Harmful Effects of UV Light on the Skin (피부에 대한 자외선의 유해작용)

  • It is well hewn that ultraviolet light(UV) penetrates into the skin depends on the wavelength and causes many biochemical reactions in the skin. The enormous amount of information reveals that UV effects sun-burn, sun-tan and photoaging etc. In addition to these, it is now recognized that UV initiates immuno-suppression, and associated with this, skin cancer. In this review, I tried to illuminate the processes of injurious effects of UV on the skin.

A study on the curing characteristics of multi-concentrating UV-LED Curable Coating (다중 집광성 UV-LED 경화형 코팅의 경화특성에 관한 연구)

  • We investigated the curing properties of cured coatings for a multi-focal UV-LED. The coatings are for LEDs that operate at multiple UV wavelengths, unlike conventional single-wavelength UV-LEDs. Using UV-LED light sources with wavelengths of 365, 395, 420, and 450 nm, we analyzed the optical characteristics such as the direction of light flux and light source. We also analyzed the curing characteristics at each UV-LED wavelength to optimize the LED for composite wavelengths. The curing performance state was predicted through computer simulation for when the multiple wavelengths of UV light sources are superimposed, and then actual LEDs were designed and fabricated. To improve the internal high-speed curing, a multi-spot module was fabricated, in which each LED is condensed, and multiple wavelengths are synthesized and condensed at the same position. The adhesive strength, surface hardness, and internal hardness of the curing agent were tested by varying the wavelength combination conditions. The surface hardening and internal hardening were compared and analyzed using a hardness tester and FT-IR analyzer. As a result, the characteristics of the surface and internal hardness were improved by a multi-spot method in which four wavelengths were overlapped in a UV-LED rather than a single wavelength.

Dose-dependent UV Stabilization of p53 in Cultured Human Cells Undergoing Apoptosis Is Mediated by Poly(ADP-ribosyl)ation

  • The effect of poly(ADP-ribosyl)ation on the stability of p53 in SK-HEP1 cells treated with UV light was examined. Intracellular levels of p53 increased in cells treated with a low dose of UV light ($20J/m^2$), whereas they increased but then declined after a higher dose of UV ($100J/m^2$). Intracellular levels of p53 in the UV treated SK-HEP1 cells were dependent on the UV dose. Use of proteasome inhibitors revealed that p53 is degraded by proteasomal proteolysis after high doses of UV light. We present evidence that, at low doses, poly(ADP-ribose)polymerase (PARP) poly(ADP-ribosyl) ates p53 and protects it from proteasomal degradation before caspase-3 is activated, whereas at high doses the cells undergo UV induced apoptosis and PARP is cleaved by caspase-3 before it can protect p53 from degradation. Destabilization of p53 by cleavage of PARP may be important in cell fate decision favoring apoptosis.

Dissolved Organic Matter (DOM) Leaching from Microplastics under UV-Irradiation and Its Fluorescence P roperties: Comparison with Natural P articles (UV 광풍화에 의한 미세플라스틱 기원 유기물 용출과 형광 특성: 자연유래 유기성 입자와의 비교)

  • Numerous studies have investigated the occurrence and fate of microplastics in the environment; however, only limited effort has been devoted to exploring the characteristics of dissolved organic matter (DOM) leached from microplastics. In microplastic (MP)-contaminated environment, MPs are typically mixed with naturally-occurring particles, which interferes with their detection in the environment. Thus, it is necessary to distinguish between the DOM leached from MPs and those leached from natural particles and also to characterize their properties. This study investigated DOM leaching behavior from MPs (polystyrene: PS, polyvinylchloride: PVC) and natural particulates (forest soil: FS, litter leaves: LL) under light, which is considered one of the main weathering processes that affect MPs in the environment. The leached DOM concentrations and fluorescence characteristics were compared under dark versus light conditions. Regardless of the origins, UV light promoted DOM release from all the particulates. More DOM was released from natural particles than from MPs under both conditions. However, the effect of promoting DOM release by UV was more pronounced for MPs than for natural particles. It was observed from fluorescence spectra that the intensity of the humic-like region was substantially reduced when MP-derived DOM was exposed to UV light, whereas the change of intensity was very little for natural particles. Under light conditions, the ratio of protein-like to humic-like fluorescence of MP-derived DOM was higher than that of DOM from natural particles. This study implies that a substantial amount of DOM could be leached from MPs even in MP-polluted environment under UV irradiation. Protein/humic fluorescence ratio could be utilized as a fast probing indicator to separate the two sources of particles under light.

Degradation of herbicide paraquat by Fenton reagent and UV light irradiation (Fenton 시약 및 UV 광 조사에 의한 제초제 paraquat의 분해)

  • This study was to investigate the potential degradation of a herbicide paraquat by Fenton reagents(ferric ion and hydrogen peroxide) under UV light irradiation(365 nm) in an aqueous solution. When $10{\sim}500$ mg/L of paraquat was reacted with either ferric ion or hydrogen peroxide in the dark or under UV light, no degradation was occurred. However, the simultaneous application of both ferric ion(0.8 mM) and hydrogen peroxide(0.140 M) in paraquat solution(500 mg/L) caused dramatic degradation of paraquat both in the dark (approximately 78%) and under UV light(approximately 90%). The reaction approached an equilibrium state in 10 hours. In the dark, when $0.2{\sim}0.8$ mM ferric ion was added, $20{\sim}70%$ paraquat of $10{\sim}500$ mg/L was degraded, regardless of hydrogen peroxide concentrations($0.035{\sim}0.140$ M), while under UV light, 95% of 10 and 100 mg/L paraquat was degraded regardless of ferric ion and hydrogen peroxide concentrations. At paraquat concentration of 200 and 500 mg/L, paraquat degradation increased with increasing ferric ion concentrations as in the dark. However the increase in hydrogen peroxide concentration did not affect the extent of paraquat degradation. The initial reaction rate constants(k) for paraquat degradation ranged from 0.0004 to 0.0314, and 0.0023 to 0.0367 in the dark and under UV light, respectively. The initial reaction rate constant increased in proportion to the increase in ferric ion concentration in both conditions. The half-lives of paraquat degradation(t1/2) were 20 - 1,980 and 19 - 303 minutes in the dark and under UV light, respectively. This study indicates that Fenton reagents under UV light irradiation are more potent than in the dark in terms of herbicide paraquat degradation in an aqueous solution.

The Changes of Chlorophyll and Glycoalkaloid Contents in Potato Tubers after Exposure of Fluorescent and UV Light (광선의 영향에 따른 감자의 Chlorophyll과 Glycoalkaloid의 함량 변화)

  • To determine changes of chlorophyll and potato glycoalkaloid(PGA) contents in potato tubers after lightening, Dejima potatoes cultivated in Korea were used and PGA was analyzed by HPLC. Potatoes were stored under fluorescent and UV light exposure or darkness for 7 days at $5^{\circ}C$. The contents of chlorophyll and glycoalkaloid in the peel of potatoes exposed to fluorescent light were increased to $84\%,\;69.5\%$ respectively compared to potatoes stored without lightening. While a marked effect of lightening on the peel of potatoes exposed to fluorescent light, UV light lead to no change of chlorophyll and less increase in glycoalkaloid than fluorescent did. The increase of chlorophyll was provoked by only fluorescent light. Cortex part did not accumulate any chlorophyll or glycoalkaloid.

The Study of Thermal Effect Suppression and Wavelength Dependence of Azobenzene-coated FBG for UV Sensing Application (UV광 측정용 아조벤젠 코팅된 FBG의 열적 효과 제거 및 파장 의존성에 대한 연구)

  • In the paper, we have demonstrated an azobenzene-coated fiber Bragg grating (FBG) for monitoring ultraviolet light (UV) intensity in remote measurement. The elasticity of the coated azobenzene polymer is changed by the UV light, which induces a center wavelength change corresponding to the change of the FBG's grating period. The wavelength shift resulting from both UV light and other light with the wavelength out of the UV range was about 0.18 nm. In order to improve the accuracy of the measurement, the center wavelength shift caused by radiant heat of the light source was sufficiently removed by using a thermal filter. The amount of the center wavelength shift was consequently reduced to 0.06 nm, compared to the result without the thermal filter. Also, the FBGs coated by using azobenzene polymer were produced by two different methods; thermal casting and UV curing. Considering temperature dependence, UV curing is more suitable than thermal casting in UV sensor application of the azobenzene-coated FBG. In addition, we have confirmed the wavelength dependence of the optical sensor by means of four different band pass filters. Thus, we found out that the center wavelength shift per unit intensity is 0.029 [arb. unit] as a maximum value at 370 nm wavelength region and that the absorption spectrum of the azobenzene polymer was very consistent with the wavelength dependence of the azobenzene-coated FBG.

Improvement in Light Extraction Efficiency of 380 nm UV-LED Using Nano-patterned n-type Gan Substrate (나노 구조의 패턴을 갖는 n-type GaN 기판을 이용한 380 nm UV-LED의 광 추출 효율 개선)

  • Ultraviolet (UV) light emitting diodes (LEDs) were grown on a patterned n-type GaN substrate (PNS) with 200 nm silicon-di-oxide (SiO2) nano pattern diameter to improve the light output efficiency of the diodes. Wet etched self assembled indium tin oxide (ITO) nano clusters serve as a dry etching mask for converting the SiO2 layer grown on the n-GaN template into SiO2 nano patterns by inductively coupled plasma etching. PNS is obtained by n-GaN regrowth on the SiO2 nano patterns and UV-LEDs were fabricated using PNS as a template. Two UV-LEDs, a reference LED without PNS and a 200 nm PNS UV-LEDs were fabricated. Scanning Electron microscopy (SEM), Transmission Electron Microscopy (TEM), X-Ray Diffraction (XRD), Photoluminescence (PL) and Light output intensity- Input current- Voltage (L-I-V) characteristics were used to evaluate the ITO-$SiO_2$ nanopattern surface morphology, threading dislocation propagation, PNS crystalline property, PNS optical property and UVLED device performance respectively. The light out put intensity was enhanced by 1.6times@100mA for the LED grown on PNS compared to the reference LED with out PNS.

Cloning and Characterization of UV-B Inducible Chalcone Synthase from Grape Cell Suspension Culture System and Its Expression Compared with Stilbene Synthase

  • We performed the cloning of a chalcone synthase (CHS) gene, the key enzyme in the anthocyanin biosynthesis, from the cDNA library constructed with grape suspension cells irradiated UV-B. The PCR fragment was used to cloning the CHS gene. One CHS cDNA clone containing an open reading frame and a partial stilbene synthase (STS)cDNA, the stilbene-type phytoalexin, were isolated. The CHS cDNA clone (VCHS) showed 87% sequence homology with VvCHS (V.vinifea) and 72.3% identity with VSTSY(V.vinifea). its amino acid sequences were longer than any other CHS genes as 454 residues. Two genes were weakly expressed in white light irradiated cells, but highly induced in UV-B irradiated condition during 32 hours. Interestingly, the STS was quickly and abundantly expressed from 2 hours when supplemented with jasmonic acid (JA) and the maximum expression was observed at 4 hours and then gradually decreased. But, the additional UV-B or white light quickly degraded the STS expression than only JA treated grape suspension cells. The CHS also was rapidly induced with JA and the synergistical effect was observed at the addigional light treatment of UV-B or white light. These results are indicated that CHS and STS have different response mechanisms against the environmental stresses.
