PROPERTIES OF CALCIUM HYDROXIDE-EUGENOL COMPOUND (수산화칼슘-유지놀 화합물의 물성)

  • When a zinc-oxide eugenol type sealer was placed in root canals treated previously with calcium hydroxide, acceleration of its setting and the yellowish discoloration were observed clinically. The purpose of this study was to evaluate the properties of calcium hydroxide-eugenol compound. Some physical properties of calcium hydroxide-eugenol compound were compared with a manufactured zinc-oxide eugenol based root canal sealer, Tubli-seal$^{(R)}$ in terms of water solubility, water sorption, film thickness and microleakage. Solubility and water sorption were determined by the use of the method described in American Dental Association Specification(ADAS) no. 57. Ten samples of each material were prepared into disks 20mm in diameter and 1.5mm in thickness. The samples were immersed in 50ml of distilled water at $37{\pm}1^{\circ}C$ for 7 days. The samples were then removed and placed in a desiccator. The values for solubility and water sorption were calculated using differences between the weights of same sample. Film thickness was determined by the use of the method described in ADAS no. 57 too. A small quantity of mixed cement was placed between two glass plates of which thickness was measured previously. 15Kg loading was applied and total thickness of the glass plates and the cement film was measured. The thickness difference was recorded as the material's film thickness. Microleakage was determined with a dye penetration method. Experimental materials were placed between the dentin surface of bovine tooth and the acrylic rod. These units were immersed in Pelican ink (W-Germany) for three days. Dye-penetrated dentin surfaces of bovine tooth were measured using the NIB Image 1.60 Macintosh program. The results are as follows: 1. Water solubility value of calcium hydroxide-eugenol compound (20.98${\pm}$2.94%) was statistically higher than those of Tubli-seal$^{(R)}$(2.52${\pm}$0.49%)(p<0.05). 2. Water sorption value of calcium hydroxide-eugenol compound (59.72${\pm}$17.75%) was statistically higher than those of Tubli-seal$^{(R)}$(3.15${\pm}$0.76%)(p<0.05). 3. Film thickness value of calcium hydroxide-eugenol compound (0.36${\pm}$0.03mm) was statistically higher than those of Tubli-seal$^{(R)}$(0.12${\pm}$0.1mm)(p<0.05). 4. Dye penetration value after 3 days-immersion of calcium hydroxide-eugenol compound(57.63${\pm}$25.85%) was statistically higher than those of Tubli-seal$^{(R)}$(28.05${\pm}$23.46%)(p<0.05).

EFFECT OF POLYPHOSPHATE IN ROOT CANAL SEALERS ON THE GROWTH OF ORAL BACTERIA (Polyphosphate가 함유된 근관충전재가 구강세균의 성장에 미치는 영향)

  • Eliminating the infecting bacteria of the root canal system and preventing reinfection must be the main objectives of all endodontic works. None of commercially available root canal sealers have the properties of desirable tissue compatibility and strong antibacterial activity. The purpose of this study is to develope an ideal root canal sealer using commercially available polyphosphate (polyP), Calgon, which is known to be antibacterial and safe. For the study. resin type AH26, zinc oxide eugenol type Tubli Seal. Ca(OH)$_2$ type Apexit as base sealers for polyP (0~3%) and para formaldehyde containing N2 as a control base were selected. Specimens (3$\times$4mm) of the sealers were prepared in a 37$^{\circ}C$ incubator for 3 and 10 days and their antibacterial activity against streptococci and black pigmented anaerobic rods was observed using an agar diffusion method. The result were as follows: 1. Among 3 day old root canal sealers. N2 as a positive control showed the strongest antibacterial effect. followed by AH26. Tubli Seal and. Apexit which barely showed antibacterial activity against the test bacteria. In contrast. 10 day old AH26 showed a greater antibacterial activity than 10 day old N2. 2. All sealer specimens showed a greater antibacterial activity against black pigmented anaerobic rods than streptococci. Three day old ones appeared to be more antibacterial than 10 day old ones except for Apexit. 3. As compared to N2, 3 day old AH26 demonstrated a similar antibacterial activity against black pig mented anaerobic rods but to a lesser extent to streptococci. Ten day old AH26 showed a greater antibacterial activity against black pigmented anaerobic rods than 10 day old N2. 4. As compared to AH26. Tubli Seal generally revealed a lower antibacterial activity but it showed a greater antibacterial activity aginst S. gordonii Challis. 5. Enhancement of antibacterial activity by polyP was more clearly observed when it was added to Ca(OH)$^{\circ}C$ based root canal sealers. Tubli Seal and N2. 6. The addition of polyP enhanced the antibacterial activity of 3 day old AH26 against S. gordonii G9B (16%) and Challis (29%), and P. gingivalis 2561 (24%) only. Moreover, polyP failed to increase antibacterial activity of 10 day old AH26 against the test strains but P. gingivalis A7A1 28(13%). 7. The addition of polyP increased the antibacterial effect of 3 day old Tubli Seal on several test bacteria including s. mutans GS 5 (50%). s. gordonii G9B (47%) and Challis (122%). and all the test strains of P. gingivalis (13~35%) except for 9 14K 1. The addition of polyP to 10 day old Tubli Seal increased antibacterial activity of the root canal sealer against most test strains. 8. 3 day old Apexit failed to show antibacterial activity. if any very little against S. mutans GS 5 and Pr. intermedia ATCC 49046. However. polyP increased its antibacterial activity by 50 and 69%, respectively. Increase of antibacterial activity of 10 day old Apexit by polyP was more clearly observed than that of 3 day old one.

EVALUATION OF APICAL MICROLEAKAGE AND ADAPTATION TO THE DENTINAL WALL OF THE GLASS-IONOMER ROOT CANAL CEMENT (근관충전용 글라스 아이오노머 시멘트의 근단 폐쇄성과 치질 적합성에 관한 연구)

  • The purpose of this study was to compare the apical leakage and adaptation to the dentinal wall of a glass ionomer root canal cement(Ketac-Endo) with those of AH-26 and Tubli-Seal. 102 single-rooted teeth were insrumented with step-back technique and randomly divided into four groups according to kinds of root canal cements: Ketac-Endo, AH-26, Tubli-Seal, and no root canal cement. Four experimental groups were as follows; Group 1 : Filling with gutta-percha and Ketac-Endo Group 2 : Filling with gutta-percha and AH-26 Group 3 : Filling with gutta-percha and Tubli-Seal Group 4 : Filling with gutta-percha without root canal cement All the specimens were obturated with lateral condensation technique, and stored in 100% humidity for 4 days. 20 teeth in each group were placed in centrifuge tube filled with India ink, and then centrifuged for 20 minutes at 3,000 rpm. And they were evaluated for linear dye leakage using a stereoscope and analyzed by one-way ANOVA. 22 teeth were split longitudinally and evaluated the adaptation to the dentinal wall using scanning electron microscope(4 teeth in each group) and stereoscope(2 teeth in 1, 2, 3 group). The obtained results were as follows ; 1. In case of not used root canal cement(Group 4), there was showed much degree of dye penetration comparerd with used root canal cements (Group 1, 2, 3)(P<0.01). 2. In the case of used root canal cements(Group 1, 2, 3), there was no significant difference in linear leakage in groups(P>0.01). 3. In the case of used root canal cements(Group 1, 2, 3), there was showed intimate adaptation to the dentinal wall independent on kinds of root canal cements. 4. Fractured surfaces indicated failure of cohesion in Ketac-Endo, and failure of adhesion to dentine in AH-26 and Tubli-Seal. 5. According to similar apical leakage and adaptability to the dentinal wall to another root canal cements, it is possible to be used glass-ionomer cement as root canal cement.

A STUDY ON THE MICROLEAKAGE OF ROOT CANAL SEALERS (각종 근관충전용 시멘트의 미세누출에 관한 연구)

  • The purpose of this study was to compare the degree of microleakage of Glass-Ionomer root canal sealer possessed several enviable properties with that of the other sealers and to evaluate clinical performance. One hundred twenty single-rooted teeth were used in this experiment. The teeth were cleaned mechanically and immersed for 24 hours in 5.25% sodium hypochlorite and clinical crowns then were removed. After the root canals were instrumented using a step-back technique. one hundred twenty single-rooted teeth were divided into five groups of 24 in each. Group 1 : Tubli-Seal(Kerr Co., MI, U.S.A/ZOE-based), lateral condensation Group 2 : Sealapex(Kerr/Sybron, Romulus, MI/ $Ca(OH)_2$-based), lateral condensation Group 3 : AH 26(De Trey Co., Zurich Switzerland/Resin-based), lateral condensation Group 4 : Ketac-Endo(ESPE GMBH & CO. KG Seefeld:oberbay. Germany/ Glass Ionomer Cement-based), lateral condensation Control group : no sealer. lateral condensation And then. the root canals were obturated by lateral condensation technique with gutta-percha and experimental sealers. The control group were obturated without sealer. The teeth were placed in a vacuum chamber for 15 minutes and immersed 2% methylene blue under vacuum for 15 minutes. The teeth were passively stained for 1 week and 2 weeks and were cleared and evaluated for linear dye leakage using Tool maker's microscope(${\times}200$). The results were as follows: 1. There were statistically significant differences in the degree of dye penetration between the control group and experimental groups(p<0.05). 2. In the experimental groups, Sealapex($1.2871{\pm}0.9180mm$) exhibited the lowest mean value of dye penetration, followed by Ketac-Endo($1.4432{\pm}0.8082mm$), AH 26($1.5030{\pm}0.7752mm$) and Tubli-Seal($1.6458{\pm}1.0292mm$)(p>0.05). 3. There were statistically significant differences in the variation of microleakage between 1 week and 2weeks in Tubli-Seal and Seal apex groups (p<0.05). 4. The degree of dye penetration of all groups were increased as the time elapsed and AH 26 showed the lowest variation(+0.11) and Tubli-Seal(+ 1.03) showed the highest variation (p<0.001).

Cytotoxicity and Antibacterial property of New Resin-based Sealer (새로운 레진 계통의 근관봉함재의 독성과 항균 작용에 대한 연구)

  • 이 연구는 기존의 레진 근관봉함재를 보완하여 개발한 근관봉함재(Adseal; 새로운 레진 계통의 근관봉함재)를 이미 상품화된 레진 계통의 근관봉함재(AH 26, AH Plus), 산화 아연 유지놀 계통의 근관봉함재(TubliSeal EWT, Pulpcanal sealer EWT), 수산화 칼슘 계통의 근관봉함재(Sealapex)와 비교하여 세포독성과 항균작용을 평가하고자 한다. 세포독성 실험은 L929 쥐의 섬유아세포를 사용하여 세포의 viable ratio를 계산한 후, Giemsa stain으로 염색하여 세포의 양상을 관찰하였고, 항균작용 실험은 Enterococcus faecalis Porphyromonas endodontalis, Porphyomonas gingivalis, Prevotella intermedia, Fusobacterium nucleatum 와 Fusobacterium necrophorum를 사용하여 agar diffusion test로 평가한다. Adseal은 다른 근관봉함재에 비해 훨씬 낮은 세포독성을 보였고, AH Plus, AH 26, TubliSeal EWT, Sealapex, Pulpcanal sealer EWT의 순으로 세포독성의 정도가 높아짐을 알 수 있었다. 또한 Adseal은 Enterococcus faecalis 에서는 낮은 항균작용을 보이지만, Black-pigmented bacteria 에서는 높은 항균작용을 보이는데, 모든 근관봉함재는 서로 다른 종에 따라 어느 정도의 항균효과를 가지고 있음을 알 수 있었다.

A STUDY OF SEALING ABILITY OF DENTIN BONDING AGENT AS ROOT CANAL SEALER (Dentin Bonding Agent의 근관폐쇄(根管閉鎖) 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • The purpose of this study was to, examine the sealing ability of the denting bonding agent, when used with gutta percha cone, as a root canal sealer. Seventy two upper and lower extracted human teeth with single root were randomly selected and instrumented in a conventional method with K-file. After instrumentation and dry the canal with paper point, there were divided into three groups and twenty four teeth in each group were filled with the following material; In group I: Scotchbond in combination with gutta percha cone. In group II: AH 26 in combination with gutta percha cone. In group III: Tubli-Seal in combination with gutta percha cone. All specimens were immersed in Indian Ink, decalcified, and cleared. The depth of dye penetration into the canals were evaluated by caliper at the intervals of 1 day, 1 week, 2 weeks. The results were as follows; 1. Depth of dye penetration for all groups increased with time. 2. As to difference in mean dye penetration at time interval of 1 day and 14 days, Scotchbond group exhibited the smallest value as 0.53mm and Tubli-Seal group exhibited the largest value as 0.70mm. 3. There was the difference of dye penetration between each group, but its difference was not significant, statistically.

A COMPARATIVE STUDY ON RADIOPACITY OF ROOT CANAL SEALERS (근관 전색재의 방사선 불투과성에 관한 비교연구)

  • This study was performed to assess the radiopacity of a variety of root canal sealers according to the specification concerning root canal sealers. Ten materials including Tubli-$Seal^{TM}$. Kerr Pulp Canal $Sealer^{TM}$, AH $26^{(R)}$, AH $plus^{(R)}$, AH plus $jet^{TM}$, Ad sea $1^{TM}$. $Sealer^{TM}$, $NOGENOL^{TM}$, ZOB $seal^{TM}$, $Epiphany^{TM}$ and dentin were evaluated in this study. In the first part, densitometric reading of an each step of aluminum step wedge on occlusal film was performed at different voltage and exposure time. In the second part, ten specimens were radiographed simultaneously with an aluminum step wedges on the occlusal films under decided condition. The mean radiographic density values of the materials were transformed into radiopacity expressed equivalent thickness of aluminum (mm Al). The following results were obtained. 1. Among the various conditions, the appropriate voltage and exposure time that meet the requirement density was 60 kVp at 0.2 s 2. All of the materials had greater radiopacity than 3 mm Al requirement of ANSI/ADA specification No. 57 (2000) and ISO No. 6876 (2001) standards. 3. The radiopacity of materials increased as thickness of materials increased. 4. The mm Al value of each specimen at 1mm in thickness has a significant difference in the statistics. It suggests that root canal sealers have a sufficient radiopacity that meet the requirement.

A COMPARATIVE STUDY ON RADIOPACITY OF CANAL FILLING AND RETROGRADE ROOT-END FILLING MATERIALS (근관 충전재 및 치근단 역충전재의 방사선 불투과성에 관한 비교 연구)

  • This study was performed to assess the radiopacity of a variety of canal filling and retrograde root-end filling materials according to the specification concerning root canal obturation materials. Ten materials including Gutta-percha pellets, amalgam, Fuji II LC, $Dyract^{(R)}$ AP, Super $EBA^{(R)}$, $IRM^{(R)}$, AH $26^{(R)}$, $Sealapex^{TM}$, Tubli-$Seal^{TM}$, and dentin were evaluated in this study. In the first part, densitometric reading of an each step of aluminum step wedge on occlusal film were performed at 60 kVp (0.2, 0.3, 0.4 s), 70 kVp (0.2, 0.3, 0.33 s) to decide appropriate voltage and exposure time. In the second part, ten specimens which are 5 mm in diameter and 0.5, 1.0, 1.5, 2.0, 2.5, 3.0 mm in thickness, were fabricated from each material studied. The specimens were radiographed simultaneously with an aluminum step wedge under decided condition (60 kVp, 0.2 s). The mean radiographic density values of the materials were transformed into radiopacity expressed equivalent thickness of aluminum (mm Al). The following results were obtained. 1. Among the various conditions including 0.2 s, 0.3 s, 0.4 s at 60 kVp and 0.2 s, 0.3 s, 0.33 s at 70 kVp, the appropriate voltage and exposure time that meet the requirement of density from 0.5 to 2.0 was 0.2 s at 60 kVp. 2. All of the materials in this study had greater radiopacity than the minimun level recommended by ISO No. 4049 standards. 3. Most of the materials had greater radiopacity than 3 mm Al requirement of ANSI/ADA specification No. 57 (2000) and ISO No. 6876 (2001) standards except for Fuji II LC and Dyract. It suggests that all experimental canal filling and retrograde root-end filling materials have a sufficient radiopacity that meet the requirement concerning root canal obturation materials except for Fuji II LC and Dyract.
