다양한 자외선(UVB) 처리조건에 의한 목이버섯의 품질 특성 (Quality Characteristics of Ear Mushroom by Various UVB (Ultraviolet B) Treatment Conditions)

  • To examine the possibility of ear mushroom (EM) as a source of natural vitamin D, the UVB (ultraviolet B) was treated according to sample drying status, drying methods before UVB treatment and harvest time. And then, vitamin D2 and ergosterol contents were investigated. According to the sample drying status, the vitamin D2 contents of fresh and freeze-dried EM (whole) increased to 4,634.4~4,780.9 ㎍/100 g D.W. (dry weight) under UVB dose 52.5~70.0 kJ/㎡ and above 18,693.1 ㎍/100 g D.W. under above 105 kJ/㎡, respectively. By drying methods before UVB treatment, vitamin D2 contents of EM powder (below 500 ㎛) that dried in the vinyl house and freeze-dryer increased to 4,886.2~5,132.9 ㎍/100 g D.W. under above 105 kJ/㎡ and 17,103.7 ㎍/100 g D.W. under 70 kJ/㎡, respectively. Ergosterol content decreased with increasing UVB dose in all experiments. According to the harvest time, vitamin D2 content under UVB dose 210 kJ/㎡ showed marked difference and in order of June, July, August, October and April. As for the results, the optimum harvest time, drying method before UVB treatment, sample size, UVB dose for the EM contained high vitamin D2 content were June, freeze-drying, whole, and 105 kJ/㎡, respectively.

The Signaling of UV-induced Apoptosis in Melanocytes

  • Ultraviolet B (UVB) radiation may activate or deteriorate cultured human epidermal melanocytes, depending on the doses and culture conditions. In this study, we examined whether apoptosis of melanocytes can be induced by physiologic doses of UVB irradiation. PI staining for DNA condensation and flow cytometric analyses demonstrated the apoptotic cell death of melanocytes after UVB irradiation. The level of p53 and Bax revealed a dose-dependent increase with increasing dose of UVB, but the level of Bcl-2 remained unchanged. Confocal microscopic examination showed that Bax moved trom a diffuse to a punctate distribution after UVB irradiation. However, there were no changes in the pattern of Bcl-2. We next examined the downstream targets of apoptosis. Our results showed that a precursor form of caspase-3 disappeared with increasing doses of UVB. We also observed cleavage of poly(ADP-ribose) polymerase (PARP) after UVB irradiation. In addition, UVB irradiation resulted in a remarkable activation of c-Jun N-terminal kinase (JNK). These results indicate that UVB may induce apoptosis via JNK activation in human melanocytes.

자외선(UVB)에 의한 염색체이상과 Tannic acid의 방어효과 (Suppressing Effects of Tannic Acid on UVB induced Chromosome Aberrations in Chinese Hamster Lung Cells)

  • We observed the frequency of chromosome aberrations induced by UVB irradiations, and the suppressing effect of tannic acid on chromosome aberrations induced by UVB irradiations in CHL cells, which is a phenolic compound, a hydrolysate of tannin and a components of green tea. UVB doses used for the frequency of chromosome aberrations were from 0.2 to 1.6 KJ/m$^2$ and tannic acid concentrations were from 1.16 $\mu$g/ml to 37.50 $\mu$g/ml. For the observation of suppressing effect of tannic acid on UVB-induced chromosome aberrations, UVB dose was 1.6 KJ/m$^2$ and tannic acid concentrations were 1.0, 2.0, 4.0 $\mu$g/ml. In our study, tannic acid was treated for 24 hours in CHL, cells after UVB irradiation without S9 mix or for 6 hours with S9 mix. From this study, we obtained the following results : (1) The frequency of chromosome aberrations UVB induced were dose-dependently increased. (2) The tannic acid did not induce chromosome aberrations in cultured Chinese hamster cells. (3) UVB-induced chromosome aberrations were suppressed by tannic acid at every concentration from 1.0 $\mu$g/ml to 4.0 $\mu$g/ml with or without metabolic activation. These results suggest that the tannic acid acts as an inhibitor to UVB-induced clastogenicity of the cultured cell.

Ultraviolet A Induces Immunosuppression, Protection or Memory Enhancement Depending on Dose, while Ultraviolet B is Immunosuppressive and Tolerogenic over a Large Dose Range

  • UVR-induced immunosuppression contributes to skin cancer. The aim was to construct accurate dose response curves for primary and secondary contact sensitivity for solar-simulated UVR (ssUVR; 290-400nm), UVA and UVB as the role of UVA in immunosuppression is controversial. We used a xenon arc source. The mice were immobilised, enabling accurate dosing. C57BL/6 mice were immunosuppressed at half the dose of ssUVR required to cause sunburn but not by higher doses (up to the sunburn dose). Thus, ssUVR causes systemic immunosuppression only over a narrow, low dose range. UVA caused suppression at low but not high doses whereas UVB induced immunosuppression at all doses tested. 8 weeks later the mice were resensitised to assess tolerance. Mice exposed to the minimum immunosuppressive dose of ssUVR prior to primary sensitisation were tolerant to re-sensitisation. However, at higher doses of ssUVR, these mice were protected from tolerance. Interestingly, while low doses of UV A caused immunosuppression, even lower doses enhanced the response to the second sensitisation. Higher doses of UVA had no affect. UVB induced tolerance in a dose related manner. Thus, ssUVR only induces immunosuppression and tolerance over a narrow dose range. Both UVA and UVB are immunosuppressive at this dose, while higher doses of UVA protect from the suppressive effects of UVB. Surprisingly very low doses of UVA enhanced memory development. Thus UVR has complex effects on the immune system depending on dose and spectrum.

  • Skin is constantly exposed to air, solar radiation, ozone and other air pollutants formulating free radicals. The reactive oxygen species(ROS), formed under these conditions, are associated with skin cancers, cutaneous photoaging, and cutaneous inflammatory disorders. In this study, we sought to establish an animal model for UVB-induced skin alteration using BALB/c mice. The level of UVB irradiation used in this model was within physiological dose. BALB/c mice were exposed to a single dose of UVB ($200mJ/cm^2$ and were sacrificed at 3, 6, 24, and 48 hours following the irradiation. The effect of a single exposure to UVB irradiation on skin catalase(CAT), superoxide dismutase(SOD), and glutathione peroxidase(GPx) activities were examined. Significant decrease in the activity of all enzymes were observed at 6 hours after irradiation(p<.05). The activity of CAT decreased more sharply than those of SOD and GPx, and then remained depressed until 48 hours after UVB irradiation, whereas the activity of GPx recovered to basal level at 48 h after UVB irradiation. Our results indicate that BALB/c mouse could be an adequate animal model of UVB irradiation experiment. These results will also provide fundamental knowledge for the effective nursing strategies in reducing UV-induced skin disorders.

UVB 조사에 의한 육미지황탕의 광노화 피부 억제에 관한 실험적 연구 (Experimental Studies on the Inhibitory Effects of Yukmijiwhang-tang on Photoaging Skin Induced by UVB Irradiation)

  • The purpose of this study is to investigate the protective effects of Yukmijiwhang-tang(YM) water extracts against the UVB irradiation on the human keratinocyte HaCaT cells. We observed the effects of YM on the oxidative stress, gene expression of pro-inflammatory cytokine such as TNF-${\alpha}$ and IL-$1{\beta}$, and matrix metalloproteinase-9 in UVB-irradiated HaCaT cells. On the effects of oxidative stress and antioxidant function on the treatment with YM, The activity of xanthine oxidase(XO) was significantly decreased by treatment of YM in all the concentrations(p<0.01). The activity of superoxide dismutase(SOD) and catalase(CAT) was significantly increased by treatment of YM in a dose dependent manner(p<0.05 and p<0.01). DPPH radical was erased by treatment of YM under dose of $500{\mu}g/m{\ell}$ concentration. Treatment of HaCaT cells with YM had also significantly reduced intracellular ROS produced by UVB irradiation in a dose dependent manner(p<0.05, p<0.01, p<0.001). Gelatin zymography assay showed that YM downregulated the MMP-9 activity in UVB-irradiated HaCaT cells. RT-PCR analysis revealed that YM suppressed the expression of IL-$1{\beta}$ and MMP-9 however, it has no effects on the expression of TNF-${\alpha}$ and MMP-3. Our study suggests that Yukmijiwhang-tang exert protective actions on the UVB-irradiated HaCaT cells largely by anti-oxidative and anti-inflammatory processes.

Guinea pid를 이용한 Glycolic acid 및 UVB의 피부 자극성 평가 (Skin Irriation Effect of Glycolic Acid and UVB in Guinea Pig)

  • Alpha-hydroxy acid(AHA) are used in cosmetic products as a pH adjuster, mild exfoliant and humectant-skin conditioner. Cosmetics containing higher concentration (30%) and lower pH (3.0) of AHA can cause side effects if it is applied without the prescription. For providing information on the safety of AHA and on human risk assessments we studied skin irritation effect of glycolic acid, one of the most commonly used AHA in guinea pigs. The skin irritation by glycolic acid was increased in a dose(10% to 70%), acidity (pH 2.5 to 5.5.) and length of exposure dependent manner (for up to 14 days), respectively. The combination treatment with UVB (0.4 or 3.0 J/$cm^2$) increased glycolic acid-induced skin irritation. Histological examination showed that hyperplasia of non-inflammatory cells in the epidermis of skin treated with high dose of glycolic acid (pH 3.0). There results show that glycolic acid increased skin irritation in a dose, length of exposure and pH dependent manner, respectively, in guinea pig, and the combination with UVB increased glycolic acid-induced skin irritation. The cell proliferation of non-inflammatory cell may be involved in high doses of glycolic acid-induced skin irritation. Long-term application of more than 30% of glycolic acid (pH 3.0) may cause skin irritation.

Meta-analysis를 이용한 UVB 조사량에 따른 피부암 발생 위해도의 예측 연구 (Prediction of the risk of skin cancer caused by UVB radiation exposure using a method of meta-analysis)

  • 성층권의 오존층 파괴로 수반되는 부작용에는 우주의 자외선이 차단되지 않고 지구표면에까지 도달하는 것을 들 수 있다. 이로 인하여 생태계 파괴를 비롯하여 기후이상 및 인체 건강장애 발생의 가능성이 높아진다고 알려져 있다. 특히 자외선 노출로 인한 피부암 발생에 관한 여러 연구결과가 보고 되었으며 이에 대한 종합적이고 신뢰성 있는 재평가 필요성이 대두되었으며 유사한 연구의 국내 수행이 제시되는 때이다. 이에 메타분석이라는 방법을 통하여 신뢰성있는 BAF 값을 추정하여 보고 이 지수를 국내 자료에 적용하여 국내 피부암 발생의 변화 양상을 자외선 증가와 함께 추정하여 보았다. 3개국의 자료를 일원화하여 추정된 UVB 계수는 지수함수 모델에 의해서는 $2.07\times10^{-6}$으로, 멱함수 모델에 의해서는 2.49로 산출되었으며, 이들 모두 통계적으로 유의한 것으로 나타났으며, 이 계수로부터 BAF 값을 추정한 결과, 서울 일부 지역에서의 UVB 조사량이 1% 증가되면, 피부암 발생률은 지수함수 모델에서는 1.90%, 멱함수 모델에서는 2.51%가 증가되는 것으로 산출되었으며, 이 연구에서 적용한 메타분석의 방법으로 제시된 위해도는 비교적 신뢰도가 높을 것으로 추정된다. 현재 우리나라에서의 비흑색종 피부암 발생율이 백만명당 11명이라고 가정할 때, UVB 조사량이 1% 증가됨으로 인해 국내 비흑색종 피부암 발생율은 백만명당 11.1명$\sim$11.3명으로 증가되는 것으로 예측된다. 이렇게 낮은 수준의 위해도에도 불구하고 자외선 노출이 피부암 발생의 원인적 요인이라는 것이 밝혀진 지금, 이에 대한 불필요한 노출을 가능한 삼가고 계속적인 관심을 두어야 할 것이다.

UVB 광손상에 대한 백작약의 보호 효과 (Protective Effect of Paeoniae Radix Alba against UVB Photodamage)

  • Objective : UVB damages skin health by causing skin redness and intense inflammation, sunburn, and skin cancer. Paeoniae Radix Alba has been used to relieve gynecological symptoms, muscle spasms, and skin ailments. This study was conducted to confirm whether it has a protective effect against UVB photodamage. Methods : Ethanol extract of Paeoniae Radix Alba (PRA) was prepared by extracting 100 g Paeoniae Radix Alba in 1 L of ethanol for 48 h. Apoptosis was monitored by the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay and expression levels of apoptosis indicator proteins, and tyrosinase activity was measured with a colorimetric commercial kit. Results : In human keratinocyte HaCaT cells, PRA reduced UVB-induced cell death through apoptosis by inhibiting PARP cleavage and caspase-3 and -9. UVB-induced increase in cellular reactive oxygen species (ROS) was suppressed by PRA pretreatment. PRA also showed dose-dependent ABTS and DPPH radical scavenging activities. Furthermore, the inhibitory effect of tyrosinase activity by PRA was confirmed. Conclusion : These results demonstrated the protective role of PRA in UVB photodamage of human keratinocytes, mainly due to its antioxidant and antiapoptotic properties. We also suggest that PRA can be considered as an effective natural agent to prevent skin photodamage.

Pueraria montana var. lobata Root Extract Inhibits Photoaging on Skin through Nrf2 Pathway

  • Pueraria montana var. lobata is a bioactive substance with various beneficial health effects and has long been extensively used as a traditional medication for the treatment of fever, acute dysentery, diabetes, and cardiovascular diseases in Northeast Asian countries. The purpose of this study was to evaluate the cytoprotective activity of Pueraria montana var. lobata ethanol extract (PLE) for ultraviolet B (UVB)-induced oxidative stress in human dermal fibroblasts (HDF). It was hypothesized that PLE treatment ($25-100{\mu}g/ml$) would reduce intracellular reactive oxygen species (ROS) levels as well as increase collagen production in UVB-irradiated HDF. The results confirmed this theory, with collagen production increasing in the PLE treatment group in a dose-dependent manner. In addition, regulators of cellular ROS accumulation, including HO-1 and NOQ-1, were activated by Nrf2, which was mediated by PLE. Hence, intracellular levels of ROS were also reduced in the PLE treatment group in a dose-dependent manner. In conclusion, PLE increases collagen production and maintains hyaluronic acid (HA) levels in human dermal fibroblasts exposed to UVB-irradiation, thereby inhibiting photoaging.
