Background: Free-living amoebae (FLA) are widely distributed in freshwater, seawater, soil, and extreme environments, and play a critical role as feeders on diverse preys in the ecosystem. Also, some FLA can become opportunistic pathogens in animals including humans. The taxa Amoebozoa and Heterolobosea are important amoeboid groups associated with human pathogens. However, the identification and habitat of amoebae belonging to Amoebozoa and Heterolobosea remain poorly reported in the Republic of Korea. This study highlights the first record for identification and source of four amoebae including putative pathogens in the Republic of Korea. Results: In the present study, four previously reported FLA were isolated from freshwaters in Sangju Gonggeomji Reservoir (strain GO001), one of the largest reservoirs during the Joseon Dynasty period, and along the Nakdong River, the largest river in the Republic of Korea (strains NR5-2, NR12-1, and NR14-1) for the first time. Microscopic observations and 18S rDNA phylogenetic trees revealed the four isolated strains to be Acanthamoeba polyphaga (strains NR5-2 and NR12-1), Tetramitus waccamawensis (strain GO001), and Naegleria australiensis (strain NR14-1). Strains NR5-2 and NR12-1 might be the same species and belonged to the morphological Group 2 and the T4 genotype of Acanthamoeba. Strain GO001 formed a clade with T. waccamawensis in 18S rDNA phylogeny, and showed morphological characteristics similar to previously recorded strains, although the species' flagellate form was not observed. Strain NR14-1 had the typical morphology of Naegleria and formed a strongly supported clade with previously recorded strains of N. australiensis in phylogenetic analysis of 18S rDNA sequences. Conclusions: On the bases of morphological and molecular analyses, four strains of FLA were newly observed and classified in the Republic of Korea. Three strains belonging to the two species (A. polyphaga and N. australiensis) isolated from the Nakdong River have the potential to act as opportunistic pathogens that can cause fatal diseases (i.e. granulomatous amoebic encephalitis, Acanthamoeba Keratitis, and meningoencephalitis) in animals including humans. The Nakdong River in the Republic of Korea may provide a habitat for potentially pathogenic amoebae, but additional research is required to confirm the true pathogenicity of these FLA now known in the Republic of Korea.
In this study we tried to determine the work-hardening exponent using continuous indentation test. Work-hardening exponent, which was determined by Hollomon equation, in tensile test, is an important parameter to determine plastic deformation and brittle/ductile property of materials. For using Hollomon equation, true stress and true strain were defined by indentation depth and indentation load. Using them the new equation, which is constituted by indentation depth, indentation load and work-hardening exponent, was induced. Indentation depth was calibrated because of elastic deflection and pile-up/sink-in phenomena. Work-hardening exponents of various steels derived by it showed good agreement to the results of tensile tests. In addition to experiments, FEM simulation was accomplished to investigate changes of real contact depth with materials properties changes. Through this simulation it is concluded that the real contact depth is changed by Y/E value which affect the early stage of indentation, and work-hardening exponent which the latter stage.

Six smooth flat tensile specimens and eighteen punch specimens with three different thicknesses were machined from steel of JIS G3131 SPHC. In addition to punch tests, incremental tensile tests were conducted to obtain average true flow stress - logarithmic true strain curves. Through parametric FE simulations for the tensile specimens, material parameters related to GTN model were identified. Using indenters with three kinds of radius, punch tests were carried out to obtain fracture characteristics of punch specimens. Numerical analyses using both fracture models, GTN and $J_2$ plasticity model, gave that the former estimated well the fracture of punch specimen but the latter did not. A new concept for critical size of plate elements was introduced based on minimum relative sharpness between contact structures. Consequently, a new criterion for critical element size was proposed to be less than 20% of minimum relative radius of interacting structures.
We investigated the effects of the chemical composition and the relative density on the plastic deformation behavior of sintered Fe-based alloys by means of compressive tests. Overall compressive stresses increased as the amount of alloying elements and the relative density were respectively increased. Addition of alloying elements except for Mo increased the yield stress regardless of the relative density. The relationship between the effects of the chemical composition and the relative density and the mean rate of the stress increase was analyzed. A constitutive equation based on the Ludwik equation with the regressed parameters was proposed to predict the compressive true stress-true strain curves of the sintered Fe-based alloys. The K and n values used in the proposed equation were regressed as a function of the alloying elements and the relative density based on the individual K and n values. The plastic deformation behavior predicted using the proposed constitutive equation showed reliable accuracy compared with experimental data.

The variations of the uniaxial compressive strength, the strains and the elastic constants with respect to the angle of anisotropy are analyzed in order to investigate the characteristics of a transversely isotropic rock experimentally. Total 35 specimens of 7 different angles from a large block of rhyolite presenting the flow structure obviously are used in tests. This study is composed of two reports; the elastic constants are mainly analyzed for the every individual angle in the report No. 1 and they will be discussed synthetically in the report No. 2. From the specimens of 0 and 90 degree, 4 independent elastic constants which can directly be obtained without the help of any other suggested equations, may be referred to the true values. Data variation in the strain measurements differs on the angle is analyzed. That of small angle specimens tends higher than that of large angle specimens. The relation of apparent Young’s modulus and angle is found to be M- or U-shaped. For small angle specimens, Saint-Venant approximation cannot be applied successfully on account of showing the non-monotonous increase, and E1 is analyzed out of the true value range. In the specimen of $\phi$ = 75, the deviation of strain measurement and strength are smallest and 4 all constants are analyzed in the true value range. Therefore, specimen of the angle of around 75 may become preferable if only one specimen should be used in test of a transversely isotropic rock.
The yeast strain 504D, isolated from salted shrimp soup, showed the best proteolytic activity under alkaline condition. The yeast formed vegetative cells in almost optimal media for yeasts, but formed only pseudohyphae in the MM medium containing citric acid and true hyphae in the MM medium containing N-acetylglucosamin and ${\beta}$-D-glucose. The yeast was classified as hemiascomycetes to form ascospores by sexual reproduction, and formed blastospores and athrospores by asexual reproduction. The yeast strain did not assimilate almost of the carbon sources, nitrate and nitrite, but some organic acids and alcohols. The fatty acids of whole cells were composed of 53.67% unsaturated fatty acids and 14.58% saturated, and, especially, C17:1 was observed in this strain but not in two control yeasts. However, almost of all results were very similar to the morphological and physiological characteristics of Yarrowia lipolytica KCCM 12495 and KCCM 35426, except for a little differences which are the composition of fatty acids and the manner of mycellium formation. Therefore, the isolated yeast strain 504D is identified as a Yarrowia lipolytica.
The high temperature deformation behavior of the activated sintered W powder compacts was investigated. The W compact showed the relative density of 94% with the average W grain size of $23\mutextrm{m}$ by activated sintering at $1400^{\circ}C$ for 1 hour. Compression tests were carried out in the temperature range of $900-1100^{\circ}C$ at the strain rate range of $10^{0}$/sec - $10^{-3}$/sec. True stress-strain curve and microstructure exhibited the grain boundary brittleness which was dependent on the compression test temperature. The activated sintered W compact showed that the maximum stress as well as the strain at the maximum stress was abruptly decreased as the test temperature increase from $900^{\circ}C$ to 1000 and $1100^{\circ}C$ regardless of the strain rate. The discrepancy of the microstructure in the specimen center was obviously observed with the increase of the test temperature. After compression test at $900^{\circ}C$ the W grain was severely deformed normally against the compression axis. However, after compression test at $1000^{\circ}C$ and $1100^{\circ}C$ the W grain was not deformed, but the microcrack was formed in the W grain boundary. The Ni-rich second phase segregated along the W grain boundary could be partly unstable over $900^{\circ}C$ and affect the poor mechanical property of the activated sintered W compact.
The development of filament stretching extensional rheometers over the past decade has enabled the systematic measurement of the transient extensional stress growth in dilute and semi-dilute polymer solutions. The strain-hardening in the extensional viscosity of dilute solutions overwhelms the perturbative effects of capillarity, inertia & gravity and the kinematics of the extensional deformation become increasingly homogeneous at large strains. This permits the development of a robust open-loop control algorithm for rapidly realizing a deformation with constant stretch history that is desired for extensional rheometry. For entangled fluids such as concentrated solutions and melts the situation is less well defined since the material functions are governed by the molecular weight between entanglements, and the fluids therefore show much less pronounced strain-hardening in transient elongation. We use experiments with semi-dilute/entangled and concentrated/entangled monodisperse polystyrene solutions coupled with time-dependent numerical computations using nonlinear viscoelastic constitutive equations such as the Giesekus model in order to show that an open-loop control strategy is still viable for such fluids. Multiple iterations using a successive substitution may be necessary, however, in order to obtain the true transient extensional viscosity material function. At large strains and high extension rates the extension of fluid filaments in both dilute and concentrated polymer solutions is limited by the onset of purely elastic instabilities which result in necking or peeling of the elongating column. The mode of instability is demonstrated to be a sensitive function of the magnitude of the strain-hardening in the fluid sample. In entangled solutions of linear polymers the observed transition from necking instability to peeling instability observed at high strain rates (of order of the reciprocal of the Rouse time for the fluid) is directly connected to the cross-over from a reptative mechanism of tube orientation to one of chain extension.
The major risk associated with the use of automated power injectors is the well known complication of contrast material extravasation at the injection site. Automated injection of computed tomography (CT) contrast media can produce the compartment syndrome. The purpose of this study was to assess the ability of this device during clinically important episodes of extravasation. The extravasation detection accessory (EDA) system was composed of a strain gage, an amplifier and a computer based system. A strain gage pliable adhesive patch was applied to the skin aver the intravenous catheter and the catheter was connected to the power injector with a cable to monitor the resolution data. If the programmed monitoring, which was developed with MS Visual C++, at the extravasation occurred, then the injection was interrupted the auto injector. CT was used to demonstrate the clinically important extravasation. This study was a prospective, observational study in which the EDA system was used to monitor the automated mechanical injection of contrast material in 7 dogs. There were two true-positive cases (range of extravasation volumes: $18{\sim}22ml$), twenty three true-negative cases, three false-positive cases and no false-negative cases. The EDA system had a sensitivity of 100% and a specificity of 88% for the detection of clinically important extravasation. The EDA system had good sensitivity for the detection of clinically important extravasation and the EDA system has the clinical potential for the early detection of extravasation of the contrast medium that is administered with power injectors. The EDA system is easy to use safe and accurate for the monitoring extravasation of the intravenous injections, and this system may prove especially useful in CT applications.
SP test has been confirmed the availability, however the application of SP test is hampered because the relation of stress-strain and load-displacement is not determined definitely. This study suggested an evaluation technique of plastic flow characteristic for X20CrMoV121 steel weldment through inverse analysis using SP test and finite element analysis(FEA). From the result, good agreement was found in load-displacement curves obtained from SP test and FEA. Also, The behavior of load-displacement curve from FEA show a rule that load is increase with increasing K(strength coefficient) and displacement is increase with increasing n(work hardening index). From the inverse analysis, true stress-strain curve could be obtained for each local structure of weldment. And the CGHAZ and WM, which showed lower load- displacement behavior, have smaller work hardening index, while FGHAZ have the largest index.
