  • The experiment was conducted to determine the effects of enhanced UV-B on growth and differential responses among cultivars in soybean. The soybean cultivars subjected to enhanced UV-B irradiation at daily dose of 11.32 kJ $m^{-2}(UV-B_{BE})$ revealed that the growth was significantly depressed. Plant height, leaf number, leaf area and dry weight were inhibited by UV-B irradiation showing differential responses among cultivars used. Danyeubkong seems to be less sensitive to the enhanced W-B irradiation, while Keunolkong more sensitive. Reduction of chlorophyll content was also found significantly greater to Keunolkong. Specific leaf weight an index of leaf thickness, and flavonoid content known as UV-absorbing compounds were significantly Increased in Danyeubkong by UV-B, but those In the other cultivars were not significantly affected. The results indicated that there are cultivar diferences in tile growth and phisiological responses to the enhanced UV-B irradiation and specific leaf weight and UV-absorbing compounds in the leaves were highly related to the sensitivity of soybean by UV-B irradiation.

Enhanced Lycopene Production by UV-C Irradiation in Radiation-Resistant Deinococcus radiodurans R1

  • Although classical metabolic engineering strategies have succeeded in developing microbial strains capable of producing desired bioproducts, metabolic imbalance resulting from extensive genetic manipulation often leads to decreased productivity. Thus, abiotic strategies for improving microbial production performance can be an alternative to overcome drawbacks arising from intensive metabolic engineering. Herein, we report a promising abiotic method for enhancing lycopene production by UV-C irradiation using a radiation-resistant ΔcrtLm/crtB+dxs+ Deinococcus radiodurans R1 strain. First, the onset of UV irradiation was determined through analysis of the expression of 11 genes mainly involved in the carotenoid biosynthetic pathway in the ΔcrtLm/crtB+dxs+ D. radiodurans R1 strain. Second, the effects of different UV wavelengths (UV-A, UV-B, and UV-C) on lycopene production were investigated. UV-C irradiation induced the highest production, resulting in a 69.9% increase in lycopene content [64.2 ± 3.2 mg/g dry cell weight (DCW)]. Extended UV-C irradiation further enhanced lycopene content up to 73.9 ± 2.3 mg/g DCW, a 95.5% increase compared to production without UV-C irradiation (37.8 ± 0.7 mg/g DCW).

Dyeability and Functionality of Chaenomelis Fructus Extract (모과 추출물의 염색성과 기능성)

  • The dying properties of fabrics with Chaenomelis Fructus extract were studied through an investigation of the characteristic of Chaenomelis Fructus colorants, the effect of dyeing conditions (dye temperature, dyeing concentration, and times on dye uptakes), effect of mordant, effect of UV irradiation, and color change in addition, antimicrobial ability and deodorant ability were estimated. In the UV-Visible spectrum, the wavelength of maximum absorption of Chaenomelis Fructus extract was 280 nm and showed that tannin is the major pigment component. From the increase of absorbance by UV irradiation, it was assumed that catechol tannin color was developed through UV irradiation. An increased dyeing concentration resulted in a larger dye uptake. Dye uptake increased as the dyeing time and temperature increased. Chaenomelis Fructus extract showed relatively good affinity to silk than cotton. Mordant, Fe and Cu were effective to increase the dye uptake of cotton fabric in addition, the dye uptake of silk fabric mordanted with Fe and K improved. UV irradiation let the color of dyed fabrics develop regardless of the UV irradiation stage however, UV irradiation on the dyed fabric was more effective than on the extract for the color development. Dyed silk fabric showed very good antimicrobial abilities of 99.9% in addition, deodorant ability improved in the fabric dyed with Chaenomelis Fructus extract.

Assays of Residual Antibiotics after Treatment of γ-ray and UV Irradiation (감마선과 UV 조사에 의한 항생제 분해)

  • The pollution of antibiotics is a major cause of spreading antibiotics resistant bacteria in the environment. Applications of ozonation, UV, and ${\gamma}-ray$ irradiations have been introduced to remove antibiotics in the effluents from wastewater treatment system. In this study, we compared the chemical (HPLC) and biological (antimicrobial susceptibility test, AMS) assays in measuring of the concentrations of residual antibiotics after ${\gamma}-ray$ and UV irradiation. Most samples were degraded by ${\gamma}-ray$ irradiation (1~2 kGy). However, lincomycin and tetracycline were not degraded by UV irradiation. The concentration of residual antibiotics, that was treated with ${\gamma}-ray$ and UV irradiation, measuring by bioassay was similar to HPLC. The concentrations of ${\gamma}-ray$ irradiated cephradine measured by AMS test were 2 times higher than that of HPLC assay, indicating AMS test is more sensitive than HPLC assay. These results indicate that ${\gamma}-ray$ irradiation technique is more useful than UV irradiation, and biological assay is more useful to detect the antibiotics and toxic intermediates in antibiotics degradation.

Impact of UV-C Irradiation on Bacterial Disinfection in a Drinking Water Purification System

  • The supply of microbiological risk-free water is essential to keep food safety and public hygiene. And removal, inactivation, and destruction of microorganisms in drinking water are key for ensuring safety in the food industry. Ultraviolet-C (UV-C) irradiation is an attractive method for efficient disinfection of water without generating toxicity and adversely affecting human health. In this study, the disinfection efficiencies of UV-C irradiation on Shigella flexneri (Gram negative) and Listeria monocytogenes (Gram positive) at various concentrations in drinking water were evaluated using a water purifier. Their morphological and physiological characteristics after UV-C irradiation were observed using fluorescence microscopy and flow cytometry combined with live/dead staining. UV-C irradiation (254 nm wavelength, irradiation dose: 40 mJ/cm2) at a water flow velocity of 3.4 L/min showed disinfection ability on both bacteria up to 108 CFU/4 L. And flow cytometric analysis showed different physiological shift between S. flexneri and L. monocytogenes after UV-C irradiation, but no significant shift of morphology in both bacteria. In addition, each bacterium revealed different characteristics with time-course observation after UV-C irradiation: L. monocytogenes dramatically changed its physiological feature and seemed to reach maximum damage at 4 h and then recovered, whereas S. flexneri seemed to gradually die over time. This study revealed that UV-C irradiation of water purifiers is effective in disinfecting microbial contaminants in drinking water and provides basic information on bacterial features/responses after UV-C irradiation.

Modification of Dyeing Property of Wool Fabrics by UV Irradiation (UV조사에 의한 양모직물의 염색성 개질)

  • Wool fabrics were treated with deep UV for 10 to 30min and then dyed with C.I. Acid Yellow 99, C.I, Acid Red 57 and C.I. Acid Blue 62. FT-IR and XPS have been used to prove the chemical composition of wool fiber surfaces. The mechanical property and colorfastness were also studied. The intencities of the peaks of C=O group at $1700^cm^{-1}$ were increased with increasing irradiation time. Relative $O_{1s},\; N_{1s}\; and\; S_{2p}$ intensities increased considerably and oxygen was incorporated in the form of CO and COO on the fiber surface. The dye uptakes of wool fabrics dyed with three acid dyes were proportional to irradiation times and in inversely proportional to irradiation distances. It was found that the tensile strength of wool fabrics were gradually deteriorated with the UV irradiation times. The colorfastness, such as washing, light and rubbing, of UV irradiated wool fabrics dyed with acid dyes were good.

  • The photomachinable glass-ceramics of Ag and CeO$_{2}$ added to Li$_{2}$O-Al$_{2}$O$_{3}$-SiO$_{2}$-K$_{2}$O glass system was investigated as a function of UV irradiation time. The temperature of optimum nucleation and crystal growth temperature were confirmed at 525.deg. C, 630.deg. C respectively using DTA and TMA. The phases of Li$_{2}$O.SiO$_{2}$ habit were lath-like and/or dendrite type and [002] direction of Li$_{2}$O.SiO$_{2}$ / Li$_{2}$O.2SiO$_{2}$ phases were changed according to the UV irradiation time by 400 W, 362 nm UV light source. Under that condition, the optimum UV irradiation time was 5 min.

Changes in Facilitated Transport Behavior of Silver Polymer Electrolytes by UV Irradiation

  • Silver species other than the silver ion were formed by UV irradiation on polymer electrolyte membranes containing silver salts and their effect on complexation behavior between the silver and olefin was investigated through the separation performance of olefin/paraffin mixtures. The ideal propylene/propane separation factor reached 350 and the separation coefficient was ca.15 due to the high loading amount of silver ions into poly(2-ethyl-2-oxazoline) (POZ) without UV irradiation. On UV irradiation either in air or under nitrogen, the silver-POZ membranes became yellow-brown initially due to the formation of colloidal silver particles, and finally black and metal-like luster. Even when Ag$^{+}$ was converted, to some extent, to Ag$^{\circ}$ by UV irradiation in air at the early stage, the separation coefficient of olefin/paraffin mixtures was maintained. This suggests that silver species other than the silver ion is active for olefin carrier for facilitated transport. Meanwhile the steady decrease of the separation coefficient was observed in the silver/POZ membranes irradiated under $N_2$. It is suggested that the reduction of silver ions in POZ goes through a different photoreduction mechanism with UV irradiation depending on the environment.t.

Effect of UV-B on fatty Acid Composition, Lipid Peroxidation and Polyamine in Kidney Bean(Phaseolus vulgaris L.) (UV-B가 강낭콩(Phaseolus vulgaris L.)의 지방산 구성, 지질과산화 및 polyamine 함량에 미치는 영향)

  • To investigate the effects of UV-B on fatty acid composition, lipid peroxidation and biochemical defense responses of plant, kidney bean (Phaseolus vulgaris L.) was subjected to enhanced UV-B irradiation [daily dose : 0.02.(No UV-B) and 11.36 (enhanced UV-B) $kJ\;m^{-2};UV-B_{BE}$] for 3 weeks. UV-B drastically inhibited both height and dry weight of kidney bean. The content of malondialdehyde significantly increased by about 50% after 3 weeks of UV-B irradiation. The ratio of unsaturated to saturated fatty acids of kidney bean was increased by UV-B irradiation. Three major polyamines of kidney bean leaves : putrescine, spermidine and spermine, were observed. All of the polyamine contents were increased with UV-B irradiation. These results suggested that enhanced UV-B radiation caused oxidative stress on lipids and biochemical protection responses might be activated to prevent from damaging effects of oxidative stress generated by UV-B irradiation.

Photo-oxidation and Dyeability of Poly Ketone by UV/O3 Irradiation (자외선/오존 조사에 의한 Poly Ketone의 광산화와 염색성)

  • Poly ketone (PK) was photo-oxidized by UV/ozone irradiation and the effect of UV energy on the surface properties of the UV-irradiated PK film was investigated by the measurement of reflectance, surface roughness, contact angles, ESCA, and ATR. Reflectance, particularly at the wavelength of 380nm, decreased with increasing UV energy. And the irradiation produced nano-scale roughness on the surface uniformly. The maximum surface roughness increased from 25.3nm for the unirradiated sample to 104.9nm at the irradiation of $42.4J/cm^2$. The improvement in hydrophilicity was caused by the introduction of polar groups such as C-O and C=O bonds resulting in higher $O_{1s}/C_{1s}$. The surface energy of PK film increased from $43.3mJ/m^2$ for the unirradiated sample to $71.9mJ/m^2$ at the irradiation of $31.8J/cm^2$. The zeta potential of the UV-irradiated PK decreased with increased UV energy and the dyeability to cationic dyes increased accordingly, resulting from the photochemically introduced anionic and dipolar dyeing sites on the PK films surfaces.
