효소를 이용한 저분자 토종 닭발 콜라겐의 제조 및 품질 특성 (Preparation and quality characteristics of low molecular weight collagen treated with hydrolytic enzymes from Korean native chicken feet)

  • 본 연구에서는 단백질 가수분해효소 0.1%와 1% Protamex를 사용하여 저분자 콜라겐을 제조하였다. 토종 닭발의 조단백질과 콜라겐의 함량은 일반 육계에 비해 높은 함량이 나타났다. 단백질 가수분해 효소농도와 반응시간이 증가할수록 낮은 분자량의 콜라겐을 얻을 수 있는 것으로 나타났다. 특히 1% Protamex로 7시간 처리한 시료가 1,000-5,000 Da의 저분자 콜라겐 함량이 55.6%로 나타났으며, 평균 분자량은 5,390 Da로 가장 낮은 분자량이 나타났다. 이는 단백질 가수분해효소 Protamex가 고분자 펩타이드 결합을 저분자 펩타이드로 분해했기 때문이다. 효소처리 콜라겐의 조직감은 고분자 펩타이드의 콜라겐이 저분자 펩타이드로 분해되어 gel을 형성하지 못하고 sol의 형태를 유지하였다. 효소농도와 효소반응시간이 증가할수록 콜라겐의 평균분자량은 작아지나 효소반응 5시간부터 평균분자량의 감소가 미미해지는 경향이 나타났다. 따라서 저분자 콜라겐 효소반응시간은 경제적으로 볼 때 5시간에서 7시간 사이가 적합하다고 할 수 있다. 이 연구결과는 향후 산업적 효소를 이용한 저분자 콜라겐 제조 및 식품 소재 활용의 기초자료로 이용할 수 있을 것이다.

타액선 종양에서 제4형 교원질과 Fibronectin 발현 (Expression of Type IV Collagen and Fibronectin in Salivary Gland Tumors)

  • Objectives: Salivary gland tumors pose considerable difficulty in diagnostic and prognostic assessment based on the histopathologic features alone. We studied the expression of type IV collagen and fibronectin in salivary gland tumors with special emphasis on the differential diagnostic significance. Materials and Methods: We did immunohistochemical stain on paraffin embedded tissues of 33 benign and 24 malignant salivary gland tumors using monoclonal antibody for type IV collagen and polyclonal antibody for fibronectin. Results: 1) Well preserved linear basement membrane-like staining of type IV collagen was detected in duct-cell-derived benign salivary gland tumors. But pleomorphic adenoma exhibited a heterogeneous pattern as focal augmentation and interruption. 2) In malignant tumors, type IV collagen was distributed in an irregular, interrupted manner or completely absent. Adenoid cystic carcinomas displayed a marked staining of the basal membrane associated substances in the pseudocysts. 3) The staining pattern of fibronectin was similar to that of type IV collagen execpt more dense in the stroma. 4) Salivary gland tumors which have a prominent myoepithelial cell component revealed a particular deposition of basement membrane materials adjacent to the myoepithelial cells. Conclusion: The study of the basal membrane substances may be helpful for differential diagnosis of benign and malignant salivary gland tumors and identifying special features of salivary gland tumors such as pseudocystic pattern of adenoid cystic carcinoma. Also we think that the myoepithelial cells contribute to the formation of basement membrane materials.

Effects of the Butanol Extact of Head of Panax Ginseng on Type II Collagen-induced Arthritis in DBA/1J Mice

  • In order to evaluate the improvement effects of head of Panax ginseng on chronic arthritis, we have investigated the activity of butanol fraction (BuOH fraction) in vitro and in vivo system. BuOH fraction showed significant inhibition on the elastase activity. Anti-arthritic activity of BuOH fraction was also examined on type II collagen-induced arthritis in DBA/1J mice. Mice were immunized with injection of type II collagen emulsified in Freund's complete adjuvant, followed by a booster injection 21 days later. BuOH fraction(BHPG) was administered at an oral dose of 500mg/kg for 2 weeks from the 1st day boost. The hind paw edema was significantly decreased in the group of treatment with BuOH fraction compared to control. In collagen-induced DBA/1J mice, BuOH fraction did not affected the collagen antibody titer but significantly inhibited the tumor necrosis factoralpha(TNF-${\alpha}$) activity. These results were confirmed with histological evaluation of joint tissues. This study may raise the possibility that the usage of BuOH fraction of head of Panax ginseng as alternative medicine for the relief and prevention of rheumatoid arthritis symptoms.

섬유아세포에서 민들레 추출물 함유제(AF-343)에 의한 제 1형 교원질 발현 증가 (Increased Expression of Type I Collagen in AF-343 Treated Human Skin Fibroblasts)

  • We previously reported that the extract of Taraxacum platycarpum (AF-343) had several biological properties such as skin hydration and anti-inflammatory effects, thereby AF-343 be a promising anti-atopic dermatitis agent. However, few studies have been conducted to evaluate its effect on modulation of extracellular matrix proteins in human skin fibroblasts. The purpose of this study was to investigate the expressions of type I collagen, MMP-1, Smad2/3, and TIMP-1 proteins in AF-343-treated human skin fibroblasts. Human skin fibroblasts were treated by various concentrations of AF-343 (0~2 mg/ml). The expressions of type I collagen, matrix metalloproteinase-1 (MMP-1), Smad2/3, and TIMP-1 proteins were analyzed by Western blot analysis. In addition, level of type I collagen mRNA was analyzed by CAT assay. Expression of type I collagen protein was increased in AF-343-treated human skin fibroblasts by dose and time-dependent manners. Consistent with this result, the expressions of phospho-Smad2/3 in skin fibroblasts were increased and MMP-1 expression was decreased by AF-343 treatment. TIMP-1 expression was not significantly changed in AF-343 treated skin fibroblasts. Extract of Taraxacum platycarpum (AF-343)-induced up-regulation of type I collagen expression was through increased expression of phospho-Smad2/3. These results were occurred combined with down-regulation of MMP-1 in skin fibroblasts. Taken together, this study indicated that AF-343 has property of the modulation of ECM in tissue as well as skin hydration and anti-inflammation.

  • The purpose of this research was to investigate effects of Bambusae Caulis in Liquamen (BCL) on the synthesis of basement membrane proteins during proliferation and differentiation of 3T3-L1 cells. BCL has been used to relieve the cough and asthma, and remove phlegm in traditional oriental medicines. In recent years. it was studied for its antiinflammatory, antiallergenic. immune-modulating and anticarcinogenic capabilities. We have previously observed that glycyrrhizin stimulates the adipose conversion of 3T3-L1 cells. To investigate effects of BCL on the basement membrane proteins during proliferation and differentiation of 3T3-L1 cells, we have analyzed synthetic amounts of basement membrane components such as type IV collagen and BM40. BCL stimulated the synthesis and secretion of type IV collagen from both 3T3-L1 preadipocytes and adipocytes. The synthesis and secretion of BM40 was not affected by BCL. The continuous addition of BCL markedly stimulated cell growth and increased cell density. These results suggest an important role for type IV collagen in adipocyte differentiation.

Complication rates in patients using absorbable collagen sponges in third molar extraction sockets: a retrospective study

  • Objectives: The purpose of this study is to retrospectively evaluate the postoperative complication rates for absorbable type-I collagen sponge (Ateloplug; Bioland) use in third molar extraction. Materials and Methods: From January to August 2013, 2,697 total patients undergoing third molar extraction and type-I collagen sponge application in the Department of Oral and Maxillofacial Surgery at Yonsei University Dental Hospital (1,163 patients) and Dong-A University Hospital (1,534 patients) were evaluated in a retrospective study using their operation and medical records. Results: A total of 3,869 third molars in 2,697 patients were extracted and the extraction sockets packed with type-I collagen sponges to prevent post-operative complications. As a result, the overall complication rate was 4.52%, with 3.00% experiencing surgical site infection (SSI), 1.14% showing alveolar osteitis, and 0.39% experiencing hematoma. Of the total number of complications, SSI accounted for more than a half at 66.29%. Conclusion: Compared to previous studies, this study showed a relatively low incidence of complications. The use of type-I collagen sponges is recommended for the prevention of complications after third molar extraction.

The Effect of Trigonella foenum-graceum L. (Fenugreek) Towards Collagen Type I Alpha 1 (COL1A1) and Collagen Type III Alpha 1 (COL3A1) on Postmenopausal Woman's Fibroblast

  • Trigonella foenum-graceum L. (fenugreek) is a phytoestrogen, a nonsteroidal organic chemical compound from plants which has similar mechanism of action to sex hormone estradiol-$17{\beta}$. This study aims to assess the effectivity of fenugreek seeds extract on collagen type I alpha 1 (COL1A1) and collagen type III alpha 1 (COL3A1) which are both decreased in aging skin and become worsen after menopause. This in vitro experimental study used old human dermal fibroblast from leftover tissue of blepharoplasty on a postmenopausal woman (old HDF). As a control of the fenugreek's ability to trigger collagen production, we used fibroblast from preputium (young HDF). Subsequent to fibroblast isolation and culture, toxicity test was conducted on both old and young HDF by measuring cell viability on fenugreek extract with the concentration of 5 mg/mL to $1.2{\mu}g/mL$ which will be tested on both HDF to examine COL1A1 and COL3A1 using ELISA, compared to no treatment and 5 nM estradiol. Old HDF showed a 4 times slower proliferation compared to young HDF (p<0.05). Toxicity test revealed fenugreek concentration of $0.5-2{\mu}g/mL$ was non-toxic to both old and young HDF. The most significant fenugreek concentration to increase COL1A1 and COL3A1 secretion was $2{\mu}g/mL$ (p<0.05).

백서의 실험적 치아 이동시 교원질 발현에 관한 면역조직화학적 연구 (The Expression of Type I Collagen in Periodontal Tissue during the Experimental Movement of Rat Incisors)

  • 견인력에 의한 치아 이동시 전반적인 치주 조직의 변화를 관찰하고 특히 제1형 교원질의 발현 정도 및 분포의 변화를 알아보고자, Sprague-Dawley계 백서 21마리를 대상으로 대조군(3마리)과 실험군(18마리)으로 나누었으며, 실험군은 양 중절치 사이에 견인력(75g)을 가한 후 12시간, 1일, 4일, 7일, 14일, 28일째에 각각 3마리씩 희생시켜, 시간에 따른 제1형 교원질의 발현과 조직학적 변화를 면역조직화학적 및 조직병리학적으로 관찰한 바 다음과 같은 결과를 얻었다. 견인력을 가한 후 28일째까지, 견인측의 치주인대 섬유는 신장되어 있었고, 압박측의 치주인대 섬유는 압축되어 있었으며 치주인대 섬유의 배열은 완전히 회복되지 않았다. 대조군에서의 제1형 교원질 발현은 구강 상피, 전상아질, 치수와 치주인대에서 경미하였으나, 치조골에 연한 조골세포, 치근단의 무세포성 백악질, 조백악세포와 악간봉합 부위에서 약양성의 발현을 보였다. 실험군에서의 제1형 교원질의 발현은, 무세포성 백악질에서 치아 이동 1일째에 중등도, 7일째부터 강양성의 발현을 보였으며, 정중 구개 봉합 부위에서는 치아 이동 1일째에 중등도, 14일째부터 강양성 발현을 보였다. 치주인대에서는 치아 이동 4일째에서 견인측이 압박측보다 제1형 교원질의 발현이 많아져 7일째에서 최고조에 달하다가 14일째부터는 측간의 차이가 없었다. 골조직에서는 조골세포가 붙어 있는 골 주변부에서 강하게 반응을 보여 대조군과 구별되었으며 특히 7일째 이후에 많은 것으로 관찰되어 골개조 현상과 관련하여 주목되었다. 이러한 관찰로 교정적 치아 이동 후 골재형성 과정은 조직 형성 세포의 분화와 이들 세포에서 형성되는 제1형 교원질과 밀접한 관계가 있음을 확인할 수 있었다.

낙석등(絡石藤)약침이 Collagen 유발 관절염에 미치는 영향 (Effect of Trachelospermi Caulis Herbal-acupuncture on the Collagen-induced Arthritis in Rats)

  • Objectives : This study was built to investigate the effect of Trachelospermi Caulis herbal-acupuncture on the Collagen-induced arthritis(CIA) in rats. Methods : Arthritis was induced by intradermal injection of Bovine type II collagen solution into base of tail. Experimental group were divided into 5 groups ; Normal(N) group, Control(C) group, Trachelospermi Caulis high(TH) group, Trachelospermi Caulis low(TL) group, Saline(S) group(n=7 for each group). Normal group was had no management. Control group was injected with Bovine type II collagen solution and taken no treatment. Trachelospermi Caulis high group was injected with Bovine type II collagen solution and taken high-intensity(10mg/kg) herbal-acupuncture treatment on $ST_{36}$. Trachelospermi Caulis low group was injected with Bovine type II collagen solution and taken low-intensity(5mg/kg) herbal-acupuncture treatment on $ST_{36}$. Saline group was injected with Bovine type II collagen solution and taken saline injection on $ST_{36}$. Body weight, paw edema volume and ankle joint thickness were measured during experimental day. On the last experimental day, we analyzed WBC count, TNF-$\alpha$ & IL-$1{\beta}$ concentration, c-fos immunohistochemistry and NADPH-d histochemistry for evaluating the effect of Trachelospermi Caulis herbal-acupuncture. Results : The results were as follows ; 1. In the change of paw edema volume, TH group only has significant difference compared with C group. 2. In the change of ankle joint thickness, TH group only has significant difference compared with C group. 3. In WBC count of serum, TH, TL groups have significant decrease compared with C group. 4. In TNF-$\alpha$ concentration of effusion, TH, TL groups have significant decrease compared with C group. and TH group has significant decrease compared with TL group. 5. In IL-$1{\beta}$ concentration of effusion, TH, TL groups have significant decrease compared with C group. and TH group has significant decrease compared with TL group. 6. In c-fos positive neurons of S1S2(cortex) region, TH, TL, S groups have significant decrease compared with C group. 7. In NADPH-d positive neurons of CPu(caudate putamen) region, TH, TL groups have significant decrease compared with C group. 8. In NADPH-d positive neurons of Tfp(transverse fibers of pons) region, TH, TL, S groups have significant decrease compared with C group. and TH group has significant decrease compared with S group. Conclusions : According to above results, we hope that Trachelospermi Caulis herbal-acupuncture may have the effect that decreases progression and development of CIA. And it can be suggested that Trachelospermi Caulis herbal-acupuncture may reduce the expression of c-fos and NOS.

다양한 세포외기질이 배양 골아세포의 이동에 미치는 영향 (The Effects of Various Extracellular Matrices on Motility of Cultured MC3T3-E1 Cell)

  • Chemotactic migration of bone forming cell, osteoblast, is an important event during bone formation, bone remodeling, and fracture healing. Migration of cells is mediated by adhesion receptors, such as integrins, that link the cell to extracellular matrix ligands, type I collagen, fibronectin, laminin and depend on interaction between integrin and extracellular ligand. Our study was designed to investigate the effect of extracellular matrix like fibronectin, laminin, type I collagen on migration of osteoblast. Migration distance and speed of MC3T3-E1 cell on extracellular matrix-coated glass were measured for 24 hours using 0.01% type I collagen, 0.01% fibronectin, 100 microliter/ml laminin. The migration distance and speed of MC3T3-E1 cell was compared using a video-microscopy system. To determine migration speed, cells were viewed with a 4 phase- contrast lens and video recorded. Images were captured using a color CCD camera and saved in 8-bit full-color mode. The migration distance on 0.01% type I collagen or 0.01% fibronectin was longer than that on $100{\mu}l/ml$ laminin-coated glass. The migration speed on fibronectin-coated glass was 68 micrometer/hour which was fastest. The migration speed on type I collagen-coated glass was similar with that on fibronectin-coated glass. The latter two migration speeds were faster than that on no-coated glass. On the other hand, the average migration speed on laminin-coated glass was 37micrometer/hour and not different from that of control group. In conclusion, the extracelluar matrix ligands such as type I collagen and fibronectin seem to play an important role in cell migration. The type I collagen or fibronectin coated scaffold is more effective for migration of osteoblast in tissue engineering process.
