Studies on Protoplast Formation and Regeneration of Coriolus versicolor (구름버섯의 원형질체(原形質體) 형성(形成)과 재생(再生)에 관한 연구(硏究))

  • To establish basic techniques for protoplast fusion of Coriolus versicolor several factors affecting protoplast formation and regeneration were investigated. Protoplast isolation was at maximum with 2.5-day cultured mycelia of C. versicolor treated with the combination of two enzymes, Novozym 234 (10 mg/ml) and cellulase Onozuka R-10 (15 mg/ml), for 3-4.5 hours at $30^{\circ}C.$ As an osmotic stabilizer for stabilizing the protoplast, 0.6 M sucrose was the best for formation and regeneration of the protoplast from the mycelia of the fungus and the regeneration frequency was 3.48%. Protoplast fusion was made by a modified method of Peberdy using PEG (M.W. 4,000). The fusion frequency between two mutants of C. versicolor was 1.86% and the fusion products showed differences in growth rate and colony morphology.

  • Silver polymer electrolytes are very promising membrane materials for the separation of olefin/paraffn mixtures. Olefin molecules are known to be transported through reversible complex formation with silver ions entrapped iii polymer matrix. However, they have poor long-term stability, which is very important fur the industrial application; the selectivity through the membrane decreases gradually with time mostly due to the reduction of silver ions ($Ag^+$) into silver nanoparticles ($Ag^0$). In this study, the stability of silver polymer electrolyte was investigated for poly(vinyl pyrrolidone) (PVP) and $AgBF_4$ system containing a surfactant, i.e. $C_{18}H_{35}(OCH_2CH_2)_{20}OH$ (Brij98) as a stabilizer. The reduction behavior of silver ions to silver nanoparticles in PVP was also investigated by atomic force microscopy (AFM) and UV-visible spectroscopy. It was found that the growth of silver nanoparticles was slower and selectivity of polymer electrolyte for propylene in propylene/propane was maintained longer time when Brij98 was added as a stabilizer.

Dual Role of Acidic Diacetate Sophorolipid as Biostabilizer for ZnO Nanoparticle Synthesis and Biofunctionalizing Agent Against Salmonella enterica and Candida albicans

  • In the present study, a yeast species isolated from CETP, Vellore, Tamilnadu was identified as Cryptococcus sp. VITGBN2 based on molecular techniques and was found to be a potent producer of acidic diacetate sophorolipid in mineral salt media containing vegetable oil as additional carbon source. The chemical structure of the purified biosurfactant was identified as acidic diacetate sophorolipid through GC-MS analysis. This sophorolipid was used as a stabilizer for synthesis of zinc oxide nanoparticles (ZON). The formation of biofunctionalized ZON was characterized using UV-visible spectroscopy, XRD, scanning electron microscopy (SEM), and Fourier transform infrared spectroscopy. The antimicrobial activities of naked ZON and sophorolipid functionalized ZON were tested based on the diameter of inhibition zone in agar well diffusion assay, microbial growth rate determination, protein leakage analysis, and lactate dehydrogenase assay. Bacterial pathogen Salmonella enterica and fungal pathogen Candida albicans showed more sensitivity to sophorolipid biofunctionalized ZON compared with naked ZON. Among the two pathogens, S. enterica showed higher sensitivity towards sophorolipid biofunctionalized ZON. SEM analysis showed that cell damage occurred through cell elongation in the case of S. enterica, whereas cell rupture was found to occur predominantly in the case of C. albicans. This is the first report on the dual role of yeast-mediated sophorolipid used as a biostabilizer for ZON synthesis as well as a novel functionalizing agent showing antimicrobial property.

A cost-effective method to prepare size-controlled nanoscale zero-valent iron for nitrate reduction

  • Nanoscale zero-valent iron (nZVI) has proved to be an effective tool in applied environmental nanotechnology, where the decreased particle diameter provides a drastic change in the properties and efficiency of nanomaterials used in water purification. However, the agglomeration and colloidal instability represent a problematic and a remarkable reduction in nZVI reactivity. In view of that, this study reports a simple and cost-effective new strategy for ultra-small (< 7.5%) distributed functionalized nZVI-EG (1-9 nm), with high colloidal stability and reduction capacity. These were obtained without inert conditions, using a simple, economical synthesis methodology employing two stabilization mechanisms based on the use of non-aqueous solvent (methanol) and ethylene glycol (EG) as a stabilizer. The information from UV-Vis absorption spectroscopy and Fourier transform infrared spectroscopy suggests iron ion coordination by interaction with methanol molecules. Subsequently, after nZVI formation, particle-surface modification occurs by the addition of the EG. Size distribution analysis shows an average diameter of 4.23 nm and the predominance (> 90%) of particles with sizes < 6.10 nm. Evaluation of the stability of functionalized nZVI by sedimentation test and a dynamic light-scattering technique, demonstrated very high colloidal stability. The ultra-small particles displayed a rapid and high nitrate removal capacity from water.

  • In this study, poly(DL-lactide-co-glycolide) nanoparticles loaded with water-insoluble vitamins such as vitamin A (Retinol) and vitamin E acetate were prepared by the emulsification diffusion method. Polymer solution was prepared by the two water-miscible organic solvent, such as ethanol and acetone. Because of its biocompatible property, polyethyleneglycol-polypropyleneglycol diblock copolymer was used as surfactant and stabilizer. The influence of some preparative variables on the nanoparticle formation and on the loading efficiency of active agents, such as the type and concentration of stabilizing agent, the stirring methods, the water/oil phase ratio and the polymer concentration were investigated in order to control and optimize the process. After preparation of nanoparticles loaded with active agent, particle size and distribution were evaluated by the light scattering particle analyzer. The loading efficiency of active agents was evaluated by the UV-visible spectroscopy. As the results, particle size were 50-200 nm and dispersibility was monodisperse. The optimum loading efficiency of active agents was observed 50-60%. It was found that the appropriate of selections of binary solvent mixtures and polymeric concentrations in both organic and aqueous phases could provide good yield and favorable physical properties of PLGA nanoparticles.

  • The optimum conditions of the 1st and 2nd distillation had been investigated to obtaine a high quality monomeric MDI and fire reactive polymeric MDI by purification of crude MDI. Effect of additives on the monomeric MDI's color change, dimerization and the reactivity of polymeric MDI with standard polyol system has been tested. When the monomeric MDI yield is approximately 32%, 4,4'-MDI content is above 98% in the monomeric MDI at the 1st distillation. When the separation ratio of initial portion and residue percentage, reflux ratio are set at respectively, approximately 20wt%, 9wt%, above 2 in order to minimize the content of 2,4'-MDI in monomeric MDI, the freezing point of final distilled monomeric MDI is above $38.4^{\circ}C$. Since the monomeric MDI is inherently unstable in the room condition, monomeric MDI easily changes it's color and conducts self-polymerization reaction. To increase the stability of monomeric MDI, the composition of antioxidant, which is composed of phenolic 1st antioxidant, phosphorus 2nd antioxidant, UV absorbent and Hindered amine light stabilizer are used, and benzoyl chloride as antipolymerization agent test are that APHA color is less than 20, dimer content is remained less than 0.36wt% after 45 days storage of monomeric MDI.

The Effect of Mg Precursors on Optical and Structural Characteristics of Sol-Gel Processed Mg0.3Zn0.7O Thin Films (졸-겔법으로 성장시킨 Mg0.3Zn0.7O 박막의 Mg 전구체의 종류에 따른 광학적·구조적 특성에 관한 연구)

  • In this study, MgxZn1-xO thin films, which can be applied not only to active layers of light-emitting devices (LEDs), such as UV-LEDs, but also to solar cells, high mobility field-effect transistors, and power semiconductor devices, are fabricated using the sol-gel method. ZnO and Mg0.3Zn0.7O solution synthesized by the sol-gel method and the thin film were grown by spin coating on a Si (100) substrate and sapphire substrate. The solutions are synthesized by dissolving precursor materials in 2-methoxyethanol (2-ME) solvent, and then monoethanolamine (MEA) was added to the mixed solution as a sol stabilizer. Zinc acetate dihydrate is used as a ZnO precursor, while Mg nitrate hexahydrate and Mg acetate tetrahydrate are used as an MgO precursor. Then, the optical and structural characteristics of the fabricated thin films are compared. The molar concentration of the Zn precursor in the solvent is fixed at 0.3 M, and the amount of the Mg precursor is 30% of Mg2+/Zn2+. The optical characteristics are measured using an UV-vis spectrophotometer, and the transmittance of each wavelength is measured. Structural characteristics are measured using X-ray diffraction (XRD) and transmission electron microscopy (TEM). Composition analyses are performed using energy dispersive X-ray spectroscopy (EDS). The Mg0.3Zn0.7O thin film was well formed at the ratio of the Mg precursor added regardless of the type of Mg precursor, and the c-axis of the thin film was decreased, while the band gap was increased to 3.56 eV.

Cell Fusion of Cellulolytic Fungi, Aspergillus sp. HB1 (섬유소 분해균 Aspergillus sp. HB 1의 세포융합에 관한 연구)

  • The author isolated high cellulolytic fungi from natural sources and determined optimal condition of protoplast formation and fusion as fundamental step for improvement of the isolated it's cellulolytic ability. Three different cellulolytic fungi, such as Aspergillus sp., Penicillium sp. and Trichoderma sp., were isolated from soil. Their cellulolytic activities were compared with that of Aspergillus niger which was useful industrially and had cellulase activity. It was Aspergillus sp. that showed the highest activity of all these four fungi. And then it was followed by Penicillium sp., Trichoderma sp., and Aspergillus niger in order. An auxotrophic mutant of Aspergillus sp. was obtained by UV mutagenesis method. Having try to produce protoplast from mycelia, the author found that ${\beta}-glucuronidase$, at pH 6.0, was effective cell-wall lytic enzyme. And the optimal concentration of this enzyme was 5,000 unit/ml. Regeneration rates of wild type, met. auxotroph and arg. auxotroph, in presence of osmotic stabilizer, were 7. 0%, 7. 5% and 5.2%, respectively. PEG with M.W. 6,000 was effective stimulator for protoplast fusion in the concentration of 30% (W IV). In such a condition, we obtained 1.2% cell fusion rate.

Thermal Stability of Polypropylene-Based Wood Plastic Composites by The Addition of Ammonium Polyphosphate (폴리인산염 첨가에 의한 폴리프로필렌 기반의 Wood Plastic Composites 열안정성)

  • In order to improve the thermal stability of wood plastic composites (WPC), thermal degradation behavior of WPC in this study was investigated by the addition of wood flour and fire retardant after hybridization of wood flour and ammonium polyphosphate (APP) into polypropylene (PP) matrix. Thermal degradation behavior of all formulations was analyzed with thermogravimetric analyzer under nitrogen environment at heating rate of $10^{\circ}C/min$. As the thermal degradation temperature of wood flour is lower than that of PP, char layer formed by the wood flour decreases the speed of heat transfer to PP. In addition, the char layer increases the 2nd thermal degradation temperature and decreases the 2nd thermal degradation speed. The WPC treated with APP increases the 1st and 2nd degradation temperatures. In the case of WPC with high loading level of wood flour, the 1st thermal degradation temperature and 2nd thermal degradation rate were increased by the addition of APP, and then the amount of remnants at high temperature was increased by the increase of the APP loading level. In the case of WPC treated with APP, the amount of the remnants at high temperature was increased with the increase of wood flour content from 10 wt% to 50 wt%, indicating that char formation of the APP and wood flour occurred at the same time, resulting in high thermal stability effect by the increase of wood flour content.

Controll over the Au@Ag Core-shell Nanoparticle 2D Patterns via Diblock Copolymer Inverse Micelle Templates and Investigation of the Surface Plasmon Based Optical Property (이중블록공중합체 역마이셀 주형을 이용한 Au@Ag 코어-쉘 나노입자 2차원 패턴 제어 및 표면 플라즈몬 기반 광학적 특성 연구)

  • We demonstrated unique inter- and intra-plasmonic coupling effects in bimetallic Au@Ag core-shell NP arrays which are regularly or randomly arranged on self-assembled block copolymer (BCP) inverse micelle monolayers. Polyvinylpyrrolidone (PVP)-stabilized Au@Ag core-shell NP arrays in regular or disordered configuration were incorporated and assembled on reconstructed PS-b-P4VP inverse micelle templates through two types of processes. The intensively enhanced LSPR coupling properties of individual and assembled Au@Ag NPs were evaluated by UV-visible spectroscopy in terms of the type of ligand stabilizer, coupling between Au and Ag, thickness of Ag shell, and type of array configuration. Finally, Au@Ag core-shell NP arrays were employed as active substrates for surface enhanced Raman spectroscopy (SERS) and a significantly enhanced signal enhancement was observed in accordance with the coupling intensity of Au@Ag NPs patterns.
