Effects of Egg Shell Membrane Hydrolysates on UVB-radiation-induced Wrinkle Formation in SKH-1 Hairless Mice

  • This study was conducted to examine the effect of egg shell membrane hydrolysates (ESMH) on wrinkle, UV, and moisture protection for cosmetic use. ESMH were fragmented as whole ESMH (before fractioning), Fraction I (> 10 kDa), Fraction II (3-10 kDa), and Fraction III (< 3 kDa). In order to test whether fractionated ESMH can be used for functional cosmetic materials, we examined not only the level of hyaluronic acid and collagen production, but also the MMP-1 activity using a HaCaT and CCD-986Sk cell line. Our study treated each sample of fractionated ESMH with different concentrations (0.01, 0.1, 1 mg/mL). In our in vivo research, we used hairless mice that had been exposed to UV-B to induce wrinkles for 7 wk, then applied Fraction I to the treatment group for 5 wk and then tested skin thickness, minimum erythema dose and moisture content. In addition, Fraction I was high in collagen and HA biosynthesis and it was better than TGF-${\beta}$ in improving of the skin. When TNF-${\alpha}$ caused MMP-1 activity in the CCD-986Sk cells, the whole ESMH and Fraction I proved to be effective in hindering the induction of collagenase depending on the concentration, and also showed outstanding effects in the suppression of skin aging. We found that the treatment group mice's UV-B radiation-induced skin damage was largely mitigated compared to that of the non-treatment group mice. Thus, we have concluded that EMSH helps to mitigate UV-B radiation-induced wrinkles, collagen, HA, MMP-1 activity and can be used for functional cosmetic materials.

Enhancement of Skin-Whitening and UV-Protective Effects of Centella asiatica L. Urban by Utrasonification Process (초음파 병행을 통한 병풀의 미백 및 자외선 차단 활성 증진 효과)

  • Enhancement effect of ultrasonification process on UV-protection and skin-whitening activities using Centella asiatica L. Urban extract was investigated. Cytotoxicity of the extracts measured on human skin fibroblast cells, CCD-986sk, and then, ultrasonification associated extracts showed 5~9% lower cytotoxicity then normal crude extracts on 1.0 mg/$m{\ell}$ of highest sample concentration. The associated extrats showed highest inhibition activity of hyaluronidase on 1.0 mg/$m{\ell}$ of concentration as 54.2%. Also, the associated extract reduced expression of MMP-1 on UV-irradiated CCD-986sk cells down to 100.2% from 136.1%, and revealed high inhibitory potency on tyrosinase as 74.6% by adding 1.0 mg/$m{\ell}$ of concentration. Ultrasonification associated extract showed strong inhibition effect of melanin production on Clone M-3 cells as 84.2% by adding 1.0 mg/$m{\ell}$ of concentration. From the preliminary observations, we considered that the extracts from C. asiatica could be potent natural materials for skin-whitening and anti-aging agent, and could enhance the activities by ultrasonification process.

Synthesis and Characterization of Mica Coated with Zinc Oxide Nanoparticles (산화 아연 나노 입자로 도포된 마이카의 합성 및 특성 규명)

  • In this work, we have prepared the nanocomposite by the reaction of mica and zinc oxide, and investigated the application of nanocomposite to UV protecting creams. Mica treated with 3-aminopropyltrimethoxysilane (APTMS) reacted with 1,4-phenylenediisothiocyanate (PDC) to give -N=C=S functionalized surface, which was further reacted with zinc oxides coated with APTMS to give mica-zinc oxide nanocomposites. The composites were characterized by EA, EDS, TGA, SEM, zeta potential measurement, powder XRD, and DRS UV/Vis analyses. Finally, we measured transmittances of ultraviolet protection creams manufactured by using mica composite covered with zinc oxides in the range of 280~400 nm. The nanocomposites developed in this work might be applicable as inorganic hybrid materials for UV protecting creams.

Function of muc Gene on Mutagenesis and DNA Repair (돌연변이와 DNA 손상회복에 미치는 muc 유전자의 기능)

  • To determine whether the muc region of pKM101 and its mutant pSL4 is sufficient for the expression of these phenotypes, muc regions of pKM101 and pSL4 were subcloned onto the highcopy number vector pKB354 and were selected the cloned pJB200 and pJB210. The recombinant plasmids pJB200 and pJB210 were introduced into umu $C36^{-}$ uvr $A6^{-}$ (TK610) strain and determined the protection effect and mutagenecity for UV and MMS. The protection effect and mutagenecity of umu $C36^{-}$ uvr $A6^{-}$ (TK610) were supressed by muc gene of the recombinant plasmids. The muc gene of pSL4 has higher effect than that of pKM101. The recombiant plasmid pJB210(inclued muc gene of pKM101) did not affect uv-mutagenesis in the $recA^{-}$ (JC2926) mutant.

Inhibitory effects of Prunus persica flower extracts on UV-induced skin damage

  • For an attempt to develop safe materials protecting UV-induced skin damage, plant extracts were evaluated for their antioxidative and free radical scavenging activities. From the results of these screening procedures, the ethanol extract of the flowers of Prunus persica was selected for further study. It was found that Prunus persica (50-200 $\mu\textrm{g}$/㎖) inhibited UVB-induced DNA damage measured by tail moment in the Single Cell Gel Electrophoresis(COMET assay) and inhibited UV-induced lipid peroxidation, expecially against UVB-induced peroxidation at higher than 10 $\mu\textrm{g}$/㎖. Also P.persica(100∼1,000 $\mu\textrm{g}$/㎖) inhibited the amount of $\^$14/C-arachidonic acid metabolites release from UVB-irradiated keratinocytes and it possessed the protective activity against UV-induced cytotoxicity of keratinocytes. All these results indicate that the flowers of P. persica extract may be beneficial for protection UV-induced skin damage when topically applied.

Betula Platyphylla var. Japonica Extract Prevent Ultraviolet C Light-induced Cell Damage in Chinese Hamster Fibroblast (V79-4) Cells

  • The present study reports the protective properties of a total methanol extract of B. platyphylla var. japonica against ultraviolet (UV)-C irradiation. Pretreatment of Chinese hamster fibroblast (V79-4) cells with a total methanol extract significantly increased cell survival following $300\;J/m^2$ of UV-C irradiation. The total methanol extract was further fractionated into 5 fractions: n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these fractions, B. platyphylla var. japonica ethylacetate, butanol and water fractions showed significant protective effects against the cellular damage induced by UV-C irradiation. In order to elucidate the mechanism underlying this protective effect, DPPH (Editor note: abbreviations should be spelled out at first use.) radical scavenging and lipid peroxidation inhibitory activity were measured. Significant radical scavenging and lipid peroxidation inhibitory activities were observed for the ethylacetate fraction. In summary, the present data demonstrate that an extract of B. platyphylla var. japonica has a significant protective effect against UV-C irradiation. The underlying mechanism of this protective effect may involve radical scavenging and inhibition of lipid peroxidation by the B. platyphylla var. japonica extract.

  • We made a multi-layer conveyor sterilizing system using UV rays to prolong a storage period of dried marine products. The system is composed of 5 layer conveyor belt of 3 m long each, thus 15 m long in total. It is designed to radiate UV rays for 10 minutes the dried marine products, and can process 300 kg/hour. Eight UV lamps of 30 cm long and 15 W of power with 254 nm wave length are installed in parallel, 15 cm high above in each belt, and 40 lamps in total. The whole system is covered by 1 mm thick metal plates to block the UV rays except a front side which is covered with 5 mm thick plastic plates coated with UV protection film to survey inside of the system. A sterile ratio is about 99.97% with 10 minutes sterilization using this system.

  • UV curing technology becomes important in various sectors of applications due to the high efficiency, environmental protection and saving energy. Above all, It is necessary to consider the formulation of glass fiber sizes in some detail to achieve this study since they are governed by the products to be made and their performance in the composites made form them. The effect of glass fiber sizes against UV curing system was investigated by the method of changing size formulation. Nanosil 700 was used in order to increase curing efficiency. Yellow effect was not observed at the 1% fiber sizer content.

Study of Photocatalytic Activity and Phostability of ZnO Particles Coated with UV-stable Polydimethylsiloxane

  • ZnO particles with a size range of 50-150 nm were coated with polydimethylsiloxane (PDMS) with a thin film thickness of 3-4 nm using a simple ambient-pressure chemical vapor deposition methods. Surfaces consisting of the PDMS-coated ZnO nanoparticles were found to be superhydrophobic with a water contact angle higher than $160^{\circ}$. The superhydrophobicity was sustained in the presence of UV light. Photocatalytic activity and photocorrosion of ZnO were nearly completely quenched in the presence of PDMS coating. It is suggested that our PDMS-coating can be of potential interest for the application of ZnO in UV protection agents and energy and electronic devices.

  • UV-screening agents have some adverse effects that raise consumers' concern. The organic agents often cause irritation and may penetrate into human body while the inorganic agents raise aesthetic issues because they often turn opaque. Organic agents with high molecular weights and nano-sized inorganic agents have been developed respectively to minimize transdermal intrusion into human body and suppress turning opaque. Recently, we reported preparation of powdery UV-screening agents made of polysilsesquioxane, an organic-inorganic hybrid material. Powders would not penetrate into epidermis and organic-inorganic hybrid nature would suppress the opaqueness problem. In this study, we continued our research on this powdery polysilsesquioxane UV-screening agent, SESQUV, regarding its chemical composition, its synergic UV-screening effects when mixed with other organic agents, and its applicability in practical formulation. Results showed SESQUV was promising UV-screening agents useful in sunscreen formulation.
