A Study on the Association between Tumor Necrosis Factor Alpha Gene Polymorphism and Sasang Constitution in Cerebral Infarction

  • Objective: Tumor necrosis factor-a $(TNF-{\alpha})$, a potent immuno-modulator and pro-inflammatory cytokine, has been implicated in many pathological processes. In this study, the author examined whether promoter region polymorphism in the $TNF-{\alpha}$a gene at position-308 affect the odds of cerebral infarction (CI) and whether genetic risk is enhanced by sasang constitutional classification. Methods: 212 CI patients and 610 healthy controls were genotyped and determined according to sasang constitutional classification. The amplified genotypes were analyzed on $8\%$ polyacrylamide gel. The alleles were visualized by ethidium bromide staining. Primers for $TNF-{\alpha}$ were designed to incorporate a polymorphic site at a position -308 bp of the $TNF-{\alpha}$ gene into an NcoI restriction site. Restriction digests generated products of 87 and 20 bp for G allele and 107 bp for A allele. Results : A significant decrease was found for the $TNF-{\alpha}$ A allele in CI patients compared with controls (P=0.033, odds ratio, O.R.: 0.622). However, there was no significant association between $TNF-{\alpha}$ polymorphism and sasang constitution in CI patients. Conclusion: My finding suggests that $TNF-{\alpha}$promoter region polymorphism is responsible for susceptibility to CI in Koreans.

  • Objectives : Tumor necrosis factor-${\alpha}{\cdot}$(TNF-${\alpha}{\cdot}$) is a proinflammatory cytokine involved in the pathogenesis of rheumatoid arthritis. This study was designed to investigate the relation between TNF-${\alpha}{\cdot}$ gene polymorphism and rheumatoid arthritis in Korean population. Methods : This study was carried out on 103 rheumatoid arthritis patients who fulfilled the American College of Rheumatology 1987 revised criteria for rheumatoid arthritis and 208 healthy control subjects. Blood samples from all subjects were obtained for DNA extraction. The extracted DNA was amplified by polymerse chain reaction(PCR). PCR products were visualized by 2% agarose gel electrophoresis. We investigated the genotyping of TNF-${\alpha}{\cdot}$ by using Pyrosequencing. Results : The genotypes of TNF-${\alpha}{\cdot}$ gene were GG, AG and AA. While the distribution of TNF-${\alpha}{\cdot}$ polymorphism in control subjects was 92.31%, 7.21%, 0.48% respectively, in rheumatoid arthritis patients was 93.20%, 6.80%, 0.00%(GG, AG. AA). There was no statistical significant allelic frequency difference between control and rheumatoid arthritis groups. Conclusion : We concluded that there was no significant association between TNF-${\alpha}{\cdot}$ gene polymorphism and rheumatoid arthritis. However, the findings of this study need to be confirmed in more patients and further studies.

  • 연구배경 : 결핵은 우리나라에서 아직도 흔하게 발생하는 질환으로 세포면역반응에 의해 육아종이 형성되며, 이 과정에서 대식세포가 분비하는 tumor necrosis factor-alpha (TNF-$\alpha$), Th1 세포가 생성하는 gamma-Interferon (INF-$\gamma$), 내피 세포가 표현하는 intercelluar adhesion molecule-1 (ICAM-1)이 중요한 역할을 할 것이라고 생각되었다. 방 법 : 저자들은 결핵의 경중(輕重)과 이러한 물질들이 관련이 있는가를 알아보기 위하여, 환자의 혈액을 채취하여 TNF-$\alpha$, INF-$\gamma$를 radioimmuno assay(RIA)로, ICAM-1이 혈액으로 유리된 형태인 sICAM-1을 enzyme linked immunosolvent assay(ELISA)로 각각 측정하였다. 또한 화학 요법에 따른 변화를 알기 위해 치료 시작후 6개월 시점에서 다시 추적검사를 실시하였다. 결 과 : TNF-$\alpha$, INF-$\gamma$, sICAM-1은 중등증과 중증의 결핵에서는 의미 있게 증가하였고, 경증에서는 의의가 없었다. 6개월간의 항결핵 치료후, sICAM-1은 임상경과에 동반하여 의미있는 감소를 보였지만. TNF-$\alpha$, INF-$\gamma$에서는 감소는 있었지만 의의는 없었다. 결 론 : 본 실험 결과 결핵의 세포 면역 매개과정에는 TNF-$\alpha$와 INF-$\gamma$가 매우 중요한 역할을 하며, 그 반응 정도가 질병의 병기에 따라 심할수록 많이 증가함을 관찰하였다. ICAM-1은 TNF-$\alpha$와 INF-$\gamma$ 농도와 비례하여 sICAM-1이 증가하였고, 질병의 병기에 따라 농도의 차이가 있을뿐 아니라, 치료경과에 비례하여 농도변화를 보여, 질병의 할동성을 나타내는 것 외에 치료 경과를 나타내는 지시자로서의 가능이 있을 것으로 생각된다.

  • To search for immunoactive natural products exerting anti-inflammatory activity, we have evaluated the effects on the water extracts of Artemisia princeps Pampanini (APP) on lipopolysaccharide-induced nitric oxide (NO), tumor necrosis factor-$\alpha$ (TNF-$\alpha$), and prostaglandin $E_2$ ($PGE_2$) production by RAW 264.7 macrophage cell line. Our data indicate that this extract is a potent inhibitor of NO production and it also significantly decreased PGE2 and TNF-$\alpha$ production. Consistent with these results, the protein and mRNA expression level of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) was inhibited by water extracts of APP in a dose-dependent manner. These results suggest that APP may exert anti-inflammatory and analgesic effects possibly by suppressing the inducible NO synthase and COX-2 expressions.

방기 전탕액의 비만세포 매개성 아나필락시반응 및 종양괴사인자알파 생성 억제효과 (Inhibitory Effect of Mast Cell-Mediated Anaphylactic Reactions and Tumor Necrosis $Factor-{\alpha}$ Production by Aqueous Extract of Sinomenium acutum stem)

  • Objectives: The root and stem of Sinomenium acutum has been used for treatment of arthritis and neuralgia in oriental medicine. To find new substances of the anti-anaphylactic drugs, we studied Sinomenium acutum. Methods: To investigate the effect of this plant, the effect on anaphylactic reaction, plasma histamine level, and tumor necrosis $factor-{\alpha}-(TNF-{\alpha})$ production were measured after the aqueous extract of Sinomenium acutum stem (SSAE) was administrated to mice and rats. Results: The SSAE (0.1 to 1000 mg/kg) dose-dependently inhibited systemic anaphylactic reaction induced by compound 48/80 in mice. Especially, SSAE reduced compound 48/80-induced anaphylactic reaction with 50% at the dose of 1000 mg/kg. SSAE (100 to 1000 mg/kg) also significantly inhibited local anaphylactic reaction activated by anti-dinitrophenyl (DNP) IgE. When mice were pretreated with SSAE at a concentration ranging from 0.1 to 1000 mg/kg, the plasma histamine levels were reduced in a dose-dependent manner. SSAE (1 to 1000 g/ml) dose-dependently inhibited histamine release from the rat peritoneal mast cells (RPMCs) activated by compound 48/80 or anti-DNP IgE. The level of cAMP in RPMCs, when SSAE was added, increased compared with that of a normal control. In addition, SSAE (0.1 g/ml) had a significant inhibitory effect on anti-DNP IgE-induced $TNF-{\alpha}$ production. Conclusions: These results indicate that SSAE inhibits mast cell-mediated anaphylactic reactions and $TNF-{\alpha}$ production from mast cells.

Saxatilin, a Snake Venom Disintegrin, Suppresses TNF-α-induced Ovarian Cancer Cell Invasion

  • Saxatilin is a disintegrin known to inhibit tumor progression in vivo and in vitro. The role of saxatilin in cancer cell invasion was examined by a modified Boyden chamber assay in MDAH 2774 human ovarian cancer cell line. Saxatilin (50 nM) significantly inhibited cancer cell invasion induced by tumor necrosis factor-$\alpha$ (TNF-a$\alpha$). Saxatilin also reduced MMP-9 mRNA levels in cancer cells in a dosedependent manner. In addition, TNF-$\alpha$-induced MMP-9 activity was reduced by the treatment of saxatilin. These results indicate that transcriptional regulation of MMP-9 is an important mechanism for the tumor suppressive effects of saxatilin in MDAH 2774 human ovarian cancer cells.

Expression of angiogenin, TGF-${\beta}$, VEGF, APEX and TNF-${\alpha}$ in oral squamous cell carcinoma

  • Purpose: The purpose of this study was to verify that the expressions of angiogenin, transforming growth factor-beta(TGF-${\beta}$), vascular endothelial growth factor(VEGF), human apurinic/apyrimidinic endonuclease(APEX) and tumor necrosis factor-alpha(TNF-${\alpha}$) were associated with the tumorigenesis of the oral squamous cell carcinoma(OSCC). Materials and Methods: Fifty-one samples of OSCC and fifteen normal oral mucosae were obtained to analyze the expression levels of above five factors. mRNA expressions were quantified by the quantitative competitive PCR(QC-PCR) method. After 2% agarose gel electrophoresis stained with ethidium bromide, the concentration of mRNA was calculated by a digital image analysis system. The expression levels of angiogenin, TGF-${\beta}$, VEGF, APEX and TNF-${\alpha}$ were compared by unpaired Student's t-tests between cancer and normal tissues. We analyzed statistically to find the cut-off values that would be useful as diagnostic markers, and the linear regression analysis between every two factors of these five factors by SAS system. Results: All of these five factors (angiogenin: P<0.0037, TGF-${\beta}$: P<0.0001, VEGF: P<0.0102, APEX: P<0.0023, TNF-${\alpha}$: P<0.0074) were significantly correlated with OSCC. In the analysis to find the cut-off values for the diagnosis, we could not find any value that had a reasonable sensitivity and specificity. In the linear regression analysis, there were correlations between angiogenin and TNF-${\alpha}$, TGF-${\beta}$ and VEGF, TGF-${\beta}$ and APEX, TGF-${\beta}$ and TNF-${\alpha}$, VEGF and APEX, VEGF and TNF-${\alpha}$, APEX and TNF-${\alpha}$. Conclusion: Our results suggest that not only angiogenin, TGF-${\beta}$, VEGF, APEX and TNF-${\alpha}$ are significantly associated with the tumorigenesis, but also the close relationship between these factors might enhance the tumorigenesis of OSCC. We can not find clinical availability for diagnosis.

Inhibitory Effect of Esculetin on the Inducuble Nitric Oxide Synthase Expression in TNF-stimulated 3T3-L1 Adipocytes

  • While nitric oxide (NO) produced by inducible nitric oxide synthase (iNOS) is beneficial for host survival, it is also detrimental to the host. Thus, regulation of iNOS gene expression may be an effective therapeutic strategy for the prevention of unwanted reactions at various pathologic conditions. During the screening process for the possible iNOS regulators, we observed that esculetin is a potent inhibitor of cytokine-induced iNOS expression. The treatment of 3T3-L1 adipocytes with the tumor necrosis factor-${\alpha}$ (TNF) induced iNOS expression, leading to enhanced NO production. TNF-induced NO production was inhibited by esculetin in a dose-dependent manner. Esculetin inhibited the TNF-induced NO production at the transcriptional level through suppression of iNOS mRNA and subsequent iNOS protein expression. These results suggest esculetin, a component of natural products, as a naturally occurring, nontoxic means to attenuate iNOS expression and NO-mediated cytotoxicity.

(ERC) on lipopolysaccharide-induced nitric oxide (NO), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, and $Interferon-{\gamma}\;(IFN-{\gamma})$ production by RAW 264.7 macrophage cell line. Our data indicate that this extract is a potent inhibitor of NO production and it also significantly decreased $IFN-{\gamma}\;and\;TNF-{\alpha}$ production. Consistent with these results, the protein level of inducible Nitric Oxide Synthase (iNOS) and cyclooxygenase-2 (COX-2) was inhibited by ethanol extracts of ERC in a dose-dependent manner. These results suggest that ERC may exert anti-inflammatory and analgesic effects possibly by suppressing the inducible NO synthase and COX-2 expressions.

치수 및 치근단병소에서 interleukin-1α, interleukin-1β, tumor necrosis factor-α의 분포에 관한 연구 (TISSUE LEVELS OF INTERLEUKIN-1α, INTERLEUKIN-1β AND TUMOR NECROSIS FACTOR-α IN PULPAL AND PERIAPICAL PATHOSIS)

  • This study was designed to examine the tissue levels of interleukin-$1{\alpha}$(IL-$1{\alpha}$), interleukin-$1{\beta}$(IL-$1{\beta}$) and tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) in inflamed human dental pulps and periapical lesions, and to determine the relationship between each cytokine and pulpal and periapical pathosis. The pulps used in this experiment, were obtained in routine endodontic treatment and the periapical lesions in periapical surgery after clinical diagnoses were performed. These specimens were divided into four groups as normal pulp group(control group, n=9), acute pulpitis group(n=g), chronic pulpitis group(n= 10) and periapical lesion group(n= 18) and stored in liquid N2. For extract preparation, tissues were finely minced with a scalpel, and the fragments were incubated in $0.5m\ell$ homogenizing buffer (0.1 mol/L potassium chloride, 0.02 mol/L TRIS; pH=7.6) for two hours and grinded with glass homogenizer. Debris was removed by centrifugation and supernatants were immediately tested with enzymelinked immunosorbent assay (ELISA, R&D Co., Minneapolis, USA). Following results were obtained; 1. The concentrations of IL-$1{\alpha}$ in all experimental groups were significantly higher than in control group(p<0.05). And the concentrations of IL-$1{\alpha}$ in periapical lesion group were somewhat higher than in two pulpitis groups, but the differences among those groups were not stastically significant (p>0.05). 2. The concentrations of IL-$1{\beta}$ in all experimental groups were significantly higher than in control group (p<0.05), and all the experimental groups expressed similar concentrations. 3. The concentrations of TNF-${\alpha}$ in all experimental groups were higher than in control group but only the differences between chronic pulpitis group and control group were statistically significant(p<0.05). And the concentrations of TNF-${\alpha}$ in acute and chronic pulpit is groups were higher than in periapical lesion group but only the differences between chronic pulpitis group and periapical lesion group were statistically significant (p<0.05). 4. There was significant correlation only between IL-$1{\alpha}$ and IL-$1{\beta}$ in periapical lesion group (p<0.05).
