Detection of Retinol-binding Protein in Bovine Yolk Sac, Chorion and Allantois by Immunoperoxidase Method

  • Bovine yolk sac at day 24 of pregnancy, and placental membranes (chorion and allantois) from days 70 and 100 of pregnancy were isolated and cultured in a modified minimum essential medium in the presence of $[^{35}S]$methionine. Proteins synthesized and secreted by isolated bovine yolk sac, chorion and allantois were analyzed by fluorography of two-dimensional polyacrylamide gel electrophoresis. Serum-like proteins,transferrin, ${\alpha}$-fetoprotein, ${\alpha}$1-antitrypsin and ${\alpha}$1-acid glycoprotein,were the major protein products of yolk sac. A 21 kDa protein produced by yolk sac was identified immunochemically as retinol-binding protein (RBP). Chorion and allantios from days 70 and 100 of pregnancy were active in protein synthesis and secretion. Both chorion and allantois did not secret serum-like proteins but secreted a number of neutral-to-acidic proteins including RBP. Secretory proteins produced by the yolk sac, chorion and allantois may play important roles in the embryonic development and the successful outcome of pregnancy. Antiserum against bovine placental RBP was employed to the immunocytochemistry by immunoperoxidase method. Immunoreactive RBP was localized in epithelial cells and island-like cell clones of yolk sac. Immunostaining for RBP was detected in simple columnar epithelium of chorion and in simple squamous epithelium of allantois. In the present study, proteins synthesized and secreted by yolk sac at day 24 of pregnancy, chorion and allantois from days 70 and 100 of pregnancy were characterized In addition, RBP was localized in yolk sac, chorion and allantois by immunoperoxidase method. The immunoperoxidase method has been proven to be a very effective technique to identify the cellular source of protein synthesis in extraembryonic membranes.

Comparison of removal torques between laser-etched and modified sandblasted acid-etched Ti implant surfaces in rabbit tibias

  • PURPOSE. The purpose of this study was to analyze the effects of two different implant surface treatments on initial bone connection by comparing the Removal Torque Values (RTQs) at 7 and 10 days after chemically modified, sandblasted, large-grit and acid-etched (modSLA), and Laser-etched (LE) Ti implant placements. MATERIALS AND METHODS. Twenty modSLA and 20 LE implants were installed on the left and right tibias of 20 adult rabbits. RTQs were measured after 7 and 10 days in 10 rabbits each. Scanning electron microscope (SEM) photographs of the two implants were observed by using Quanta FEG 650 from the FEI company (Hillsboro, OR, USA). Analyses of surface elements and components were conducted using energy dispersive spectroscopy (EDS, Horiba, Kyoto, Japan). RESULTS. The mean RTQs were $12.29{\pm}0.830$ and $12.19{\pm}0.713$ Ncm after 7 days (P=.928) and $16.47{\pm}1.324$ and $16.17{\pm}1.165$ Ncm after 10 days (P=.867) for LE and modSLA, respectively, indicating no significant inter-group differences. Pore sizes in the LE were $40{\mu}m$ and consisted of numerous small pores, whereas pore sizes in the modSLA were $5{\mu}m$. In the EDS analysis, Ti, O, and C were the only three elements found in the LE surfaces. Na, Ca, Cl, and K were also observed in modSLA, in addition to Ti, O, and C. CONCLUSION. The implants showed no significant difference in biomechanical bond strength to bone in early-stage osseointegration. LE implant can be considered an excellent surface treatment method in addition to the modSLA implant and can be applied to the early loading of the prosthesis clinically.

Residue Dissipation Patterns of Neonicotinoid Acetamiprid and Thiamethoxam in Swiss Chard for the Harvest Periods under Greenhouse Conditions (시설재배 근대 중 Neonicotinoid계 살충제 Acetamiprid 및 Thiamethoxam의 생산단계 잔류특성)

  • BACKGROUND: Dissipation of acetamiprid and thiamethoxam in greenhouse grown chard samples was evaluated at 5 intervals including the pre-harvest interval after application. This study was conducted to determine the residue levels, the biological half-lives and dissipation rate of acetamiprid and thiamethoxam in chard under controlled conditions. METHODS AND RESULTS: Acetamiprid and thiamethoxam were applied in accordance with good agricultural practices for chard. Chard samples were collected at 0, 1, 2, 3, 5, 7, 10 and 14 days after application. Quantitaion was performed by HPLC-DAD system with C18 column. Limit of quantification (LOQ) of acetamiprid and thiamethoxam were both 0.02 mg/kg for chard. The recovery of acetamiprid and thiamethoxam were 77.8~107.5% and 94.3~108.6% at two concentration levels. The half-lives of pesticide dissipation in chard for two fields were 11.9 and 8.2 days for acetamiprid and 3.6 and 3.3 days for thiamethoxam respectively. The dissipation rate of acetamiprid and thiamethoxam were estimated according to the statistics method with a 95% confidence. CONCLUSION: Dissipation of acetamiprid and thiamethoxam in chard were determined under greenhouse. The concentration of acetamiprid and thiamethoxam in chards at 0 days after application were below specified by Korean MRL. Dissipation rate constant will be useful to set the pre-harvest residue limit for public health and consumer protection.

Comparison of Lines From Anther and Maternally-derived Dihaploids in Flue-cured Tobacco(Nicotiana tabacum L.)

  • The present study was conducted to compare the relative efficiency of two different haploid breeding methods in tobacco varietal development. A single FI hybrid plant from cross of two flue-cured cultivars of Nicotiana tabacum L., Bright Yellow4(BY4) and NC95, was used to develop the 30 anther derived dihaploid (ADH) lines and maternally-derived dihaploid (MDH) lines utilizing Nicotiana africana, respectively. As compared with mid-parent, mean of ADH lines showed increase in number of leaves, delay in days to flower and narrow in width of leaf. However, no significant differences in the other investigated characters were recognized. MDH lines also showed narrow width of leaf, while no significant differences in the other characters were observed. The variations of the investigated characters were generally greater in ADH than MDH lines. MDH lines had higher height of plant and shorter days to flower than ADH lines, while the other characters did not show remarkable differences. The degree of heritability for each of the characters observed between ADH and MDH was almost the same. The characters showing high heritability value were height of plant, number of leaves, days to flower, and yield, while those showing relatively low value were length of leaf, width of leaf, and total alkaloid content. Predicted gains from selection for increased yield were calculated for both populations(ADH, MDH) and correlated responses associated with selection for yield were estimated. Height of plant, width of leaf, days to flower, and concentration of reduceing sugar would be expected to improve with selection for yield much faster in the MDH population than in the ADH.

Effects of Cervi Cornu Aqua-Acupuncture on Lipid Metabolisms and Osteoporosis in Ovariectomized Rats (녹용약침자극(鹿茸藥鍼刺戟)이 난소적출(卵巢摘出) 흰쥐의 지질(脂質) 및 골대사(滑代謝)에 미치는 영향(影響))

  • In order to study the effect of aqua-acupuncture with Cervi Cornu (C.C.) extract solution manufactured by water-alcohol method on the lipid metabolism and osteoporosis, C.C. aqua-acupuncture was carried out every two days for 60 days on the corresponding bilateral loci of Shinsu (BL23), Taejo (BL11), and Hyonjong (GB39) from the 10th day after the ovariectomy for induced osteoporosis and thereafter the level of serum triglyceride, LDL, Ca, phosphorus and osteocalcin, as well as urine hydroxyproline were measured in the ovariectomized rats. The rats being studied were divided into a normal group (sham operation group), a control group (ovariectomized group), and a C.C. aqua-acupuncture group (group with C.C. aqua-acupunctured to the points of Shinsu, Taejo, and Hyonjong every two days for 60 days from the 10th day after the ovariectomy). The following results were obtained: 1. The serum triglyceride indicated an inhibited increase with statistical significance in the C.C. aqua-acupuncture group compared with the control group. 2. The serum LDL indicated an inhibited increase without any statistical significance in the C.C. aqua-acupuncture group compared with the control group. 3. The serum Ca showed an inhibited decrease without any statistical significance in the C.C. aqua-acupuncture group compared with the control group. 4. The serum phosphorus showed an inhibited decrease with statistical significance in the C.C. aqua-acupuncture group compared with the control group. 5. The serum osteocalcin indicated an inhibited decrease without any statistical significance in the C.C. aqua-acupuncture group compared with the control group. 6. The urine hydroxyproline showed an inhibited increase with statistical significance in the C.C. aqua-acupuncture group compared with the control group.

Comparative Analysis between Early Minithoracotomy and Conventional Treatment for Empyema (농흉치료에 대한 조기 Minithoracotomy 와 흉관삽관술의 비교연구)

  • Fifty one patients with empyema thoracic were managed at the Kyung Hee University Medical Center during 5 years between December, 1982, and December, 1987. The patients were classified into two groups; group A [early minithoracotomy-9 patients] and group B[conventional chest tube insertion-42 patients]. Each group was retrospectively analyzed to compare the results in terms of leukocyte count change, body temperature change, duration of hospitalization, elapsed time to chest tube removal and the need for subsequent decortication and tube change. There was no statistical difference between two groups in terms of etiology, age and sex. l. In the group A, mean preoperative leukocyte count [19,300/mme] decreased to 8,688/mme postoperatively. In the group B, leukocyte count changed from 16,985/mme to 14,433/mme. Their differences were significant [P< 0.05]. 2. In the group A, mean preoperative body temperature [38.5] decreased to 36.7. In the group B, body temperature changed from 38.1oC to 37.5 oC. Their differences were significant [P < 0.05]. 3. Mean duration of Hospitalization; 18.2 days [group A], 30.2 days [group B]. Their differences were significant [P < 0.01]. 4. Mean elapsing time for chest tube removal; 15.2 days [group A], 28.5 days [group B]. Their differences were significant [P < 0.01]. 5. There was no need for subsequent decortication and chest tube change in the group A. There were 22 cases [52.3 %] for subsequent decortication and 12 cases [28.6 %] for chest tube change in the group B. Early minithoracotomy in treating empyema thoracis resulted in a shorter hospital stay and a shorter period of tube drainage than conventional method.

Controlling Sacbrood Virus Disease in Apis cerana Colonies with Biological Methods in Korea

  • As Sacbrood virus (SBV), a causative agent of larval death and colony collapse in Apis cerana honey bee, is prevalent and poses one of the most significant threats to the Korean apiculture, development of methods to counter this viral disease is urgently needed. In this study we tested some SBV controlling methods, such as requeen, shook swam, adding Apis mellifera and spraying yogurt to SBV inoculated colony. Colony size measured by number of sealed brood and adult were evaluated every 15 days until two months while instances of recurrence were recorded up to five months after applying treatment methods. We also test the effects of yogurt on healthy and SBV-infected larvae at both in vitro reared larvae and colony level. Our result showed that all SBV controlling methods had similar success rates with respect to elimination of SBV clinical symptom up to 30 days post treatment. Mix-species and spraying yogurt method had similar pattern of sealed brood and adult number and higher than that of other SBV controlling methods up to 45 days post treatment. These two groups also showed the lower percentage of SBV recurrence (50% and 66.7%) at 120 days post treatment than other group that mostly colony had clinical symptom. Result on in vitro reared larvae challenged with yogurt showed that yogurt have neither harmless on healthy larvae nor remedial effect on SBV infected larvae. However, at colony level, colony in group received yogurt treatment removed significantly more SBV-infected larvae, SBV-killed larvae, and even healthy larvae in comparison to the control, suggested that yogurt could trigger the hygienic behavior of nurse bee. Our results recommended that it is practical in beekeeping by adding A. mellifera and spraying yogurt to control SBV in A. cerana colony.

Persistence and Degradation Pattern of Acequinocyl and Its Metabolite, Hydroxyl-Acequinocyl and Fenpyroximate in Butterburs (Petasites japonicus Max.)

  • Persistence and degradation patterns of acequinocyl and its metabolite, hydroxyl-acequinocyl (acequinocyl-OH) and fenpyroximate in butterburs (Petasites japonicus Max.) were investigated after pesticide application. Butterburs, one of the minor crops in South Korea, was planted in two plots (plot A for double and plot B for single application) in a greenhouse. Butterburs samples were also planted in a separate plot without pesticide treatment, as the control. A commercial pesticide containing acequinocyl and fenpyroximate was applied to the foliage of butterburs at hourly intervals after dilution. Recoveries of acequinocyl and acequinocyl-OH were 78.6-84.7% and 83.7-95.5%, respectively; the relative standard deviation of the two compounds were less than 5%. The method limit of quantification was 0.01 mg/kg. The total (Ʃ) acequinocyl residues in butterburs reduced by 96.0% at 14 days and 75.9% at 7 days, in plot A and B, respectively, after final pesticide applications. The biological half-life (DT50) of Ʃ acequinocyl and fenpyroximate, calculated using the dissipation rate, was 3.0 days and 4.0 days, respectively. These data were used to set up maximum residue and safe standard levels when the pesticides are applied to control pests during butterbur cultivation. Risk assessment results showed that the maximum % acceptable daily intake was 7.74% for Ʃ acequinocyl and 0.16% for Ʃ fenpyroximate. The theoretical maximum daily intake of Ʃ acequinocyl and fenpyroximate was 26.3% and 35.8%, respectively. In conclusion, the concentrations of Ʃ acequinocyl and fenpyroximate in butterburs pose no significant health risks to Koreans.

Effect of Ensiling Density on Fermentation Quality of Guineagrass (Panicum maximum Jacq.) Silage during the Early Stage of Ensiling

  • This study is to evaluate the effect of different levels of ensiling density on the fermentation quality of guineagrass silages during the early stage of ensiling. Guineagrass at the milky ripe stage was chopped and ensiled into a small-scale laboratory silo at two ensiling density levels (high density at 95 g/silo and low density at 75 g/silo). Three silos per level were opened after six ensiling periods (0.5, 1, 1.5, 2, 3 and 7 days of ensiling) and the fermentation qualities were analyzed. Within the initial 1.5 days of ensiling there were not significant (p>0.05) differences in the fermentation qualities between two density levels silages, and an almost constant pH and no or only small amounts of lactic acid, acetic acid and total volatile fatty acids were detected. However, the high density silage significantly (p<0.05) increased the rate and extent of fermentation after 1.5 days of ensiling, which was well reflected in significantly (p<0.05) faster and larger pH decline and lactic acid production at each elapsed time as compared with the low density silage. This resulted in significantly (p<0.05) lower finial pH and significantly (p<0.05) higher lactic acid content at the end of the experiment. Moreover, there was higher AA content relative to LA in both the H-D and L-D silages during the full fermentation course, and resulted in the AA-type silage. There were generally somewhat or significantly (p<0.05) higher acetic acid, volatile fatty acids and ammonia-N/total nitrogen in the high density silage than in the low density silage during the initial 3 days of ensiling. However, there were higher (p>0.05) ammonia-N/total nitrogen and significantly (p<0.05) higher butyric acid content in the low density silage at day 7 of ensiling. The silages of two density levels showed an initial increase in glucose between 0.5 and 1 day for the high density silage and between 1 and 1.5 days for the low density silage, respectively, thereafter showed a large decrease until the end of the experiment. There were not large differences (p>0.05) in ethanol content between the low density and high density silages that showed small amounts within initial 3 days of ensiling. However, the low density silage had a significantly (p<0.05) higher ethanol content than the high density silage at the end of experiment. From the above results it was suggested that the increase in ensiling density was an effective method to improve the fermentation quality, especially for tropical grasses.

THE SHEAR BOND STRENGTHS OF COMPOSITE RESINS TO GLASS IONOMER CEMENTS BY SURFACE TREATMENT AND ELAPSED TIME (광중합 GIC충전후 경과시간 및 표면처리에 따른 복합레진과의 결합강도에 관한 연구)

  • For the purpose of establishing the most appropriate method of bonding between glass ionomer liners and composite resin and comparing the materials for sandwich technique, an experiment was performed to measure the shear bond strengths between the two with the variables in the surface treatment of liners and elapsed time till composite buildup. Materials used were Vitrebond and Fuji II LC, each as the restorative and liner respectively, and each group was subdivided by surface treatment (acid etching and sandblasting) and time elapsed from GIC filling to composite buildup (immediately, 1 day, 7 days), consisting 12 groups as a whole. Each subgroup was composed of 10 specimens and the shear bond strength between GIC liners and composite resin was measured under UTM and analyzed. The result were as follows: 1. The shear bond strength between two materials was highest when initially filled Fuji II LC was sandblasted after 1 days and composite built-up (Group FS1). And the lowest value was found when GIC was acid-etched after 7 days and composite built-up (Group FE7). Significant difference was found between the two groups. (P<0.01) 2. In regard of surface treatment of GI liners, acid-etched group (VE) showed higher bond strength than sandblasted group (VS) for Vitrebond. But, the reverse was true for Fuji II LC. (P<0.05) 3. In regard to the time elapsed from GI filling to composite buildup, the group of 1 day elapse showed relatively higher strength for Vitrebond. On the contrary, immediate buildup group (FE0) was stronger for acid-etched group and 1 day elapse group(FS1) was higher for sand-blasted group in Fuji II LC. (P<0.05)
