  • To search for immunoactive natural products exerting anti-inflammatory activity, we have evaluated the effects on the water extracts of Artemisia princeps Pampanini (APP) on lipopolysaccharide-induced nitric oxide (NO), tumor necrosis factor-$\alpha$ (TNF-$\alpha$), and prostaglandin $E_2$ ($PGE_2$) production by RAW 264.7 macrophage cell line. Our data indicate that this extract is a potent inhibitor of NO production and it also significantly decreased PGE2 and TNF-$\alpha$ production. Consistent with these results, the protein and mRNA expression level of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) was inhibited by water extracts of APP in a dose-dependent manner. These results suggest that APP may exert anti-inflammatory and analgesic effects possibly by suppressing the inducible NO synthase and COX-2 expressions.

In-vitro Anti-inflammatory Activity of Rubus coreanus Miq. on Nitric Oxide, $Interferon-\gamma$, Cycloxygenase-2, and Tumor Necrosis $Factor-\alpha$ Production in the Macrophage like Cell Line RAW 264.7 Activated by Lipopolysccharide

(ERC) on lipopolysaccharide-induced nitric oxide (NO), tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$, and $Interferon-{\gamma}\;(IFN-{\gamma})$ production by RAW 264.7 macrophage cell line. Our data indicate that this extract is a potent inhibitor of NO production and it also significantly decreased $IFN-{\gamma}\;and\;TNF-{\alpha}$ production. Consistent with these results, the protein level of inducible Nitric Oxide Synthase (iNOS) and cyclooxygenase-2 (COX-2) was inhibited by ethanol extracts of ERC in a dose-dependent manner. These results suggest that ERC may exert anti-inflammatory and analgesic effects possibly by suppressing the inducible NO synthase and COX-2 expressions.

류마티스 관절염에 있어 종양괴사인자 다형성에 대한 연구 (Study on Tumor Necrosis Factor- ${\alpha}$ Gene Polymorphism in Rheumatoid Arthritis)

  • Objectives : Tumor necrosis factor-${\alpha}$(TNF ${\alpha}$) is a proinflammatory cytokine involved in the pathogenesis of rheumatoid arthritis. This study was designed to investigate the relation between TNF-${\alpha}$ gene polymorphism and rheumatoid arthritis in Korean population. Methods : This study was carried out on 103 rheumatoid arthritis patients who fulfilled the American College of Rheumatology 1987 revised criteria for rheumatoid arthritis and 208 healthy control subjects. Blood samples from all subjects were obtained for DNA extraction. The extracted DNA was amplified by polymerse chain reaction(PCR). PCR products were visualized by 2% agarose gel electrophoresis. We investigated the genotyping of TNF-${\alpha}$ by using Pyrosequencing. Results: The genotypes of TNF-${\alpha}$ gene were GG, AG and AA. While the distribution of TNF-${\alpha}$ polymorphism in control subjects was 92.31%, 7.21%, 0.48% respectively, in rheumatoid arthritis patients was 93.20%, 6.80%, 0.00%(GG, AG, AA). There was no statistical significant allelic frequency difference between control and rheumatoid arthritis groups. Conclusions : We concluded that there was no significant association between TNF-${\alpha}$ gene polymorphism and rheumatoid arthritis. However, the findings of this study need to be confirmed in more patients and further studies.

  • 연구배경 : 종양괴사인자(tumor necrosis factor ; TNF)는 다양한 생물학적 기능을 가지고 있는 바, 그 중 생체 외에서 증명된 뚜렷한 항암 효과로 말미암아 최근 항암 유전자요법의 중요한 대상으로 관심을 모으고 있다. 현재 유전자 이입의 기술적 문제로 생체 외에서 암세포에 유전자 이입을 시행한 후 이를 다시 환자의 생체내로 이식하는 방법이 연구의 주종을 이루고 있다. 그러나 저자들의 과거의 연구를 포함한 여러 연구에서 TNF가 이입된 암세포는 TNF에 대해 내성을 보이는 이에는 새로이 방어 단백질을 합성하는 것이 관여할 것이라는 시사가 있었다. 이 획득내성의 기전을 밝히는 것이 종양생물학의 이해를 넓히고 보다 효과적인 항암 유전자요법을 개발하기위한 매우 중요한 과제로 생각된다. 저자들은 TNF 유전자 이입에 따른 암세포의 TNF에 대한 획득내성에, 일부 세포에서 TNF에 의해 발현이 유도된다는 것이 밝혀진, 항산화효소의 하나인 MnSOD의 발현의 변화가 관여하는 지를 규명하고자 본 실험을 수행하였다. 방 법 : TNF에 다양한 감수성을 보이는 인체 및 생쥐 기원의 4가지 암세포주(WEHI164, NCI-H2058, A549, ME180)에 TNF-$\alpha$ 유전자를 retroviral 이용하여 이입하고 TNF의 발현을 시도하여 PCR, ELISA, MTT assay로 확인하였고, TNF 유전자가 이입된 세포(WEHI164-TNF, NCI-H2058-TNF, A549-TNF, ME180-TNF)는 TNF에 내성을 보이는지 역시 MTT assay로 검증하였다. TNF 유전자 이입 전후의 MnSOD mRNA 발현의 차이는 Northern blot analysis를 통하여 비교하였다. 결 과 : 1) TNF-$\alpha$ 유천자 이입 및 발현 확인 PCR을 시행한 결과 TNF 유전자가 이입된 각 세포 790 base pair 표기의 진한 DNA band를 보인 반면 모세포주는 보이지 않아서 retroviral vector를 이용한 유전자 이입이 DNA 수준에서 이루어 있었다. 그리고 TNF 유전자가 이입된 세포의 배양상층액에서 TNF양을 ELISA로 측정한 결과 TNF를 세포에 따라 1.91ng/24hr/$10^6\;cells$에서 3.91ng/24hr/$10^6\;cells$ 생산함을 알 수 있었다. 2) TNF 유전자 이입 전후, 암세포의 TNF에 대한 감수성 비교 TNF 농도 100ng/ml에서 WEHI164-TNF와 ME180-TNF 세포는 통계적으로 유의하게 (p<0.01) TNF에 대한 내성을 획득함을 알 수 있었다. 3) TNF 유전자 이입 전후외 MnSOD mRNA 발현양상 TNF에 감수성을 보이는 WEHl164와 ME180세포는 TNF 유전자 이입 후에 MnSOD mRNA 발현이 증가되지 않았으며, TNF에 내성을 보이는 NCIH2058과 A549 세포는 TNF 유전자 이입 후에 MnSOD mRNA 발현의 증가가 관찰되었으나 이는 모세포에 외부에서 TNF를 주었을 때도 관찰되었다. 결 론 : 세포주에 TNF 유전자를 이입하여 TNF를 발현하게 하였을때 세포 자신은 TNF에 대해 내성을 하게 되는데, 이 획득내성은 MnSOD 발현 능력의 향상에 의한 것은 아닐 것으로 판단되나 각 세포주의 자연 상태의 TNF에 대한 감수성 여부는 MnSOD의 발현 차이가 관련될 것으로 생각된다.

가와사키병 환자에서 면역글로불린 투여 전 후 호모시스테인, tumor necrosis factor-alpha 혈중 농도에 대한 연구 - 가와사키병 환아에서 호모시스테인, TNF-α 혈중 농도 비교 분석 - (Serum homocysteine and tumor necrosis factor-alpha levels after intravenous gammaglobulin treatment in patients with Kawasaki disease)

  • 목 적 : 호모시스테인은 성인에서 관상동맥 질환을 포함한 심혈관 질환의 독립적인 위험 인자로서, 혈관 내피세포 손상과 혈액 응고 촉진, 혈전 형성 등의 기전으로 동맥경화증의 위험을 증가시킨다고 알려져 있으나 이러한 호모시스테인이 소아에 미치는 영향은 아직까지 연구된 바가 없다. $TNF-{\alpha}$는 전염증성 사이토카인으로서 혈관 내피세포 손상과 관상동맥의 염증 반응을 초래한다. 본 연구에서는 호모시스테인이 가와사키병의 관상동맥 합병증에 미치는 영향과 $TNF-{\alpha}$의 연관성에 대해 알아보고자 하였다. 방 법 : 가와사키병 환아 24명(관상동맥 확장이 없는 군(1군) : 8명, 관상동맥 확장이 있는 군(2군) : 16명)에 대한 임상 양상과 면역글로불린 투여 전과 후, 회복기에서의 호모시스테인과 $TNF-{\alpha}$ 혈중 농도를 비교하였다. 대조군은 같은 시기에 내원한 열이 없는 정상 소아(3군 : 10명)와 열성 질환 소아(4군 : 11명)로 정하였다. 결 과 : 면역글로불린 투여 전의 호모시스테인은 1군($14.0{\pm}7.0{\mu}mol/L$)에서 3군($5.9{\pm}1.1{\mu}mol/L$)에 비해 유의하게 높았으며(P<0.05), 2군($17.4{\pm}8.0{\mu}mol/L$)에서는 3군($5.9{\pm}1.1{\mu}mol/L$)과 4군($9.2{\pm}2.0{\mu}mol/L$) 각각에 비해 유의하게 높았다(P<0.05). $TNF-{\alpha}$에서도 면역글로불린 투여 전 2군($858.4{\pm}934.0pg/mL$)이 3군($8.0{\pm}1.1pg/mL$)과 4군($54.1{\pm}146.3pg/mL$)에 비해 유의하게 높았다(P<0.05). 2군에서 시간 경과에 따른 호모시스테인과 $TNF-{\alpha}$를 비교해 보면 면역글로불린 투여 전에 각각 $17.4{\pm}8.0{\mu}mol/L$, $858.4{\pm}934.0pg/mL$로 가장 높았고, 면역글로불린 투여 후와 회복기로 갈수록 감소하였다. 또한 가와사키병 급성기에서 호모시스테인과 $TNF-{\alpha}$는 양의 상관관계를 가진다(r=0.488, P<0.05). 결 론 : 호모시스테인의 혈중 농도가 높을수록 가와사키병에서 관상동맥 병변이 발생할 가능성이 더 높았으며 $TNF-{\alpha}$와 같은 사이토카인도 관상동맥 병변의 형성에 중요한 역할을 할 것으로 생각된다.

대식세포에서 폐렴구균 협막 다당류에 의한 TNF-$\alpha$ 및 Nitric Oxide 생성 (Pneumococcal Capsular Polysaccharides Induce the Production of TNF-$\alpha$ and Nitric Oxide in Murine Peritoneal Macrophages)

  • Capsular polysaccharides (CPs) from Streeptococcus pneumoniae were examined for the ability to induce secretory responses in a pure population of peritoneal macrophages. The highly purified CPs were able to affect the macrophage, ie, secretion of tumor necrosis factor-alpha (TNF-$\alpha$) and nitrite. As after stimulation with CPs, secretion of TNF-u induced by these CPs reached its maximum within the first few hours of the interaction, while secretion of nitrite was increased with time. In addition, production of TNF-$\alpha$ and nitrite was increased in a dose-dependent manner. In the presence of indomethacin, CP-stimulated TNF-$\alpha$ production was not altered. In contrast, LPS with indomethacin stimulated 24.5% more TNF-$\alpha$ than LPS alone, suggesting that the intracellular signaling processes for TNF production are differentially stimulated by CP and LPS. The results demonstrate that CPs are potent inducer of macrophage secretory activities.

Prosuction of Cytokine and NO by RAW 264.7 Macrophages and PBMC In Vitro Incubation with Flavonoids

  • Flavonoids, a group of low molecular weight phenylbenzopyrones, have various pharmacological properties including antioxidant activity, anticancer, and immunomodulatory effects. In the present study, lipopolysaccharide (LPS) and phorbol 12-myristate 13-acetate/phytohemagglutinin (PMA/PHA) were used as stimulants for RAW 264.7 macrophages and human peripheral blood mononuclear cell (hPBMC), and tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-2 productions were measured. In addition, flavonoids were examined for their effects on LPS-induced NO production in RAW 264.7 macrophages. The results showed that all compounds were not strongly cytotoxic at the tested concentrations on hPBMC and RAW 264.7 macrophages. On immunomodulatory properties, catechin, epigallocatechin (EGC), naringenin, and fisetin repressed NO production and TNF-${\alpha}$ secretion. Furthermore, catechin, epigallocatechin gallate (EGCG), epicatechin (EC), luteolin, chrysin, quercetin, and galangin increased IL-2 secretion while EGC, apigenin, and fisetin inhibited the secretion. These results indicated that flavonoids have the capacity to modulate the immune response and have a potential anti-inflammatory activity. There was no obvious structure-activity relationship regard to the chemical composition of the flavonoids and their cell biological effects.

Genipin Selectively Inhibits TNF-${\alpha}$-activated VCAM-1 But Not ICAM-1 Expression by Upregulation of PPAR-${\gamma}$ in Human Endothelial Cells

  • Vascular inflammation process has been suggested to be an important risk factor in the development of atherosclerosis. Recently we reported that induction of peroxisome proliferator-activated receptor-${\gamma}$ (PPAR-${\gamma}$) selectively inhibits vascular cell adhesion molecule-1 (VCAM-1) but not intercellular cell adhesion molecule-1 (ICAM-1) in tumor necrosis factor (TNF)-${\alpha}$-activated human umbilical vein endothelial cells (HUVEC). In this study, we investigated whether genipin inhibits expression of cellular adhesion molecules, which is relevant to inflammation. Pretreatment with genipin reduced reactive oxygen species (ROS) production and expression of VCAM-1, but not ICAM-1 in TNF-${\alpha}$-activated HUVEC. Genipin dose- and time-dependently increased PPAR-${\gamma}$ expression and inhibited TNF-${\alpha}$-induced phosphorylation of Akt and PKC with different degrees. Finally, genipin prevented TNF-${\alpha}$-induced adhesion of U937 monocytic cells to HUVEC. Taken together, these results indicate that upregualtion of PPAR-${\gamma}$ by genipin selectively inhibits TNF-${\alpha}$-induced expression of VCAM-1, in which regulation of Akt and/or PKC play a key role. We concluded that genipin can be used for the treatment of cardiovascular disorders such as atherosclerosis.

Hyperglycemia increases the expression levels of sclerostin in a reactive oxygen species- and tumor necrosis factor-alpha-dependent manner

  • Purpose: Sclerostin, an inhibitor of Wnt/${\beta}$-catenin signaling, exerts negative effects on bone formation and contributes to periodontitis-induced alveolar bone loss. Recent studies have demonstrated that serum sclerostin levels are increased in diabetic patients and that sclerostin expression in alveolar bone is enhanced in a diabetic periodontitis model. However, the molecular mechanism of how sclerostin expression is enhanced in diabetic patients remains elusive. Therefore, in this study, the effect of hyperglycemia on the expression of sclerostin in osteoblast lineage cells was examined. Methods: C2C12 and MLO-Y4 cells were used in this study. In order to examine the effect of hyperglycemia, the glucose concentration in the culture medium was adjusted to a range of levels between 40 and 100 mM. Gene expression levels were examined by quantitative reverse transcription-polymerase chain reaction and Western blot assays. Top-Flash reporter was used to examine the transcriptional activity of the ${\beta}$-catenin/lymphoid enhanced factor/T-cell factor complex. Tumor necrosis factor-alpha ($TNF{\alpha}$) protein levels were examined with the enzyme-linked immunosorbent assay. The effect of reactive oxygen species on sclerostin expression was examined by treating cells with 1 mM $H_2O_2$ or 20 mM N-acetylcysteine. Results: The high glucose treatment increased the mRNA and protein levels of sclerostin. High glucose suppressed Wnt3a-induced Top-Flash reporter activity and the expression levels of osteoblast marker genes. High glucose increased reactive oxygen species production and $TNF{\alpha}$ expression levels. Treatment of cells with $H_2O_2$ also enhanced the expression levels of $TNF{\alpha}$ and sclerostin. In addition, N-acetylcysteine treatment or knockdown of $TNF{\alpha}$ attenuated high glucose-induced sclerostin expression. Conclusions: These results suggest that hyperglycemia increases sclerostin expression via the enhanced production of reactive oxygen species and $TNF{\alpha}$.
