) and ultrasonic treatment(400W, 20kHz, 15, 20, 25, 30, 35, 40, 50, 60min). In the best conditions of each treatment, the SCOD ratio(%) of treated/untreared samples were increased 102% in acid(pH5), 986% in alkali(pH13), 959% in thermal($200^{\circ}C$) and 1123% in ultrasonic(35min) treatment. As the result, the ultrasonic treatment was most effective, followed by alkali, thermal, acidic treatment. In the effects of total gas productivity, the thermal($200^{\circ}C$) pretreatment was the highest, followed by thermal($150^{\circ}C$), ultrasonic(90min), alkaline(pH9) and ultrasonic(50min).
