The characteristics of ice packing factor (IPF) at the ice slurry system using one line type are compared with the system using two lines type. The installation space for one transporting line is saved at the one line system. For the one line type, the ice packing factor is reduced along the downstream, but for the two lines type, the ice packing factor is fixed. For the one line system, mass flow rate in the main line is fixed along the down-stream, but for two lines system, the mass flow rate in the main line is reduced along the downstream. For one line system, along the down stream after IPF=0, the temperature at the main steam is increased, and the extracted mass flow is increased. The initial IPF, at which the IPF is not arrived at zero upto the final node, is proposed for the B area.
A new design based on the appropriate geometry of molded part and type of runner system under the optimal processing conditions was proposed to minimize the micro weld lines on the sub deco surface molded by two shot molding. Theoretical and experimental studies were conducted to examine the cause of the weld lines during the overmolding process in two shot molding. Various dimensions and geometries of substrate$(1^{st}shot)$ and the wall thickness of overmold$(2^{nd}shot)$ have been proposed to avoid the weld lines which are the most inevitable appearance defects occurred on the sub deco. The each design proposal was analyzed by mold flow analysis after part modeling. The analysis results were compared with molded part from mass production tool. It could be seen that from the analysis that the proper geometry of plastic part and type of runner system considering pressure drop under the optimal processing conditions were the most influential factors to avoid weld lines occured on the sub deco.
Background: The roots of Panax ginseng contain two types of tetracyclic triterpenoid saponins, namely, protopanaxadiol (PPD)-type saponins and protopanaxatiol (PPT)-type saponins. In P. ginseng, the protopanaxadiol 6-hydroxylase (PPT synthase) enzyme catalyses protopanaxatriol (PPT) production from protopanaxadiol (PPD). In this study, we constructed homozygous mutant lines of ginseng by CRISPR/Cas9-mediated mutagenesis of the PPT synthase gene and obtained the mutant ginseng root lines having complete depletion of the PPT-type ginsenosides. Methods: Two sgRNAs (single guide RNAs) were designed for target mutations in the exon sequences of the two PPT synthase genes (both PPTa and PPTg sequences) with the CRISPR/Cas9 system. Transgenic ginseng roots were generated through Agrobacterium-mediated transformation. The mutant lines were screened by ginsenoside analysis and DNA sequencing. Result: Ginsenoside analysis revealed the complete depletion of PPT-type ginsenosides in three putative mutant lines (Cr4, Cr7, and Cr14). The reduction of PPT-type ginsenosides in mutant lines led to increased accumulation of PPD-type ginsenosides. The gene editing in the selected mutant lines was confirmed by targeted deep sequencing. Conclusion: We have established the genome editing protocol by CRISPR/Cas9 system in P. ginseng and demonstrated the mutated roots producing only PPD-type ginsenosides by depleting PPT-type ginsenosides. Because the pharmacological activity of PPD-group ginsenosides is significantly different from that of PPT-group ginsenosides, the new type of ginseng mutant producing only PPD-group ginsenosides may have new pharmacological characteristics compared to wild-type ginseng. This is the first report to generate target-induced mutations for the modification of saponin biosynthesis in Panax species using CRISPR-Cas9 system.
To select new germplasm for salinity tolerance from new plant type (NPT) breeding lines, the sixty F$_4$ lines selected from the crosses between Korean cultivars and IRRI's NPT lines were evaluated for salinity tolerance at the seedling stage with salinized culture solution (EC=12 dS/m) in the controlled conditions. Two NTP lines derived from a cross between 'Ilmibyeo' and 'IR66152-AC5-1', 'HR15258-7-1' and 'HR15258-27-1', were found to have good tolerance. The salinity tolerance of the lines was compared to their parents and the sensitive ('IR29') and tolerant ('Pokkali') checks in three salinity levels, no salinity (control) and an EC of 12 and 16 dS/m. Visual salinity score, shoot Na+ and Na-K ratio in two NPT lines was significantly low compared with the parents and IR29. Indicating that salinity tolerance of the lines might be derived from a transgressive segregation. The relative water content of the lines was higher than Pokkali, and the dry weight of shoot and root was proportionally decreased to salinity score and salinizing concentration. The visual salinity scores were significantly correlated with shoot Na concentration, Na-K ratio, relative water content, and reduction of dry weight (P<0.01). Their tolerance was attributed to root and shoot characteristics that led to high shoot water content, thus diluting the toxic effect of salts.
The genetic makeup could be the most important among many factors affecting yield and quality of tobacco(Nicotiana tabacum L.). The mammoth gene found in N. tabacum is associated with greater leaf number and poor leaf quality. This study was carried out to obtain the basic information about the inheritance of mammoth gene and white flower color. Two flue-cured breeding lines, KF 9373-2 and KF 8832-85, F$_1$, F$_2$, two parents backcrossed with F$_1$, and F$_3$ lines derived from cross of above two lines were investigated for flowering type(mammoth gene) and flower color. All plants of F$_1$ population revealed normal flowering type and pink flower color. The progeny of F$_2$ generation was segregated into the phenotypic ratio of 9 : 3 : 3 : 1 with normal flowering type and pink flower color, normal and white, non flowering type(NF) and pink, and NF and white, respectively. Among the progenies of back-crossing populations, the flowering type showed a segregation ratio of 1 : 1 as normal and NF in BP$_1$ and flower color did also 1 : 1 as pink and white in BP$_2$. All lines have the mammoth gene in F$_3$. that were selected in F$_2$ progeny as non flowering. But 9 lines among 14 were segregated with 3 : 1 as pink and white flower in F$_3$. which were selected in F$_2$ as pink flower color. These results indicated that the characters of mammoth gene and white flower were controlled by a pair of recessive genes, respectively.
Motivated by the need for developing the more efficient lighting system for light culture of strawberries in the greenhouse, this paper aims at acquiring and suggesting more concrete and scientific foundation of illuminating position, numbers of light source by investigating the types of lighting system and illumination distribution in the greenhouse for light culture of strawberries. The results of investigation and experiment are summarized as following below: 1. The types of lighting system used in the greenhouse producing strawberries were classified as 1 line and 2 lines lighting system. 2. As for the arranging types of artificial light, 2 lines lighting system, were classified as Z-type, N-type and W-type. (Refer Fig. 3) 3. The results of illumination distribution for Z-type, N-type and W-type of 2 lines illuminating system in the greenhouse with a small size tunnel measured at the height of 1.5m from the ground with 220V, 100W lamp in 6m light gap showed that maximum illuminance are 961x, 1211x, 1251x, minimum illuminance are 4.41x, 4.71x, average illuminance are 33.71x, 43.11x, 44.51x and standard deviations are 28.31x, 35.41x, 38.31x at each types. 4. Proportion of the area below optimal illuminance to floor area at the two lines illuminating system of Z-, N-, and W-type in greenhouse were appeared as 39.4%, 26.0% and 26.3%, respectively. Also proportion of the area over optimal illuminance to floor area at the two lines illuminating system of Z-, N-, W-type in greenhouse were appeared as 16.8%, 14% and 14.7%, respectively. Thus N-type was superior to the others from the view points of optimal illumination distribution and energy saving.
The factory under this study consists of mixed-model assembly lines and workcenters which provide parts to the main lines. Parts produced by the workcenter have different specifications for different product models. The workcenters fabricate parts in batches, and they are divided into two types. A type 1 center supplies parts only to the main line that is designated to the center while type 2 center provides parts to all the main lines. The purpose of this study is to develop a scheduling scheme for the workcenter, and the main objective of the schedules is to provide parts for the main lines without delay. The facts that make the scheduling challengeable are that 1) the different models existing together on a main line request different parts, 2) the spaces for part inventories are limited and 3) set up times are sequence dependent and long in some cases. This study presents developed scheduling schemes for the type 1 center and explains the scheduling and control structure used.
This paper presents a scheme for a hierarchical production scheduling and control system for a refrigerator factory with mixed model assembly lines. The setting of the factory is as follows. There are three mixed-model assembly lines called main line A, B and C and two batch lines that supply parts to the main lines. For each of the main lines, three work-centers are dedicated to them. The sub-lines and work-centers produce parts in batch type. An incoming production order from the master planner is characterized by its product type, amount, and due date. Under this situation, the proposed scheme has several features to schedule and control the above mentioned factory; 1) select the starting time and the place (assembly line) for an order processing, 2) devise a way to control orders to be processed as scheduled, and 3) reschedule orders when something unexpected happen. Finally, this paper provides a case study where the proposed scheme is applied to.
The aim of this study was to compare the growth performance, fiber characteristics of the pectoralis major muscle, and meat quality characteristics in the heavy weight (HW) and random bred control (RBC) quail lines and genders. The HW male exhibited more than two times greater body (245.7 vs 96.1 g, p<0.05) and pectoralis major muscle (PMW; 37.1 vs 11.1 g, p<0.05) weights compared to the RBC female. This growth performance in the HW line was associated with a greater muscle fiber area (1,502 vs $663.0{\mu}m^2$, p<0.001) compared to the RBC line. Greater muscle mass of the HW male was accompanied by a higher percentage of type IIB fiber compared to the HW female (64.0% vs 51.0%, p<0.05). However, muscle fiber hyperplasia (increase in fiber number) has had a somewhat limited effect on PMW between the two lines. On the other hand, the HW line harboring a higher proportion of type IIB fiber showed rapid pH decline at the early postmortem period (6.23 vs 6.41, p<0.05) and lighter meat surface (53.5 vs 47.3, p<0.05) compared to the RBC line harboring a lower proportion of type IIB fiber. There were no significant differences observed in the measurement of water-holding capacity including drip loss (2.74% vs 3.07%, p>0.05) and cooking loss (21.9% vs 20.4%, p>0.05) between the HW and RBC lines. Therefore, the HW quail line developed by selection from the RBC quail, was slightly different in the meat quality characteristics compared to the RBC line, and a marked difference was found in growth performance between the two quail lines.
With the advent of smart clothing for health care and sports, the sophisticated designs with curved seams are drawing attention. One of the problems in those clothing is to determine the design curves in 2D pattern, such that it corresponds to the lines on the intended 3D body. Moreover, the difficulty increases when the original pattern needs to be changed for various sizes and body types. We compare two methods of pattern enlargement in this paper: one is the offset/projection type, and the other is the split grading type. For the enlarged pattern with offset/projection type, the 3D surface offset was first adopted to transform the standard lower body to the target larger size; next, the design lines were projected to the new 3D surface, following which the 3D pattern was developed from the newly transformed 3D surface. In the second method, the enlarged pant patterns were developed by the split grading method. Here, a 3D pattern was developed from the initial body, and then enlarged to the target size by the conventional split grading method. Two feminine pants patterns were examined by 3D virtual fitting. We observed that the 3D offset/projection pants pattern was well fitted, having an evenly distributed surplus, as compared with the sample developed using the split grading method. The difference between the two patterns were apparent at the location where several curved lines merged.
