다양한 낙농 발효유제품에서 HPLC를 이용하여 탄수화물과 유기산의 동시 검출에 관한 연구 (Simultaneous Determination of Carbohydrates and Organic Acids in Various Cultured Dairy Foods by High-Performance Liquid Chromatography: A Preliminary Study)

  • 본 연구에서는 다양한 낙농 발효유제품에 있어서 다양한 유기산과 탄수화물의 분석이 HPLC 방법으로 동시에 잘 진행되었다. 이 방법의 주요 장점으로는 (1) 간단한 시료 준비와 단일 추출이 가능, (2) 유동상(mobile phase)과 추출 용매에 있어서 편리성과 경제성, (3) 추출제와 유동상에 사용되는 황산의 농도가 매우 낮기에 친환경적인 실험, (4) 하나의 샘플당 30분 안에 HPLC의 분석이 완료 가능, (5) 다양한 농도범위에서도 분석 및 정량 가능, (6) 몇 가지 물질을 제외하고 모든 화합물의 변동계수가 낮음, 그리고 (7) 회수율이 대체적으로 높다는 것이다. 또한, $60{\sim}65^{\circ}C$에서 분석 column을 사용하였기에 column 효율면에서도 손실이 극히 적은 것으로 관찰되었다. 따라서 본 실험에서 분석된 다양한 낙농 발효유제품에서의 acid, 탄수화물, diacetyl, acetoin 등이 HPLC 방법은 매우 신속하고 정확하게 검출되었다. 하지만 다양한 향미의 변화를 최소화하면서 다양한 향미를 효과적으로 추출할 수 있는 HPLC 방법들이 지속적으로 개발되어야 다양한 낙농 발효유제품의 품질개선에 기여할 것으로 사료된다. 왜냐하면 향미 성분의 분석은 성분을 정성하는 것뿐만 아니라, 정성된 화합물이 그 식품의 향에 어느 정도 기여하는지를 중요하게 결정하기 때문이다. 또한 본 실험에 사용된 HPLC 방법으로 다양한 유산균주의 일정한 배양시간동안 유당분해효소(lactase) 능력이 조사 되었는데, 각 Probiotic lactic acid bacteria마다 다양한 유당분해효소(lactase)의 능력을 보였으며, 같은 균종에서도 차이가 있는 것으로 나타났다. 본 실험에서는 Bifidobacterium spp.가 Lactobacillus spp.와 Streptococcus spp.보다 높은 유당분해효소(lactase)의 능력을 보였다. 하지만 유산균주의 유당분해효소 능력을 정확하게 파악하기 위해서는 각 Probiotic lactic acid bacteria의 exo- 및 endo-lactase에 대한 연구가 추가적으로 필요하다. 결론적으로 다양한 낙농발효식품 및 기타 발효식품 등에서 HPLC 방법으로 동시에 유기산과 탄수화물 분석이 공인방법으로써 인정되기 위해서는 더 많은 기초자료 확립과 재현성에 관한 지속적인 연구가 진행되어야 할 것으로 사료된다.

식품 중 인공감미료의 분석법에 관한 연구 (A Study on the Analytical Method of Artificial Sweeteners in Foods)

  • 본 연구는 인공감미료의 사용확대에 따른 실태 파악 및 실제 섭취량 조사를 통해 현행 사용기준의 안전수준에 대한 안전성을 확보하는 연구의 기초자료로서 우리나라에서 식품첨가물로 허용되어 있는 인공감미료인 삭카린나트륨, 아스파탐, 아세설팜칼륨 및 수크랄로스에 대한 식품 중 분석법을 확립하였으며 결과는 다음과 같다. 먼저 투석이나 정제과정 없이 보다 간편하고 짧은 시간에 효율적으로 시료를 전처리할 수 있는 방법을 시료의 성상에 따라 확립하였다. 고속액체크로마토그래프(HPLC)의 최적 분석조건을 검토한 결과, 삭카린나트륨, 아스파탐 및 아세설팜칼륨의 3종 인공감미료의 분석에 컬럼은 Symmetry $C_{18}(3.9mm\;i.d{\times}150mm,\;5{\mu}m)$, 이동상은 0.005M tetrapropylammonium hydroxide가 함유된 0.01M $KH_{2}PO_{4}$: acetonitrile(9:1, pH 3.5). 측정파장은 210mm로 설정하였다. 수크랄로스의 분석조건은 컬럼은 Symmetry $C_{18}(3.9mm\;i.d{\times}150mm,\;5{\mu}m)$, 이동상은 water: methanol(7:3)을 사용하였고 검출기는 굴절율 검출기(RI), sensitivity=16호 설정하였다. 검출한계는 삭카린나트륨, 아스파탐 및 아세설팜칼륨은 각각 0.1ppm, 수크랄로스는 25ppm으로 측정되었다. 이와 같이 결정된 인공감미료의 최적 분석조건으로 회수율을 측정한 결과 아스파탐 92.5%, 아세설팜칼륨 97.3%, 삭카린나트륨 96.5%, 수크랄로스 93.4%로 양호한 결과를 얻었다. 시중에서 유통되고 있는 제품 중 총 17종 151품목을 다상으로 4종의 인공감미료 함량을 정량한 결과, 아스파탐은 탄산음료 2품목에서 $180.8{\mu}g/g$, 발효음료 4품목에서 $65.3{\mu}g/g$, 껌 2품목에서 $232.5{\mu}g/g$, 사탕 1품목에서 $1,672.0{\mu}g/g$, 혼합제제식품첨가물 2품목에서 $5,259.0{\mu}g/g$이 검출되었으며 아세설팜칼륨은 탄산음료 2품목에서 $110.8{\mu}g/g$, 껌 3품목에서 $250.3{\mu}g/g$, 혼합제제식품첨가물 1품목에서 $2,362.1{\mu}g/g$, 삭카린나트륨은 어묵 1품목에서 $42.3{\mu}g/g$, 수크랄로스는 껌 1품목에서 $120.1{\mu}g/g$이 검출되었으며 검출된 인공감미료는 표시사항과 일치하였다.

중국산 참가리비, Patinopecten yessoensis의 인공종묘 생산 및 부착치패 중간양성에 관한 연구 (A Study on the Production of Artificial Seed and Intermediate culture for Attached Spats of the Chinese Stock of a Scallop, Patinopecten yessoensis)

  • 중국산 참가리비 어미패의 GSI 월 변화를 보면 2월 중순 17.2 이었고, 3월 중순에는 중국산은 20.2로 채란 가능한 상태였으나 국내산 참가리비의 GSI는 2월 6.9, 3월 10,8로 미숙상태를 보였다. 어미패를 음건 및 자외선 조사 해수를 이용하여 3월 10일 및 15일 2차례에 걸쳐 채란한 결과 총 22,800만개 수정란을 채란하였고, 이중에서 17,728만마리의 D상 부유유생(부화율 77.8%)을 확보하였으며, 실내에서 25일간 사육한 후 안점이 형성된 유생 4,750만마리를 대상으로 채묘기를 넣어 총 185만마리의 부착치패를 채묘하였다. 부착치패는 강릉시험포에서 5-60일간 5단계로 나누어 실내 사육한 후 양양군 수산항내 부착치패 중간양성장으로 이동하여 성장과 생존율을 조사한 결과 실내에서 12일간 사육하여 이동한 부착치패의 생존율이 13.0%로 가장 높았다. 이 기간을 제외하면 실내사육기간이 길수록 생존율이 높았으나 성장차이는 실내사육 기간의 장단과는 상관없이 크지 않았다. 강릉시 사천항 연안에 있는 가리비 양식장에서 인공산 및 자연산 부착치패의 성장 및 생존율을 조사한 결과 양양 수산항 내에서 중간양성을 거치고 7월 10일에 본 장소에 수하한 부착치패의 경우 평균각고는 0.9 mm에서 12월 16일에 24.7 mm로 성장하였고 생존율은 85.0%로 나타난 것에 비하여 현지에서 채묘하여 수하한 자연산의 경우는 시작시 평균각고는 0.6 mm에서 종료시에는 23.9 mm로 성장하였으며 생존율은 85.7%로 인공산과 비슷한 결과를 보였다.

패 추출물의 항산화능 및 이화학적 특성에 미치는 감마선 조사의 영향 (Effects of γ-Irradiation on Antioxidant and Physicochemical Properties of Ishige okamurai Extracts)

  • 감마선 조사는 식품의 부패방지 및 안정성 향상을 위해 그 사용 범위가 확대되고 있다. 이에 본 연구에서는 패 분말과 70% 발효주정 추출물에 3, 7 및 20 kGy로 감마선을 조사하여 항산화능의 변화를 알아보았다. 패 분말에 감마선 조사하여 70% 발효주정으로 추출한 뒤 수율, 총 페놀화합물 함량 및 DPPH 라디칼 소거능의 변화를 알아본 결과, 조사선량이 증가함에 따라 수율은 증가하여 20 kGy 조사구에서는 19%의 수율을 보였으나, 총 페놀화합물 함량 및 DPPH 라디칼 소거능에 있어서는 유의적인 차이를 보이지 않았다. 패 70% 발효주정 추출물에 감마선 조사하여 총 페놀화합물 함량, 항산화능 및 이화학적 특성 변화를 알아본 결과, Rancimat에 의한 항산화도에 있어서는 비조사구 및 3, 7, 20 kGy 조사구 각각 1.07, 0.74, 0.91 및 1.22의 AI값을 보여 3과 7 kGy 조사구는 감소하였으며, 20 kGy 조사구는 증가하였다. 총 페놀화합물 함량과 DPPH 라디칼 소거능은 분말에 감마선 조사하였을 때와 같이 유의적인 차이를 보이지 않았다. 색도는 감마선 조사선량이 증가함에 따라 유의적으로 색이 감소하는 경향을 보였고, pH의 경우 조사구가 비조사구보다 다소 감소한 값을 보였다. 따라서 감마선 조사된 패 70% 발효주정 추출물은 본래의 항산화 활성을 유지하면서 비조사구보다 패 추출물 본래의 색이 옅어져 식품에 첨가시 식품자체의 색에 큰 영향을 미치지 않아 식품 천연 항산화제로서 적용이 용이할 것으로 사료된다.

  • 잣나무 유묘(幼苗)와 쥐똥나무 삽목묘(揷木苗)를 인공산성우(人工酸性雨)로 처리(處理)하고 엽내(葉內) 엽록소(葉綠素) a와 b의 함량(含量)을 분석(分析)하여 양(量)의 증감(增減)에 따른 산도(酸度) 수준(水準)의 영향(影響)을 논의(論議)하였다. 처리(處理)된 시험수종(試驗樹種)은 1985년(年) 봄에 pot에 재배(栽培)되고 해당지역(該當地域)의 30년간(年間)의 강우(降雨) 양식(樣式)을 모의(模擬)해서 산성우(酸性雨)(pH 수준(水準) 6.5, 4.0 및 2.0)를 5월(月)부터 8월(月)까지 4개월간(個月間)에 걸쳐 살포(撒布)하고, 동년(同年) 9월(月) 18일(日) 성숙(成熟)된 엽(葉)을 채취(採取)하여 엽중(葉中)의 엽록소(葉綠素)를 분석(分析)하였다. 분석(分析) 결과(結果)에 의(依)하면, 양(兩) 수종(樹種) 공통적(共通的)으로 산성우(酸性雨)의 산도(酸度)가 강(强)해짐에 따라 엽록소(葉綠素) a와 b의 함량(含量)은 감소(減少)하는 경향(傾向)을 보였고. 엽록소(葉綠素) a에 있어서는 감소(減少) 경향(傾向)이 통계적(統計的)으로 유의(有意)했고, 엽록소(葉綠素) b의 경우(境遇)는 유의(有意)하지 않았으나 감소(減少) 경향(傾向)이 있는 것으로 받아들일 수 있었다. 산성우(酸性雨)의 pH 수준(水準)에 따른 엽록소(葉綠素) a와 b에 대한 감소(減少) 효과(効果) 차이(差異)에 있어서는 양(兩) 수종(樹種) 모두 pH 수준별(水準別)로 감소(減少) 지수(指數)가 유사(類似)하였으며, 따라서 엽록소(葉綠素) a 대(對) b의 함량비(含量比)는 산성우(酸性雨)의 pH 수준(水準) 여하(如何)에 불구(不拘)하고 일정(一定)한 값을 유지(維持)하고 있었다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. For higher concentration than 300mg/41 about glucose, a saturation state of abforbance was observed at 500, 550 and 570nm. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. To synthesize microspheres for biomedical application, "two stage shot growth method" was developed. The uniformity ratio of synthesized microspheres was always smaller than 1.05. And the surface charge density (or the number of ionizable functional groups) of the microspheres synthesized by "two stage shot growth method" was 6~13 times higher than that of the microspheres synthesized by conventional seeded batch copolymerization. As a previous step for biomedical application, adsorption experiments of bovine albumin on microspheres were carried out under various conditions. Our department has developed an information system for routine hematology using down-sized computer system. We have used an IBM 486 compatible PC with 16MB main memory, 210 MB hard disk drive, 9 RS-232C port and 24 pin dot printer. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. The authors suggests that this system based on down-sized computer could provide a progressive approach to total LIS based on local area network, and the implemented system could serve as a model for other hospital's LIS for routine hematology. 30609 T00401030609 ^x To develop an artificial bone substitute that is gradually degraded and replaced by the regenerated natural bone, the authors designed a composite that is consisted of calcium phosphate and collagen. To use as the structural matrix of the composite, collagen was purified from human umbilical cord. The obtained collagen was treated by pepsin to remove telopeptides, and finally, the immune-free atelocollagen was produced: The cross linked atelocollagen was highly resistant to the collagenase induced collagenolysis. We propose a simple adaptive algorithm to increase capability of noise cancelation in QRS complex detection with two stage adaptive filter. At the first stage, background noise is removed and at the next stage, only spectrum of QRS complex components is passed. Two adaptive filters can afford to keep track of the changes of both noise and QRS complex. Each adaptive filter consists of prediction error filter and FIR filter The impulse response of FIR filter uses coefficients of prediction error filter.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer.
