Utilization of Ascidian(Halocynthia roretzi) Tunic -2. Optimum Level of Carotenoid Extracts from Ascidian Tunic for the Pigmentation of Rainbow Trout, Oncorhynchus mykiss- (우렁쉥이 껍질성분의 이용에 관한 연구 -2. 무지개송어 육색 개선을 위한 우렁쉥이 껍질 추출물의 최적 첨가량-)

  • This study was designed to determine the optimum pigment concentration supplemented in diet and feeding periods on the pigmentation of rainbow trout(Oncorhynchus mykiss) by using acetone-extracts of ascidian tunic as a natural pigment source. The eight pigmented diets contained carotenoid of ascidian tunic extracts at concentrations of 0, 200, 400, 800, 1,600 and 3,200mg/kg of diet, carophyll pink at concentration of 800mg/kg and commercial diet. No difference in pigment concentration was found between the ascidian extracts group and the control group until 4 weeks, but the redness of muscle and integument in the 1,600, 3,200mg/kg diet and carophyll pink was increased in the dorsal and caudal areas of fish from 6 weeks of age. In the sensory panel test, fish fed the ascidian tunic extracts diet were similar to those fed the carophyll pink diet. The optimum concentration and feeding periods for pigmentation of rainbow trout was found to be ascidian tunic extracts of 1,600mg/kg diet for 8 weeks.

Effect of Dissolved Oxygen on Occurrence of Tunic Softness Syndrome in Sea Squirt Halocynthia roretzi, Tongyeong, South Coast of Korea (멍게의 물렁증 발생에 미치는 용존산소의 영향)

  • This study analyzed the occurrence of tunic softness, survival rate, metabolic rate and histopathologic changes arising from the effect of hypoxic environment in order to find the causes of occurrences of tunic softness, which manifests as the key phenomenon of mass mortality of Halocynthia roretzi. Regarding the survival of H. roretzi with reduction in dissolved oxygen, all the entities died on the 4th day of exposure to the dissolved oxygen concentration of $2mg\;L^{-1}$ while 50% mortality was observed on the 5th day of exposure to the dissolved oxygen concentration of $3mg\;L^{-1}$. Therefore the 5 days-$LC_{50}$ was found to be $3.55mg\;L^{-1}$ (1.86~$4.96mg\;L^{-1}$). However, occurrence of tunic softness was not observed during the period of exposure to low oxygen concentration. The oxygen consumption rate significantly decreases at the dissolved oxygen concentration of less than $5mg\;L^{-1}$ in comparison to the control group. Therefore, it is presumed that H. roretzi controls the respiration rate for prescribed period of time when exposed to hypoxic environment. Regarding the histopathologic changes in the gill, digestive gland and cyst of H. roretzi due to hypoxic environment, necrosis of epithelial layer, in filtration of blood cells, and condensation of nucleus that compose each of the organs were observed. Regarding morphological changes, the decrease in volume with shrinking of the tunic, discoloration of the internal organs and necrosis of gill and hepatopancreas were observed.

  • Rainbow trout, Oncorhynchus mykiss, were cultured with different levels of carotenoids in odor to investigate the feeding effect of ascidian tunic extracts on the liver fatty acid compositions of them. Dominant monoenoic fatty acids of the ascidian tunic extracts were 16:1n-7($5.9\%$), 18:1n-9($21.9\%$), and 18:1n-7($3.5\%$) and polyenoic fatty acids of them were linoleic(18:2n-6, $14.2\%$), eicosapentaenoic(20:5n-3, $3.5\%$), and docosahexaenoic acid(22:6n-3, $8.3\%$). The compositions of fatty acid in the liver lipids were affected by the tunic extract levels during the feeding. The percentage of monoenoic acids in extract diets was decreased, and that of n-3PUFA was increased during feeding 2 weeks. But the n-3PUFA contents were decreased in 4 weeks. The 20:4n-6 content in rainbow trout fed extract diet was higher than that in the control and pink diet groups. The rainbow trout fed with ascidian tunic extracts showed an increase of essential fatty acids in the fish tissue, compared to the control or pink diet feeding groups.

  • Components of polysaccharides isolated from ascidian tunic were measuerd by gel filtration, electrophoresis and chemical analyses. The sulfated polysaccharides consisted in sulfate, protein, uronic acid and amino sugars. Hexosamines were composed of arabinose, xylose, glucose, galactose, glucuronic acid, N-acetylgalactosamine and N-acetylglucosamine by gas chromatography analysis. The galactose was predominant hexose after autoclave and nutrase digestion followed by DEAE-cellulose ion exchange chromatography and gel-permeation chromatography on Sephadex G-100 and G-25. FT-IR spectra of isolated polysaccharides from ascidian tunic and standard chondroitin sulfates have similar functional groups of the type of vibration and frequency. Molecular weight of isolated polysaccharides by autoclave represented more than 40 kDa by polyacrylamide gel electrophoresis. But the neutrase treatment group divided into three band. The highest molecular band group was shown more than 100 kDa, and the two low molecular band group were shown about 22 kDa and 5 kDa, respectively, compare to standard materials.

  • A relatively rare and puzzling tumor of the testicular tunic is reported. The tumor, so called a fibrous pseudotumor of testicular tunics, is presented because of the clinical dilemma this rare entity causes urologists and pathologists. This report demonstrates the necessity for familiarity with testicular pseudotumors in order to avoid an unnecessary orchiectomy.

Preparation and Characteristics of Low-salt Soy Sauce with Anti-hypertensive Activity by Addition of Miduduk Tunic, Mulberry, and Onion Extracts (미더덕 껍질, 뽕잎, 양파 추출물을 첨가한 항고혈압 저염간장의 제조 및 특성)

  • Extracts of Styela clava (Korean name: miduduk) tunic, mulberry, and onion were selected by pre-screening to develop low-salt soy sauce (12% salt content) with anti-hypertensive activity. After choosing the formula for low-salt soy sauce, extracts were added separately or by mixture in combination. In the case of anti-hypertensive activity, low-salt soy sauce containing extracts of miduduk tunic and onion showed 30% increased inhibitory activity towards angiotensin I converting enzyme, compared to normal salt soy sauce (15% salt content). Addition of extracts also significantly increased DPPH radical scavenging activities of the soy sauces. These results suggest that natural resources such as miduduk tunic, mulberry, and/or onion might be potential candidates for development of low-salt soy sauce with anti-hypertensive activity.

Change of Characteristics during Organic Liquid Fertilizer Processing using Ascidian Tunic (우렁쉥이 껍질을 이용한 유기 액비 제조시 발효액의 특성 변화)

  • This study was carried out to investigate the characteristics of the organic liquid fertilizer and find out optimum fermentation conditions of Effective Microorganisms (EM) including ascidian tunic. During the EM fermentation by adding ascidian tunic, electrical conductivity (EC) was increased, contrast to decrease the pH value, on related to the initial dosage molasses rate. Additionally, the total nitrogen quantity was shown to be increased on EM fermentation and the most effective increasing was recorded up to 220% on more than 15% molasses dosage condition. The phosphorus quantity was to be maximum rate on 21th of EM fermentation, and other contents, such to potassium, calcium, magnesium, sodium, had be shown an increasing patten during the fermentation period. After the EM fermentation, the concentration of hazardous material (Zn, As, Cd, Cu, Cr, Hg, Ni, pb) was measured below than the official criteria for commercial fertilizer. As a effective material in fermentated fertilizer, the 29 kinds free amino acids were detected and their total concentration was measured to 7080.94 mg/L.

  • To utilize the ascidian tunic as a natural pigment and dietary sources for rainbow trout (Oncorhynchus mykiss), juvenile were fed on experimental diets containing enzymatic hydrolysates of ascidian tunic treated with three commercial mined enzymes (ultrazyme, cellulase, viscozyme) for 12 weeks. From the results of feeding experiment, similar growth rate was checked in the enzymatic hydrolysis group compared with control, and those were a higher than of ascidian tunic powder group. The total acetone extractable pigment in muscle of the enzymatic hydrolysates group was lower than that of the ascidian extracts group and carophyll pink group until 8 weeks, but the level of those pigment of the enzymatic hydrolysates was similar to the ascidian extracts and carophyll pink group after 12 weeks. The lipid content was increased with the pigment concentration in the all experimental group. But the ascidian tunic pigment did not influence on the composition of the fatty acids in the muscle and liver. From the consideration of results for pigmentation, the enzymatic hydrolysates of ascidian tunic were suitable for both a natural pigment and dietary protein and carbohydrates sources as a substitute synthetic pigment for aquaculture use.

  • The major purpose of this study is to obtain the evident and visual structure of costume of priest with symbol through the Old Testament. Based on corroborative the Old Testament, this thesis analyzes the costume of priest. This compares Aaron with his sons. This compares the two onyx stones on the shoulder pieces with four rows of precious stones on the breastpiece. This compares the robe with the woven tunic. This era falls on about from the 15th century before Christ. The crafts men whom God have endowed with skill make the vestments. Due to the christian religion, the body was entirely concealed. The costume of priest were not only used as dignity but also grandeur. According to the Old Testament the colour of the costume has signified not only the colour sense but also important symbolic meanings. The priest wore, over undergarments, a woven tunic, and he wore over the tunic, a robe, an ephod, a breastpiece, a sash, using a turban as head covering. Gold, and blue, purple and scarlet yarn, and fine linen are used. Aaron and his sons must wear them whenever they enter the Tent of Meeting or approach the altar to minister in the Holy Place, so that they will not incur guilt and die.

Quality Properties of Fish Paste Prepared with Refined Dietary Fiber from Ascidian (Halocynthia roretzi) Tunic (우렁쉥이 껍질로부터 정제된 섬유소 첨가 어묵의 품질특성)

  • Fish paste was prepared to enhance physiological functions by adding 2.5, 5 and 10% dietary fiber isolated from ascidian (halocynthia roretzi) tunic. Hardness, adhesiveness, gumminess, chewiness and shear force of the fish paste were increased with addition of the dietary fiber. Water activity and Hunter's color values of the fish paste were not significantly changed by addition of the dietary fiber. Results of sensory evaluation indicated that no difference was observed in color, texture and overall acceptance (p<0.05). However, the fish paste with 5% dietary fiber scored the highest and was generally preferred by sensory panels.
